Pregnancy-associated changes in morphological and histochemical characteristics of the uterine cervix of the native Korean cattle

This study was performed to investigate the effect of gestation on the histological and histochemical changes of the uterine cervix of the native Korean cattle. A total of 110 cows obtained from the chonbuk abattoir were divided into six groups. The 1st group was of 9 non-pregnant cows and 101 singleton pregnant cows were grouped into 5 groups from pregnant I to pregnant V according to gestation periods by means of crown rump length measures. For light microscopy the tissues were fixed in 10% neutral formalin and processed routinely for paraffin sections. The 6um sections were taken and stained with H-E, Alcian blue pH 1.0, Alcian blue pH 2.5, Alcian blue pH 2.5/PAS, PAS reaction, toluidin blue, and trichrome. The cervical lengths and widths were increased in relation to advancing gestation. The cervical mucosubstance was composed of mixed mucopolysaccharides; the acid mucus was increased from the early pregnant state but the neutral mucus was found after pregnant IV, and the mucus was stained deeply in each staining in the fold cavity in according to gestation state.

Regional distribution and relative frequency of endocrine cells in ten portions of the gastrointestinal tract of the Korean native cattle were observed by immunocytochemical methods using specific antisera against chromogranin, serotonin, somatostatin, glucagon, bovine pancreatic polypeptide (BPP), motilin, gastric inhibitory polypetide (GIP), neurotensin, secretin, gastrin and substance P. In the abomasum, chromograin-, serotonin-, somatostatin-, motilin-, glucagon-, gastrin-, and substance P-immunoreactive cells were found. Chromogranin- and serotonin- and serotonin-immunoreactive cells were more numerous in the fundic region than pyloric region. Somatostatin- and gastrin- immunoreactive cells were numerous in the pyloric region than in the fundic region. In the large intestine, chromogranin-, serotonin- and BPP-immunoreactive cells were widely distributed and most numerous in the rectum. Normal swine brains at 1 to 70 days after birth were used to investigate the presence and morphology of the subependymal layer (SL) in the ventricle walls. The brain samples were taken from 27 pigs of 4 swine breeds. SLs were observed on the walls of the lateral ventricle (LV) but none were observed on the walls of the 3th and 4th ventricles. SLs of the LV walls were composed of mainly 3-to 10-cell layers in thickness. The thinnest region of SLs was composed of only 1-to 2-cell thick on the dorsal and ventral walls, and the thickest region was composed of 250-to 300-cell thick on extension region of the SLs into the angle between the corpus callosum and caudate nucleus. SLs were extensively thick in young age but gradually decreased in size and cell number with age after 20-day age. SL layers were composed of mainly oligodendrocytes, astrocytes and immature cells of them. Morphological differences of SL in different breeds of pigs were not observed.

In order to investigate the morphological characteristics of dendritic cells in the mammary gland, the appearance on the clear cells(CLs) or ATPase-positive dendritic cells(APDCs) have been observed by the light microscope. CLs were observed in the mammary tissues of the experimental animals, such as mice, rats, guinea pigs, rabbits, cats, dogs, pigs, cows and Korean native goats, and these CLs were confirmed as the ATPase-positive cells of typical dendritic appearance(APDCs). The APDCs were distributed in between the secretory epithelial cells, between the secretory epithelial cells and the myoepithelial cells, the basal area of the secretory epithelial cells, the interalveolar and interlobular connective tissues, and in between the epithelial cells of secretory duct. The APDCs were observed more frequently during the middle period of lactation than the other periods, and were irregularly or uniformly distributed according to the location. The mean difference between the inbred and the noninbred counts in the mice was statistically significant(P0.0001), and the similar result was presented in the rats(P0.0000)
