Bem-Estar Animal consiste no estado do animal em sua tentativa de adaptação ao meio ambiente envolvente, podendo ser medido e melhorado com a ação do ser humano. Nos últimos anos o tema “Bem-Estar Animal” tem adquirido importância, resultante das grandes preocupações com o manejo dos animais em vida e constatação da influência que este tem na qualidade da carne post mortem. Este trabalho teve como objetivo determinar o efeito do lote de suínos transportados, respectivo tamanho, tipo de trajeto e do tempo decorrido entre a origem e o abate na ocorrência de lesões, consideradas indicadores mínimos de bem-estar animal em matadouro. Nos matadouros de suínos da região norte de Portugal, foram monitorados 834 animais com relação à ocorrência de lesões compatíveis com lacerações, eritemas cutâneos, hematomas e fraturas, em cinco diferentes lotes. Pela análise global dos dados, verificou-se que as lacerações estavam presentes em 72% dos animais e foi o tipo de lesão mais frequentemente observado. Seguiram-se os eritemas cutâneos em 20%, os hematomas em 11% e as fraturas em apenas 1% dos animais. O tamanho do lote de animais transportados, a duração da viagem e o tempo de permanência no matadouro influenciaram significativamente a ocorrência das lesões monitoradas, nomeadamente lacerações, eritemas e hematomas.

The aim of this work was to report the potential of MALDI-MS for the characterization of lipid species present in a single equine embryo, and to study some lipid structures detected by collision induced dissociation (CID) experiments. In the positive ion mode spectrum, we could observe mostly protonated and sodiated species of sphingomyelins (SM), phosphatidylcholines (PC) and triacylglycerols (TAG). In the negative ion mode, we observed phosphatidylethanolamines (PE) and phosphatidylinositols (PI). MS/MS spectrum of most intense lipid ions was performed to show MALDI-MS/MS structural information potential. MS/MS spectrum in the positive mode of m/z 760.6 (attributed as PC34:1) depicted characteristic PC fragments of m/z 184.1 (choline polar head), and the neutral loss (NL) of 183 (phosphorylcholine). For the ion of m/z 766.6 (attributed as PE 38:5), we observed the NL of 140, characteristic of PE. For the ion of m/z 808.7 (attributed as PC 38.5), besides the fragment at m/z 184.1 at the NL of 183, it was possible to observe the loss of trimethylamine (ion of m/z 749.6), and the cyclophosphane (ion of m/z 147.0). Finally, for the negative ion mode, we isolated and fragmented the ion at m/z 863.6, which was attributed as PI 36:1 due to the presence of m/z 153 (glycerol phosphate – H2O-H), 223 (phospho inositol – 2H2O-H), 241 (phospho inositol – H2O-H), 281 (oleic acid), and 581.3 (lysophosphoinositol – H2O-H). We conclude that MALDI-MS allowed the detection of a broad range of PC, SM, PE, PI and TAG lipid species, as well as a fast and confident characterization of lipid structures from a single equine embryo.

Potamotrygonidae is a family of freshwater stingray endemic in South America. We determined the hematological and serum biochemistry parameters of Potamotrygon motoro and Potamotrygon falkneri from Parana Basin and Potamotrygon orbignyi and Potamotrygon scobina from the Amazon Basin. Blood was collected from 55 specimens of Potamotrygon sp., and these parameters were evaluated: red blood cell count, hematocrit percent, hemoglobin concentration, leucocytes count, trombocytes count, total protein concentration, albumin, uric acid, urea, creatinine, triglycerides, cholesterol, high-density lipoprotein HDL, calcium, phosphorus, calcium/phosphorus relationship, creatine kinase CK, gamma-glutamyl transpeptidase GGT, Aspartate transaminase AST, alanine transaminase ALT, globulin, albumin/globulin relationship, alkaline phosphatase AP, sodium, chloride, potassium, triiodothyronine T3, and thyroxine T4 hormones. The hematological indices were similar for all species, except for levels of heterophils in P. orbignyi and P. scobina that were higher. Calcium, phosphorus, calcium/phosphorus relationship, sodium, potassium, chloride, AST, ALT, GGT, CK, albumin/globulin relationship, uric acid, creatinine, urea, triglycerides, HDL, T3, T4 levels had no significant difference between the species. These results suggest that there is low influence of habitat and feeding habits. AP, total protein, albumin, globulin, glucose, and total cholesterol had significant differences between the studied species. These results support the theory that stingrays migrated from the Atlantic Ocean, adapting to different conditions over time and placed themselves geographically distant from each other.

Immunohistochemical expression of proliferating cell nuclear antigen (PCNA) was evaluated in the endometrium of mares during estrus and at early diestrus. Three samples were collected by endometrial biopsy from 10 mares, on estrus/ second day, in the ovulation day and seven days after the ovulation day. PCNA expression was high in luminal epithelium and low in endometrial glands on samples taken on estrus/second day and on the ovulation day (p < 0.05). For samples collected on the seventh day following ovulation, the averaged PCNA immunostaining was higher in glandular epithelium (p < 0.05). The study revealed that luminal epithelial cells exhibit higher proliferation during estrus and glandular epithelial cells exhibited higher proliferation during diestrus.

As doenças renais em equinos, geralmente estão associadas a problemas respiratórios e digestórios, entretanto não são frequentes as intervenções cirúrgicas neste órgão para estes animais. O estudo dos vasos sanguíneos relativos aos rins nos animais domésticos e selvagens é de fundamental importância para o entendimento de diversos aspectos de sua fisiologia e nutrição. Neste trabalho, foram utilizados 30 pares de rins de equinos Puro Sangue Inglês, dos quais 15 machos e 15 fêmeas, de diferentes faixas etárias, provenientes de doação do Jockey Club de São Paulo. Durante a necropsia dos animais, os rins unidos por meio de suas artérias e veias renais foram retirados em bloco. Em seguida, foram fixados em solução de formol a 10%, por aproximadamente 48h, para a dissecação e individualização dos componentes dos pedículos renais. Para a análise dos resultados foram demarcados os quadrantes cranioventral, craniodorsal, caudoventral e caudodorsal, com duas linhas uma craniocaudal e a outra dorsoventral. Os resultados obtidos revelaram que: os ramos arteriais da região hilar são observados no rim direito em número de dois em 50%, no esquerdo em número de dois (33,3%), os ramos arteriais na região justahilar são computados no rim direito em número de quatro a cinco em 20% e no rim esquerdo em número de seis (16,7%), e nos ramos dispostos na região extrahilar são registrados no rim direito em número de três (26,7%) e no esquerdo em número de três (20%).

Environmental changes affect fish homeostasis, turning them more susceptible to diseases. In Brazil, outbreaks of Streptococcus agalactiae infection have been reported in Nile tilapia when they are outside of their thermal comfort zone. This investigation evaluated mortality rate and which were the most infected organs at temperatures that naturally occurred in southern of Brazil. Forty Nile tilapia (Oreochromis niloticus) were infected with S. agalactiae and distributed in four groups (n=10) and each group was exposed to a different temperature: G1: 24°C, G2: 26°C, G3: 28°C, and G4: 32°C. Fish were monitored for 10 days. In this period, fish that presented irreversible clinical signs were sacrificed and samples of brain, liver, and kidney were collected for bacteriological and molecular analysis. Signs compatible with a streptococcal infection were observed in all groups. Highest mortality rates occurred at 24°C and 32°C. The brain was the most affected organ with the highest percentage of isolation of S. agalactiae by both methods of diagnosis. The results suggest that, as in mammals, temperatures that are further away from the comfort zone influence fish homeostasis, increasing susceptibility to bacterial infections.

Tissue repair is a response reaction to lesions and aggressions that constitutes a dynamic process to maintain the integrity of the organism. Wound healing experiments have used several approaches in order to assess and compare treatment methods, and these discrepancies hamper comparisons among assays. This study assessed three different methods of wound measurement commonly used in healing assays: clock method, graph paper method, and computer-assisted image analysis. We used 30 Wistar rats, kept in appropriate conditions for animal well-being. After anesthesia, and using an eight-millimeter punch, two lesions were made in the back region of each rat. The wounds were assessed on days four, seven, and 14 after infliction. At four days, all methods generated similar results. By day seven, the clock method had lost precision, likely due to wound shrinkage, and yielded greater means compared to the other two methods. On the last assessment, the computer-assisted method appeared to have more precise results, with the other two generating statistically higher means. Computer-assisted image analysis seems to have maintained wound measuring precision throughout this experiment, even when faced with small lesions. Considering these results, the authors recommend the use of computer-assisted measurements in future experiments.

The increase in the population’s acquisition power in emerging countries like Brazil has resulted in increased consumption of meat, milk and their derivatives, and a consequent growing surveillance regarding the responsibility of maintaining the quality of these food products. The total or partial replacement by other than the species declared on the product label in meat, milk or derived products compromises the nature and quality of these products, hurting consumer choice rights, which may be based on medical and nutritional recommendations, the economic value of the product or habits and/or dietary restrictions of each specific culture. Species identification in dairy and meat products is important in food traceability. Although food matrices are complex and variable, biomolecular techniques are gradually being applied for species identification, having proven increasingly reliable, fast, specific and highly sensitive, even in mixed samples. For these reasons, this review intends to show the main molecular methods applied to adulteration detection in dairy and meat derivatives, including an already established method, such as the polymerase chain reaction (PCR), as well as more advanced technologies, such as real-time PCR, next-generation DNA sequencing methods and DNA biochip or DNA microarray, which have been gradually applied to the detection and quantification of exogenous DNA in food samples, even if present in small amounts.

Carnivore semen cryopreservation procedures started with semen washing and centrifuging in culture media for seminal plasma removal and microorganisms elimination. The objective of this study was to perform coatis semen cryopreservation comparing the effects between two extenders Ham’s F-10 and M199 for washing and centrifugation before cryopreservation using Dilutris medium. Semen samples (n = 36) were collected by electroejaculation from six adult male coatis (Nasua nasua) between May and October of 2008 at the Universidade Federal de Mato Grosso Zoo. Sperm total motility (%), progressive sperm motility (0-5), plasma membrane integrity spermatozoa rates (%), and acrosome integrity (%) were analyzed. These fresh semen samples were divided in two fractions, diluted in 1 ml of Ham’s F-10 (Ham’s F-10, Nutricel S.A., Brazil) or M199 (M199, Nutricel S.A., Brazil) and centrifuged at 300 g for 10 min. The supernatant was discarded and pellets resuspended in 1 ml of Dilutris (Dilutris, Minitube®, Brazil), stored at 5ºC for 3 hours, transferred to 0.25 ml straws, placed in liquid nitrogen vapor for 20 min, and immersed in liquid nitrogen. The means/SD for fresh semen and cryopreserved semen using Ham’s F-10/Dilutris and M199/Dilutris were, respectively: 84.28 ± 11.57, 45.38 ± 27.26, and 44.61 ± 25.03 for total motility; 3.64 ± 1.44, 2.15 ± 1.14, and 2.07 ± 1.03 for progressive sperm motility; 92.76 ± 3.46, 84.69 ± 15.77, and 89.76 ± 13.97 for live spermatozoa rate; and 94.76 ± 2.89, 92.35 ± 4.73, and 90.58 ± 7.17 for acrosome integrity. No significant difference (P < 0.05) were observed between the values obtained from the Ham’s F-10/Dilutris or M199/Dilutris treatments. Both treatments demonstrated to be suitable for freezing semen from this species.

Caprine arthritis encephalitis (CAE) is a chronic disease caused by a small ruminant lentivirus (SRLV), which causes significant losses in goat breeding. The actual state of animal infection with SRLV is difficult to determine due to a complex pathogenesis of the virus, including factors such as delayed or intermittent seroconversion in serological tests. Several serological techniques are available for disease diagnosis, such as screening or confirmation tests, which are different in sensitivity and specificity. Regarding the choice of the test to be applied, availability of commercial immunoreagents, team training, antigen used, and cost of techniques must be considered. This review presents the serological methods available for use in different stages of CAE control and eradication programs, and management measures to be adopted in conjunction with serological diagnosis of the disease.
