Detection of sunflower and maize used in food and biofuel production.

Oil crops are an important source of energy and are widely used in food and biofuel production. Accurate detection of oilseeds is important for food authentication, quality and safety assessment, correct labeling, and efficient biofuel manufacture. The study was conducted on two important oilseed crops such as sunflower (Helianthus annuus) and maize (Zea mays L.). DNA-based polymerase chain reaction (PCR) method was selected as the most reliable and sensitive technique for food traceability. This study presents PCR methods for reliable detection of sunflower and maize. The study included several stages, such as design of oligonucleotide primers using bioinformatics tools; Genomic DNA extraction; development and optimization of PCR systems; assessment of PCR products by agarose gel electrophoresis. Seeds, flours and various processed food products were investigated, including cold-pressed and refined cooking oils. New efficient PCR primers targeting maize zein and invertase genes as well as sunflower helianthin gene were identified. The obtained results revealed the best DNA extraction and PCR amplification methods for sunflower and maize. Testing of food products demonstrated that the new PCR methods developed in this study are useful for the reliable detection of sunflower and maize in foods. Tab. 1, Fig. 2, Ref. 6.
