Effect of exercise intensity on plasma prostaglandin concentrations in horses

Exertion has an effect on plasma, serum, and/or urine prostanoid concentrations in many species. We investigated the effect of exercise intensity on plasma prostaglandin concentrations during and after exercise in horses. Six Thoroughbreds completed 4 trials: 3 exercise trials (low-, medium-, and high-speed) and 1 nonexercise (control) trial on a high-speed treadmill. Blood samples were collected from a jugular catheter before, during and after exercise. The PCV and blood lactate, plasma protein, plasma prostacyclin (6-keto-PGF1 alpha), thromboxane B2 (TXB2), and prostaglandin E2 (PGE2) concentrations were measured before, during, and after exercise. Exercise significantly (P = 0.001) increased plasma TXB2 concentration during and after exercise in the low-, medium-, and high-speed trials, although effect of exercise intensity was not detected. Exercise was associated with an increase in PCV and blood lactate and plasma protein concentrations. There was no effect of exercise on plasma 6-keto-PGF1 alpha concentrations; PGE2 was not detected in plasma from any horse in any trial.

The pharmacokinetic properties of enrofloxacin were determined in broiler chickens after single IV and orally administered doses of 10 mg/kg of body weight. After IV and oral administrations, the plasma concentration-time graph was characteristic of a two-compartment open model. The elimination half-life and the mean +/- SEM residence time of enrofloxacin for plasma were 10.29 +/- 0.45 and 9.65 +/- 0.48 hours, respectively, after IV administration and 14.23 +/- 0.46 and 15.30 +/- 0.53 hours, respectively, after oral administration. After single oral administration, enrofloxacin was absorbed slowly, with time to reach maximal plasma concentration of 1.64 +/- 0.04 hours. Maximal plasma concentration was 2.44 +/- 0.06 micrograms/ml. Oral bioavailability was found to be 64.0 +/- 0.9%. Statistically significant differences between the routes of administration were found for the pharmacokinetic variables-half-lives of the distribution and elimination phase and apparent volume of distribution and volume of distribution at steady state. In chickens, enrofloxacin was extensively metabolized into ciprofloxacin. Residues of enrofloxacin and the major metabolite ciprofloxacin in fat, kidney, liver, lungs, muscles, and skin were measured in chickens that received an orally administered dose of 10 mg/kg once daily for 4 days. The results indicate that enrofloxacin and ciprofloxacin residues were cleared slowly. Mean muscle, liver, and kidney concentrations of the metabolite ciprofloxacin ranging between 0.020 and 0.075 micrograms/g persisted on day 12 in chickens after dosing. However, at the time of slaughter (12 days), enrofloxacin residues were only detected in liver and mean +/- SEM concentration was 0.025 +/- 0.003 micrograms/g.

The Cooper isolate of bovine herpesvirus 1 (BHV1) was used to produce a thymidine kinase-negative (TK-) recombinant by insertion of a beta-galactosidase (bgal) expression cassette into the TK coding region. The recombinant virus (TK- bgal+) was tested for abortifacient activity in cattle by inoculation of 5 pregnant heifers at 25 to 29 weeks gestation. Five additional heifers were inoculated with the Cooper TK-positive (TK+) virus to serve as controls. After inoculation, both groups of heifers developed similar febrile responses and neutralizing antibody titers. Virus was isolated from blood of all heifers during the first postinoculation (PI) week, and isolation frequencies were similar for both groups. In contrast, whereas virus was isolated from many of the nasal and vaginal swab specimens of heifers inoculated with TK+ virus, only rare virus isolations were made from the heifers given TK- bgal+ virus. All heifers inoculated with TK+ virus aborted between PI days 19 and 35. The finding of characteristic microscopic lesions and viral antigen in fetal tissues indicated that the abortions were caused by BHV-1 infection. Virus was isolated from 3 fetuses, and all isolates were TK+. Two heifers inoculated with TK- bgal+ virus aborted at PI days 25 and 39. Fetal tissues had typical BHV-1 microscopic lesions and viral antigen. Virus was isolated from blood of both fetuses, and the isolates were TK- bgal+. Results of this study indicate that inactivation of the TK gene reduces, but does not eliminate, the abortifacient activity of BHV-1.

To examine the postnatal development of equine small intestine, biopsy specimens of jejunal mucosa from 8 ponies, between 6 and 28 weeks old, were subjected to analytical subcellular fractionation and assay of organelle marker enzymes. Fractionation revealed a reduction in the particulate brush border component of beta-galactosidase (lactase) activity between 6 and 28 weeks, and a corresponding increase in soluble activity, although the reduction in mean specific activity was not significant. There also was a decrease in the proportion of brush border to soluble aminopeptidase N activity, a relative loss of brush border gamma-glutamyltransferase activity, and a considerable decrease in the specific activity of alkaline phosphatase throughout the gradient fractions. In contrast, there were marked increases in activities of (alpha-glucosidase (maltase) and sucrase in the older ponies, accompanied by considerable changes in the intracellular distribution of particulate alpha-glucosidase activity, which was predominantly associated with endoplasmic reticulum at 6 weeks, whereas the large increase in activity observed by 28 weeks was clearly associated with the brush border. The modal density of brush borders also increased with age, suggestive of an increase in the glycoprotein-to-lipid ratio of the microvillar membrane. In contrast to these brush border changes, there was relatively little alteration in the activities or density distributions of marker enzymes for endoplasmic reticulum, basolateral membranes, mitochondria, or lysosomes. These findings indicate that maturation of equine intestinal epithelium during the first few months of life results in major changes in the properties and enzyme composition of enterocyte brush borders.

Activities of diagnostically important enzymes were measured in serum and lysates of liver, kidney, skeletal muscle, heart, intestine, lung, and pancreatic tissues from wild-caught yellow rat snakes, Elaphe obsoleta quadrivitatta. All samples were analyzed for alkaline phosphatase, lactate dehydrogenase (LD), aspartate transaminase (AST), alanine transaminase, gamma-glutamyltransferase, and creatine kinase (CK) activities. The major enzyme activities found in the liver were LD and AST. The kidney had moderate activities of LD, AST, alanine transaminase, and CK. Skeletal muscle and heart contained high CK activity. Intestine, lung, and pancreas had low activities for most enzymes analyzed. Little to no gamma-glutamyltransferase activity was found in serum or tissues analyzed. Serum enzyme activities in yellow rat snakes were similar to those described for other reptile species, except for serum CK activity, which was increased in rat snakes.

Serum alpha-mannosidase activity and swainsonine concentration were determined in 4 cattle and 15 sheep (3 groups of 5 each) that were administered ground locoweed (Oxytropis sericea or Astragalus lentiginosus) containing swainsonine at dosages of approximately 0.8 mg/kg of body weight/d (cows, 30 days each) and 0, 1.0, and 1.5 mg/kg/d (sheep, 11 days each). The cattle developed mild clinical signs of locoism, including signs of depression, lethargy, and slight intention tremors. Clinical signs of toxicosis were not observed in the sheep. Within 24 hours of initial treatment, serum alpha-mannosidase activity in cows and sheep, measured by the release of 4-methylumbelliferone from an artificial substrate, was markedly decreased to 28 and 40 micromoles of 4-methylumbelliferone/L, respectively. Mean serum alpha-mannosidase activity of control cows and sheep was 400 +/- 94 and 422 +/- 42 (mean +/- SD), respectively. In the treated animals, decreased serum alpha-mannosidase activities returned to normal or higher activities within 6 days after treatment was discontinued. Using a jack bean alpha-mannosidase assay, increased swainsonine activity (153, 209, and 381 ng/ml, respectively) was detected in the serum of cattle and of sheep in the high- and low-dose groups within 24 hours after treatment with locoweed. Swainsonine concentration remained high, with mean concentrations of 204, 432, and 395 ng/ml (cows and 2 sheep groups, respectively) during the treatment period. After treatment, swainsonine was rapidly cleared, with estimated serum half-life of 16.4, 17.6, and 20.3 hours (cows, and high- and low-dose sheep groups, respectively). Significant differences in either alpha-mannosidase activity or swainsonine concentration were not detected between the 2 groups of treated sheep. These results suggest that serum alpha-mannosidase and swainsonine values are sensitive indicators of locoweed intoxication in cattle and sheep. Furthermore, it suggests that swainsonine is rapidly absorbed, resulting in rapid inhibition of serum alpha-mannosidase activity, leading to high serum swainsonine concentration. After exposure is eliminated, swainsonine is rapidly cleared from the serum, with serum alpha-mannosidase activity returning to normal values shortly thereafter.

A rapid subjective method for estimating the extent of gross pneumonia lesions in slaughtered pigs was compared with dissection of lungs in 51 slaughtered pigs. After standardization for prevalence in the regional industry, regression analysis indicated that the subjective method was highly predictive of the extent of pneumonic lesions (R2 = 0.88). Part of the error with the subjective method was attributed to approximations used for the relative proportions of lung lobes, which result in overestimation of the affected tissue by approximately 90%. Retrospective analysis of data from a slaughter monitoring program revealed strong associations (R2, 0.54 to 0.91) between prevalence, mean, median, and maximal lung scores in groups of pigs. Maximal lung score was biased by sample size, but prevalence and mean or median lung scores could be used to describe pneumonia severity in groups of pigs. Our results indicate that error in measurement of the extent of pneunomic tissue in slaughtered pigs is unimportant if the time of onset, clinical severity, and duration of disease are not quantified.

Six horses, highly trained for dressage competition, were used to study the stride kinematics of the walk, and to compare the kinematics of the collected, medium, and extended walks. Horses were filmed in a sagittal plane at a rate of 150 frames/s; temporal, linear, and angular data were extracted from the films. Results of ANOVA and Duncan's multiple range test indicated that the speed of the collected walk (1.37 m/s) was significantly (P < 0.01) slower than that of the medium (1.73 m/s) and extended (1.82 m/s) walks, values for which were not significantly different from each other. The increase in speed was associated with a significant increase in stride length, from 157 cm in the collected walk to 193 am in the extended walk. This was a result of an increase in the over-tracking distance, whereas there was no significant difference in the distance between lateral placements of the limbs. Stride duration decreased (P < 0.01) from the collected walk (1,159 ms) to the extended walk (1,064 ms). Angles of the metacarpal and metatarsal segments, measured on the palmar/plantar aspect, were higher at impact and lower at lift off in the collected than in the extended walk (P < 0.01). This indicated greater range of angular motion of this segment during the stance phase in the extended walk. Only 1 of the 6 horses had a regular 4-beat rhythm of the footfalls, with equal time elapsing between the lateral and diagonal footfalls.

Pharmacokinetic variables of bupivacaine (BPV) were determined after IV and epidural administrations in the same 6 dogs. Plasma BPV concentration curves after IV administration were adjusted to biexponential kinetics: a rapid distribution phase was followed by a slower elimination phase, with half-life of 34.5 +/- 7.8 minutes. Mean plasma clearance was 20.2 +/- 7.4 ml/min/kg of body weight, and mean volume of distribution at steady state was 0.7 +/- 0.2 L/kg. After epidural administration, absorption was rapid. Peak plasma concentration, 1.4 +/- 0.4 microgram/ml, was detected approximately 5 minutes after BPV administration. The half-life corresponding to epidural administration (179 +/- 33.6 minutes) was 5 to 6 times longer than that observed after IV administration, possibly because of the slow release of BPV from the epidural space. Induction times were short (2.3 +/- 2.2 minutes); anesthesia quickly began and lasted for more than 2 hours (158 +/- 48.8 minutes). During that period, BPV plasma concentration ranged between 1.4 and 0.2 microgram/ml. Changes in systolic blood pressure and heart rate were correlated to high plasma concentration of BPV. These modifications were observed for the first 30 minutes, reaching baseline values after 60 minutes.

Intraocular pressure (IOP) was measured, using applanation tonometry, in both eyes of 20 horses after topical application of 0.5% proparacaine to the cornea. Ultrasonic pachymetry was used to measure central, mid-peripheral, and peripheral corneal thickness (CT) in all 4 quadrants of both eyes of 25 horses. All measurements were repeated after auriculopalpebral nerve block, sedation by IV administration of xylazine, or combination of nerve block and sedation. Mean IOP after topical anesthesia of the cornea was 20.6 +/- 4.7 mm of Hg for the left eye and 20.35 +/- 3.7 mm of Hg for the right eye. Mean central CT was 793.2 +/- 42.3 micrometers. The peripheral part of the cornea was significantly (P < 0.05) thicker, on average, than the central part of the cornea. Auriculopalpebral nerve block had no significant effect on IOP or CT. Intravenous administration of xylazine resulted in a significant (P < 0.05) decrease in IOP, but had no effect on CT.
