Immunohistochemical Detection of Pro-Inflammatory and Anti-Inflammatory Interleukins in the Lungs of Sheep with Jaagsiekte

In this study, it was aimed to evaluate the levels of interleukins such as IL-1β, IL-6, IL-10 and IL-12β in sheep with Jaagsiekte by immunohistochemical methods. In this way, it will be revealed whether interleukins are effective in the progression of Jaagsiekte and how useful they are in the diagnosis of the disease. The material of the current study consisted of lung tissues of 26 sheep (Control, n=6 and Jaagsiekte, n=20) brought to the Department of Pathology for routine histopathological diagnosis. Tissue samples taken were fixed in 10% buffered formaldehyde solution. 5 µm-thick sections were taken from the paraffin blocks prepared after routine tissue follow-up procedures. Hematoxylin & Eosin staining was applied to the sections in order to detect histopathological changes. Sections were examined and photographed under a light microscope. The routine streptavidin–biotin peroxidase complex method was used.In sheep with Jaagsiekte, tumoral foci with large and small acinar or papillary growths were observed in the alveolar and bronchiole lumens. The control group was negative for IL-1β, IL-6, IL-10 and IL-12β immunoreactivity. IL 1β-6-10 and 12β levels were dramatically increased in the Jaagsiekte group compared to the control group.It was determined that interleukins were produced from tumoral cells and tumor microenvironment elements, and these interleukins showed pro-inflammatory effects, except for IL-10.

This study was to examine the opinions of first-year veterinary faculty students about the use of plastinated anatomic prosections in addition to wet cadavers in anatomy practical lessons. The students were shown plastinated organs and organs fixed in formaldehyde. Then a questionnaire comprising 7 questions was administered to the 100 student participants. The questionnaire responses were analyzed statistically using SPSS vn. 22.0 Frequencies software.In response to question 1, 58.4% of the students thought that there was no anatomic difference between the plastinated organs and the organs fixed in formaldehyde. In contrast to the strong smell of formaldehyde, 88% of the students stated that the plastinates were odourless. The use of plastinated organs was thought to improve the quality of education by 89.1% of the students, compared to the negative aspects of formaldehyde. It was stated by 84.2% of the students that plastinated organs should be included in anatomy lessons due to the thoughts of the majority of the students that plastinated tissues could make a greater contribution to anatomy lessons. In response to the final question, 92.1% of the students stated that they felt no abhorrence of the plastinated organs.Plastination may be especially useful for educational institutions without access, space, or the financial resources for dissection, and can emphasize unique or pathological samples. The results of this study demonstrated that plastinated samples were perceived as a useful addition to traditional resources in the teaching of anatomy.

Apoptosis, also known as programmed cell death, has become a target for treating many diseases, especially cancer. Many factors are influential in the cell's pathway to apoptosis. The defects in these pathways may transform the cell to become malignant, and the organism may face a lethal outcome such as cancer. Understanding apoptosis will provide clues in guiding the pathogenesis of diseases. Two main pathways leading to apoptosis, intrinsic and extrinsic, take an active role. The granzyme B pathway is also considered an apoptotic pathway, and this pathway is activated by enzymes secreted by immune cells such as T and NK. Many caspase molecules have initiator and enforcer roles and are active at critical points in the cell's apoptosis process. In cancer treatments, activating molecules in these pathways and repairing disrupted pathways are among the target approaches. This review discusses target strategies for inhibiting apoptotic pathways and molecules in cancer cells and activating these apoptotic pathways.

This study; It was aimed to compare serum and rumen content, trace element and serum vitamin levels and rumen content in indigestion cattle with healthy animals. Materials and Methods: The study was conducted on a total of 30 cattle, 10 healthy (control group) and 20 indigestion group. Indigestion diagnosis in animals was determined by anamnesis information, clinical and rumen content examination.Hematologically, there was no statistical difference between indigestion and control groups. Serum magnesium (Mg), calcium (Ca), zinc (Zn) levels of the measured trace elements decreased in the group with indigestion (p<0.05), while the levels of cobalt (Co) increased significantly (p<0.01). While there was a significant increase (p<0.5) in cattle with indigestion in rumen content Ca values compared to healthy cattle, serum levels were decreased (p<0.05). A positive correlation (p<0.01) was determined between rumen content levels of indigestion cattle and serum Mg levels of control group and rumen content of control group. In addition, a negative correlation was found between serum Ca and rumen content values of cattle with indigestion. In serum vitamin levels, Vitamin B1 (Vit B1) decreased statistically (p<0.05), while Vitamin B12 (Vit B12) was found to increase non-statistically (p>0.05).As a result, it was concluded that the decrease in serum Mg, Ca and Zn values in indigestion animals is important and these trace elements should be used in treatment.

Indiscriminate use of antibiotics in poultry farms increases the chance of antibiotic resistant and extended-spectrum β-lactamase (ESBL) producing bacteria in Bangladesh. Therefore, the study was under taken to detect ESBL producing Escherichia coli (E. coli) in chickens. Materials and methods: A total of 60 cloacal swab samples (20 from commercial layer, 20 from commercial broiler and 20 from commercial sonali chickens) were collected from Rajshahi district of Bangladesh. The E. coli was isolated from these samples and identified based on cultural, staining, and biochemical characteristics. The disk diffusion method was used to assay the antibiotic resistant/sensitivity patterns of the isolated E. coli. Phenotypc detection of ESBL producing E. coli was also done.The prevalence of E. coli in chickens was 61.67% in Rajshahi district of Bangladesh, where the prevalence was 60%, 60%, and 65% in commercial layer, commercial broiler, and commercial sonali chickens, respectively. The antibiotic sensitivity assay of E. coli isolated from commercial layer chickens showed 100%, 80%, 50%, 40%, and 40% resistant to amoxicillin, tetracycline, cefotaxime, ciprofloxacin, and ceftazidime, respectively. E. coli isolated from commercial broiler chickens showed 100%, 100%, 60%, 50%, and 40% resistant to amoxicillin, tetracycline, cefotaxime, ceftazidime, and ciprofloxacin, respectively. E. coli isolated from commercial sonali chickens showed 90%, 70%, 50%, 50%, and 40% resistant to amoxicillin, tetracycline, cefotaxime, ciprofloxacin, and ceftazidime, respectively. In phenotypic detection, the overall prevalence of ESBL producing E. coli was 43.33%, where 40%, 50%, and 40% in the commercial layer, commercial broiler, and commercial sonali chickens, respectively in Rajshahi district of Bangladesh.These results indicated that chickens are a potential reservoir for ESBL producing E. coli and their antibiotic resistances are obviously significant. These findings will help us to make proper guideline for the treatment, prevention and control of E. coli prevalent in chickens in Bangladesh.

In this case report, the clinical and radiographic results of the treatment of Salter Harris Type II fracture in the left humerus of a 10-month-old female and crossbred dog with parallel pin technique was evaluated. The dog with complaining of lameness was brought to Siirt University, Faculty of Veterinary Medicine, Clinic of Surgery Department and local fracture findings were found in the distal 1/3 of the left humerus. On radiological examination, it was found that the distal physeal line of the left humerus was detached. Also, it was seen that the integrity of the bone cortex was disrupted through in a line that included the metaphysis at the medial angle. In the operation, following the reduction of the fracture fragments, 2 krischner pins with 2 mm in diameter parallel to each other were applied from the medial cortex of the humerus to the lateral side of the distal condule and fixation was achieved. After the operation, the limb was taken to a backed bandage. In the radiological examination of the case on post-op 3rd week, it was found that the formation of the collus began. On the post-op 21st day, the bandage was removed and physical therapy applications were started to apply. On the post-op 4th week, it was seen that the dog used the extremity functionally and it was discharged. As a result, it was concluded that parallel double pin applications can be used successfully in the treatment of Salter Harris Type II fractures that are formed in the distal of dog's humerus.

This study was planned to assess the possible relationships with pregnancy-associated glycoproteins (PAG) concentrations by the determination of the biomarkers of oxidative stress in the plasma of dairy cattle during the early period of gestation. Materials and Methods: Blood samples were collected from coccygeal vessels in pregnant (n = 54) and nonpregnant (n = 45) cows. Measurement of biomarkers of oxidative stress (LPO, GSH and SOD) was carried out in females using spectrophotometric method.Plasma PAG concentrations increased and continuously over the both periods investigated. There were significant differences between pregnant and non-pregnant groups (P < 0.001). The concentration of SOD were significantly lower (P< 0.05) in pregnant females from day 25 to 35 (7.08 ±0.31 U.ml-1) and day 36 to 50 after AI (6.6 ± 0.29 U.ml-1) compared with non-pregnant cows (7.59 ± 0.35 U.ml-1). Concerning the concentrations of LPO and GSH, the values obtained were also significant lower (P < 0.05) in pregnant females in the period 25-35 days post AI (122.7 ±10.27 μM and 6.46 ±1.24 μmol/min.ml-1, respectively) and 36-50 days post AI (108.05±6.17 μM and 6.2±0.77 μmol/min.ml-1, respectively) than in the non-pregnant females (124.8 ± 12.16 μM and 6.96 ± 0.92 μmol/min.ml-1, respectively).It was observed that the markers of oxidative stress tended to be higher in non-pregnant females compared with pregnant females during the early period of gestation in dairy cattle. Our results suggest the existence of a relationship among the concentration of oxidative stress markers and PAG during early pregnancy.

In this case report, clinicopathological findings and certain biochemical parameters were presented in quails with the pendulous crop. The animal material of the study consists of a total of 3 Japanese quails (Coturnix coturnix japonica). The quails were examined clinically first, followed by the post-mortem examinations. The investigations of Na, K, Mg, total TP and Alb were conducted using autoanalyzer. The plasma levels of sodium, potassium, magnesium, total protein and albumin in the blood samples collected from the animals were found to be higher than the averages for quails. Postmortem examination of quails has revealed that the veins of their crops were prominently pronounced and that the crops were filled with a yellowish, foulodor liquid containing whole grain particles. Furthermore, the crop mucosa of an animal contained ulcerative lesions. As a result, it was found that certain mineral levels and protein profiles of animals were affected by the pendulous crop phenomenon, and that laboratory findings should be considered alongside physical findings when dealing with it.

The aim of this study was to determine the effects of lycopene administration as a protective agent against necrotic damage of NaF, a fluoride compound found to have high cytotoxic effects in the renal epithelial cell. Material- Method: The renal epithelial cell was cultured in DMEM high glucose medium, containing 10%FBS, 1%L-Glutamine (2mM) and 1% penicillin/streptomycin. With the MTT viability test, the non-toxic dose of lycopene (1 µM) and the IC50 value of NaF at the 24th hour was determined to be 3200 µM. The study groups were divided into four as control, NaF, lycopene and NaF+lycopene (the combination of NaF and lycopene). After the total mRNA obtained from these groups were converted to cDNA, expression levels of the identified necrotic genes were determined by real-time PCR method.While the Ripk1 gene did not change in the group given lycopene at the 24th hour, it was found that it increased 2.6 times in the group that received only fluoride, while it increased 7 times in the group treated with NaF+lycopene. A significant difference was detected between the groups in terms of gene expression pattern. While the Ripk3 gene increased slightly in the 24th hour applied lycopene group, it was observed that only NaF applied group increased 8 times and NaF+lycopene applied group increased in the 9 times.Based on the results obtained from this study, it was seen that activation of necrotic genes is important in explaining the molecular basis of cell death from NaF, which is applied as fluoride source, in revealing the molecular basis of the necrotic pathway. It was found that the decrease in cell viability due to NaF increased with lycopene, but the use of lycopene with fluoride also increased necrotic gene expression.

The study was conducted to determine the occurrence and distribution patterns of diseases of goat in Dhaka, Bangladesh during the period of January 2018 to December 2018. Materials and Methods: A total of 452 goats were recorded during the study period. The diseases were diagnosed based on clinical signs, patient owners complain and laboratory findings. The prevalence was measured according to season and age of the goats which were studied. Statistical analysis was done by Chi-square test and P-value was calculated by using SPSS 25 for windows (SPSS, Inc., Chicago, IL).Results: In this study, 381 (84.29%) goats were found to be diseased from 452 visited goats at Central veterinary Hospital (CVH), Dhaka. The highest prevalence was found with worm infestation (31.42%) followed by PPR (13.72%), miscellaneous (ruminal acidosis, disuria, repeat breeding) (12.16%), pneumonia (10.18%), dermatitis (4.20%), enteritis (3.76%), urolithiasis (3.09%), bloat (2.65%), mastitis (1.55%), tetanus (1.11%) and protozoal diseases (0.44%). Rainy season represented the highest (86.11%) prevalence of goat diseases followed by winter season (82.85%) and summer season (82.44%). PPR was significantly (p<0.05) more prevalent in winter season than other two seasons. In this study, Goats had significantly higher (p<0.01) prevalence of worm infestation in the summer season (29.00%) and the rainy season (39.35%) and in all three (0-12 months, 13-24 months and >24 months) age groups with 21.64%, 45.06%, and 32.20% respectively. Prevalence of mastitis was lowest in 0-12 months (0.43%) and 13-24 months (0.62%) age group but the prevalence of tetanus was lowest in above 24 months (1.69%) age group.These findings will help to know about age-wise and season wise variation of different diseases of goat in this area and will help to execute proper preventive measures against those diseases.
