Capsular serogroup of Pasteurella multocida isolated in VRI, Malaysia from year 2014 to 2016

A total of 65 Pasteurella multocida were isolated and identified from various animal’s samples received by Veterinary Research Institute (VRI) during the period of 2014 to 2016. These animals comprises of cattle, goat, pig, chicken, duck and rabbit. The serogroup of Pasteurella multocida were carried out using designation system of Carter’s capsular typing and molecular serogrouping method. Based on cases submitted to VRI, the prevalence of pasteurellosis in Malaysia ranging from 1.0% to 3.2% (2014 to 2016). It is low compared to previous reports and the pattern of predominant serogroups and animal hosts were found to be changing every year. In 2014, 80% (12/15) of the isolates were Pasteurella multocida Carter’s type D where all were isolated from goats. In 2015, the predominant serogroup changed to Pasteurella multocida Carter’s type A with a prevalence rate of 40.6% (13/32) which were mostly isolated from duck and cattle. While for Pasteurella multocida Carter’s type D, the prevalence in 2015 reduced to 21.9% (7/32) compared to the previous year and it was isolated from various animal species. Interestingly, in 2015 there was one isolate of Pasteurella multocida Carter’s type B isolated from goat with no reported history of outbreak. In 2016, the prevalence of Pasteurella multocida Carter’s type A increased to 72.2% (13/18), with a high percentage (92.3%) infection in young calves showing clinical signs with high mortality and morbidity in infected farms. Furthermore, during these 3 years of study, 3 isolates of Pasteurella multocida serogroup F were also identified each from pig, goat and chicken, respectively. In conclusion, this study revealed that pasteurellosis had become sporadic in Malaysia and the distribution of serogroups were diverse in all species of animal with no definitive host.

In Peninsular Malaysia, footand-mouth disease (FMD) has been reported since early 1860 which then became sporadic, causing outbreaks every year. Since then, Peninsular Malaysia has become endemic with FMD. The aim of this study is to provide findings of the current FMD occurrence and its serotyping in Peninsular Malaysia. An identification of Foot and Mouth Disease serotype was carried out in Peninsular Malaysia by the Regional Veterinary Laboratory Kota Bharu (RVLKB) only. Epithelial tissue samples were received from 10 states throughout Peninsular Malaysia from 2012 until 2016. Indirect sandwich ELISA was performed using ELISA kit for FMDV antigen detection supplied from the Institute for Animal Health, Pirbright Laboratory. All findings and results in this paper were based on samples received by RVLKB and does not reflect overall cases reported to State DVS or to DVS Malaysia. From the results, 2013 had the highest samples positive for FMDV (35% from 43 samples), followed by 2014 (31% from 80 samples), 2012 (24% from 122 samples), 2015 (21% from 39 samples) and the lowest is 2016 (17% from 194 samples). The FMDV serotypes detected throughout 2012 to 2016 from 110 positive samples were Serotype O (80%), followed by Serotype A (20%) and none from Serotype Asia 1. Strict regulation, FMD vaccine evaluation by LPB ELISA and strict animal movement shall be considered to achieve FMD free for upcoming Year 2020.

Monitoring of veterinary drug residues in chicken, swine and cattle was conducted in Peninsular Malaysia from 2010 to 2016. Tissue samples were collected from slaughterhouses and processing plants. A total of six groups of veterinary drugs were analysed in 8,708 samples using bioassay or immunoassay and LCMS method. The average violation rate was 3.4%, 2.5%, 1.9%, 0.8%, 1.6% and 2.7% for year 2010, 2011 and 2012, 2013, 2014, 2015 and 2016, respectively

Most studies on Simulum damnosum in Nigeria has focused mainly on the problem of onchocerciasis from the ecological standpoint. However, this study is aimed at viewing it from the human angle, with focus placed on the ignorance of the rural dwellers about the influences of their environment on the increasing incidences of onchocerciasis in their population. The premise is that, as the government and the scientists are working towards finding a lasting solution to the epidemic, the rural dwellers need to wake up to the realities of their own destiny. They need to come out and collaborate with other good efforts to form a strong united force that will rid their society of this menace. A study on the level of ignorance was carried out. It is assumed that their ignorance has kept the villagers relatively inactive and passive about freeing themselves. A descriptive method was used for this research and about 450 villagers were randomly selected and verbally interviewed. A questionnaire was designed and used as the instrument of the research. The research was carried out in two different villages of Asejire and Asepari. Findings were encouraging. Some of the villagers did not know about the existence of the disease even when some of their close relatives showed the symptoms. This high level of ignorance shows that it will be difficult to attain any cooperation from them in any attempt to make their environment unfavourable for the vector.

Spotted fever group rickettsioses are a group of tick-borne zoonotic diseases caused by intracellular bacteria of the genus Rickettsia. The diseases are widely reported amongst international travellers returning from most sub-Saharan Africa with fever, yet their importance in local populations largely remains unknown. Although this has started to change and recently there have been increasing reports of the diseases in livestock, ticks and humans in Kenya, they have not been investigated in wildlife. We examined the presence, prevalence and species of Rickettsia present in wildlife in two regions of Kenya with a unique human-wildlife-livestock interface. For this purpose, 79 wild animals in Laikipia County and 73 in Maasai Mara National Reserve were sampled. DNA extracted from blood was tested using the polymerase chain reaction (PCR) to amplify the intergenic spacer rpmE-tRNAfMet and the citrate synthase-encoding gene gltA. Rickettsial DNA was detected in 2 of the 79 (2.5%) animals in Laikipia and 4 of the 73 (5.5%) in Maasai Mara. The PCR-positive amplicons of the gltA gene were sequenced to determine the detected Rickettsia species. This revealed Rickettsia sibirica in a Topi (Damaliscus lunatus ssp. jimela). This is the first report of spotted fever group rickettsioses in wildlife and the first to report R. sibirica in Kenya. The finding demonstrates the potential role of wild animals in the circulation of the diseases.

The transmission of diseases between livestock and wildlife can be a hindrance to effective disease control. Maintenance hosts and contact rates should be explored to further understand the transmission dynamics at the wildlife-livestock interface. Bovine tuberculosis (BTB) has been shown to have wildlife maintenance hosts and has been confirmed as present in the African buffalo (Syncerus caffer) in the Queen Elizabeth National Park (QENP) in Uganda since the 1960s. The first aim of this study was to explore the spatio-temporal spread of cattle illegally grazing within the QENP recorded by the Uganda Wildlife Authority (UWA) rangers in a wildlife crime database. Secondly, we aimed to quantify wildlife-livestock interactions and cattle movements, on the border of QENP, using a longitudinal questionnaire completed by 30 livestock owners. From this database, 426 cattle sightings were recorded within QENP in 8 years. Thirteen (3.1%) of these came within a 300 m-4 week space-time window of a buffalo herd, using the recorded GPS data. Livestock owners reported an average of 1.04 (95% CI 0.97-1.11) sightings of Uganda kob, waterbuck, buffalo or warthog per day over a 3-month period, with a rate of 0.22 (95% CI 0.20-0.25) sightings of buffalo per farmer per day. Reports placed 85.3% of the ungulate sightings and 88.0% of the buffalo sightings as further than 50 m away. Ungulate sightings were more likely to be closer to cattle at the homestead (OR 2.0, 95% CI 1.1-3.6) compared with the grazing area. Each cattle herd mixed with an average of five other cattle herds at both the communal grazing and watering points on a daily basis. Although wildlife and cattle regularly shared grazing and watering areas, they seldom came into contact close enough for aerosol transmission. Between species infection transmission is therefore likely to be by indirect or non-respiratory routes, which is suspected to be an infrequent mechanism of transmission of BTB. Occasional cross-species spillover of infection is possible, and the interaction of multiple wildlife species needs further investigation. Controlling the interface between wildlife and cattle in a situation where eradication is not being considered may have little impact on BTB disease control in cattle.

The aim of the study was to determine the species spectrum of ixodid ticks that infest horses and donkeys in South Africa and to identify those species that act as vectors of disease to domestic livestock. Ticks were collected opportunistically from 391 horses countrywide by their owners or grooms, or by veterinary students and staff at the Faculty of Veterinary Science, University of Pretoria. Ticks were also collected from 76 donkeys in Limpopo Province, 2 in Gauteng Province and 1 in North West province. All the ticks were identified by means of a stereoscopic microscope. Horses were infested with 17 tick species, 72.1% with Rhipicephalus evertsi evertsi, 19.4% with Amblyomma hebraeum and 15.6% with Rhipicephalus decoloratus. Rhipicephalus evertsi evertsi was recovered from horses in all nine provinces of South Africa and R. decoloratus in eight provinces. Donkeys were infested with eight tick species, and 81.6% were infested with R. evertsi evertsi, 23.7% with A. hebraeum and 10.5% with R. decoloratus. Several tick species collected from the horses and donkeys are the vectors of economically important diseases of livestock. Rhipicephalus evertsi evertsi is the vector of Theileria equi, the causative organism of equine piroplasmosis. It also transmits Anaplasma marginale, the causative organism of anaplasmosis in cattle. Amblyomma hebraeum is the vector of Ehrlichia ruminantium, the causative organism of heartwater in cattle, sheep and goats, whereas R. decoloratus transmits Babesia bigemina, the causative organism of babesiosis in cattle.

The objective of the study was to record the tick species collected from three species of tortoise, each in a different province of South Africa. Ticks were collected from leopard tortoises, Stigmochyles pardalis, in the southern region of the Kruger National Park, Mpumalanga province; from hingeback tortoises, Kinixys zombensis, in the Enseleni Nature Reserve, KwaZulu-Natal province and from angulate tortoises, Chersina angulata, in the West Coast National Park, Western Cape province. Of the 63 leopard tortoises examined, 58 were infested with Amblyomma marmoreum and 49 with Amblyomma hebraeum, and all stages of development of both species were recovered. Amblyomma nuttalli was collected from 25 hingeback tortoises, and all stages of development were present. All 24 angulate tortoises examined were infested with Amblyomma sylvaticum, and large numbers of larvae, nymphs and adults were collected. Three snake species and a sand lizard were also infested with A. sylvaticum. The adults of A. marmoreum, A. nuttalli and A. sylvaticum were identified as specific parasites of the family Testudinidae, whereas all stages of development of A. hebraeum were classified as generalists.

Campylobacter spp. are common pathogenic bacteria in both veterinary and human medicine. Infections caused by Campylobacter spp. are usually treated using antibiotics. However, the injudicious use of antibiotics has been proven to spearhead the emergence of antibiotic resistance. The purpose of this study was to detect the prevalence of antibiotic resistance genes in Campylobacter spp. isolated from chickens and human clinical cases in South Africa. One hundred and sixty one isolates of Campylobacter jejuni and Campylobacter coli were collected from chickens and human clinical cases and then screened for the presence of antimicrobial resistance genes. We observed a wide distribution of the tetO gene, which confers resistance to tetracycline. The gyrA genes that are responsible quinolone resistance were also detected. Finally, our study also detected the presence of the blaOXA-61, which is associated with ampicillin resistance. There was a higher (p < 0.05) prevalence of the studied antimicrobial resistance genes in chicken faeces compared with human clinical isolates. The tetO gene was the most prevalent gene detected, which was isolated at 64% and 68% from human and chicken isolates, respectively. The presence of gyrA genes was significantly (p < 0.05) associated with quinolone resistance. In conclusion, this study demonstrated the presence of gyrA (235 bp), gyrA (270 bp), blaOXA-61 and tetO antimicrobial resistance genes in C. jejuni and C. coli isolated from chickens and human clinical cases. This indicates that Campylobacter spp. have the potential of resistance to a number of antibiotic classes.

A retrospective study of clinical bovine dermatophilosis outbreaks and cases for the period 1995-2014 was conducted, using data obtained from the Division of Veterinary Services (DVS). A total of 3856 outbreaks and 26 659 cases of dermatophilosis were reported countrywide during this period. The post rainy season accounted for 37.9% of the outbreaks followed by the rainy season (26.7%), cold dry season (22.1%) and the hot dry season (13.2%). A retrospective space-time scan statistic in SaTScan™ was used to detect clusters. From this study, it was evident that dermatophilosis was spreading from the north-west of Zimbabwe through the central to the north-east during the period 2010-2014. Five clusters were identified mainly in the central and north-western regions of Zimbabwe. The primary cluster was centred at Ungwe, Gokwe district in Midlands; the second, third, fourth and fifth likely clusters were centred at Bonga (Mashonaland Central), ARDA (Mashonaland West), Nsenga (Matabeleland North) and Zanda in Gokwe, respectively. The findings of this study suggest the continued spread of dermatophilosis across the country; as such the Department of Livestock and Veterinary Services are advised to develop measures aimed at managing this spread such as dipping, quarantine, movement control and raising farmer awareness.
