PHYSICO-CHEMICAL PROPERTIES OF SOME LISTERIA PHAGES

Listeria monocytogenes is a food borne pathogenic bacteria and caused a dangerous infection of humans. Six lytic bacteriophages specific to L. monocytogenes were isolated from irrigation and sewage waters and named ØLG, ØLA, ØLM, ØLD, ØLN and ØLP. The phages were propagated, and then purified by the ultracentrifugation. Morphological properties of Listeria phages showed that the phages were tailed phages, varied in their sizes and assigned to be a member of Siphoviridae (ØLG, ØLA, ØLM) and Myoviridae (ØLN, ØLD, ØLP) families. All Listeria phages were highly stable under different temperature conditions and its thermal inactivation point was reached to 80°C. The longevity in vitro of the phages was up to 60 days, as well as phages were active at pH values ranging from 4.0 to 12.0. Listeria phages did not lose their infectivity after exposure to UV for 90 min at 35 and 53 cm distances. Results of SDS-PAGE showed that phages had 5-6 protein fragments with molecular weights of 66, 45, 37, 35, 33 and 28 kDa distributed among the six phages.

Black cumin is one of the important medicinal plant and well known to flok remedy. Balady, Sori and Turki cultivars belong to black cumin are variated in chemical composition and antiviral activity. The results reflected significant heritability between the three cultivars in length and number of capsule per plant and number of branches per plant, while non-significant values of plant height, number of locules capsule, and seed yield per plant. Eight fatty acids methyl esters were detected in seeds of three cultivars by GLC. It was found change quantitatively of fatty acid between three cultivars. SDS-PAGA showed change qualitatively of poly peptides content accompanying three cultivars. A similarity of about 90% was found between Balady and Sori cultivars in relation to heritability. Water seed extracts, in vitro reduced ToMV infectivity to 6.73; 6.78 and 5.08% of Balady, Sori and Turki respectively. Four hours post and pre ToMV inoculation were most sensitive period to ToMV replication for three cultivars. On the other hand, the antiviral event changed in conformation and chemical structure of virion, coat protein and nucleic acid of ToMV with black cumin seed extract by spectroscopy. Balady, Sori and Turki; black cumin cultivars were varieted based on heritiability, chemical composition according to fatty acid GLC and polypeptide fraction SDS-PAGE analysis as well as antiviral activities against tomato mosaic tobamovirus.

Pollen grain morphology and seed coat characters of 11 cultivars belonging to two species of genus Hibiscus (Family Malvaceae) namely H. esculentus (H. Abelmoschus) and H. sabdariffa were investigated. This study was carried out using light microscope (LM) and Scanning Electron Microscopy (SEM). Pollen morphology of this genus is fairly uniform. Generally radially symmetrical apolar, mostly spheroidal, pantoporate. Seed exomorphic characters revealed four types of ornamentations; reticulate, ocealate,foveolate andruminate. Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (SDS-PAGE) was employed to characterize those taxa. Thirty one bands of seed protein profiles have been constructed from the gel. The produced dendrograms were analyzed by STATISCA program using UPGMA clustering method showed a close affinity among the seven H. esculentus cultivars and the four H. sabdariffa cultivars.
