Combined effects of fasting and diet on interferon production and virus replication in calves infected with a vaccine strain of infectious bovine rhinotracheitis virus

A study was undertaken to investigate the combined effects of fasting and different diets on interferon (IFN) production and virus replication measured in nasal secretions of calves inoculated with a vaccine strain of infectious bovine rhinotracheitis virus. Four groups of calves were inoculated intranasally with infectious bovine rhinotracheitis virus. Two groups were inoculated 24 hours after onset of a 3-day fast; upon refeeding, 1 group was fed a maintenance diet (M diet) of hay, and the other was fed a higher energy diet (HE diet) of hay and concentrate. Nonfasted control groups were fed the M diet or the HE diet. Overall IFN production was highest (P less than 0.01) in nonfasted calves fed the M diet throughtout the study and lowest in nonfasted calves fed the HE diet. Fasted calves refed the HE diet produced consistently and significantly more IFN than did nonfasted calves fed this diet. Fasted calves refed the M diet, however, produced significantly less IFN, compared with control calves fed the M diet throughout the study. Overall mean virus excretion was similar in all groups; therefore, the amount of virus replication per se did not account for the differences in IFN production, nor did greater IFN production result in less virus excretion. Serum cortisol concentrations and immune responses were not significantly affected by fasting or diet.

Amendments to the Animal Welfare Act (PL 99-198) require that an exercise program for dogs be established by the attending veterinarian. A 6-week study was conducted to determine the effects of a moderate exercise program in purpose-bred Beagles. Sixteen male Beagles (4/group) were maintained as follows: (1) standard cage without exercise; (2) standard cage with individual exercise periods (35 minutes, 3 times/week); (3) large cage without exercise; and (4) standard cage with group-release exercise periods. Blood samples were collected for CBC, serum biochemical analysis including determination of serum cortisol concentration, and immune function (lymphocyte transformation assay). Group-released dogs interacted with each other during most of the exercise time. Fighting in these dogs occurred only during the third week. Dogs had little inclination to exercise when released along into the exercise area. Regardless of the size of the cage, dogs did not exercise unless human beings were present in the room. There were no significant differences in laboratory findings among dogs in the 4 groups. This moderate exercise program had no demonstrable effects. Similarly, continuous cage housing, without a formal exercise program, could not be determined to be detrimental to the physiologic or health status of dogs.

A significant (P less than 0.0001) positive correlation was demonstrated between left ventricular internal chamber dimension in diastole or systole and body weight, body surface area, cycle length, and the square root of cycle length. On the basis of adjusted coefficients of determination, multiple regression analysis, using body weight or body surface area and cycle length or the square root of cycle length, was superior to separate simple regression with these variables in accounting for variations in left ventricular internal chamber dimensions. Shortening fraction had a significant (P less than 0.0001) negative correlation and left ventricular free wall measurements had a significant (P less than 0.0001) positive correlation to body weight and body surface area. For these echocardiographic variables, correlation to the square root of cycle length was insignificant (P greater than 0.05), and a multiple regression model was not helpful in developing confidence intervals. Septal wall measurements were not correlated with body weight, body surface area, cycle length, or the square root of cycle length.

Twenty-four male domestic shorthair cats were used to evaluate the acute and chronic effects of a single, toxic but sublethal, orally administered dose of chlorpyrifos. A dose of 10 mg/kg of body weight did not induce clinical signs of toxicosis, but a dosage of 40 mg/kg induced clinical signs of toxicosis, and 1 of 12 cats died. Chlorpyrifos given at a dosage of 0.1 mg/kg to 2 cats reduced whole blood and plasma cholinesterase (Che) activities to values obtained after cats were given doses that induced clinical signs of toxicosis. Regeneration time for whole blood and plasma Che activities ranged from 7 to 28 days. Brain Che activity was considerably decreased in 1 cat that died 4.5 hours after dosing, but was normal in all others at 28 days after dosing. Other than decreased Che activity, significant changes were not seen in hematologic or serum biochemical values. Toxin-related lesions were not seen during macroscopic or microscopic examination.

Normal sensory nerve conduction velocity (SNCV) values in 8 ponies and 8 horses were compared by use of a percutaneous signal-averaging technique. Nerve fibers evaluated included those in the medial and lateral palmar and plantar digital nerves. Mean SNCV values were significantly slower (P < 0.0002) for horses, compared with those values for ponies. Animal height and nerve segment length were inversely related to SNCV consistently. The SNCV values were affected by surface skin temperature by a factor of approximately 1.2 m/s change for 1 degrees C change in temperatures from 35 C. The ability to calculate warning limits to define those SNCV values in normal and abnormal ranges were developed from these data for both ponies and horses.

Tracheas from 15 toy breed dogs with normal tracheas and 6 dogs with collapsed tracheas were examined histologically and histochemically. Tracheal cartilage was analyzed for chondroitin sulfate by means of alcian blue (CEC method) and for calcium with alizarin red S. Cartilage arcs from dogs with collapsed tracheas had areas that were apparently hypocellular, and some had other areas that appeared like fibrocartilage or fibrous tissue. Histochemically, collapsed tracheal cartilage had less chondroitin sulfate and calcium than did normal tracheal cartilage. Cartilage arcs from the collapsed tracheas were not uniformly affected to the same degree, and parts of a given tracheal arc appeared normal, whereas other parts had an abnormal histologic appearance.

Bilateral kidney translocation was performed on 7 adult sheep. In each sheep, we took 9 renal biopsy specimens (40 mg each) percutaneously between 36 and 110 days after surgery. The serum urea nitrogen and creatinine concentrations remained normal, at least through post-operative day 66 (7 biopsy specimens). The 7 sheep were euthanatized and necropsied 113 days after surgery; 75 to 90% of each kidney was normal. The only abnormal areas of each kidney were attributable to resolving biopsy lesions. This surgical model may allow for fewer animals to be used for tissue residue or nephrotoxicity studies.

Autologous canine neutrophils were labeled with technetium 99m and reinjected in 7 dogs with experimentally induced focal abscessess to determine the ability of scintigraphy to localize a focus of sepsis (abscess). Good localization of labeled cells in an abscess was achieved; however, a large portion of the technetium 99m eluted from the neutrophils.

Clinical signs of respiratory tract disease were observed in chickens that were inoculated intratracheally with 1 x 10(6) oocysts of Cryptosporidium baileyi at 2 or 14 days of age (10 chickens/group), but not in chickens inoculated at 28 or 42 days of age (10 chickens/group). Orally inoculated chickens in all age groups (10 chickens/group) did not develop clinical signs of disease. Orally and intratracheally inoculated chickens in all age groups were infected, as determined by the finding of cryptosporidia in tissue sections of the trachea, bursa of Fabricius, and cloaca, and by the recovery of oocysts from their feces. Chickens inoculated at 2 and 14 days of age excreted oocysts for a longer period and had greater numbers of cryptosporidia in their tissues, compared with chickens inoculated at 28 and 42 days of age.

Three Beagles with chronic anemia and reticulocytosis were studied. The dogs originated from a large breeding colony and appeared clinically normal with the exception of splenomegaly. The PCV ranged from 30 to 39% (normal, 46 to 56%), with reticulocyte indices of 2.3 to 9.9. Red blood cells were morphologically normal, and examination of marrow aspirates revealed erythroid hyperplasia. Shortened chromium-51 RBC life-spans (7.2 to 15.4 days in anemic dogs; 22.2 to 25.2 days in control dogs) documented a hemolytic anemia. Acquired causes of hemolytic anemia were ruled out. Red blood cells had normal glycolytic enzyme activities, no evidence of unstable or abnormal hemoglobin, and had altered osmotic fragility curves. The breeding of 2 anemic dogs resulted in off-spring with anemia and reticulocytosis. Polyacrylamide gel electrophoresis revealed no abnormalities in RBC membrane cytoskeletal proteins in all anemic adult dogs and in 3 offspring.
