Objective: To determine whether a novel optimized plasmid carrying the porcine growth hormone–releasing hormone (GHRH) wild-type cDNA administered at a lower dose was as effective at eliciting physiologic responses as a commercial GHRH plasmid approved for use in Australia. Animals: 134 gilts. Procedures: Estrus was synchronized and gilts were bred. Pregnant gilts were assigned to 2 treatment groups (40 gilts/group) or 1 untreated control group (24 gilts). Gilts in one of the treatment groups received the commercial GHRH plasmid, whereas gilts in the other treatment group received a novel optimized GHRH plasmid; both plasmids were administered IM in the right hind limb, which was followed by electroporation. Sow and litter performance were monitored for the 3 gestations after treatment. Results: A significant increase in insulin-like growth factor-I concentrations, decrease in perinatal mortality rate, increase in the number of pigs born alive, and increase in the weight and number of pigs weaned were detected for both groups receiving the GHRH-expressing plasmids, compared with values for the control group. Additionally, there was a significant decrease in sow attrition in GHRH-treated females, compared with attrition in the control group, during the 3 gestations after treatment. Conclusions and Clinical Relevance: Both of the GHRH plasmids provided significant benefits for sow performance and baby pig survivability for pregnant and lactating sows and their offspring during the 3 gestations after treatment, compared with results for untreated control gilts. Use of a novel optimized plasmid reduced the effective plasmid dose in these large mammals.

To determine the effect of swine hepatitis E virus (HEV) infection on pregnant gilts, their fetuses, and offspring, 12 gilts were intravenously inoculated with swine HEV. Six gilts, who were not inoculated, served as controls. All inoculated gilts became actively infected and shed HEV in feces, but vertical transmission was not detected in the fetuses. There was no evidence of clinical disease in the gilts or their offspring. Mild multifocal lymphohistiocytic hepatitis was observed in 4 of 12 inoculated gilts. There was no significant effect of swine HEV on fetal size, fetal viability, or offspring birth weight or weight gain. The offspring acquired anti-HEV colostral antibodies but remained seronegative after the antibodies waned by 71 days of age. Swine HEV infection induced subclinical hepatitis in pregnant gilts, but had no effect on the gilts' reproductive performance, or the fetuses or offspring. Fulminant hepatitis associated with HEV infection was not reproduced in gilts.

Objective: To isolate and characterize neural stem and progenitor cell populations in the brain of adult dogs. Animals: 7 healthy adult dogs. Procedures: Dogs (age, 10 to 60 months) were euthanized for reasons unrelated to the study. The subventricular zone (SVZ) adjacent to the lateral ventricles and subgranular zone (SGZ) of the hippocampus were isolated and used to generate single cell suspensions for nonadherent culture. The resulting primary neurospheres were serially passaged to assess self-renewal capacity. Neurospheres were differentiated by the withdrawal of growth factors and the addition of serum. Differentiated and undifferentiated neurospheres were analyzed via reverse transcriptase PCR assay or immunocytochemical staining for markers of pluripotency and neural lineage. Results: Neurospheres were generated from the SVZ and SGZ in all dogs. The SVZ generated more primary neurospheres than did the SGZ. Serial passage was successful, although few neurospheres could be generated after the fifth passage. Undifferentiated neurospheres were positive for SOX2, nestin, and glial fibrillary acidic protein (GFAP) and negative for OCT4 and NANOG. After differentiation, GFAP, neuronal class III β-tubulin, and 2′, 3′-cyclic nucleotide 3′-phosphodiesterase–positive progeny were noted migrating out of the neurospheres. Conclusions and Clinical Relevance: Results suggested the persistence of SOX2-positive, nestin-positive, GFAP-positive, OCT4-negative, and NANOG-negative neural progenitor cells in the SVZ and SGZ regions of mature canine brains, which are capable of producing multiple cell lineages. This study may serve as a basis for future studies investigating the role of these cells in various disease processes, such as neoplasia, or for regenerative purposes.

Humoral immune responses and protective efficacy by various doses of Salmonella ghost cells carrying enterotoxigenic Escherichia coli (ETEC) fimbrial antigens for protection against piglet colibacillosis were studied. All groups were orally primed and boosted at 11 and 14 wk of pregnancy, respectively. Group A sows were inoculated with phosphate-buffered saline (PBS), and groups B, C, and D sows were immunized with 2 × 10(9) 2 × 10(10), and 2 × 10(11) ghost cells, respectively. Serum immunoglobulin (Ig)G, and colostrum (Ig)G and (Ig)A levels of groups C and D sows were significantly higher than those of group A sows. In addition, serum (Ig)G and (Ig)A levels in group C and D piglets were significantly increased compared to those of group A piglets. After challenge with wild-type ETEC, diarrhea and mortality were not observed in group C and D piglets, while diarrhea was observed in 88.9% and 58.8% of groups A and B piglets, respectively, and 16.7% mortality was observed in group A piglets. These findings indicate that oral immunization of sows with 2 × 1010 or 1011 ghost cells can effectively protect their offspring from colibacillosis.

Objective-To determine whether cats exposed at a residence were infected with Mycobacterium bovis, whether the tuberculin skin test can identify cats infected with M bovis, and whether an ELISA could identify tuberculosis-infected cats. Animals-20 domestic cats exposed to a cat with laboratory-confirmed disseminated M bovis infection. Procedure-Cats were administered a tuberculin skin test and monitored for 72 hours. Blood and fecal samples were collected. Cats were then euthanatized, and postmortem examinations were performed. Tissues were examined grossly and histologically for signs of mycobacteriosis. Pooled tissue samples and fecal samples were submitted for mycobacterial culture. Blood samples were examined for evidence of tuberculosis by use of a comparative ELISA. Results-4 cats had positive responses for the ELISA, and 2 cats had suspicious responses. All tuberculin skin tests yielded negative results. No gross or histologic lesions of tuberculosis were detected in any tissues, and mycobacteria were not isolated from tissues or feces obtained from the 20 cats. Conclusions and Clinical Relevance-All cats that had positive or suspicious responses for the ELISA were offspring of the cat with tuberculosis. Evidence of tuberculosis was not seen in other cats at the residence, the owner, or the attending veterinarian. The most likely source of tuberculosis for the infected cat was through the consumption of M bovis-infected wildlife carcasses or offal. Because M bovis is endemic in wildlife in northeastern Michigan, there is a risk of exposure to tuberculosis in companion animals, their owners, and attending veterinarians.

Serum IgA, IgG, and IgM concentrations were determined for Beagle sires and dams of 717 matings to assess the relationship of parental immunoglobulins with the morbidity and mortality of their pups. A significant relationship was not found between parental immunoglobulins and pup mortality. Pups born to dams with low serum IgA (P < 0.001) and IgM (P < 0.02) concentrations, however, were found to have an increased incidence of sneezing, coughing, nasal discharge, and conjunctivitis. Thirty-eight percent of pups born to dams with IgA less than or equal to 40 mg/dl developed these same conditions during the first 18 weeks of life, compared with 32% of pups of dams with IgA of 41 to 65 mg/dl and 27% of pups of dams with IgA > 65 mg/dl. Similarly, 41% of pups born to dams with low IgM (less than or equal to 135 mg/dl) developed abnormal respiratory tract signs, compared with 34% and 30% of pups born to dams with medium (136 to 175 mg/dl) and high (> 175 mg/dl) IgM, respectively. Serum IgA concentrations of the sires were also associated with abnormal respiratory tract signs in pups, but this influence was evident only at 10 to 18 weeks of age. To determine biologic variability of serum IgA, 60 Beagle dams were selected from 3 serum IgA categories, low (10 to 21 mg/dl), medium (60 to 80 mg/dl), and high (125 to 210 mg/dl). A second serum IgA was determined from a sample taken 2 years later. The intraclass correlation coefficient (rI) indicated considerable biologic variability in all 3 groups: rI = - 0.24, rI = 0.09, and rI = 0.46, for low, medium, and high IgA categories, respectively. In contrast, minimal variability was noticed between observers (rI = 0.98) and in the radial immunodiffusion test itself (rI = 0.96).

Kedah-Kelantan cattle (KK) being an indigenous breed are highly adapted to the hot-humid Malaysian climate and can survive in harsh, marginal environments. This makes the KK a valuable genetic resource, given the challenges of climate change and the changing demands of the livestock sector. Hitherto there is no comprehensive programme to genetically improve the purebred KK. Genetic improvement of the KK would be to fulfill the breeding objectives of increasing lean meat growth rate, enhancing meat quality, raising feed efficiency, improving fertility and maintaining adaptability. The breeding structure proposed is a 2-tier breeding structure, with a nucleus tier followed by a commercial tier below it. The nucleus tier would comprise of a number of pedigree farms run as a community&#xD;breeding project. A sire reference scheme is proposed, where progeny of reference sires are used as genetic links between pedigree herds and between years. Some guidelines are offered on the establishment&#xD;and implementation of the scheme. Modern breeding technology such as BLUP using an Animal Model, artificial insemination, embryo transfer, tissue scanning, MAS and MAI could be used as tools to support the KK genetic improvement programme. To address the problem of genetic erosion, emphasis should be given to the conservation and sustainable utilization of the KK. The programme is expected to have a high impact on the livestock sector. Substantial investments are needed to develop infrastructure and human capital associated with the KK breeding programme. The establishment of a KK breed society will improve awareness concerning the KK and protect the interests&#xD;of the KK breeding community.

First lactation milk yield per day of lactation length of 379 Sahiwal cows, collected from cattle breeding farm, Chak Ganjaria, Lucknow were utilized for the present study. The optimum number of progeny size to achieve the accuracy attained by different methods of sire evaluation (Daughter's average (DA), contemporary comparison (CC), Least-squares (LS), Simple Regressed Least-squares (SRLS) and Best linear unbiased prediction (BLUP» ranged from 47.615 (CC) to 161.069 (BLUP). Similar results were also obtained in the ratio ofoptimum progeny size estimated by different methods with that of actual progeny size per sire available ranging from 5.270 (CC) to 17.828 (BLUP). Present findings revealed that BLUP method showed highest increase in accuracy of 207.72% among all the methods studied.

Influence of 33 imported heads Landrace breed boars of foreign selection on feeding performance and meat productivity of swine of the Belarusian Meaty breed was studied in the conditions of the Republic of Belarus for the enhancement of Belarusian breeds genealogical structure. There was stated the positive influence of Landrace Breed on feeding performance and meat productivity of mixed bred stores obtained after the introductive crossing with sows of Belarusian Meaty breed which was expressed in the predominancy of mixed bred hogs over the purebred ones at the stage of fattening performance test: at the age of reaching 100 kg body weight – on 0,7%; at average daily weight gain - on 1,5%; at feed consumption - on 1,2%. Mixed bred stores also had thinner back fat (on 0,9 mm) and bigger loin (on 0,4 square cm). There was stated the one-sided influence of boars of Landrace Breed on feeding performance and meat productivity indices of mixed breeds. It was established, that hybrids of four lines (Zaboj, Zalet, Zont and Bison) with regard to control groups showed the effect of heterosis at age of achievement of body weight of 100 kg, at a daily average weight gain and at forage consumption in the following rates: 1,5% (Р less than 0,05), 2,8% and 1,5% (Р less than 0,05) for Zaboj line; 2,3%, 4,9% and 2,1% (Р less than 0,01) for Zalet lines; 1,4%, 3% and 1,5% (Р less than 0,05) for Zont lines; 3% (Р less than 0,001), 6,7% and 2,7% for a line of Bison. In meat productivity indices there was stated the in-between character of inheritance which caused the decreasing of fat thickness and increasing of loin eye area at descendants of trial groups of lines: Zaboj, Zalet, Zenit, Zont on 0,88 mm (Р less than 0,05) and 0,6 square cm, 1,7 mm (Р less than 0,01) and 2,1 square cm (Р less than 0,001), 1,3 mm (Р less than 0,01) and 0,9 square cm, 1,4 mm
