Evaluation of recombinant LipL32 and LipL41 antigens of Leptospira interrogans serovar Canicola by ELISA for serodiagnosis of bovine leptospirosis.

Recombinant LipL32 and LipL41 outer membrane proteins of Leptospira interrogans serovar Canicola were produced, and used as a pooled antigen in enzyme-linked immunosorbent assay (ELISA) to detect leptospiral antibodies in bovine sera samples. The optimum concentration of the pooled antigen was found to be 50ng of each antigen per well by using known positive and negative cattle sera. Using a total of 500 bovine sera samples the sensitivity, specificity and accuracy of pooled antigen based ELISA as compared to microscopic agglutination test (MAT) were 100%, 88.1% and 91.6%, respectively. The results suggested that antigen in ELISA could be preferred for detection of all those cases, which might have remained undiagnosed by performing MAT.

This study aimed to evaluate a system that identifies cartilage turn over and/or degradation through measurement of a new keratan sulfate (KS) epitope concentration in equine sera. Blood samples were collected from 30 horses, 1 (n=15) and 2 year-olds (n=15). Serum samples were analyzed for an epitope of keratan sulfate by 1/20/5D4 (KS5D4) and new epitopes of keratan sulfate using high sensitive keratan sulfate (HSKS), measured by two respective enzyme-linked immunosorbant assays (ELISAs). There was no correlation in serum concentration of KS evaluated using 5D4 and HSKS. Age had no significant effect on concentrations of KS measured with KS5D4 while 1 year-old horses showed significantly higher amounts than 2 year-olds with HSKS. Results suggest that HSKS could detect early signs of cartilage metabolic changes.

Apesar da descrição de vários casos de leishmaniose felina no mundo, pouco se sabe sobre a importância do gato como um reservatório da doença. O objetivo do presente estudo foi determinar a ocorrência de leishmaniose em gatos provenientes de uma área endêmica para leishmaniose visceral no Brasil. Duzentos gatos foram incluídos no estudo. A presença de infecção foi determinada por meio do achado de formas amastigotas do parasita em esfregaços obtidos por punção biópsia aspirativa de linfonodos, medula óssea, baço e fígado, e pela presença de anticorpos anti-L. chagasi pelo teste de ELISA indireto. Nossos resultados demonstraram uma prevalência de infecção em 14,5% (31/200) dos gatos na população estudada, sendo 4% (8/200) por diagnóstico parasitológico e 11,5% (23/200) por sorologia. | Despite the description of several cases of feline leishmaniasis around the world, little information is available about the importance of the cat as a reservoir of the disease. The aim of the present study was to determine the occurrence of leishmaniasis in cats from an endemic area for visceral leishmaniasis in Brazil. Two hundred cats were included in this study. Infection was evaluated through the presence of amastigotes in stained smears from fine-needle aspirates of lymph nodes, bone marrow, spleen and liver, and by antibody reactivity against Leishmania chagasi using indirect ELISA. Our results showed a prevalence of infection in 14.5% (31/200) of the feline population studied, with 4% (8/200) of positivity by parasitological diagnosis and 11.5% (23/200) by serology.
