Nigella (Nigella sativa) plants and oil for centuries in Africa, Asia and the Middle East today is by the United States and millions of people in Europe "to promote health" was a plant used and has been a material referenced in terms of health. As a result of scientific research, it was suggested that the seeds of vitamins, active substances and essential fatty acids should be consumed by scientists due to acid. Nigella sativa has significantly increased the total lymphocyte amount and has been supported by studies that have an immunomodulatory effect on both humans and animals. Healthy life support issues to the forefront of popular Cumin seed, which is an important component of thymoquinone (TQ) has been preferred due to potential medicinal properties as a source of healing. In the scientific researches, seedlings of black seed, vitamins, active substances and essential fatty acids may be useful to be consumed due to the products.

This study was planned to evaluate the adaptive and innate immune responses by preparing and evaluating the immunizing and protective efficacy of an inactivated whole gram-negative bacteria and their crude antigens with natural adjuvant Nigella sativa (N.S) oil compared with Freund’s adjuvant (FA) then detected differential and total white blood cell count to each group in addition to cytokines responses to humoral and cellular immune responses determined by ELISA.Results of this study showed that the total leukocyte count was increased in all adjuvant groups compared with a control group and the leukocyte differential count was performed and the result indicated that the lymphocyte was increased in the group of N.S. oil adjuvant, and it was statistically significant in N. S. adjuvant with killed bacteria (G4). In contrast, monocyte percentage has increased in the two groups of Freund's vaccine adjuvant.Concerning adaptive immune responses, Freund's vaccine adjuvant directs the immunity toward cell-mediated immunity, on the other hand, the N. S. vaccine adjuvant directs the immunity toward humoral immunity as revealed by the results of ratio of IFN-γ /IL-4.In conclusion, the uses of N. S. vaccine adjuvant directed the immunity toward TH2 responses, on the other hand, Freund's vaccine adjuvants guide the immunity toward TH1 immune responses.

To investigate the antihyperglycemic potent of thymoquinone (TQ), this study has been conducted in streptozotocin-induced diabetic male rats.Diabetes was induced by single injection with streptozotocin (60 mg/kg b.w., i.p.). Rat ≥ 200 mg/dl of blood glucose was used as diabetic.Sixty five adult male rats (aged 56 days and weighted 138±8.8g) were divided into five groups, non-diabetic control (were drenched with drinking water) and four diabetic groups(DM, TQ50,TQ100, and DMI) were drenched with drinking water,TQ (50 mg/kg, bw), TQ (100 mg/kg, bw), and injected with insulin (4 IU/animal), respectively, for 42 days. During the experiment,body weight gains were recorded and blood samples were obtained weekly for assessment of plasma glucose and insulin concentrations. TQ treated male rats showed normal activity and body health throughout the experiment. Significant decrease of body weight gain has been recorded in untreated diabetic (DM) and insulin treated diabetic (DMI) groups as compared with that of intact control (C) and TQ treated diabetic (TQ50 and TQ100) groups, started from the fourth day of experiment, while DM group registered the lowest body weight gain among the experimental groups. Results of blood glucose concentrations referred to significant elevation in diabetic groups as compared with intact control. While in comparison between the diabetic groups, blood glucose concentration decreased significantly TQ50, TQ100, and DMI groups compared with DMI group. It has been found that insulin treated (DMI) and TQ treated (TQ50 and TQ100) male rats recorded no significant difference in serum insulin concentration when compared with each other but they were significantly lower than that of intact control male rats (C), but the average means of these four groups were significantly higher than that of non-treated diabetic male rats (DM).These changes were time dependent during the studied experimental period. It can be concluded that drenching of 100 mg/kg of TQ has potent hypoglycemic effect in experimentally-induced diabetic male rats.

In this study, the effects of three different ozonated oils (Sesame, Nigella sativa and Hypericum perforatum) on wound closure rate, healing process and possible complications were examined macroscopically and microscopically. Twenty-one adult Wistar albino female rats were used in the study. Subjects were divided into three groups, early wound healing, (7 days), medium wound healing, (14 days) and late wound healing (21 days). Four full-thickness skin wounds of equal size (10 mm in diameter) were formed on the back regions of all rats. This region was chosen for preventing self-inflicted injuries and reducing external irritation. The wound was left open during the healing process. While the first wound (control) received no treatment in the second wound, ozonated Sesame oil, in the third wound ozonated Nigella sativa oil and in the fourth wound ozonated Hypericum perforatum oil were used. No group obtained parenteral drug administration. First, second and third main groups were euthanized on days 7, 14 and 21, respectively. The wound healing was assessed macroscopically daily. Wound sizes of individual rats were measured with a caliper and digitally photographed every day from the day of injury. After euthanasia, all wound sites of the subjects were evaluated histopathologically.There were no significant differences in wound healing between treatments in the first day 7. On they 14, it was found that the healing was better in the group applied Nigella sativa and Sesame oils (p≤0.05). On day 21 wound healing was completed in all subjects with a resultant of better outcome in Nigella sativa oil treatment compared with others (p≤0.01).In this study the best wound healing outcome was achieved with Nigella sativa oil and Sesame oil where three different minced oils were used.

Nigella sativa (family Ranunculaceae ) is an annual plant that has been traditionally used on the Indian subcontinent and in Middle Eastern countries. In this study, we investigated the effect of N. sativa oil on the drug-metabolizing cytochrome P450 (CYP) enzymes and whether it has a protective effect against the acute hepatotoxicity of CCl4. Intraperitoneal injection of rats with CCl4 drastically decreased CYP2E1, CYP2B, CYP3A2, CYP2C11, and CYP1A2 mRNA and protein expressions. Oral administration of 1 ml/kg N. sativa oil every day for one week prior to CCl4 injection alleviated CCl4-induced suppression of CYP2B, CYP3A2, CYP2C11, and CYP1A2. Moreover, CCl4 increased iNOS and TNFalpha mRNA, while N. sativa oil administration for one week prior to CCl4 injection downregulated the CCl4-induced iNOS mRNA and up-regulated IL-10 mRNA. These results indicate that N. sativa oil administration has a protective effect against the CCl4-mediated suppression of hepatic CYPs and that this protective effect is partly due to the downregulation of NO production and up-regulation of the anti-inflamnatory IL-10.
