ARTs in wild felid conservation programmes in Poland and in the world

With the exception of the domestic cat, all felid species (Felidae) are currently threatened with extinction in their natural habitat. To develop effective and optimal wild cat conservation programmes with assisted reproductive technology (ART) it is necessary to combine advances from different disciplines of science, starting from the biology of the species, through research into the population and habitat, assisted reproductive technologies, establishment of gene banks, developing bioinformatic systems, and ending with biodiversity and endangered species management. In the last few years knowledge of felid reproduction has expanded considerably thanks to comparative studies utilising the domestic cat as a research model for endangered wild cats. Basic reproductive techniques utilised in both domestic cat breeding and rescuing wild felid populations that are threatened with extinction include semen collection and cryopreservation, artificial insemination, oocyte collection, in vitro maturation, in vitro fertilisation, somatic cloning, and embryo transfer. The main directions in which assisted reproductive technologies are being developed in wild cat conservation implementations and the contribution of Polish research centres in advancing these methods are presented.

All effects taken together, bovine tuberculosis (bTB) has a long-term detrimental effect on bovine herds and many wildlife species in South Africa. The disease is not only found in domestic cattle but also in African buffaloes and has to date been diagnosed in 21 wildlife species, including several rare and endangered species, thus having a potentially serious effect on conservation and biodiversity. In cattle, bTB is mostly characterised by sporadic outbreaks, but bovine herds chronically infected with the clinical disease are not uncommon. Presently, the recognised bTB control strategy in South Africa is based on 'test and slaughter', using the intradermal tuberculin test, followed by the slaughter of animals that have tested positive. Affected herds are placed under veterinary quarantine with movement restrictions until the outbreak is eradicated; this can take several years or last indefinitely if the outbreak cannot be eradicated. The same measures apply to infected buffalo populations, often with no prospect of ever being eradicated. This strategy is neither practical nor viable in the context of a communal farming system and becomes unethical when dealing with valuable wildlife reservoir hosts. Transmission of bTB between wildlife and cattle has been demonstrated and emphasises the need for an effective, affordable and culturally acceptable control strategy to curb the spread of bTB in South Africa. In countries with similar challenges, vaccination has been used and found to be promising for treating wild and domestic reservoir species and may hence be of value as a complementary tool for bTB control in South Africa.

Elephant Training Center (ETC) Way Kambas National Park (WKNP) was built to support human-elephant mitigation conflict. The small population of captive sumatran elephant in ETC WKNP need a comprehensive strategy in order to maintain the genetic variation of each individual and avoid inbreeding drive. Currently, genetic studies have opened new field studies in ecology, included conservation ecology. Patterns in variation of population has been investigated by molecular method supporting species conservation effort. The captive sumatran elephant’s ID Card is a necessary in database building, which included morphology, health status, and genetic profile. Genetic profile in each ID Card was filled by cytogenetic and molecular profile for RADP result, that initiated with DNA isolation. The DNA sources collected by blood sampling protocol described by Asiyah et al. (2016) from captive sumatran elephant in ETC, WKNP, and be carried to laboratory in cold condition. The DNA sources stored at 4oC and isolated following commercial protocol. The result of DNA isolation stored at -20oC until amplification analysis. DNA isolation was successfully done, for further individual genetic ID building.

To obtain plasma biochemistry values, blood was collected for 47 nesting females of apparently healthy Eretmochelys imbricata sea turtles using sodium heparin as an anticoagulant. Blood samples were collected in April-Jun for two years (nesting season). Hematologic characteristics, including packed cell volume, white blood cell counts, red blood cell count and hemoglobin level; and plasma chemistry values, including creatinine, blood urea nitrogen, uric acid, triglyceride, total cholesterol, and glucose were measured. The data generated from this study may be useful for clinical assessment of health and disease of wild hawksbill sea turtles on nearshore habitats in the Gulf of Mexico, thus contributing to a conservation of this species.
