Standardization of an enzyme-linked immunosorbent assay for detection of antibody to avian reticuloendotheliosis virus

Enzyme-linked immunosorbent assay (ELISA) for detection of antibodies to reticuloendotheliosis virus (REV) at single serum dilution was standardized. REV HI, one of the Korean field isolates, was inoculated into chicken embryo fibroblast (CEF) cells and was harvested from the culture fluids and cells after 10 to 12 days. Viruses were purified by centrifugation at the 107,000×g for 12 hours on 20, 30, 45% (W/V) sucrose gradient. Virus specific fraction was collected and used as ELISA antigen. To standardize ELISA, the optimal concentration of coating antigen (1 ㎍/well) and conjugate (1/1000) was determined by corrected OD (OD value of positive serum-OD value of negative serum) and P/N ratio (OD value of positive serum/OD value of negative serum).

A 6-month-old male Poodle was referred with chief complaint of dysponea and cough. Cyanosis was not detected. This dog was diagnosed as a case of canine patent ductus arteriosus (PDA) by X-ray and ultrasonography. Diuretics and bronchodialators were administered for 4 weeks. Clinical symptoms were not improved. Thoracotomy was done at right recumbent position. The length of ductus arteriosus was 8 mm and the diameter was 6 mm, respectively. Double ligation was performed in surgery. Continuous cardiac murmur, cough and strong femoral pulsation were disappeared after surgery. The diameters of the pulmonary artery and vein found to normal by X-ray on 10 days after operation.

The purpose of this study is to establish the analgesic effects of electroacupuncture for Korean native goat. Electroacupuncture was applied to the 6 Korean native goats. In 3 of them, rumenotomy was performed, and in the other 3, laparotomy was done. The analgesic induction time was 15 to 30 minutes. The acupoints used were Tian-ping (Celestial Peace, GV-5), Bai-hui (Hundred Meetings, GV-20), left 13th thoracic nerve and left 3rd lumbar nerve. Electroacupuncture was performed in lateral recumbency. Needles were inserted 1-2 cm deep, and connected to the electroacupuncture apparatus. The electrical stimulation condition was 30 Hz and 2-6 volts.

In this report, we described a case of acute colic with uterine rupture in a Thoroughbred broodmare during parturition. A 14-year-old broodmare showed acute colic characterized by rolling just after delivering of a healthy foal. Large intestine was prolapsed through vulva, contaminated with soil and its serosa was heavily damaged. We performed a euthanasia according to the owner's request. Post mortem examination revealed about 15 cm rupture on ventral side of the uterus. In addition, the orphan foal follow his owner after necropsy on his dam.

Three hundred and eighty-four samples of leaf litter, soil, faeces from domestic and game animals, compost and aqueous cultures of infective nematode larvae contaminated with unidentified fungi were plated out on water agar, baited with pure infective larvae of Haemonchus contortus, incubated and examined for the presence of nematophagous fungi. Duddingtonia flagrans was isolated from five samples, and 73 samples were positive for other nematophagous fungi.

Seasonal surveys were conducted at the Vaal Dam between April 2000 and January 2001. Twenty smallmouth yellowfish (Labeobarbus aeneus) and 20 largemouth yellowfish (Labeobarbus kimberleyensis) were collected with the aid of gill nets. Surface water quality variables were included. The cestodes were identified as either Bothriocephalus acheilognathi Yamaguti, 1934 or "other cestode spp.". The majority (99.8 %) of the cestodes found in both yellowfish species were identified as B. acheilognathi (Asian tapeworm). The prevalence, mean intensity and abundance of B. acheilognathi in both yellowfish species were calculated. Ecological parameters including species specificity, seasonality, gender specificity and relationships between fish size and the Asian tapeworm prevalence were also included. In this study, B. acheilognathi preferred L. kimberleyensis over L. aeneus although a low intensity was observed in smallmouth yellowfish. Furthermore, the infection (in terms of prevalence, abundance and mean intensity) in largemouth yellowfish was markedly higher. Seasonal patterns observed in the Asian tapeworm's infection of smallmouth yellowfish are attributed to breeding and subsequent feeding patterns of this fish species with relatively high infections recorded in winter and spring. For L. kimberleyensis no explanation can be given regarding the seasonal patterns observed for the mean intensity and abundance of B. acheilognathi. The maximum and minimum mean intensity and abundance values in largemouth yellowfish were recorded in autumn and spring, respectively. In addition, the prevalence of B. acheilognathi was consistently high in all four seasons.

Considerable difficulties are associated with the isolation of Yersinia enterocolitica from food particularly milk and milk products. Most methods are time consuming require enrichment steps and are unable to differentiate pathogenic isolates from non pathogenic ones. The purpose of this study was to evaluate the detection of Yersinia enterocolitica in milk by both polymerase chain reaction (PCR) and conventional culturing methods. Fifty milk samples were collected from Shami goats in North Sinai governorate. Two primers (DG26 and DG63) were used in PCR and the size of the PCR-product was 440bp. The results obtained by PCR technique were in good agreement with that obtained by conventional culturing method. Five samples (10%) were positive by PCR while 4 samples were positive by conventional culturing method. Interestingly, PCR results are obtained within few hours. Moreover, it solved the problem of interpretation of classical biochemical and serological typing in one step without necessity of using additional examinations. This makes diagnosis in food control laboratories much faster and more efficient.

The present study was designed in a trial to use serum replacement instead of the newborn calf serum in propagation of BHK-21 cell cultures with subsequent reducing the cost of foot and mouth disease (FMD) vaccine production. Two batches of BHK-21 cell culture were prepared where the medium of the first batch was supplemented with newborn calf serum while the medium of the second batch was supplemented with serum replacement. FMD virus was propagated 7 passages using BHK-21 cell culture. Both virus titration and complement fixation titer (CF) revealed that propagation of FMD virus in cell cultures supplemented with newborn calf serum yields a titre higher than that in case of cells supplemented with serum replacement. Also two batches of FMD inactivated vaccine were prepared from the virus propagated in the two-mentioned cell culture batches. Two groups of susceptible calves were vaccinated with these vaccines. Both of virus neutralization test (VNT) and enzyme linked immunosorbent assay (ELISA) revealed that higher antibody levels were induced in calves vaccinated with the vaccine prepared from cells supplemented with calf serum than those vaccinated with vaccine prepared from cells grown with serum replacement. BHK-21 cell culture supplement with newborn calf serum is most susceptible for FMD virus propagation yielding higher titer of the virus. Moreover, the growth pattern of the used cell culture was much better when the newborn calf serum was used.

The lectin-binding characteristics of the epithelial lining of the thoracic air sacs of the chicken were determined. Con A, LCA and PSA bound to the apical membrane as well as to the cytoplasm distal to the nucleus of the surface epithelium, indicated the presence of a-linked mannose as well as Nacetylchitobiose- linked a-fucose residues in the glycoproteins. GSL I bound to the apical membrane and cytoplasm distal to the nucleus, but not to the cilia of the epithelium, where-as MPL, DBA and RCA120 bound to the apical membrane, cilia and cytoplasm, indicated the presence of a-linked Nacetylgalactosamine residues. However, neither SJA or SBA showed any binding, indicating the absence of b anomers of galactosyl (b1.3)N-acetylgalactosamine and b-linked N-acetylgalactosamine residues. UEA I bound to the apical membrane and cilia, as well as to the cytoplasm of a few cells, indicated the presence of a-linked fucose residues. PNA bound to the apical membrane of some, but not all, surface epithelium cells, indicated the presence of galactosyl (b1.3)N-acetylgalactosamine residues. WGA bound to the apical membrane and cilia, as well as to the cytoplasm of a few cells, indicated the presence of neuraminic acid residues.

Poisoning with cardiac glycoside-containing plants is collectively the most important plant-associated poisoning of livestock in southern Africa. As a diagnosis of this significant poisoning is currently based on circumstantial evidence, a practical chemical procedure indicating the presence of cardiac glycosides in plants and animal specimens would be of considerable benefit. The fluorescence polarization immunoassay (FPIA) method, used to determine digoxin plasma levels in humans and dogs, was adapted to estimate cardiac glycoside levels in known cardiac-glycoside- containing plants as well as in the rumen and organs of dosed sheep. Positive FPIA values were obtained with bufadienolide-containing plants, while negative results were obtained with plants not known to contain cardiac glycosides. The FPIA has aided in the diagnosis of cardiac glycoside poisoning in livestock and game in 30 outbreaks examined at the Division of Toxicology, Onderstepoort Veterinary Institute. Each outbreak is briefly described. As a result of this assay, a better understanding of cardiac glycoside poisoning has been reached.
