Aggregate risk study of exposure to endophyte-infected (Acremonium coenophialum) tall fescue as a risk factor for laminitis in horses

Loline and ergot alkaloids found in endophyte-infected (Acremonium coenophialum) tall fescue (EITF) cause vasoconstriction of equine vessels in vitro. An aggregate risk study was used to evaluate the association between horses exposed to EITF and development of laminitis. Veterinary teaching hospitals participating in the Veterinary Medical Data Base were grouped by whether equine accessions were likely to have been at high, moderate, or low risk for exposure to EITF. From 1980-1990, there were 185,781 accessions, of which 5,536 had diagnosis of laminitis. Proportion of equine accessions with laminitis reported by veterinary teaching hospitals for high, moderate, and low risks, were 3.41, 3.04, and 2.00 cases/100 accessions, respectively (P < 0.0001). Comparison of the proportion of accessions with laminitis in the high- and moderate-risk groups with that in the low-risk group revealed significant differences between risk groups over all months (P = 0.063) and differences from month to month within risk groups (P = 0.0001). If the difference among risk groups is attributed entirely to exposure to EITF, the population-attributable risk is 7 cases/1,000 admissions, or 15% of all admissions for laminitis at veterinary teaching hospitals in our data base. Preliminary data support an association between hoses exposed to EITF and increased risk of laminitis; however, studies at the individual animal level are indicated to confirm this hypothesis.

Aflatoxin (AF)-contaminated and fumonisin B1 (FB1)-contaminated (culture material from Fusarium moniliforme) diets were fed singly and in combination to growing cross-bred barrows. Six barrows (3 replicates of 2 each; mean body weight, 17.5 kg) per group were fed: 0 mg of AF and 0 mg of FB1/kg of feed (control); 2.5 mg of AF/kg of feed; 100 mg of FB1/kg of feed; or 2.5 mg of AF plus 100 mg of FB1/kg of feed for 35 days. The effects on production performance, serum biochemical, hematologic, immunologic, and pathologic measurements were evaluated. Body weight, gain, and feed consumption were significantly (P < 0.05) decreased by AF and AF plus FB1 diets. The FB1 diet decreased feed consumption, and although body weight was numerically decreased, it was not statistically significant. Aflatoxin increased serum gamma-glutamyltransferase (GGT) activity and total iron concentration and decreased urea nitrogen concentration and unsaturated iron-binding capacity. The FB1-alone diet increased serum GGT activity, whereas the AF plus FB1 diet increased serum aspartate transaminase, cholinesterase, alkaline phosphatase, and GGT activities, increased RBC count, triglycerides, and total iron concentrations, and decreased unsaturated iron-binding capacity and urea nitrogen concentration. For the most part, the effects of the AF plus FB1 diet on body weight and hematologic measurements could be considered additive. However, the effect of the AF plus FB1 diet on cholinesterase and alkaline phosphatase activities was greater than additive and was a synergistic response. One pig in the FB1-diet group and 2 pigs in the combination-diet group died. Postmortem lesions in pigs of the FB1-diet group consisted of ascites and increased liver weight. Observations at necropsy for pigs of the AF plus FB1-diet group consisted of hydrothorax, ascites, pulmonary edema, gastric erosions and ulceration, and increased liver and spleen weights. The AF diet increased relative liver weight and resulted in liver that was pale, rubbery, and resistant to cutting. Histologic lesions consisted of hepatic necrosis or degeneration, or both, with variable degrees of bile duct proliferation in barrows of the AF-diet groups. Renal tubular nephrosis was observed in barrows of the FB1 diet group, but this was not consistent in the AF plus FB1-diet group. Cell-mediated immunity, as measured by mitogen-induced lymphoblastogenic stimulation index, was decreased in barrows of the AF and FB1-diet groups, and values in barrows given the combination diet were significantly decreased from those in barrows given the single toxin diets. It was concluded that AF and FB1 (from culture material), singly or in combination, can adversely affect clinical performance, serum biochemical, hematologic, and immunologic values and induce lesions in growing barrows. For most of the variables we evaluated under our study conditions and dosages of toxins, measurements were affected more by the combination diet than by either single toxin diet, and the toxic responses could be described as additive or more than additive, particularly for induction of liver disease.

Health and ruminal variables were intensively measured during adaptation to grain-based diets in 6 beef cattle with fistulated rumens. The cows had been maintained on prairie grass hay-supplemented diets, and were converted to a grain-based finishing ration by feeding each successive diet (diets 1-4, respectively) for a period of 7 days. Each cow was evaluated and samples were obtained 3 times each day for the first 5 days that each diet was fed. Health variables monitored were rectal temperature, pulse, respiratory and rumen motility rates, fecal consistency, demeanor, blood pH, and blood glucose and L(+) lactate concentrations. Ruminal variables monitored were pH and glucose, DL-lactate, and volatile fatty acid concentrations of rumen contents. Data were analyzed by use of a multivariate ANOVA. We determined that most of the health variables were within reference rang limits throughout the adaptation period; however, analysis of pulse and respiratory rates indicated that diets 2 and 4 were stressful. Although blood pH continually decreased during feeding of the 4 diets (7.38 to 7.30), blood L(+) lactate and glucose concentrations had large increases only within diet 4. The pH of ruminal contents decreased progressively from 6.8 to 5.3. Rumen glucose concentration was low (< 1 micromole/ml), except with diet 4 in which values were 8 times higher than for other diets. By the end of the study, the ruminal contents of all animals were acidic (pH < 5.5), and, on the basis of higher than background amounts of ruminal glucose and DL-lactate, it was determined that rumen microbial equilibrium had not yet been achieved. Analysis of results of this study suggested that ruminal imbalance could be evaluated by monitoring pulse and respiratory rates, blood pH, and blood glucose concentrations. Assessment of the rumen alone could be accomplished by monitoring the variables of rumen pH, rumen glucose, and DL-lactate concentrations. Respiratory rate, blood and rumen content pH, and blood L(+) lactate concentrations were significantly (P < 0.001) affected by time after feeding.

Growth indices were examined in 24 identically managed tanks, each containing 120 diploid juvenile rainbow trout (initial mean body weight, 9.3 g), during a 12-week study to examine tank effects associated with tank location in a multi-user research facility. Growth indices included mean body weight, feed intake, feed conversion index, and specific growth rate. The null hypothesis that tank effect had no effect on growth over the 12-week period was rejected (P = 0.038), and mean weight in individual tanks differed by as much as 18.7%. During the study, it was determined that the proximity of tanks to common-use walkways in the facility could affect growth indices. This was indicated by significant differences in the mean fish weights among blocks of tanks served by different header tanks after 4 (P = 0.001) and 8 (P = 0.024) weeks. The block containing tanks of fish with the highest mean weight was nearest to the 2 common-use walkways in the facility. Fish in this block of tanks, compared with those in other blocks, had significantly greater feed intake but no significant differences in conversion efficiency. Compensatory growth, a well known growth attribute in fishes, diminished the difference in mean weight between these blocks of tanks by the end of the study. Comparison of paired tanks within header tank blocks indicated that fish in those located nearest to walkways had higher feeding rates over the 12-week period (P = 0.048), but less efficient teed conversion (P = 0.040) than did fish in matched tanks located farthest from walkways. However, there were no differences in mean weight of fish. Results of this trial document the risks involved in identifying fish in a tank as the experimental unit when treatments are administered to the tank of fish, the latter being the true experimental unit.

A longitudinal survey of 820 cats in 73 households was conducted over a period of 6 years to establish the fate of pet cats that were seropositive after natural exposure to feline coronavirus (FCoV). In particular, their risk of developing feline infectious peritonitis (FIP) was determined. The seropositive cats were assigned to 1 of 3 groups: cats from households in which FIP had recently been diagnosed; cats from households in which FIP had not been diagnosed, but from which kittens had been relocated and subsequently died of FIP; and cats from households in which FIP had not been diagnosed. Cats in the first group were not at greater risk of developing FIP than were cats in the other 2 groups. Consequently, any household in which seropositive cats live must be considered a potential source of FCoV that can cause FIP. There was no evidence that the enhanced disease, which has been described after experimentally induced infection of seropositive cats, exists in nature. Thus, analysis of the survival of the seropositive cats over periods of up to 36 months indicated that their risk of developing FIP decreased with time, suggesting the development of immunity rather than increased susceptibility to disease. In addition, of 56 cats deemed to have been naturally reinfected because their anti-FCoV antibody titers decreased and subsequently increased, only 3 developed FIP.

Newborn foals of mares grazing on Acremonium coenophialum-infected fescue pasture throughout gestation or from gestation day 300 to parturition had increased gestation duration and decreased serum triiodothyronine concentration. Pregnant mares were allotted to 4 treatments: grazing continuously on endophyte-free (E-) fescue, grazing continuously on endophyte-infected (E+) fescue, grazing on E+ fescue from gestation day 300 to parturition, and grazing on E+ fescue from conception to gestation day 300. Morphometric studies indicated that foals born to mares exposed to endophyte late in gestation had large, distended thyroid follicles lined by flat cuboidal epithelial cells. Mean triiodothyronine concentration in foals exposed to endophyte (395.2 ng/dl) was decreased (P < 0.01), compared with mean values in control foals (778.0 ng/dl). Thyroxine and reverse triiodothyronine concentrations were not significantly different among groups. Foal organ weight as a percentage of foal body weight was not significantly different among experimental groups.

Growing dogs were fed diets containing soy oil or poultry fat as the main fat source and soybean meal or meat meal as the main protein source to examine the effects of types of dietary fat and protein on fatty acid concentrations in serum and skin and on serum cholesterol concentrations. Dogs fed diets containing soy oil had higher serum linoleic acid concentrations and lower serum oleic acid, arachidonic acid, and cholesterol concentrations than dogs fed diets containing poultry fat. The type of dietary protein had marginal effects on fatty acid concentrations and did not affect serum cholesterol. Similar differences were found in cutaneous fatty acid concentrations, with soy oil-fed dogs having significantly (P < 0.05) higher linoleic acid and lower oleic acid concentrations in their skin than had poultry fat-fed dogs. This study suggested that dietary fat source influences serum and cutaneous fatty acid concentrations and serum cholesterol concentrations in dogs, irrespective of dietary protein source.

Indices of renal function and damage were measured in 12 healthy male adult llamas fed a diet of mixed alfalfa/grass hay (mixed hay) and water ad libitum. Using a collection bag fitted over the preputial area, urine samples were collected at 6, 12, and 24 hours. Serum samples were obtained concurrently to determine endogenous creatinine clearance (CL), total (TE) and fractional excretion (FE) of electrolytes (Na, K, Cl, P), electrolyte CL, urine and serum osmolality, urine enzyme activities (gamma-glutamyltransferase and N-acetyl-beta-D-glucosaminidase), and urine protein concentration. Urine production was quantified. Three months later, 10 of the 12 llamas were fed a grass hay diet and water ad libitum. Similar samples were obtained, and similar measurements were made. Urine production was higher when the llamas were fed the mixed hay diet. Total urine volume for llamas fed mixed hay ranged from 628 to 1,760 ml/24 h, with a median of 1,307.5 ml/24 h, compared with a range of 620 to 1,380 ml/24 h and a median of 927.50 ml/24 h for llamas fed grass hay. Median urine osmolality was higher in llamas fed mixed hay (1,906 mOsm/kg of body weight, with a range of 1,237 to 2,529 mOsm/kg), compared with llamas fed grass hay (1,666 mOsm/kg with a range of 1,163 to 2,044 mOsm/kg). Creatinine CL did not vary significantly over time for either diet. Median creatinine CL was higher for llamas fed mixed hay, compared with llamas fed grass hay--0.78 ml/min/kg with a range of 0.20 to 1.83 ml/min/kg vs 0.45 ml/min/kg with a range of 0.13 to 3.17 ml/min/kg. Clearances for K and Cl varied significantly among the periods. However, median CL for Na and P did not vary over time for either diet. Overall values for these electrolytes in llamas fed mixed hay and grass hay diets were: CL(Na), 0.001 and 0.002 ml/min/kg and CL(P), 0.0006 and 0.0004 ml/min/kg respectively. The FE rates of K, Cl, and P did not vary significantly over time for either diet. Median respective FE for these electrolytes in the llamas fed mixed hay and grass hay diets include: FE(K), 84.90 and 63.10%; FE(Cl), 0.85 and 1.30%; and FE(P), 0.10 and 0.10%. Fractional excretion of Na varied over time for both diets and could not be expressed accurately as an overall median. Median respective TE of electrolytes for llamas fed the mixed hay and grass hay diets were: TE(Na), 0.007 and 0.03 mEq/kg/h; TE(Cl), 0.04 and 0.06 mEq/kg/h; and TE(P), 0.0002 and 0.00 mg/kg/h; TE(K) varied significantly (P < 0.05) over time for both diets. Urine gamma-glutamyltransferase activity changed significantly (P < 0.05) over time. Urine N-acetyl-beta-D-glucosaminidase activity was influenced by an interaction between diet and time. Median urine protein concentration was 26.0 mg/dl, with a range of 11.0 to 73.0 mg/dl for llamas fed mixed hay, and was 28.0 mg/dl, with a range of 16.0 to 124.0 mg/dl for llamas fed grass hay.

Effects of selenium (Se) deficiency and supplementation on production of colostral immunoglobulins by beef cows and transfer of antigen-specific and nonspecific immunoglobulins to their calves were examined. Eighty beef cows, with marginal to deficient Se status (blood Se concentration, 50 micrograms/L), were allotted by breed and age to 1 of 4 Se treatment groups (n = 20/group): no supplemental Se; parenteral administration of 0.1 mg of Se and 1 mg of vitamin E/kg of body weight; ad libitum consumption of 120 mg of Se/kg of salt-mineral mix (SMM); and parenteral administration of 0.1 mg of Se and 1 mg of vitamin E/kg plus ad libitum consumption of 120 mg of Se/kg of SMM. All cows were inoculated IM with lysozyme. Cows consumed Se-deficient pastures or hay (21 to 62 micrograms/kg) during the study that began at mid-gestation and ended at postpartum hour 24. Although the concentration of specific lysozyme antibodies was not affected, cows given 120 mg of Se/kg of SMM (treatments 3 and 4) had higher colostral IgG concentration (P < 0.002) than did Se-deficient cows (treatments 1 and 2). Calves from cows in treatments 3 and 4 had higher postsuckle serum concentrations of IgG (P < 0.01) than did calves from cows in treatments 1 and 2. Colostral IgM and calf serum IgM concentrations did not differ among treatments.

Reduced prevalence of diarrhea and mortality has been reported after dietary supplementation with zinc compounds in swine with naturally acquired colibacillosis and those challenge-exposed with Serpulina hyodysenteriae; however, the usefulness of this approach for control of enteric diseases of swine remains to be determined. To examine the effect of dietary zinc-containing compounds on the colonization and development of cecal lesions associated with S hyodysenteriae infection, a defined diet alone or with added ZnO, ZnSO4, or Zn-methionine complex to a final concentration of approximately 6,000 mg of Zn2+/kg of complete feed was fed ad libitum to 156 female mice (strain C3H/HeN) for 10 days prior to oral inoculation either with S hyodysenteriae or sterile trypticase soy broth. Rations were continued for 42 days, while at weekly intervals, 3 mice/group were necropsied for determination of body weight, cecal weight, liver zinc concentration, presence of S hyodysenteriae in the cecum, and gross and histologic assessments of cecal lesions. From postinoculation day 0 to 42, the liver zinc concentration of mice fed the zinc-supplemented diets was approximately twice that of mice fed the basal diet, irrespective of the source of zinc. From postinoculation day 7 through 42, the overall recovery rate of S hyodysenteriae in infected mice fed the basal diet was 77.8%. In contrast, recovery rates of S hyodysenteriae from S hyodysenteriae-inoculated mice fed the zinc-supplemented diets were 0% for Zn-methionine and ZnO and 16.7% for ZnSO4. Mice fed the basal diet had significantly (P < 0.05) higher weight gain than mice fed the zinc-supplemented diets. However, the severity of cecal lesions, as determined by histologic examination and quantitative determination of longitudinal crypt length of the cecum was significantly (P < 0.05) less in mice fed the zinc-supplemented diets than in mice fed the basal diet. Data from this study indicate that dietary supplementation with 6,000 mg of zinc/kg of feed significantly reduced the recovery rate of S hyodysenteriae and provided partial protection against development of cecal lesions in mice inoculated with S hyodysenteriae. However, at this concentration, zinc had a deleterious effect on growth of the mice.
