BACKGROUND: Free cyanide is a potent toxic agent in the aquatic environment.  Freshwater fish are the most cyanide-sensitive group with high mortality at free cyanide concentrations above 20 μg/L. Exposure to cyanide ions can cause stress, increased mortality and place an appreciable metabolic load on fishes. Rhodanese is a ubiquitous mitochondrial enzyme in both prokaryotes and eukaryotes that detoxifies cyanide (CN-) by converting it to thiocyanate (SCN). OBJECTIVES: The purpose of this investigation was to determine and compare the pattern of tissue distribution of Rhodanese in different tissues of four native Barbus fish including Mesopotamichthys sharpey, Tor grypus, Luciobarbus xanthopterus and Luciobarbus barbulus. METHODS: Fishes (10 from each species) with length of 32.5 ± 6.5 and weight of 440 ± 110   were collected from five major fishing reservoirs of Karoun River including Gotvand, Shushtar, Molasani, Darkhoine and Ahvaz. Rhodanese activity was assayed by the method of Sorbo in the liver, kidney, gill and intestine. The unit of enzyme activity was defined as micromoles thiocyanate formed per minute at 37 °C and pH 9.2 and enzyme activity was expressed as U/mg protein. RESULTS: Rhodanese activity was detected in all tissues studied, albeit in different amounts. Specific activities of Rhodanese (U/mg protein) in different tissues ranged from 0.135 to 0.337 in the liver, 0.113 to 0.262 in the kidney, 0.121 to 0.157 in the gill, and 0.094 to 0.162 in the intestine, respectively. CONCLUSIONS: The highest activity of Rhodanese in all four species was observed in the liver and kidney, followed by the gill and intestine. Our results suggest that Rhodanese may be functional in many physiological activities in these species which needs to be clarified in detailed.

BACKGROUND: Safflower plant can be used in fish due to its antioxidant properties. In the present study, the side effects of intraperitoneal injection of safflower extract in rainbow trout have been investigated. OBJECTIVES: The effect of the intraperitoneal injection of Safflower (Carthamus tinctorius) extract on Aspartate aminotransferase, Alanine aminotransferase and Alkaline phosphatase as tissue damage indicators and also the histopathologic analysis of kidney and liver tissues in rainbow trout (Oncorhynchus mykiss) have been investigated. METHODS: To this end, rainbow trout with an average weight of 100 ±5 gr were supplied and intraperitoneally injected with different levels of Safflower extract. In this regard, one negative control group (with no injection), one positive control group (injected with 0.2 ml normal saline) and three treatment groups (injected with 50, 100 and 200 mg/kgbw of safflower extract, respectively) were considered. Blood samples were taken on the third, seventh and tenth days after injection, in order to isolate blood serums and analyze the ALP, ALATA and ASAT activities. Kidney and liver tissue samples were also taken on the seventh-day post injection. RESULTS: The levels of ALP, ALATA and ASAT activities significantly increased in all treatment groups that received safflower extract compared to control groups in all samples (sig<0.05). In histological analysis typical pathologic effects were recorded in kidney and liver tissue sections. CONCLUSIONS: Intraperitoneal injection of Safflower extract at dosages of 50-200 mg/kgbw led to damage in the liver and kidney tissues, so that the concentration of 200 mg / L had severe histological complications in these tissues. Hence some limitations must be taken into account for using this extract as immune-stimulant or vaccine adjuvant.

BACKGROUND: Cobalt is an essential nutrient for ruminants which is required for the synthesis of vitamin B12 by the ruminal microflora. Cobalt deficiency can lead to a deficiency of vitamin B12, which plays a major role in many metabolic reactions in the body. OBJECTIVES: This study was conducted to evaluate cobalt status in the serum and liver of cattle. METHODS: In different seasons, the blood serum and liver concentration of cobalt in slaughtered cattle in Urmia city were measured by atomic absorption in 151 and 196 cattle, respectively.   RESULTS: The results of cobalt measurement indicated that in 12.6% of serum and 21.4% of liver samples the concentration of cobalt was less than normal range with the Mean ± SD value of 0.64 ± 0.25 µg/dl and 0.15 ± 0.04 µg/gDM, respectively. The seasonal variations of cobalt values were assessed in serum and liver samples and following results were obtained: spring (2.18 ± 1.64 µg/dl, 0.23 ± 0.13 µg/gDM), summer (3.42 ± 1.04 µg/dl, 0.42 ± 0.27 µg/gDM), autumn (3.45 ± 0.37 µg/dl, 0.47 ± 0.38 µg/gDM) and winter (2.75 ± 0.59 µg/dl, 0.35 ± 0.09 µg/gDM). CONCLUSIONS: According to the results of this study, it seems that there is a subclinical deficiency of cobalt in cattle of this region and it is recommended that preventive measures be considered to prevent the complications and disorders caused by its clinical deficiency.

BACKGROUND: The histogenetic study is a useful and practical laboratory method for obtaining basic and effective information in order to reveal the process of histogenesis and development of organs in different stages during the embryonic period. This applied method helps us to understand the formation time of each organ and its tissue structure. OBJECTIVES: The aim of the present study was to study the histogenesis and histochemistry of the pheasant liver during the embryonic period. METHODS: Sixty fertile eggs were placed in the incubator and sampling was performed from day 5 to the end of incubation period. The liver samples were fixed in 10% Buffered Formalin and the slides were stained with Hematoxylin and Eosin (H&amp;E), Periodic Acid Schiff (PAS) and Masson Trichrome (MT). RESULTS: In this study, liver parenchyma, changes in the hepatocytes and their glycogen storage, as well as the appearance time of canaliculi, biliary ducts, central veins and port spaces were investigated. CONCLUSIONS: Differences in the time of the formation of the organs and structures in various birds relate to different incubation period or species variations. In this research, as the first study on the liver histogenesis of the pheasant, the formation of this organ from the fifth day to the end of the fetal period was examined and it was observed that the evolution and tissue development of the liver is completed until the eighteenth day of incubation period.

Parafasciolopsis fasciolaemorpha is a liver fluke typically parasitising moose in Central and Eastern Europe. The aim of our studies was to describe a case of fatal moose parafasciolopsosis, with special emphasis on the histopathological changes caused in the liver tissue by around 10,000 flukes. A male moose, found dead in Polesie National Park, eastern Poland, was subjected to parasitological necropsy. Macroscopic and histopathological examination of the liver was performed. Over 10,000 flukes identified as P. fasciolaemorpha were isolated from the liver parenchyma. Histopathological examination of the liver revealed the presence of multiple cavities, which were filled with flukes and cellular detritus and encysted with a layered capsule of connective tissue. Extensive liver fibrosis with signs of incomplete septal cirrhosis was also observed. Parafasciolopsosis with accompanying diarrhoea was the most probable reason for the moose’s death. However, it is possible that most moose are able to survive extremely intensive P. fasciolaemorpha infection by formation of extensive fibrosis, which isolates flukes from the liver parenchyma and therefore retards the failure of the organ. To the best of our knowledge, this is the first histopathological description of changes in the liver of a moose infected with P. fasciolaemorpha.

The study of histopathological changes caused by influenza A (H5N8) viral infection in bird species is essential for the understanding of their role in the spread of this highly infectious virus. However, there are few such studies under natural conditions in minor gallinaceous species. This article describes the pathomorphological findings in Colchis pheasants infected naturally with H5N8 during an epizootic outbreak in Bulgaria. Samples of internal organs of 10 carcasses were collected for histopathological and immunohistochemical evaluation, virus isolation and identification, and nucleic acid detection. Consistent macroscopic findings were lesions affecting the intestine, heart, lung, and pancreas. Congestion and mononuclear infiltrate were common findings in the small intestine, as were necrosis and lymphoid clusters in the lamina propria of the caeca. Congestion with small focal necrosis and gliosis with multifocal nonpurulent encephalitis were observed in the brain. Myocardial interstitial oedema and degenerative necrobiotic processes were also detected. Immunohistological analysis confirmed systemic infection and revealed influenza virus nucleoprotein in all analysed organs. Variable necrosis was observed in the brain, liver, trachea, heart, small intestine, and caeca. Viral antigen was commonly found in the brain, heart, lung and trachea. Contact with migrating waterfowls was suspected as a reason for the outbreak.

Introduction: The clinical symptoms of portosystemic shunts (PSSs) and hepatic microvascular dysplasia (HMD) – portal vein hypoplasia (PVH) in dogs are similar. PSSs are abnormal vascular connections between the portal vein system and systemic veins. HMD is a very rare developmental vascular anomaly, recognisable during histopathological examination. The study aim was to assess the prevalence of HMD–PVH and hepatocellular and vascular pathologies in the liver. Material and Methods: Liver biopsies from 140 dogs (of different breeds and both sexes) arousing clinical suspicion of PSS were examined histopathologically. Results: An initial PSS diagnosis was confirmed in 125 dogs (89.29%). HMD–PVH was found in 12.32% of dogs, as an isolated disease in 9.29%, especially in Yorkshire terriers, and with extrahepatic PSS in 6.67%. Histopathological analysis of muscles around sublobular veins showed that HMD cases presented hypertrophy or hypertrophy with fibrosis. In 2.17% of all dogs with liver vascular developmental disorders calcification was visible around vessels (without correlation by degenerative changes in those vessels), suggesting prior onset of deep metabolic disorders. Clinical suspicion of PSS was also formed upon quite different pathological processes in young dogs. Conclusion: Histopathological findings diagnosed the type of vascular anomalies (PSS or HMD–PVH) or other pathological changes conclusively, therefore detailed hepatic histopathology is an indispensable component of the clinical diagnostic process.

The aim of this study was to evaluate potential protective effects of propolis on furan-induced hepatic damage by assessing the levels of malondialdehyde (MDA) and reduced glutathione (GSH), antioxidant enzyme activities, and histopathological changes in the liver. Albino Wistar rats were divided into six groups: a control, propolis-treated (100 mg/kg b.w./day), low-dose furan-treated (furan-L group; 2 mg/kg b.w./day), high-dose furan-treated (furan-H group; 16 mg/kg b.w./day), furan-L+propolis treated, and furan-H+propolis treated group. Propolis and furan were applied by gavage; propolis for 8 days, and furan for 20 days in furan-L groups and 10 days in furan-H groups. While MDA levels were elevated in furan-treated groups, levels of GSH and activities of antioxidant enzymes decreased (p < 0.001). The levels of MDA and GSH and activities of antioxidant enzymes were normal in the furan+propolis groups, especially in the furan-L+propolis group (p < 0.001). While the aspartate transaminase, alanine transaminase, alkaline phosphatase, and lactate pdehydrogenase activities were elevated in the furan-H treated group (p < 0.05 and p < 0.001), they were unchanged in the furan-L treated group. Histopathologically, several lesions were observed in the liver tissues of the furan-treated groups, especially in the higher-dose group. It was determined that these changes were milder in both of the furan+propolis groups. The results indicate that propolis exhibits good hepatoprotective and antioxidant potential against furan-induced hepatocellular damage in rats.

Introduction: Clinical doses of anaesthetic agents were administered to rabbits and effects on the brain, heart, and liver were investigated biochemically and histopathologically. Material and Methods: The rabbits were randomly divided into three main groups (16 rabbits each) and each group into study (n = 8) and control (n = 8) groups. All study group rabbits received 3 mg/kg of midazolam (M) intramuscularly. Group 1.1 (M) received nothing further, group 2.1 (MK) also received 25 mg/kg of ketamine, and group 3.1 (MKI) besides ketamine was also given 2% isoflurane to induce anaesthesia for 30 min. NaCl solution in the same volume as midazolam and ketamine was injected into the controls. Results: In clinical evaluation significant differences were detected in respiratory and heart rates. In blood gas analysis the PO2 and PCO2 values showed statistical differences in anaesthesia intervals. Significant biochemical value changes were recorded in creatine kinase-Mb, glucose, and total protein. Histopathological liver examinations revealed higher total apoptotic and normal cell numbers in the MK than in the M and MKI groups. Apoptotic cell numbers were statistically significant in M and MK groups. Conclusion: Anaesthetic agents may increase programmed apoptosis. The MKI anaesthetics combination was found to cause less cell destruction in general than the other study groups. It was indicated that MKI was the safer anaesthetic combination in rabbits.

OBJECTIVE To evaluate 3 doses of gadoxetic acid (Gd-EOB-DPTA) for hepatic CT and cholangiography in cats and to determine optimal timing for hepatobiliary image acquisition and evaluation of the contrast-enhanced hepatobiliary anatomy. ANIMALS 6 healthy cats. PROCEDURES Cats were anesthetized; sequential CT scans were performed 0, 5, 25, 45, 65, and 85 minutes after IV administration of Gd-EOB-DTPA at low (0.0125 mmol/kg), medium (0.1 mmol/kg), and high (0.3 mmol/kg) doses. Hepatobiliary enhancement for each dose was objectively assessed over time and by use of a subjective semiquantitative visual assessment score. RESULTS No contrast-related adverse effects were detected. Each increase in dose of contrast medium resulted in a significant increase in HU across the hepatobiliary system. The liver had a significantly higher number of HU at 45 minutes, with homogenous enhancement at all doses of contrast medium. Contrast-enhanced cystic and bile duct HU were significantly higher and maximal at 65 minutes. Contrast-enhanced gallbladder HU did not plateau by 85 minutes. At a high dose of contrast medium, 12 of 60 (20%) biliary tract scores indicated no enhancement, 34 (57%) indicated poor enhancement, and 14 (23%) indicated moderate enhancement. No cat had excellent enhancement of the biliary tract at any dose. CONCLUSIONS AND CLINICAL RELEVANCE Gd-EOB-DTPA–enhanced hepatic CT and cholangiography in cats were safely performed and provided good hepatic enhancement but poor to moderate enhancement of the biliary tract. This technique may be useful for assessing the liver parenchyma in cats, but its value for assessing the biliary tract is questionable.
