Long-term persistence of Toxoplasma gondii in tissues of pigs inoculated with Toxoplasma oocysts and effect of freezing on viability of tissue cysts in pork

To study the distribution of tissue cysts in porcine tissues, 16 pigs were fed oocysts of 4 strains of Toxoplasma gondii (4 pigs/strain). Pigs were euthanatized between postinoculation days 103 and 875 and portions of 5 to 14 organs were bioassayed in mice and/or cats for T gondii. For bioassays, 50- to 100-g portions of tissue were incubated in acidic pepsin solution to free bradyzoites from cysts in parenchyma, and washed sediment from the digests of each specimen was inoculated SC into mice (6 mice/organ). For bioassays in cats, a 500-g portion or whole organ was fed to Toxoplasma-free cats (1 cat/organ). Toxoplasma gondii was recovered from tissues of 14 of the 16 pigs (from the brains of 12, hearts of 11, tongues of 10, and diaphragms of 6). Toxoplasma gondii was isolated from commerical cuts of meat from 5 infected pigs, from the arm picnic and ham of 3, Boston butt, spareribs, and tenderloin of 2, and bacon and tailbone of 1. Regarding the 4 pigs euthanatized between postinoculation days 759 and 865, cats shed T gondii oocysts after the ingestion of hearts of all 4; tongues of 3; bacons, hams, arm picnics, Boston butts, spareribs, and diaphragms of 2; and livers, kidneys, and tenderloins of 1. Toxoplasma gondii was found to be inconsistently distributed among the organs and muscles, but overall, tongue and heart were more heavily infected than were other tissues. Tissue cysts in pork were rendered nonviable at -12 C for 3 days.

Ponies with recurrent airway obstruction (principal ponies) and their controls were given aerosolized Micropolyspora faeni antigen via endotracheal tube during a period when the principal ponies were in disease remission. In both groups of ponies, we performed bronchoalveolar lavage (BAL) and measured pulmonary function at base line, and 5 hours after aerosol administration of 30 ml of 0.9% NaCl solution or 30 ml of 1% w/v particulate M faeni antigen in 0.9% NaCl solution. In both groups of ponies, aerosolized M faeni antigen increased WBC count, neutrophil numbers, and albumin concentration in BAL fluid, but macrophage numbers decreased. In the principal ponies, BAL mast cell numbers were decreased 5 hours after administration of M faeni antigen. The M faeni antigen had no effect on the mechanical properties of the lungs or on gas exchange in the control ponies, but did increase respiratory frequency minute ventilation and pulmonary resistance, and decreased arterial oxygen tension in the principal ponies. Changes in pulmonary function were apparent only in the principal ponies, which suggests that neutrophils, per se, do not cause pulmonary dysfunction and that M faeni may be one of the etiologic agents involved in chronic obstructive pulmonary disease.

Intramammary devices (IMD) were abraded with medium-grade emery cloth or were left smooth. One IMD of each type was inserted into a mammary quarter of each of 5 lactating cows. The remaining 2 quarters served as controls. Quarter foremilk, bucket milk, and stripping milk samples were collected for 3 consecutive days at 2 weeks after IMD insertion, and milk somatic cell counts (SCC) were determined. Milk samples also were collected immmediately after and 0.5, 1,2,4,6,8, and 11 hours after milking. All quarters were challenge exposed with 250 colony-forming units of Streptococcus uberis at 2 months after IMD insertion. Foremilk and stripping milk samples were collected for bacteriologic culture and SCC at the next 10 milkings. Mean foremilk, bucket milk, and stripping milk SCC (X 10(6) cells/ml) were 0.18, 0.07, and 0.91, respectively, for quarters with abraded IMD; 0.06, 0.05, and 0.43, respectively, for quarters with smooth IMD; and 0.03, 0.03, and 0.15, respectively, for control quarters. Mean SCC after milking (X 10(6) cells/ml) for the various intervals were 0.70, 1.29, 0.70, 0.97, 1.15, 1.17, 0.77, and 0.85 for quarters with abraded IMD; 0.43, 0.62, 0.61, 0.45, 0.64, 0.60, 0.31, and 0.26 for quarters with smooth IMD; and 0.15, 0.24, 0.15, 0.19, 0.15, 0.15, 0.14, and 0.06 for control quarters. After challenge exposure, 2 of 5 of the quarters with abraded IMD, 4 of 5 of the quarters with smooth IMD, and 8 of 9 of control quarters became infected. Results indicated that abraded IMD increased SCC in stripping milk to concentrations that provided 60% portection against challenge eexposure with S uberis.

To obtain synchronous infection, 10 cats were inoculated with feline herpesvirus-1 (FHV-) on the oral, nasal and conjunctival mucosa. Swab specimens of the nasal conjunctival, and pharyngeal mucosa were obtained for virus isolation from each cat before inoculation and at 3-day intervals thereafter until postinoculation day 21. Recovery of virus and evidence of clinical signs were used to document FHV-1 infection. Serum was obtained from blood samples collected sequentially from each cat between day 0 and postinoculation day 90. Virus-neutralizing antibody titer was determined in all serum specimens. Immunoprecipitation with [35S]methionine- and [14C]glucosamine-labeled viral antigens, followed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, was performed on each specimen. Three precipitation bands with approximate molecular weights of 105,000, 68,000, and 60,000 were separated from [14C]glucosamine- and [35S]methionine-labeled immunoprecipitates. The concurrent detection of virus-neutralizing antibody glycoprotein-specific immunoprecipitins implied that in cats, the FHV-1 glycoproteins were important in the induction of virus-neutralizing antibodies to FHV-1.

Cardiovascular effects and pulmonary gas exchange were compared during conventional mechanical ventilation (CMV) and interrupted high-frequency, positive-pressure ventilation (IHFPPV) in 6 anesthetized ponies in dorsal recumbency. When the peak airway pressure (Paw) was held constant at control values attained during CMV (18 to 20 cm of H2O), and the ventilator frequency of IHFPPV was varied over the range, 2.5 to 12.5 Hz, significant (P less than 0.05) changes from control values were observed only in the ratio of dead-space volume to tidal volume (VD/VT) and in the respiratory minute volume (VE). The mean +/- SEM) carbon dioxide excretion (VCO2) was 2.12 +/- 0.1 ml/kg/min during IHFPPV. Dead-space ventilation ranged from 40 to 73.7% of total ventilation and increased directly with increasing frequency. The VE also increased, from 89 ml/kg/min at a ventilatory frequency of 2.5 Hz to 145 ml/kg/min at a frequency of 12.5 Hz. Maintaining the frequency of IHFPPV constant at 12.5 Hz and increasing the Paw over the range of 5 to 30 cm of H2O caused significant (P less than 0.05) changes in arterial partial pressure of O2 (PaO2), VCO2, pulmonary shunt fraction (QS/QT), VE, arterial-alveolar differences in oxygen tension (AaDO2), VD/VT, and cardiac output, compared with CMV. The PaO2 and the VCO2 increased linearly with increasing Paw. With increasing Paw, VD/VT decreased directly with increasing Paw from 98 to 69.3%. Gas exchange at a Paw of 15 cm of H2O during IHFPPV was equivalent to conditions at Paw of 20 cm of H2O during CMV. At a higher Paw during IHFPPV, improvements over control values were observed in gas exchange.

A study was undertaken to investigate the combined effects of fasting and different diets on interferon (IFN) production and virus replication measured in nasal secretions of calves inoculated with a vaccine strain of infectious bovine rhinotracheitis virus. Four groups of calves were inoculated intranasally with infectious bovine rhinotracheitis virus. Two groups were inoculated 24 hours after onset of a 3-day fast; upon refeeding, 1 group was fed a maintenance diet (M diet) of hay, and the other was fed a higher energy diet (HE diet) of hay and concentrate. Nonfasted control groups were fed the M diet or the HE diet. Overall IFN production was highest (P less than 0.01) in nonfasted calves fed the M diet throughtout the study and lowest in nonfasted calves fed the HE diet. Fasted calves refed the HE diet produced consistently and significantly more IFN than did nonfasted calves fed this diet. Fasted calves refed the M diet, however, produced significantly less IFN, compared with control calves fed the M diet throughout the study. Overall mean virus excretion was similar in all groups; therefore, the amount of virus replication per se did not account for the differences in IFN production, nor did greater IFN production result in less virus excretion. Serum cortisol concentrations and immune responses were not significantly affected by fasting or diet.

The effect of glucocorticoids on early follicular growth in sows undergoing normal estrous cycles was evaluated by administration of dexamethasone during the middle of the luteal phase. Plasma specimens were obtained for measurement of luteinizing hormone (LH), follicle-stimulating hormone (FSH), progesterone, and estradiol-17 beta concentrations. Fifteen sows were used. Control sows (n = 5) were given physiologic saline solution twice daily from day 9 to day 14 of the estrous cycle. Sows of the second group (n = 5) were given dexamethasone (30 microgram/kg of body weight, IM) similarly, and those of the third group (n = 5) were given dexamethasone plus gonadotropin-releasing hormone (GnRH+ 50 microgram at 6-hour intervals, IV). Plasma specimens, obtained twice daily from day 8 through day 26, indicated that progesterone production and luteal regression were not inhibited by any of the 3 treatment regimens. Although preovulatory plasma estradiol concentration increased in control sows, such was not observed in the sows treated with dexamethasone or dexamethasone plus GnRH (P less than 0.01). Ovulation, with formation of corpora lutea, occurred in gilts given saline solution. Dexamethasone administration resulted in persistence of 19 to 41 follicles/ovary (2 to 4 mm in diameter), and dexamethasone-plus-GnRH treatment resulted in 6 to 18 follicles/ovary (5 to 6 mm in diameter). Plasma was obtained at 15-minute intervals for 12 hours to compare the effect of treatmenton hormone concentrations on day 12 of the estrous cycle with the values on day 8. Glucocorticoid administration had no significant effect on mean concentration, final concentration excluding those hormone concentrations that constituted part of a pulse (referred to as base line), number of pulses, pulse amplitude, and area under the pulse for either gonadotropin. Addition of GnRH to dexamethasone treatment significantly (P less than 0.01) increased all plasma LH values, but only base-line concentration of FSH. For estradiol, pulse amplitude and mean pulse area were increased (P less than 0.05), and although the frequency of pulses was not significantly altered, base-line concentration in glucocorticoid-treated sows was significantly reduced, compared with that of control sows. In sows treated with GnRH plus dexamethasone, the pulse frequency of estradiol was significantly (P less than 0.01) increased, but pulse area and amplitude were similar to those of sows given saline solution. Dexamethasone treatment was associated with an increase in mean and base-line concentrations of progesterone. The results suggest that high midcycle glucocorticoid concentrations (1) do not inhibit luteal function or regression, (2) have little influence on LH and FSH secretion during the middle of the luteal phase, (3) alter the pattern of estradiol secretion, (4) are associated with the persistence of small ovarian follicles, and (5) result in the development of fewer but larger follicular structures when GnRH is administered concurrently.

Twenty-four male domestic shorthair cats were used to evaluate the acute and chronic effects of a single, toxic but sublethal, orally administered dose of chlorpyrifos. A dose of 10 mg/kg of body weight did not induce clinical signs of toxicosis, but a dosage of 40 mg/kg induced clinical signs of toxicosis, and 1 of 12 cats died. Chlorpyrifos given at a dosage of 0.1 mg/kg to 2 cats reduced whole blood and plasma cholinesterase (Che) activities to values obtained after cats were given doses that induced clinical signs of toxicosis. Regeneration time for whole blood and plasma Che activities ranged from 7 to 28 days. Brain Che activity was considerably decreased in 1 cat that died 4.5 hours after dosing, but was normal in all others at 28 days after dosing. Other than decreased Che activity, significant changes were not seen in hematologic or serum biochemical values. Toxin-related lesions were not seen during macroscopic or microscopic examination.

Concentrations of serum IgA, IgG, and IgM were determined for 829 adult Beagles from a commercial kennel in which several IgA-deficient dogs had been identified previously (index kennel). These values were compared with measurements of 100 adult dogs from another Beagle kennel (control kennel). After adjustment for differences in the ages and gender of the dogs, dogs from the index kennel had significantly (P less than 0.0001) lower IgA concentrations (mean, 46 mg/dl). Regardless of kennel, males had significantly (P less than 0.01) higher IgA concentrations than did females. Dogs in the control kennel had significantly (P less than 0.04) higher IgG concentrations (mean, 2,649 mg/dl) than did dogs in the index kennel (mean 2,478 mg/dl), and female dogs in the control kennel had significantly (P less than 0.05) higher IgM concentrations (mean, 189 mg/dl) than dogs of either sex in the index kennel (mean, 162 mg/dl) or male dogs in the control kennel (mean, 163 mg/dl). For both sexes, concentrations of IgA, IgG, and IgM increased with age.

Effects of age and season on type and occurrence of sperm abnormalities were examined in semen samples collected from 3 groups of Nubian bucks at ages of 4 to 9 months, 10 to 21 months, and 39 to 50 months. The average total percentage of sperm abnormalities at the onset of puberty (141 +/- 4 days) was 64.6 +/- 14.8% (head, 19.5 +/- 13.6%; middle piece, 17.2 +/- 9.3%; and proximal protoplasmic droplets, 14.6 +/- 10.5%), but this improved rapidly and was reduced to 12.5 +/- 7.5% by 8 months of age (head, 1.9 +/- 4.5%; middle piece, 4.6 +/- 2.8%). Further increase in age, at least up to 4 years, did not reveal a significant effect (P less than 0.05) on the type of percentage of total abnormalities. Similar to age, a comparison of data among seasons did not reveal a significant effect on the type or occurrence of sperm abnormalities in 10- to 21-month-old or 39- to 50-month-old bucks. Seemingly, Nubian bucks started producing good quality semen at 8 months of age, and season did not influence sperm abnormalities.
