Early metabolic profiling in the periparturient period reduces the occurrence of postparturient metabolic diseases in cows

Objective: To study the impact of early metabolic profiling and intervention measures in the peri¬parturient period on the occurrence of postparturient metabolic diseases in cows. Materials and Methods: Using a cohort of dairy cows from two selected areas of Bangladesh, we routinely tested the serum calcium level and ketone bodies in the urine at periparturient periods. In addition, milk samples were tested for the presence of mastitis at different stages of lactation. Animals showing reduced serum calcium levels, high ketone bodies in the urine or the presence of clinical and subclinical mastitis received appropriate therapeutic intervention immediately after detection. After the intervention, the number of animals that got sick with diseases or conditions like milk fever, ketosis, mastitis, and dystocia, which are caused by metabolic problems, was recorded. Results: In the periparturient period, most of the animals had lower serum calcium levels (8.13 ± 1.2 mg/dl), which were significantly increased by the following intervention (10.05 ± 1.4 mg/dl). On the other hand, there was a higher number of ketosis-affected animals (33.9%, 20/59) during the periparturient period, which decreased (18.6%, 11/59) during the postparturient period. Similarly, the number of mastitis-affected cows was also decreased in postparturient cows (30.9%, 13/42) than in periparturient cows (59.5%, 25/42), following improved hygienic measures. After early intervention, the number of study animals with metabolic diseases decreased from 51.7% before intervention to 15.3% after intervention. Conclusions: Early metabolic profiling significantly reduced the occurrence of metabolic diseases in cows. Therefore, we recommend regular metabolic profiling of dairy cows and receiving early intervention measures to reduce the occurrence of metabolic diseases on the farm. Objective: Porcine circovirus type 2 (PCV2) is one of the most important causative agents of swine diseases that pose a global economic threat. Presently, there is little or no information on the perception and awareness of PCV2 and its associated effects among pig farmers in Nigeria. Therefore, this research was carried out to describe pig farmers' views, awareness, and likely impact of PCV2 and its associated postweaning multisystemic wasting syndrome (PMWS) on pig production in the southwestern region of Nigeria. Materials and Methods: A cross-sectional survey of pig farmers in Oyo and Ogun states, Southwest Nigeria, was carried out with the help of a self-administered questionnaire. Results: A total of 111 farms out of the 385 required took part in the study, resulting in a total response rate of 28.8%. 89 (79.2%, 95% CI = 70.8–85.8) pig farmers who participated were unaware of PCV2, while 46 (41.4%, 95% CI = 32.7–50.7) had heard about PMWS. The level of awareness was generally poor, with an average score of 1.43 (SD ± 1.25; 23.9%). Only 23% (25/111) of the participants had a high level of awareness. To promote awareness about PCV2/PMWS, participants' most preferred sources of information were seminars, extension services (especially by veterinary and agricultural extension officers), social media (WhatsApp and YouTube), and mobile telephone (through calls or text messages). Conclusions: The present study showed a gap in the level of farmers' awareness about PCV2/ PMWS, and to bridge the gap, more scientific-based evidence is needed to promote targeted educational programs and policy formulations. Also, with the dearth of information about PCV2, it is necessary to determine its prevalence and the characteristics of the virus possibly circulating within the swine herds in Nigeria. Objective: Rodlet cells produce secretions of glycoproteins in nature. This study investigated the microscopic morphology, histochemical and immunohistochemical reactions, and distribution of the rodlet cells in the gut of Binni fish (Mesopotamichthys sharpeyi). Materials and Methods: Thirty samples were obtained from the cranial, middle, and caudal por¬tions of Binni intestine immediately after being euthanized, fixed in Bouin's solution for 18 h at 24°C, and had undergone routine histological processing, different conventional histochemical stains, and immunostaining with TNF-α and S100 protein antibody. Results: The intestine of Binni fish showed different stages of rodlet cells classified into three distinctive forms: vesicular, granular, and mature cells. Rodlet cells are poorly stained with hema¬toxylin and eosin. Their secretory granules have a weak positive reaction with periodic acid-Schiff (PAS) and Alcian blue (AB), and react positively to combined AB and PAS. Rodlet cells were stained lightly with Safranin O, observed pink in color by Giemsa stain, and showed reactivity to Masson's and Mallory trichrome stains. Rodlet cells were immunostained positively against TNF-α and S100 antibodies, indicating that they have an immune function. Conclusions: Rodlet cells, with their neutral glycoprotein secretions, play a crucial role in the immunity of Binni fish intestine. [J Adv Vet Anim Res 2022; 9(2.000): 282-289]

Objective: The purpose of this research is to detect the antibacterial properties of lactic acid bacteria (LAB) against pathogenic bacteria. Materials and Methods: Isolation and determination of Lactobacillus spp. Testing of the antibacterial activity of LAB was conducted using filtrate and nonfiltrate forms. The lactic acid bacterial isolates were confirmed to be identified through Gram staining, cell shape, catalase testing, and motility testing. Results: The results of the analysis of the LAB inhibition zone using filtrate and nonfiltrate forms against the bacteria Bacillus cereus were included in the very strong category. The results of the analysis of the LAB inhibitory zone using filtrate and nonfiltrate forms and the agar well method against Staphylococcus aureus bacteria were classified into the very strong category. The results of the LAB inhibitory zone analysis using filtrate and nonfiltrate forms with the well method against Escherichia coli bacteria are included in the very strong category, whereas the results from the LAB inhibitory zone analysis using the filtrate and nonfiltrate forms with the agar diffusion method (disks) are included in the strong category. Conclusion: Based on the results, LAB isolated from Sumba mare's milk displayed antibacterial activity in the strong and very strong categories against pathogenic bacteria such as B. cereus, S. aureus, and E. coli. Objective: This study aimed to determine the anticoccidial potential of Tinospora rumphii ethan¬olic stem extract, resulting in an effective reduction in oocysts per gram counts of Eimeria species in goats in comparison to the commercial coccidiostat toltrazuril. Materials and Methods: Eighteen goats that were naturally infected with coccidia were identified as experimental animals. The experimental animals were grouped and were administered a single dose of T. rumphii extract at concentrations of 300, 600, 1,000, and 2,000 mg extract/kg body weight. The fecal samples were collected before treatment, during the first treatment, and every week thereafter for 28 days posttreatment. Fecal examination was carried out using a modified McMaster technique to determine the oocysts per gram of feces, and the mean efficacies of the extracts were calculated. Results: Stem extracts of T. rumphii were able to lower the oocysts per gram count than pre¬treatment for all concentrations, but the efficacy, in general, was still lower than toltrazuril. A significant difference existed in the efficacy of the extracts among the treatments on day 14. Administering 1,000 mg/kg extract resulted in the highest efficacy rate (95.25%) at 14 days post¬treatment and was comparable to that of toltrazuril (89.89%). Conclusion: The results show that the T. rumphii stem extract has the potential to reduce the Eimeria species oocysts per gram counts of naturally infected goats. Objectives: The research aims to analyze the catabolic strength of different hydrolytic enzymes in assessing the biological conversion potential of lignocellulose parts of agricultural biomass wastes into functional edible sugars and biofuels. Materials and Methods: The enzymes' hydrolytic properties—versatile peroxidase, manganese peroxidase, and lignin peroxidase were used to identify their complexing strength with the lignin substrate, whereas endoglucanase cel12A, acidocaldarius cellulase, and Melanocarpus albomyces endoglucanase were tested on the cellulose gel substrate. Because the biodegradation properties are heavily influenced by the "enzyme-substrate complexing energy level," proper molecular optimization and energy minimization of the enzymes and substrates were carried out, as well as the identification of the enzyme's active sites prior to complexing. comprehensive molecular dynamic simulation was run to study their—alpha carbon, root-mean-square deviation (Å), molecular surface area (Å2), root-mean-square fluctuation (Å), radius of gyration (nm), hydrogen bonds with hydrophobic interactions, and solvent accessible surface area (Å2) values for 50 ns. The simulated data mining was conducted using advanced programming algorithms to establish the final enzyme-substrate complexing strength in binding and catalysis. Results: Among the lignin-degrading enzymes, versatile peroxidase shows promising catalytic activity with the best docking pose and significant values in all the dynamic simulation parameters. Similarly, Melanocarpus albomyces endoglucanase shows the best activity in all aspects of molecular docking and dynamics among the cellulose-degrading enzymes. Conclusion: The lignin content of biomass wastes can be degraded into cellulose and hemicellulose using lignin-degrading enzymes. The cellulose can be further degraded into glucose and xylose sugars following the cellulose-degrading enzyme activity. These sugars can be further degraded into biofuel through anaerobic fermentation. Systematic bioconversion of the lignocellulosic components can ensure sustainable biomass management, creating an alternative food and energy source for human beings to face the challenges of global hunger where the enzymes can pave the way. Objective: The study aimed to develop and assess an Android app designed for farmers with a low educational status that can formulate a least-cost ration. Materials and Methods: First, a computer-android-based app named BLRI FeedMaster was developed to guide users in formulating a balanced ration at the least cost. A survey was conducted on 30 livestock officers and 18 farmers with 50 cattle to evaluate its efficacy at the field level. The study outcomes were milk yield, feeding cost, milk composition, time, and cost for management before and after using the BLRI FeedMaster app. Descriptive statistics and paired sample t-tests were used to analyze the data. Results: After adopting the BLRI FeedMaster app, a significant increase was observed in daily average milk yield (9.39 ± 0.32 l from 8.37 ± 0.36 l), while a considerable decrease was observed in daily average feed quantity (4.88 ± 0.15 kg from 5.60 ± 0.17 kg) and feed cost (BDT 28.00 ± 0.50 from BDT 29.75 ± 0.49). Besides, the number of visits, time, and cost for seeking professional services regarding feed, health care, and other information was significantly minimized. The number of visits decreased to 0.36 ± 013 from 3.07 ± 0.38, and the consumed time was reduced from 270 ± 34.30 to 235.71 ± 59.42 min (p < 0.05) after adopting the app. Conclusion: Hence, this app was very beneficial for farmers with a low economic and educational background and may ultimately help farmers with profitable animal farming and sustainable pro¬duction in the least developed countries like Bangladesh. Objective: This study focuses on the orocutaneous fistula (OCF), a pathological channel between the buccal cavity and the outer surface (skin) of the face, causing leakage of saliva and fluid from the oral cavity to the face externally and also the ectopic incisors (EIs) erupted in a rare position in a day-old calf. The surgical interventions for these congenital defects were further described in this study. Materials and Methods: The calf was presented with an abnormal and external opening in the left submandibular region, having congenitally exposed jawbone and muscles with the resemblance of a linear groove, and was clinically examined based on the problem of swallowing milk due to lateral passing out of liquid through the unnatural tract before entering into the digestive tract. Clinical observation revealed an OCF with four EIs that abnormally erupted in the externally exposed groove of the fistula. Reconstructive surgery (RS) was performed along with a thorough guided approach to repair the defect, emphasizing normal functionality of the buccal cavity connecting to the pharynx and cranial esophagus, and smooth extraction of the EIs was ensured without making any deep wound. Results: After 2 weeks of postoperative care with supportive medications, the calf was quite recovered, having no further complications in the submandibular region along with no visible defect in swallowing. Conclusion: OCF in calves can be fruitfully treated by RS before getting severely infected and complicated. Outside the oral cavity, submandibular EIs can be easily removed, ensuring no further bleeding and defect. [J Adv Vet Anim Res 2022; 9(3.000): 509-515]

Objective: Gastrointestinal parasites (GIPs) negatively impact small ruminant production and pro¬ductivity nationwide, particularly in tropical and sub-tropic regions. Amongst other nematodes, Haemonchus contortus, Trichostrongylus colubriformis, and Teladorsagia circumcincta are the most common species in small ruminants animals. Thus, this study aimed to investigate communal sheep farmer's knowledge and attitudes toward GIPs in the Eastern Cape Province, South Africa. Materials and Methods: A cross-sectional survey was conducted between September and November 2018 from three agro-ecological zones, namely, arid region, semi-humid, and humid. All data from this study were analysed with the Statistical Analysis System. Results: Of the total of 107 farmers who participated in the study, 69% were males, and 38% were females. Most livestock owners (85%) were aged >46 years old across all the study areas. The majority of farmers (83%) perceived that their animals are susceptible to wireworm (H. contortus) during the hot-wet season, followed by the hot-dry season (14%), with relatively low during the cold season (2%). Most farmers (85%) interviewed revealed that lambs are more exposed to para¬sitic infection, than mature sheep (15%) across all agro-ecological zones. An insignificant number of farmers (8%) with knowledge about GIPs life cycle and its biology (92%) across all agro-eco¬logical zones. This study reveals a significant increase in the occurrence of GIPs over the past few years across all agro-ecological, with largely (67%) attributed to the resistance of the strain to deworming remedies and changes in climatic weather patterns (33%). The farming experience was strongly (p < 0.05) associated with the farmer's gender and age. Helminths were reported significantly higher (p < 0.05) in humid zones than in other agro-ecological zones. Conclusions: This study concludes that most farmers perceived lambs as more susceptible to GIPs than old sheep. Therefore, farmers should be enlightened about the infection and transmission dynamics of the GIPs to develop appropriate control measures against worm infection to boost sheep production in the study area. It was also suggested that farmers should adhere to remedial instruction and adopt rotational deworming programs to avoid anthelmintic resistance. [J Adv Vet Anim Res 2022; 9(3.000): 351-358]

Objective: This study aims to develop and validate an Attenuated Total Reflectance–Fourier Transform Infrared Spectroscopy (ATR-FTIR) spectroscopic technique combined with a partial least squares regression (PLSR) model for rapid quantification and monitoring of aflatoxins in chicken feeds and food grains. Materials and Methods: A model of ATR-FTIR-PLSR was developed using ATR-FTIR spectra of mixed aflatoxin standards in 100% acetonitrile (112 samples) and 75% methanol (112 samples), validated by testing its prediction on 125 feed/food samples spiked with variable concentrations of aflatoxins, and applied to screen 660 samples of commercial chicken feeds and food grains from Nigerian and Malaysian markets for total aflatoxins, for which the dietary exposure risks to aflatoxins (DERA) and associated hepatocellular carcinoma (HCC) risks were evaluated for both countries. Results: The ATR-FTIR-PLSR model demonstrated excellent prediction power [R2 = 99.59%, p = 0.001, root mean square error of calibration (RMSEC) = 1.69, RMSE p = 1.98, bias = −0.26], sensi¬tivity (limit of quantitation and limit of the method < 5.0 ng/gm), precision (coefficient of variation = 0.97–1.72), and accuracy (% recovery of 88%–106%) in all the spiked samples. The model's pre¬diction was statistically reliable (R2 = 99.8%, p < 0.05) when compared with a high-performance liquid chromatography method. Levels of aflatoxins in the commercial samples signify high DERA (0.92–138.2 ng of aflatoxins/kg BW/day) and HCC risk (1.07%–159.91% of HCC/100,000 people/ year) in the exposed populations. Conclusions: Results feature the conceivable implementation of the proposed ATR-FTIR-PLSR model for rapid, accurate determination and monitoring of aflatoxins in commercial chicken feeds and food grains; and the need to strengthen aflatoxin control/prevention strategies in the study populations.
