Molecular screening of Fragaria L. genotypes according to the genes of fruit flavor | Молекулярный скрининг генотипов рода Fragaria L. по генам аромата плодов

英语. The aroma of fruits plays a significant role in consumer preferences and is increasingly becoming the focus of attention in strawberry breeding programs. However, researchers face significant difficulties, since the aromatic profile of the crop is quite complex and includes more than 350 volatile compounds. Only a few strawberry genes that control the desired aromatic compounds can be identified with certainty, among which mesifurane (FaOMT gene) and γ-decalactone (FaFAD1 gene) are of particular importance. Mesifurane is responsible for the fruity and caramel flavor, while γ-decalactone is responsible for the peach-like, fruity, sweet one. The aim of the research was the molecular screening of Fragaria L. genotypes using the FaOMT and FaFAD1 genes to identify promising forms in breeding for fruit flavor improvement. The objects of the study were 24 strawberry cultivars of various ecological and geographical origin from the bioresource collection of the Russian Research Institute of Fruit Crop Breeding (VNIISPK), Orel region, Russia. Genomic DNA was extracted from young strawberry leaves using the CTAB protocol with Porebski's modifications for plants containing high polysaccharide and polyphenol components. DNA markers, codominant FaOMT-SI/NO and dominant FaFAD1, were used to identify genotypes with high levels of mesifurane (FaOMT+ gene) and γ-decalactone (FaFAD1 gene) accumulation, respectively. The functional, active allele (FaOMT+) was detected in 17 of the tested samples (70.8%), while the non-functional (inactive) allele (FaOMT-) – in four samples (16.7%). A combination of active and inactive alleles of the FaOMT gene (FaOMT+FaOMT–) was identified in three cultivars (12.5%). The presence of the FaFAD1 marker (500 bp target product) was identified in seven genotypes (29.1%). For the purpose of fruit flavor breeding, the most promising genotypes are those combining the functional alleles of the FaOMT+ and FaFAD1 genes. In the analyzed collection, five cultivars: 'Dezy', 'Аsia', 'Siria', 'Sara', and 'Vima Kimberly' (20.8%) were identified as the most promising due to their specific combination of characteristics.

俄语. Аромат плодов играет важную роль в предпочтениях потребителей и всё чаще становится предметом внимания в программах селекции земляники. Однако исследователи сталкиваются со значительными трудностями, поскольку ароматический профиль культуры достаточно сложен и включает более 350 летучих соединений. С уверенностью можно идентифицировать лишь несколько генов земляники, контролирующих желаемые ароматические соединения, среди которых важное значение имеют мезифуран (ген FaOMT) и γ-декалактон (ген FaFAD1). Мезифуран отвечает за фруктовый и карамельный, а γ-декалактон – за персикоподобный, фруктовый, сладкий аромат. Цель исследований – молекулярный скрининг генотипов рода Fragaria L. по генам FaOMT и FaFAD1 для идентификации перспективных форм в селекции на улучшение аромата плодов. Объекты исследования – 24 сорта земляники садовой биоресурсной коллекции ВНИИСПК (Орловская область) различного эколого-географического происхождения. Геномная ДНК для анализа была выделена из молодых листьев методом CTAB с модификациями Porebski для выделения ДНК из растительной ткани с высоким содержанием полисахаридов и фенольных соединений. Для идентификации генотипов с высоким уровнем накопления мезифурана (ген FaOMT+) и высоким уровнем накопления γ-декалактона (ген FaFAD1) применяли ДНК-маркеры – кодоминантный FaOMT-SI/NO и доминантный FaFAD1 соответственно. В анализируемой коллекции сортов земляники FaOMT+ (функциональный, активный аллель) был выявлен у 17 протестированных образцов (70,8%), а нефункциональный (неактивный) аллель (FaOMT–) – у четырех (16,7%). Комбинация активных и неактивных аллелей гена FaOMT (FaOMT+FaOMT–) была обнаружена у трех сортообразцов (12,5%). Маркер FaFAD1 (целевой продукт 500 п.н.) был идентифицирован у семи генотипов (29,1%). Для селекции на аромат плодов наиболее перспективными являются генотипы, совмещающие функциональные аллели генов FaOMT+ и FaFAD1 и соответственно сорта Dezy, Аsia, Siria, Sara, Vima Kimberly (20,8%).
