peer reviewed | The purpose of this study was to assess the impact of freezing and thawing on MR images of equine feet examined ex vivo. Nine equine cadaver digits were first imaged at room temperature (T0). Among the 9 digits, 3 (group 1) were imaged in a 3 Tesla MR system after one and after 2 freezing-thawing cycles. Digits of group 1 were thawed in a cold room at 4°C for 36h. Three other digits (group 2) were imaged after one freezing-thawing cycle. Digits of group 2 were thawed in a cold room at 4°C and then rescanned after 24h at room temperature. The last 3 digits (group 3) were scanned after one freezing-thawing cycle. Digits of group 3 were thawed at room temperature for 24h. Sequences used were Spin Echo (SE) T1, Turbo Spin Echo (TSE) T2 and proton density (PD), Short Tau Inversion Recovery (STIR), Double Echo Steady State (DESS), 3D Gradient Echo (GE) T1 and 2D GE T2*. Images obtained on the fresh limbs at room temperature were subjectively compared side by side to images obtained at the different freezing-thawing cycles. A quantitative analysis to assess signal change between examinations was realized by measuring signal to noise ratio (SNR). Visibility and margination of the anatomical structures of the foot and overall image quality were subjectively considered unchanged except for the hoof where the lamina was considered less visible distally after freezing and thawing in the GE T2* and in TSE T2 and PD sequences. Quantitative analysis demonstrated SNR changes in the bone marrow only in the distal phalanx in the SE T1 sequence when the feet were thawed at room temperature. When the feet were thawed in a cold room at 4°C, bone marrow SNR changes were present in the SE T1, GE T1 and TSE PD sequences. Signal changes were significant in the synovial recess when the thawing process was made at 4°C and not when the thawing process was at ambient temperature. The soft tissue structures and the hoof capsule showed significant changes with an increase of SNR, except in STIR, after freezing and thawing at 4°C and at room temperature. SNR changes in the soft tissues were mainly present in GE sequences.

1992
Nachreiner, R.F. | Oschmann, S.J. | Edquist, L.E. | Richards, J.I.
Five studies were performed to determine factors affecting progesterone concentration in skim milk. Results of the first study indicated that progesterone concentration was higher in skim milk of samples kept 16 hours in an ice bath (0 C) than of those left at room temperature (21 C). In the second study, this temperature effect was found to be reversible, with skim milk progesterone concentration increasing when whole milk samples were cooled prior to centrifugation. In the third study, [3H]-labeled progesterone was used to determine the relationship between fat content of foremilk (the first milk obtained from the teats), midmilk (milk obtained through milking), and strippings (milk obtained immediately after milking machines have been removed) samples and temperature (4 C and 21 C) on the percentage of progesterone in the skim milk fraction. The relationship between percentage of butterfat and percentage of progesterone in skim milk was linear when the log of these variables was used for calculations. In the fourth study, assayable progesterone in the skim milk fraction of foremilk, midmilk, and strippings was affected by temperature. In the fifth study, a multiple-regression procedure was used to determine the amount of variation in percentage of radioactive progesterone in the skim milk fraction. Independent variables (whole milk butterfat and temperature of incubation [1, 3, 13, 22, 37, and 50 C]) and the natural log of each variable, were entered into a stepwise multiple-regression analysis. The log of the temperature and percentage of butterfat of whole milk at the time of centrifugation accounted for 89.2% (r2 = 0.892) of the variation in the log of the progesterone concentration in the skim milk fractions. The equation describing this relationship was: log percentage of progesterone in the skim milk fraction = 4.046 - 0.144 X (log of temperature of whole milk sample) - 0.688 X (log percentage of butterfat in whole milk sample). The loss of progesterone from skim milk fractions of warm whole milk samples is possibly a physical phenomenon dependent on the temperature of the sample and its percentage of butterfat. A nomograph was created to allow others to use these variables in making adjustments in progesterone concentrations.

Biogas plant is an anaerobic digester that produces biogas fromorganic materials. The general purpose of implementing biogas plant in an animal farm is to obtain biogas from animal manure and the effluent of biogas digester after fermentation process can be used as organic fertiliser. There are 15 biogas plants based on livestock waste that have been developed throughout Malaysia until 2018. Two more biogas plants are planned to be built in Segamat and Keningau. All biogas plants built were originally aimed for domestic use which is as source of energyused for cooking and incinerator as well as to generate electricity for farm use. Out of 15 biogas plants, 7 plants were built on cattle farms, 4 plants on pig farms, 3 plantson poultry farms and only 1 plant on buffalo farm. Total construction cost for each biogas plant is between MYR4,500 to MYR30 million. Based on the survey conducted, it revealed that only 40% of the plants are still functional. However, 60% were dormant or nonfunctional due to poor maintenance, design errors, lack of technical knowledge to run the system consistently, improper planning and monitoring or insufficient livestock waste to continuously producing biogas.

Objective: Hydrogen peroxide (H2O2) was used for the activation of lactoperoxidase system on preservation of milk. Materials and methods: Milk samples were collected from Bangladesh Agricultural University dairy farm. The collected milk samples were added with 0.02, 0.04, 0.06, 0.08, 0.10, 0.12 and 0.14% of 10% H2O2 along with control one. All milk samples were kept at room temperature (28-310C). Flavor, clot on boiling (COB), acidity %, and methylene blue reduction (MBR) test were observed every one hour interval. Results: The milk samples were acceptable in terms of flavor up to 18 h at 0.14% H2O2. Similarly, this milk sample took maximum time (19 h) to give COB positive test. Acidity % was within normal range for 0.02, 0.04, 0.06, 0.08, 0.10, 0.12 and 0.14% H2O2 treated milk samples up to 8, 9, 11, 12, 12, 14, 17 and 18 h, respectively. The objective of this study was to evaluate the efficacy of EK extender alone or incorporation with CoenzymeQ10 (CoQ10) and/or soyphosphatidylcholine (SPC) in poultry semen and their effects on seminal traits during temporal storage at 4⁰C for different time intervals (12 h, 24 h, and 36 h). Heterospermic pooled semen samples diluted (1:4) with EK, EK + SPC, EK+ CoQ10 and EK + SPC + CoQ10 extenders separately, preserved and different spermiogram were assessed. Various seminal traits within the same extender differ significantly (p<0.05) among different groups and with different time intervals of storage. CoQ10 and SPC in the EK extender exhibited favorable synergistic effect on sperm quality and were able to protect the male gametes against cold-stress up to 36h at 4⁰C. In this study, we concluded that incorporation of SPC and CoQ10 together in EK extender possess novel potentiality to maintain seminal quality during liquid storage of poultry semen at 4⁰C and for their safe transportation and further use for Artificial Reproductive technologies (ARTs).
