Protective effects of bisoprolol against cadmium-induced myocardial toxicity through inhibition of oxidative stress and NF-κΒ signalling in rats

The aim of the study was to investigate the mitigative effects of bisoprolol (BIS) in cadmium-induced myocardial toxicity on oxidative stress and its inhibitive effect on nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB) signalling in rats. Male albino Wistar rats were assigned to control, Cd, BIS 2 (2 mg/kg b.w.) and BIS 8 (8 mg/kg b.w.) groups with nine rats in each. Over four weeks, the control group was administered 1% gum acacia, all other groups received 3mg/kg b.w. CdCl₂ dissolved in distilled water, and the BIS groups were additionally given bisoprolol in gum acacia. Blood samples were collected for biochemical estimations. Blood pressure and serum biomarker (lactate dehydrogenase, aspirate transaminase, alanine transferase and creatine kinase-MB, enzyme (superoxide dismutase, lipid hydroxy peroxidase, catalase and malondialdehyde), and tumour necrosis factor alpha (TNF-α) concentrations were measured. Western blot analysis was conducted for NF-κB and glutathione S-transferase (GST). After sacrificing the rats, cardiac tissue samples were examined histopathologically. Our findings pointed to a significant decrease (P < 0.05) in the studied serum biomarkers and levels of the relevant enzymes in the BIS 8 group compared to the Cd group. A significant decrease (P < 0.05) in NF-kB p65 expression and TNF-α levels was noted in the BIS 8 group relative to the BIS 2 and Cd groups, indicating a reduction at a higher dose. In microscopy, histopathological changes in the cardiac muscles of the BIS 8 group were evident compared to those of the Cd group. BIS seemed to have protective effects against cardiac injury induced by cadmium and could be considered a novel therapeutic drug and prognostic biomarker in the pathology of the many cardiovascular diseases caused by heavy metal intake.

The aim of this study was to evaluate potential protective effects of propolis on furan-induced hepatic damage by assessing the levels of malondialdehyde (MDA) and reduced glutathione (GSH), antioxidant enzyme activities, and histopathological changes in the liver. Albino Wistar rats were divided into six groups: a control, propolis-treated (100 mg/kg b.w./day), low-dose furan-treated (furan-L group; 2 mg/kg b.w./day), high-dose furan-treated (furan-H group; 16 mg/kg b.w./day), furan-L+propolis treated, and furan-H+propolis treated group. Propolis and furan were applied by gavage; propolis for 8 days, and furan for 20 days in furan-L groups and 10 days in furan-H groups. While MDA levels were elevated in furan-treated groups, levels of GSH and activities of antioxidant enzymes decreased (p < 0.001). The levels of MDA and GSH and activities of antioxidant enzymes were normal in the furan+propolis groups, especially in the furan-L+propolis group (p < 0.001). While the aspartate transaminase, alanine transaminase, alkaline phosphatase, and lactate pdehydrogenase activities were elevated in the furan-H treated group (p < 0.05 and p < 0.001), they were unchanged in the furan-L treated group. Histopathologically, several lesions were observed in the liver tissues of the furan-treated groups, especially in the higher-dose group. It was determined that these changes were milder in both of the furan+propolis groups. The results indicate that propolis exhibits good hepatoprotective and antioxidant potential against furan-induced hepatocellular damage in rats.

The present study has been carried out to investigate the effect of three neurotransmitters (Glutamate, L-Arginine and GABA) on some aspects of the reproductive performance of mature male Albino rats. For this purpose, a total of 100 mature male Albino rats were used. Rats were divided into 4 comparable groups; the first consists of 10 rats, was left as control. The second was administered glutamate 10 mg/ kg, the third group was injected by L- Arginine 20 mg/ kg while the fourth was injected by GABA 1 mg / rat. The results showed that administration of glutamate was concomitant with increase in synthesis and release of pituitary LH causing increase in its serum level as well as decrease serum level of testosterone. On the other hand, prolonged L-Arginine administration led to remarkable elevation in both pituitary and serum LH and significant decrease of serum testosterone. While, GABA administration led to remarkable decrease in pituitary and serum LH with significant decrease in serum testosterone level.

Two various kinds of olive mill solid residues (Crude and partly destoned) were used to investigate the adsorbing property of olive mill solid residues (OMSR) to heavy metals. 100 adult albino rats were classified into four groups. One group served as (–ve) control group (10 rats), received balanced ration and supplied tap water; in addition to three equal groups (each of 30 rats) received water polluted with1/100LD50 of either lead, cadmium or copper at concentration levels( 107.2 mg/L, 8.8mg/L or 58.4mg/L respectively). Each main group was divided into 3sub- groups, one (+ve) control and two experimental groups fed either 20 % crude olive mill or 20% partly destoned olive mill. Polluted water administration extended for 1 month, meanwhile feeding 20% crude or partly destoned (OMSR) continued for further 30 days after cessation of polluted water as a withdrawal period. Samples of serum, muscle, liver and kidney were collected at one month of the experiment and at the end of the withdrawal period after one month. Level of metals were determined by atomic absorption spectrophotometer. Results indicate that crude olive mill exhibit higher adsorption capacities to Pb, cd, and cu than partly destoned especially at withdrawal period. In conclusion, the main advantage of this process is the conversion of this waste product to a useful adsorbent material with low coast in minimizing the toxic hazard of environmental pollution with heavy metals.

Histopathological changes were studied in Swiss albino mice (N:36)which were challenged with the South Indian local strain of Trypanosoma evansi. Each animal was infected with 5×105 trypanosomes intraperitoneally. The animals were examined daily for development of clinical signs and infection status by wet blood-films made from the tail veins. The infected mice were dull and depressed from two days post-infection (DPI) onwards. Systematic post-mortem examination of the infected mice was performed and pathological changes were recorded. The different tissue samples were collected in 10% formalin and were used to study the histopathological changes. Postmortem examination from 3-4 DPI (the maximum period of observation) revealed splenomegaly, hepatomegaly, marked congestion of lungs, presence of fl uid in peritoneal cavity. Histopathologically, heart muscles showed hyaline degenerative changes and haemorrhages. Liver parenchyma revealed congestion of central vein and sinusoids, binucleated hepatocytes and fatty change of hepatic cells. Thickening of interstitial space with mononuclear infiltration, areas of collapse, areas of emphysema, edema and dilated and congested blood vessels were the histopathological changes noticed in the lungs of the infected mice. In the spleen, giant cells aggregation, hyperplasia, thickening of capsule and trabecule were the changes indicating irreversible degeneration. The affected kidney showed inter-tubular hemorrhages in the cortex, medullary hemorrhages, congested glomerulus, atrophied glomerulus, desquamated tubular epithelium and disruption of renal tubules at some places.
