Plasma disposition, concentration in the hair, and anthelmintic efficacy of eprinomectin after topical administration in donkeys

Objective—To investigate plasma disposition, concentration in the hair, and anthelmintic efficacy of eprinomectin after topical administration in donkeys. Animals—12 donkeys naturally infected with strongyle nematodes. Procedures—The pour-on formulation of eprinomectin approved for use in cattle was administered topically to donkeys at a dosage of 0.5 mg/kg. Heparinized blood samples and hair samples were collected at various times between 1 hour and 40 days after administration. Samples were analyzed via high-performance liquid chromatography with fluorescence detection. Fecal strongyle egg counts were performed by use of a modified McMaster technique before and at weekly intervals for 8 weeks after treatment. Results—Plasma concentration and systemic availability of eprinomectin were relatively higher in donkeys, compared with values reported for other animal species. Concerning the anthelmintic efficacy against strongyle nematodes, eprinomectin was completely effective (100%) on days 7 and 14 and highly effective (> 99%) until the end of the study at 56 days after treatment. No abnormal clinical signs or adverse reactions were observed for any donkeys after treatment. Conclusions and Clinical Relevance—Eprinomectin had excellent safety. The relatively high plasma concentration after topical administration could result in use of eprinomectin for the control and treatment of parasitic diseases in donkeys.

Hematologic and serum biochemical values were determined in blood samples from 217 donkeys (Equus asinus). Donkeys were classified on the basis of size, sex, age, and whether they were domestic or feral. Parametric (mean +/- 2 SD) and nonparametric (2.5th to 97.5th percentile) reference ranges were calculated for each analyte. For all donkeys, 26 of 46 analytes significantly departed from gaussian distribution. Serum lactate dehydrogenase activity in miniature donkeys was higher than that in other donkeys. Differential leukocyte counts in feral donkeys differed from those in other types in ways that suggested that the former had smaller parasite loads or experienced greater stress. Erythrocyte, lymphocyte, and platelet counts and fibrinogen, glucose, inorganic phosphorus, and potassium concentrations decreased with age. Eosinophil counts, mean corpuscular volume, and plasma protein, serum protein, and serum globulin concentrations increased with age. Female donkeys had significantly (P < 0.05) higher mean corpuscular hemoglobin concentration and leukocyte and neutrophil counts than did male donkeys. Mean corpuscular hemoglobin increased with age, and females had higher values than did males of all age groups. An interaction between age and sex was observed for alkaline phosphatase activity, with a trend for decreased activity with age.

Objective-To determine the disposition of a bolus of meloxicam (administered IV) in horses and donkeys (Equus asinus) and compare the relative pharmacokinetic variables between the species. Animals-5 clinically normal horses and 5 clinically normal donkeys. Procedures-Blood samples were collected before and after IV administration of a bolus of meloxicam (0.6 mg/kg). Serum meloxicam concentrations were determined in triplicate via high-performance liquid chromatography. The serum concentration-time curve for each horse and donkey was analyzed separately to estimate standard noncompartmental pharmacokinetic variables. Results-In horses and donkeys, mean +/- SD area under the curve was 18.8 +/- 7.31 microgram/mL/h and 4.6 +/- 2.55 microgram/mL/h, respectively; mean residence time (MRT) was 9.6 +/- 9.24 hours and 0.6 +/- 0.36 hours, respectively. Total body clearance (CL(T)) was 34.7 +/- 9.21 mL/kg/h in horses and 187.9 +/- 147.26 mL/kg/h in donkeys. Volume of distribution at steady state (VD(SS)) was 270 +/- 160.5 mL/kg in horses and 93.2 +/- 33.74 mL/kg in donkeys. All values, except VD(SS), were significantly different between donkeys and horses. Conclusions and Clinical Relevance-The small VD(SS) of meloxicam in horses and donkeys (attributed to high protein binding) was similar to values determined for other nonsteroidal anti-inflammatory drugs. Compared with other species, horses had a much shorter MRT and greater CL(T) for meloxicam, indicating a rapid elimination of the drug from plasma; the even shorter MRT and greater CL(T) of meloxicam in donkeys, compared with horses, may make the use of the drug in this species impractical.

In contrary to most domestic livestock species, the common donkey (Equus asinus) is widely known as an animal with marked seasonality in reproductive activity. The annual cycle of daily photoperiod has been identified as the determining factor in sexual activity. A synthesis of the particularities of donkey reproduction is important and constitutes a basis of scientific reflection for managing asine livestock farms and establishing a well conservation plan for the different breeds around the world. It is necessary to have a perfect knowledge of the seasonal physiological changes in order to optimize the reproductive characteristics of donkeys. The objective of this paper is to review the current state of knowledge on the reproduction seasonality of common donkeys. We start with a remainder of some anatomical of the genital apparatus and sexual behavior aspects. Moreover, the investigations undertaken by many authors reveal the influence of the season on testicular biometry, histology, seminal and hormonal parameters in male donkeys populations. In conclusion, despite scientific controversy on the reproductive seasonal character in donkeys, it could likely be influenced by several factors mainly the photoperiod.

Objective: To establish and validate an objective method of radiographic diagnosis of anatomic changes in laminitic forefeet of donkeys on the basis of data from a comprehensive series of radiographic measurements. Animals: 5 donkeys with and 85 without forelimb laminitis for baseline data determination; a cohort of 44 donkeys with and 18 without forelimb laminitis was used for validation analyses. Procedures: For each donkey, lateromedial radiographic views of 1 weight-bearing forelimb were obtained; images from 11 laminitic and 2 nonlaminitic donkeys were excluded (motion artifact) from baseline data determination. Data from an a priori selection of 19 measurements of anatomic features of laminitic and nonlaminitic donkey feet were analyzed by use of a novel application of multivariate statistical techniques. The resultant diagnostic models were validated in a blinded manner with data from the separate cohort of laminitic and nonlaminitic donkeys. Results: Data were modeled, and robust statistical rules were established for the diagnosis of anatomic changes within laminitic donkey forefeet. Component 1 scores ≤ −3.5 were indicative of extreme anatomic change, and scores from −2.0 to 0.0 denoted modest change. Nonlaminitic donkeys with a score from 0.5 to 1.0 should be considered as at risk for laminitis. Conclusions and Clinical Relevance: Results indicated that the radiographic procedures evaluated can be used for the identification, assessment, and monitoring of anatomic changes associated with laminitis. Screening assessments by use of this method may enable early detection of mild anatomic change and identification of at-risk donkeys.

The donkey population has remained unchanged in the last two decades despite a decrease in the overall population of equids, emphasizing the usefulness of the donkey as a draught and pack animal. Piroplasmosis in donkeys, caused by Theileria equi and Babesia caballi, has been recognized as a serious problem of major economic importance as the affected animals manifest decreased working capacity, loss of appetite, etc. In tropical countries, T. equi infections are more wide-spread and pathogenic than those caused by B. caballi. Donkeys usually remain asymptomatic carriers with positive antibody titres throughout life. Transmission of infection occurs from animal to animal through ticks such as Hyalomma spp. Rhipicephalus spp. and Dermacentor spp. The clinical form of the disease is diagnosed by peripheral blood smear examination, but in carrier donkeys it is very difficult to demonstrate the parasite in stained blood smears as the parasitaemia is extremely low. For diagnosis of such low grade infection or carrier animals, serological tests and DNA-based molecular diagnostic techniques, which are discussed in the present review, have become mandatory. Currently, there is no suitable pharmacotherapy available to clear the T. equi infection from affected donkeys, though some new drugs and drug combinations used against this disease condition have been discussed. In the present situation, there is an urgent need for international cooperation and coordination for development of sensitive molecular diagnostic tools and effective pharmacotherapies for curtailment of the disease condition. Hence, it is imperative to develop and exchange reagents and technology developed through human resource sharing in the interest of sustainability of donkey husbandry.
