Use of indirect enzyme-linked immunosorbent assay with hot saline solution extracts of a variant (M-) strain of Brucella canis for diagnosis of brucellosis in dogs

An ELISA, using hot saline solution extracts (HSS) of a less-mucoid variant (M -) strain of Brucella canis as antigen, was developed for detection of antibodies against B canis in dogs. The test was applied to 177 field serum samples previously tested by use of the 2-mercaptoethanol rapid slide agglutination test, 2- mercaptoethanol-tube agglutination test, and agar gel -immunodiffusion containing HSS and cytoplasmic antigens of B canis. Results indicated that this ELISA seems to be highly specific (95.6%) and slightly less sensitive (93.8%). The HSS obtained from B canis wild-type RM 6/66 also have been used, but in our study, it seemed to be unsuitable for use in ELISA because of the high background values observed for sera with negative test results.

Akabane virus (AKV) strain OBE-1 was inoculated IV into 17 pregnant sheep. Ten fetuses infected at 29 to 45 days of gestation and examined 29 to 30 days later had AKV antigen in the following groups of cells: neuroglial cells in the brain and spinal cord, ganglion cells in the cranial and abdominal ganglia, layer of ganglion cells in the retina, ganglion cells (Auerbach's plexus) in small intestine, hepatocytes, cells in the arterial wall of mesenteric membrane, and trophoblast cells in the placenta. Prior to detection of circulating virus-neutralizing antibody, immunoglobulin-containing cells were found initially at 59 days of gestation in the peripheral portion of white pulp tissue in the spleen. After that, numbers of immunoglobulin-containing cells gradually increased. These results indicated that AKV may have strong affinity for neuronal and ganglional cells in infected fetuses and immunoglobulin-containing cells might be considered the earliest immunologic response to AKV replication in the fetus.

The ionophore A23187 was used to facilitate release and continued development of Anaplasma marginale in short-term erythrocyte cultures. Addition of 10 micromolar A23187 to the cultures resulted in significant decrease in percentage of parasitized erythrocytes (PPE) by 24 hours after treatment; further development and increase in PPE was not observed. In contrast, the PPE of untreated cultures, those treated with dimethyl sulfoxide (DMSO) only and with 1 micromolar A23187 increased slightly during that time. Total erythrocyte count decreased in treated cultures in excess of that expected after samples of the medium were taken for analysis. The greatest cell loss and increased hemoglobin concentration in culture medium was observed in cultures treated with 10 micromolar A23187 and with an equivalent volume of DMSO. The DMSO appeared to cause hemolysis of some erythrocytes, but not of infected cells selectively. Release of A. marginale inclusion bodies was seen by electron microscopy in samples from the 10 micromolar A23187-exposed cultures. At 30 minutes after treatment, free initial bodies were frequently seen. Inclusion body membranes and individual A. marginale were associated with membranes of adjacent erythrocytes. Individual rickettsiae were seen in cell depressions and appeared to be entering erythrocytes. However, neither further invasion nor development of the parasite in erythrocytes was observed. Ionophore A23187 appeared to promote release of A. marginale from erythrocytes, but did not enhance infection of erythrocytes or development of organisms in vitro.

Using polyclonal rabbit and monoclonal mouse anti-dog IgE antibodies, we developed ELISA for measurement of ragweed-specific IgE in canine serum. In the ELISA, microtitration plates were coated with ragweed extract and sequentially incubated with canine serum, purified monoclonal or polyclonal anti-dog IgE, and conjugated goat antibody to mouse IgG or rabbit IgG. Serum ragweed-specific IgE values were measured by the 2 ELISA in serum samples from 60 ragweed-allergic dogs and in serum from 10 control dogs. Passive cutaneous anaphylaxis (PCA) tests were performed on these sera to compare results with those of the ELISA, Mean coefficient of variation between assays was 0.20 +/- 0.10 for the assay using the polyclonal antibody and was 0.17 +/- 0.10 for that using monoclonal antibody. Sensitivity was 0.6 U/ml for the ELISA, using polyclonal antibody, and 2.5 U/ml for the ELISA, using monoclonal antibody. Serum ragweed-specific IgE values measured by the 2 ELISA strongly correlated with PCA titers (P < 0.0000), but the ELISA using polyclonal antibody had higher correlation with PCA titer (r = 0.84) than the ELISA using monoclonal antibody (r = 0.59). The geometric mean ragweed-specific IgE value measured by the 2 ELISA and by PCA testing, was significantly higher (P < 0.0000) in allergic dogs than in control dogs. The 2 ELISA were specific, sensitive, and reproducible for measurement of ragweed-specific IgE in canine serum.

Bordetella avium is an important respiratory tract pathogen of turkeys. In common with other pathogenic bordetellae, B avium manifests a tissue tropism for cilia of the respiratory tract epithelium. To determine the molecular characteristics of the host cell receptors for B avium, we used hemagglutination and in vivo adherence assays. Carbohydrates, mucus, sialic acid-specific lectin, and other glycoconjugates were evaluated for their ability to competitively inhibit binding of B avium to host cells. The gangliosides, GD(1a) and GT(1b), completely inhibited hemagglutination, whereas N-acetylneuraminic acid (sialic acid) partially inhibited hemagglutination. Adherence to turkey tracheal mucosa in vivo was significantly (P < 0.01) inhibited by GD(1a) and GT(1b) gangliosides, N-acetylneuraminic acid, bovine sub-maxillary mucin, and horseshoe crab (Limulus polyphemus) lectin. Treatment of the tracheal mucosa with neuraminidase also inhibited adherence of B avium. We conclude that N-acetylneuraminic acid and the gangliosides, GD(1a) and GT(1b) may be important components of the tracheal mucosa receptor for B avium in turkeys.

Procedures were developed to count neutrophils in milk using a flow cytometer. Milk samples from 2 experiments were counted: 1 with 4 noninfected cows and a second with 5 noninfected cows that were injected with endotoxin in 2 mammary quarters. Thus, the procedures were evaluated on normal milk and on that with high somatic cell count. Flow cytometric procedures involved fluorescence detection (from the dye carboxydimethylfluorescein diacetate) to distinguish intact and viable from fragmented cells, forward light scatter to detect cell size differences, and right-angle side scatter to detect cellular granularity. High fluorescence, large size, and high degree of granularity identified viable neutrophils. For all samples, neutrophils were also counted manually, using the cytologic centrifugation approach to create the slides; manual counts were used as the standard for comparison. In experiment 1 (normal milk), mean values for percentage of viable neutrophils estimated by manual and flow cytometry procedures agreed closely (26% vs 25.8% for foremilk and 28.8% vs 26.6% for bucket milk). Sources of variation in manual and flow cytometric estimates of percentage of neutrophils were examined. Cow variation was significant (P < 0.01) for manual and flow cytometric counts, but was larger for flow cytometric counts. Day-to-day variation in counts on milk from the same cow was negligible for manual counts, but was significant (P < 0.01) for flow cytometric counts. Coefficients of variation were considerably Larger for manual counts than for flow cytometry. In experiment 2 (milk with high cell count, foremilk), agreement between mean values obtained by flow cytometry and by manual counting was somewhat less. However, predicting manual percentage of neutrophils, using the flow cytometric estimate, had R2 of 0.77. Regression of the manual percentage of neutrophils value on the flow cytometric percentage of neutrophil value was close to 1.0, with only a small negative intercept. Some additional refinement of flow cytometric procedures may be required before flow cytometric estimates of percentage of neutrophils can be accepted without simultaneous validation by use of manual counting. In particular, causes of day-to-day variation in flow cytometric results should be identified and reduced.

The IgG and IgM classes of antibodies to a water-soluble antigen preparation derived from microsporum canis were determined by ELISA in the sera of 79 cats with dermatophytosis confirmed by results of fungal culture, and of 46 specific-pathogen-free-derived, barrier-maintained cats with no previous exposure to dermatophytes. Of the 79 cats with dermatophytosis, the species isolated were: M canis from 72, M gypseum from 6, and Trichophyton mentagrophytes from 1. Concentrations of soluble M canis antigen-specific IgG and IgM were significantly (P < 0.05) higher in the cats with dermatophytosis than in the control cats. The IgG concentration was larger than or equal to 2.0 ELISA units/ ml in 71% of the cats with dermatophytosis and in 9% of the control cats, whereas IgM concentration was greater than or equal to 4.0 ELISA units/ml in 38% of the cats with dermatophytosis and in 11% of control cats. There was no significant difference in either IgG or IgM values between the cats with M canis infection and those with other non-M canis dermatophyte infections.

Chicken egg yolk IgG can be absorbed and transferred as efficiently as colostral antibodies in the blood of neonatal pigs. Egg yolk IgG has a half-life of 1.85 days in newborn pig serum. This is shorter than the reported half-life (12 to 14 days) of homologous IgG in serum of pigs. Similar to colostral antibodies, egg yolk IgG absorption from intestine ceased at about 34 hours of age, after a logarithmic decrease in absorption rate from birth. Egg yolk IgG absorption inhibition time in the gastrointestinal tract took 1.73 hours to decrease by half. Egg yolk IgG was protective against experimentally induced diarrhea in pigs when it was administered at high dose, and multiple dosing was instituted. Adverse effects were not observed when chicken egg yolk IgG was administered orally to pigs.

A serologic survey was conducted to determine the prevalence and seroconversion rates for ovine adenovirus (OAV) serotypes 1-4 and bovine adenovirus (BAV) serotypes 2, 3, and 7 in sheep in Iowa and in surrounding states. For 2 consecutive years, paired serum samples were obtained from 1- to 2-month-old lambs as they entered a ram test station and, again, 2 months later. Sera were tested for adenovirus antibodies by use of a microtitration serum virus-neutralization test. At the time of entry, high prevalence of antibody (titer greater than or equal to 2) was detected to all tested adenoviruses except BAV-3. All adenoviruses were active in the ram test station both years, as indicated by greater than or equal to fourfold increase in adenovirus antibody titer (seroconversion) in some of the lambs. The prevalence and seroconversion rate for OAV-1 was 94.0 and 7.2%, respectively; for OAV-2, 98.6 and 15.1%; for OAV-3, 86.5 and 11.0%; for OAV-4, 98.4 and 13.2%; for BAV-2, 97.6 and 22.4%; for BAV-3, 11.4 and 3.8%; and for BAV-7, 81.6 and 4.5%. The results indicate that adenovirus infections were widespread in the sheep population and that the prevalence of active infection based on seroconversion rates was approximately 45%.

Strength in bending was determined for inter. dental fixation apparatuses applied to hemimandibles obtained from 24 canine cadavers. Hemimandibles were osteotomized perpendicular to the long axis between the third and fourth premolars, and segments were stabilized with 1 of 5 interdental fixation apparatuses: Erich arch bar (EAB, n = 6); Stout loop (SL, n = 6); acrylic A, n = 6); Stout loop and acrylic (SLA, n = 24); and Erich arch bar and acrylic (EABA, n = 6). Ultimate strengths (mean +/- SEM) of EAB, SL, A, SLA, and EABA were 395 +/- 48; 523 t 57; 1,106 +/- 102; 1,306 +/- 156; and 2,707 +/- 504 N.m, respectively. Stiffness (mean +/- SEM) of EAB, SL, A, SLA, and EABA were 2,944 t 357; 6,322 +/- 2,201; 16,010 +/- 5,017; 15,777 +/- 1,026; and 27,079 +/- 5,576 N.m/ radian, respectively. Yield strengths (mean +/- SEM) Of EAB, SL, A, SIA, and EABA were 66 +/- 6; 264 +/- 19; 911 +/- 126; 1,114 +/- 159; and 1,855 +/- 401 N.m, respectively. There were no significant differences in acrylic weight, cross-sectional area of the acrylic, or area moment of inertia of acrylic at the osteotomy site among A, SIA, and EABA; and there were no significant differences in osteotomy surface area and area moment of inertia at the osteotomy site among all apparatuses (P > 0.05). The EABA apparatus had significantly higher mean ultimate strength, mean stiffness, and mean yield strength compared to other interdental fixation apparatuses. There were no significant differences in the mean ultimate strength, mean stiffness, or mean yield strength between EAB and SL (P > 0.05). Apparatuses that combine acrylic with metal reinforcement (SLA, EABA) were significantly stronger and stiffer than those that used metal alone (EAB, SL) or acrylic alone (A).
