Neutrophil activation associated with increased neutrophil acyloxyacyl hydrolase activity during inflammation in cattle

Acyloxyacyl hydrolase (AOAH) is a lysosomal enzyme found in neutrophils and macrophages that acts to partially deacylate the lipid-A component of the endotoxin of gram-negative bacteria rendering it less toxic, yet maintaining much of its immunostimulatory potential. We have found that the activity of neutrophil AOAH per cell increased during localized inflammation. The purpose of this study was to determine the mechanism(s) responsible for these increases in neutrophil AOAH activity. Because changes in neutrophil maturity commonly are associated with inflammation, intravascular infusion of purified gram-negative bacterial lipopolysaccharide and SC injection of bovine recombinant granulocyte colony-stimulating factor was used to induce large numbers of circulating immature neutrophils. Immature neutrophils were found to have AOAH activity equal to that of mature cells; however, when neutrophils were stimulated in vitro with known activators, AOAH activity of activated cells was more than that of unstimulated cells. The increase in AOAH activity was inversely related to prestimulation activity. Increases in AOAH activity after neutrophil activation were not a result of de novo synthesis of the enzyme, because cycloheximide did not prevent activation-induced increases in activity.

The toxicity of zinc ethylene-bis-dithiocarbamate (zineb), a widely used fungicide, was studied in four 4-week-old Friesian calves with immature rumen function. Calves were first subjected to liver biopsy, and thereafter, 3 of them were orally administered 200 mg of zineb/kg of body weight daily for 80 days, whereas the fourth calf served as control and remained untreated. Clinical, hematologic, and pathologic (including ultrastructural) findings were recorded. The distribution in body fluids and tissues of the parent compound and one of its main metabolites, ethylenethiourea (ETU), also was examined. Treated calves had unthrifty appearance and reduction in weight gain. They also had remarkable impairment of thyroid function, as reflected by reduction in serum concentrations of triiodothyronine and thyroxine and increase in weight of the thyroid gland associated with epithelial vacuolization and foci of hyperplasia. Moderate increase in liver glycogen content and impairment in maturation of germ cells were recorded consistently. Whereas zineb was widely distributed in body tissues, ETU accumulated mainly in the liver and the thyroid gland, although noticeable concentrations also were attained in muscle. Data were consistent with involvement of ETU mainly in the pathogenesis of thyroid gland lesions, and indicate that unweaned calves given zineb develop a clinicopathologic syndrome that does not differ qualitatively from that already described in adult cattle exposed to zineb.

Functional capabilities of morphologically mature (segmented) and immature granulocytes (neutrophils and eosinophils) from bone marrow from cows were studied and compared with similar activities of segmented granulocytes from blood. Phagocytosis of Escherichia coli and postphagocytic oxidative metabolic stimulation, measured by nitroblue tetrazolium (NBT) reduction, were evaluated simultaneously. Phagocytosis was observed readily in segmented neutrophils, neutrophilic bands, and metamyelocytes and rarely in myelocytes. Phagocytosis was not seen in promyelocytes and myeloblasts. Neutrophilic bands and metamyelocytes were phagocytically less active than were segmented neutrophils. Washed segmented bone marrow neutrophils possessed phagocytic activity similar to that of blood neutrophils, whereas the activity of unwashed segmented bone marrow neutrophils was markedly less than that of blood neutrophils. Reduction of NBT was observed only in blood segmented neutrophils and bone marrow segmented neutrophils; the magnitude of NBT reduction was significantly (P = less than 0.005) less in bone marrow neutrophils than in blood neutrophils. Eosinophils were phagocytically less competent than were neutrophils. The NBT reduction was observed only in eosinophils from blood, but not in eosinophils from bone marrow.

Five mature Holstein cows and 6 first-lactation Holstein cows were administered 100 mg of glucose/kg of body weight, IV, over a 20-minute period on postpartum day 30. A series (preinfusion, glucose infusion, and postinfusion) of blood samples was collected at -15, -10, -5, 5, 10, 15, 20, 30, 45, 60, 75, 90, 105, and 120 minutes from the start of the infusion. Serum was obtained and was assayed for glucose, immunoreactive insulin (IRI) growth hormone (GH), and free fatty acid concentrations. Baseline glucose and free fatty acid concentrations were similar in cattle of both groups throughout the sample collection period. Both groups of cattle disposed of the infused glucose in a similar manner. The first-lactation cows secreted significantly (P < 0.0001) more IRI to utilize the glucose load than did the mature cows, 71 +/- 13 microU/ml vs 38 +/- 7 microU/ml, respectively (mean +/- SEM). Preinfusion and glucose infusion GH concentrations were similar in cattle of both groups. In the postinfusion period, GH values were significantly (P < 0.0002) higher in the first-lactation cows (8.7 +/- 1.8 ng/ml) than in the mature cows (5.8 +/- 1.6 ng/ml). Compared with that in the mature cows, the higher IRI concentration required by the first-lactation cows to utilize approximately the same glucose load suggested that first-lactation cows were insulin resistant. The increased insulin response to increased glucose concentration may be one reason first-lactation cows produce less milk than do mature cows. Other factors, such as variation in the ability of the mammary gland to synthesize milk cannot be excluded.

Labor and delivery stimulate increased release of catecholamines and endogenous opioid peptides in neonates. Catecholamines promote adaptation to the extrauterine environment after birth. Enkephalins are stored together with catecholamines in the adrenal medulla and have an inhibitory effect on catecholamine release. We investigated the influence of labor and neonatal hypoxia on epinephrine, norepinephrine, and met-enkephalin release in calves. Blood samples were taken from the umbilical artery before rupture of the umbilical cord and from the jugular vein repeatedly after birth. Highest plasma norepinephrine concentration was found in calves delivered at the end of gestation (term calves) before umbilical cord rupture. In calves delivered before the physiologic end of gestation (preterm calves), norepinephrine values increased after cord rupture, but remained lower than values in term calves. Epinephrine release followed a similar pattern, but norepinephrine was clearly predominant. In term calves, met-enkephalin values were significantly higher than values in preterm calves. In calves of both groups, met-enkephalin release increased after cord rupture. During birth, the increase in catecholamine release seems to take place earlier than that of enkephalins. Norepinephrine-dominated stimulation during expulsion of the calf might be followed by increasing enkephalinergic inhibition after cord rupture and onset of respiration. Reduced release of catecholamines and enkephalins in preterm calves may be connected with delayed adaptation to the extrauterine environment.

Studying of the comparative productivity of various species and varieties of perennial grasses for the selection of the most productive species and increasing of production capacity of well-balanced feeds with low prime cost was realized in the conditions of sod-podzolic loamy soils of Vitebsk region of the Republic of Belarus. In course of study there were analyzed the following grasses: meadow clover (Trifolium pratense); creeping trefoil (Trifolium repens); great trefoil (Medicago sativa); timothy grass (Phleum pratense); meadow fescue grass (Festuca pratensis); chewing fescue (Festuca rubra); cock's-foot (Dactylis glomerata); common ryegrass (Lolium perenne); meadow foxtail (Alopecurus pratensis); awnless brome (Bromus inermis Leyss), and hybrid of fescue grass and perennial ryegrass. Studying of soil and climatic conditions in the analyzed area showed that leguminous grasses had higher yielding capacity than the cereal ones. After the 4-year observation the studied leguminous grasses provided higher yielding indexes in comparison with the cereal ones per 1 gram: dry matter – on 14,9%; available energy - on 29,7%; crude protein – on 44,7%. For the production of green mass in the conditions of Vitebsk region into the basis of planning of green and raw materials conveyor system there should be taken into consideration terms of beginning of hay cutting ripening of perennial leguminous and bluegrass grasses. Cereal grasses showed higher indexes of sugar content in 1,8 times in comparison with leguminous ones. For the provision of standard content of sugars and digestible protein in energy fodder unit it was reasonable to form grass mixtures on the basis leguminous and bluegrass grasses in ratio 60% and 40% respectively
