Effect of sodium hexametaphosphate on dental calculus formation in dogs

A series of studies was conducted to identify a practical measure for preventing dental calculus formation in dogs. The studies involved a colony of 27 Beagles that received an initial dental prophylaxis. The dogs were then stratified on the basis of their normal rate of calculus formation and randomly assigned to parallel groups within each strata. During 4-week test periods, a variety of experimental regimens were instituted, followed by clinical assessments of calculus. Major observations were that a crystal growth inhibitor, soluble pyrophosphate, incorporated into a dry dog food modestly reduced calculus formation when used at high concentrations; anticalculus effects attributable to this agent were significant (P < 0.05) only when it was used as a surface coating the coating of dry dog chow or plain biscuits with a calcium sequestrant, sodium hexametaphosphate (HMP), provided the greatest benefit and resulted in significant (P < 0.05) reductions in calculus formation of about 60 to 80%, depending on the dosage regimen; and the feeding of a single daily snack of 2 HMP-coated plain biscuits (0.6% HMP) decreased calculus formation by nearly 80%. We concluded that the coating of dry dog chow or plain dog biscuits with HMP is an effective means of reducing calculus formation in dogs.

Health and ruminal variables were intensively measured during adaptation to grain-based diets in 6 beef cattle with fistulated rumens. The cows had been maintained on prairie grass hay-supplemented diets, and were converted to a grain-based finishing ration by feeding each successive diet (diets 1-4, respectively) for a period of 7 days. Each cow was evaluated and samples were obtained 3 times each day for the first 5 days that each diet was fed. Health variables monitored were rectal temperature, pulse, respiratory and rumen motility rates, fecal consistency, demeanor, blood pH, and blood glucose and L(+) lactate concentrations. Ruminal variables monitored were pH and glucose, DL-lactate, and volatile fatty acid concentrations of rumen contents. Data were analyzed by use of a multivariate ANOVA. We determined that most of the health variables were within reference rang limits throughout the adaptation period; however, analysis of pulse and respiratory rates indicated that diets 2 and 4 were stressful. Although blood pH continually decreased during feeding of the 4 diets (7.38 to 7.30), blood L(+) lactate and glucose concentrations had large increases only within diet 4. The pH of ruminal contents decreased progressively from 6.8 to 5.3. Rumen glucose concentration was low (< 1 micromole/ml), except with diet 4 in which values were 8 times higher than for other diets. By the end of the study, the ruminal contents of all animals were acidic (pH < 5.5), and, on the basis of higher than background amounts of ruminal glucose and DL-lactate, it was determined that rumen microbial equilibrium had not yet been achieved. Analysis of results of this study suggested that ruminal imbalance could be evaluated by monitoring pulse and respiratory rates, blood pH, and blood glucose concentrations. Assessment of the rumen alone could be accomplished by monitoring the variables of rumen pH, rumen glucose, and DL-lactate concentrations. Respiratory rate, blood and rumen content pH, and blood L(+) lactate concentrations were significantly (P < 0.001) affected by time after feeding.

The main objective of the study reported here was to determine whether signs typical of exocrine pancreatic insufficiency (EPI) are alleviated when affected dogs are fed a diet with low fat content, compared with feeding ordinary commercial dog food or food prepared by the owner. The most cost-effective amount of enzyme supplement also was estimated. The study consisted of 6 test periods. Duration of the first and third periods was 4 weeks, and that of the others was 2 weeks. During the first 2 periods, the dogs were fed their original diet. The amount of enzyme supplement was reduced by half between the first and the second period. During the last 4 periods, the dogs were fed only the low-fat diet, and amount of the enzyme supplement was reduced stepwise. During the entire study, owners were asked to assess daily the severity of 9 signs typical of EPI. A new index was established by adding the daily scores of each individual EPI sign. This index was designated the EPI index and was used as a measure of the general well-being of the dog. When the mean EPI indexes of the original diet periods were compared with those of the corresponding low-fat diet periods, there were no statistically significant differences by use of Turkeys test or the paired t-test. There was considerable variability between dogs, however. The fat content of the original diet did not correlate with the difference in EPI signs when the dogs were fed the low-fat diet. According to our study, feeding a low-fat diet to dogs with EPI did not significantly alleviate clinical signs of the disease. Decreasing the enzyme supplementation by 50% of the recommended of dose did not significantly increase severity of the cumulative EPI score. Decreasing the enzyme supplement by three-fourths of the recommended dose was excessive, and the severity of the clinical signs increased significantly (P < 0.05) The cost of the low-fat diet, compared with that of the original diet, was high.

To study their role in sulfate reduction, anaerobic bacteria were cultured from rumen fluid samples of cattle fed high-carbohydrate, short-fiber diets with and without added sulfate. The steers fed the diet with added sulfate developed polioencephalomalacia. Microbiological methods included colony-type profiles, molybdate sensitivity, presence of desulfoviridin, sulfate reduction rates of pure and mixed cultures, and incubation time effects on sulfate reduction. Colony-type profiles indicated decreased diversity, but no relative change in numbers of sulfate-reducing bacteria in rumen fluid from cattle fed diets with and without added sulfate. Thirteen bacterial isolates were selected for further study on the basis of colony type, sulfate-reducing activity, and growth in lactate, sulfate, and yeast extract media. Seven of the isolates had Desulfovibrio-like characteristics (ie, they were gram-negative, motile rods that reduced sulfate, were inhibited by molybdate, and contained the pigment desulfoviridin). The remaining 6 isolates were gram-negative, nonmotile rods. Four of these released sulfide from cysteine, and 2 generated only limited amounts of sulfide from sulfate or cysteine. The 7 sulfate-reducing isolates generated sulfide in rumen fluid broth medium at greater rates than those observed in fresh rumen fluid. Sulfate reduction could be sustained in cultures for prolonged incubation times if the gas phase containing hydrogen sulfide was replaced at frequent intervals. Variations in the amount of sulfate reduced by the pure cultures were most pronounced at short incubation times. Sulfate reduction was not inhibited in mixed cultures of sulfate-reducing and nonsulfate-reducing bacteria.

Indices of renal function and damage were measured in 12 healthy male adult llamas fed a diet of mixed alfalfa/grass hay (mixed hay) and water ad libitum. Using a collection bag fitted over the preputial area, urine samples were collected at 6, 12, and 24 hours. Serum samples were obtained concurrently to determine endogenous creatinine clearance (CL), total (TE) and fractional excretion (FE) of electrolytes (Na, K, Cl, P), electrolyte CL, urine and serum osmolality, urine enzyme activities (gamma-glutamyltransferase and N-acetyl-beta-D-glucosaminidase), and urine protein concentration. Urine production was quantified. Three months later, 10 of the 12 llamas were fed a grass hay diet and water ad libitum. Similar samples were obtained, and similar measurements were made. Urine production was higher when the llamas were fed the mixed hay diet. Total urine volume for llamas fed mixed hay ranged from 628 to 1,760 ml/24 h, with a median of 1,307.5 ml/24 h, compared with a range of 620 to 1,380 ml/24 h and a median of 927.50 ml/24 h for llamas fed grass hay. Median urine osmolality was higher in llamas fed mixed hay (1,906 mOsm/kg of body weight, with a range of 1,237 to 2,529 mOsm/kg), compared with llamas fed grass hay (1,666 mOsm/kg with a range of 1,163 to 2,044 mOsm/kg). Creatinine CL did not vary significantly over time for either diet. Median creatinine CL was higher for llamas fed mixed hay, compared with llamas fed grass hay--0.78 ml/min/kg with a range of 0.20 to 1.83 ml/min/kg vs 0.45 ml/min/kg with a range of 0.13 to 3.17 ml/min/kg. Clearances for K and Cl varied significantly among the periods. However, median CL for Na and P did not vary over time for either diet. Overall values for these electrolytes in llamas fed mixed hay and grass hay diets were: CL(Na), 0.001 and 0.002 ml/min/kg and CL(P), 0.0006 and 0.0004 ml/min/kg respectively. The FE rates of K, Cl, and P did not vary significantly over time for either diet. Median respective FE for these electrolytes in the llamas fed mixed hay and grass hay diets include: FE(K), 84.90 and 63.10%; FE(Cl), 0.85 and 1.30%; and FE(P), 0.10 and 0.10%. Fractional excretion of Na varied over time for both diets and could not be expressed accurately as an overall median. Median respective TE of electrolytes for llamas fed the mixed hay and grass hay diets were: TE(Na), 0.007 and 0.03 mEq/kg/h; TE(Cl), 0.04 and 0.06 mEq/kg/h; and TE(P), 0.0002 and 0.00 mg/kg/h; TE(K) varied significantly (P < 0.05) over time for both diets. Urine gamma-glutamyltransferase activity changed significantly (P < 0.05) over time. Urine N-acetyl-beta-D-glucosaminidase activity was influenced by an interaction between diet and time. Median urine protein concentration was 26.0 mg/dl, with a range of 11.0 to 73.0 mg/dl for llamas fed mixed hay, and was 28.0 mg/dl, with a range of 16.0 to 124.0 mg/dl for llamas fed grass hay.

Urine activity product ratios of uric acid, sodium urate, and ammonium urate and urinary excretion of metabolites were determined in 24-hour samples produced by 6 healthy Beagles during periods of consumption of a low-protein, casein-based diet (diet A) and a high-protein, meat-based diet(diet B). Comparison of effects of diet A with those of diet B revealed: significantly lower activity product ratios of uric acid (P = 0.025), sodium urate (P = 0.045), and ammonium urate (P = 0.0045); significantly lower 24-hour urinary excretion of uric acid (P = 0.002), ammonia (P = 0.0002), sodium (P = 0.01), calcium (P = 0.005), phosphorus (P = 0.0003), magnesium (P = 0.01), and oxalic acid (P = 0.004); significantly (P = 0.0001) higher 24-hour urine pH; and significantly (P = 0.01) lower endogenous creatinine clearance. These results suggest that consumption of diet A minimizes changes in urine that predispose dogs to uric acid, sodium urate, and ammonium urate urolithiasis.

Hyperxanthinuria and xanthine uroliths have been recognized with increased frequency in dogs with ammonium urate uroliths that had been given allopurinol. We hypothesized that dietary modification might reduce the magnitude of uric acid and xanthine excretion in urine of dogs given allopurinol. To test this hypothesis, excretion of metabolites, volume, and pH were determined in 24-hour urine samples produced by 6 healthy Beagles during periods of allopurinol administration (15 mg/kg of body weight, PO, q 12 h) and consumption of 2 special purpose diets: a 10.4% protein (dry matter), casein-based diet and a 31.4% protein (dry matter), meat-based diet. Significantly lower values of uric acid (P = 0.004), xanthine (P = 0.003), ammonia (P = 0.0002), net acid (P = 0.0001), titratable acid (P = 0.0002), and creatinine (P = 0.01) excreted during a 24-hour period were detected when dogs consumed the casein-based diet and were given allopurinol, compared with the 24-hour period when the same dogs consumed the meat-based diet and were given allopurinol. For the same 24-hour period, urine pH values, urine volumes, and urine bicarbonate values were significantly (P = 0.0004, P = 0.04, and P = 0.002, respectively) higher during the period when the dogs were fed the casein-based diet and given allopurinol than when they were fed the meat-based diet and given allopurinol. Endogenous creatinine clearance was significantly (P = 0.006) lower when dogs were fed the casein-based diet and given allopurinol than when they were fed the meat-based diet and given allopurinol. Significantly lower concentrations of plasma uric acid (P = 0.0001), plasma xanthine (P = 0.01), and serum urea nitrogen (P = 0.0001) were detected when dogs consumed the casen-based diet and were given allopurinol than when they consumed the meat-based diet and were given allopurinol. On the basis of these results, use of the casein-based diet and allopurinol in protocols designed for dissolution of urate uroliths may be beneficial in preventing hyperxanthinuria and xanthine urolith formation.

Urine activity product ratios of uric acid (APRua), sodium urate (APRna), and ammonium urate (APRau), and urinary excretion of 10 metabolites were determined in 24-hour urine samples produced by 6 healthy Beagles during periods of consumption of 4 diets containing approximately 11% protein (dry weight) and various protein sources: a 72% moisture, casein-based diet; a 10% moisture, egg-based diet; a 72% moisture, chicken-based diet; and a 71% moisture, chicken-based, liver-flavored diet. Significantly (P < 0.05) higher APRua, APRna, and APRau were observed when dogs consumed the egg-based diet, compared with the other 3 diets; there were no differences in these ratios among the other 3 diets. Twenty-four-hour urinary excretions of chloride, potassium, phosphorus, and oxalic acid were significantly (P < 0.05) higher when dogs consumed the egg-based diet. Twenty-four-hour urinary excretions of sodium were significantly (P < 0.05) higher when dogs consumed the egg-based diet, compared with the casein-based diet and the chicken-based, liver-flavored diet, but were not significantly different between the egg-based diet and chicken-based diet. Twenty-four-hour urine volume was similar when dogs consumed the 4 diets. Twenty-four-hour endogenous creatinine clearance was significantly (P < 0.05) lower when dogs consumed the casein-based diet; there were no differences among the other 3 diets. Although consumption of all diets was associated with production of alkaline urine, the 24-hour urine pH was significantly (P < 0.05) higher when dogs consumed the egg-based diet. These results suggest that use of diets containing approximately 10.5% protein (dry weight) and 70% moisture in protocols designed for dissolution and prevention of urate uroliths may be beneficial. The source of dietary protein in canned formulated diets does not appear to significantly influence the saturation of urine with uric acid, sodium urate, or ammonium urate.

Holstein steers were fed carbohydrate-rich, short-fiber basal diets with and without added sodium sulfate. Steers fed the high-sulfate diet developed the CNS disorder polioencephalomalacia (PEM). The onset of signs of PEM was associated with increased sulfide concentration in the rumen fluid. Over the course of the disease, anaerobic rumen bacteria were enumerated in roll tubes by use of the Hungate method to determine the effect of dietary sulfate on sulfate-reducing bacterial numbers. Media used included a general type for total counts and sulfate-containing media with and without cysteine to assess sulfate-reducing bacteria. Changes in total and sulfate-reducing bacterial numbers attributable to dietary sulfate content were not observed. The capacity to generate hydrogen sulfide from sulfate in fresh rumen fluid in vitro was substantially increased only after steers had been fed the high-sulfate diet for 10 to 12 days, which coincided with the onset of signs of PEM. The low capacity for hydrogen sulfide production of rumen fluid taken at earlier times in the feeding period suggests that rumen microorganisms must adapt to higher dietary sulfate content before they are capable of generating potentially toxic concentrations of sulfide.
