Objective: To study the impact of early metabolic profiling and intervention measures in the peri¬parturient period on the occurrence of postparturient metabolic diseases in cows. Materials and Methods: Using a cohort of dairy cows from two selected areas of Bangladesh, we routinely tested the serum calcium level and ketone bodies in the urine at periparturient periods. In addition, milk samples were tested for the presence of mastitis at different stages of lactation. Animals showing reduced serum calcium levels, high ketone bodies in the urine or the presence of clinical and subclinical mastitis received appropriate therapeutic intervention immediately after detection. After the intervention, the number of animals that got sick with diseases or conditions like milk fever, ketosis, mastitis, and dystocia, which are caused by metabolic problems, was recorded. Results: In the periparturient period, most of the animals had lower serum calcium levels (8.13 ± 1.2 mg/dl), which were significantly increased by the following intervention (10.05 ± 1.4 mg/dl). On the other hand, there was a higher number of ketosis-affected animals (33.9%, 20/59) during the periparturient period, which decreased (18.6%, 11/59) during the postparturient period. Similarly, the number of mastitis-affected cows was also decreased in postparturient cows (30.9%, 13/42) than in periparturient cows (59.5%, 25/42), following improved hygienic measures. After early intervention, the number of study animals with metabolic diseases decreased from 51.7% before intervention to 15.3% after intervention. Conclusions: Early metabolic profiling significantly reduced the occurrence of metabolic diseases in cows. Therefore, we recommend regular metabolic profiling of dairy cows and receiving early intervention measures to reduce the occurrence of metabolic diseases on the farm. Objective: Rodlet cells produce secretions of glycoproteins in nature. This study investigated the microscopic morphology, histochemical and immunohistochemical reactions, and distribution of the rodlet cells in the gut of Binni fish (Mesopotamichthys sharpeyi). Materials and Methods: Thirty samples were obtained from the cranial, middle, and caudal por¬tions of Binni intestine immediately after being euthanized, fixed in Bouin's solution for 18 h at 24°C, and had undergone routine histological processing, different conventional histochemical stains, and immunostaining with TNF-α and S100 protein antibody. Results: The intestine of Binni fish showed different stages of rodlet cells classified into three distinctive forms: vesicular, granular, and mature cells. Rodlet cells are poorly stained with hema¬toxylin and eosin. Their secretory granules have a weak positive reaction with periodic acid-Schiff (PAS) and Alcian blue (AB), and react positively to combined AB and PAS. Rodlet cells were stained lightly with Safranin O, observed pink in color by Giemsa stain, and showed reactivity to Masson's and Mallory trichrome stains. Rodlet cells were immunostained positively against TNF-α and S100 antibodies, indicating that they have an immune function. Conclusions: Rodlet cells, with their neutral glycoprotein secretions, play a crucial role in the immunity of Binni fish intestine. [J Adv Vet Anim Res 2022; 9(2.000): 282-289]

Objective: The purpose of this research is to detect the antibacterial properties of lactic acid bacteria (LAB) against pathogenic bacteria. Materials and Methods: Isolation and determination of Lactobacillus spp. Testing of the antibacterial activity of LAB was conducted using filtrate and nonfiltrate forms. The lactic acid bacterial isolates were confirmed to be identified through Gram staining, cell shape, catalase testing, and motility testing. Results: The results of the analysis of the LAB inhibition zone using filtrate and nonfiltrate forms against the bacteria Bacillus cereus were included in the very strong category. The results of the analysis of the LAB inhibitory zone using filtrate and nonfiltrate forms and the agar well method against Staphylococcus aureus bacteria were classified into the very strong category. The results of the LAB inhibitory zone analysis using filtrate and nonfiltrate forms with the well method against Escherichia coli bacteria are included in the very strong category, whereas the results from the LAB inhibitory zone analysis using the filtrate and nonfiltrate forms with the agar diffusion method (disks) are included in the strong category. Conclusion: Based on the results, LAB isolated from Sumba mare's milk displayed antibacterial activity in the strong and very strong categories against pathogenic bacteria such as B. cereus, S. aureus, and E. coli. Objective: Evaluate the effect of arrival weight on feedlot growth performance and carcass characteristics of Holstein steers of similar age. Material and Methods: Three hundred calf-fed Holstein steers (age 113 ± 1-day) were distributed in a completely randomly unbalanced design and divided into five categories (105, 112, 117, 123, and 129 kg) of shrunk initial weight (SIW). Calves were weighed on days 1, 112, 224, and 361. Calves were fed steam-flaked corn-based diets. Growth performance and dietary energy were evaluated for each period and the study as a whole (1–361-day). Results: During the rearing period, average daily gain (ADG) increased (linearly effect, p < 0.01) with increasing birth weight. Birthweight was positively associated (p < 0.05) with feedlot arrival weight (R2 = 0.47) and final harvest weight (R2 = 0.36). Overall ADG increased (p < 0.01) with increasing SIW. Dry matter intake increased linearly during the first 224-day but quadratically during the last 137 days. Overall, there was a quadratic effect (p < 0.05) of SIW on gain-to-feed and observed-to-expected dietary NE, with lower efficiencies (4%) for steers in both the lightest and heaviest SIW. Hot carcass weight, Longissimus muscle area, marbling score, and fat thickness increased (linear effect, p ≤ 0.03) as SIW increased, whereas kidney-pelvic-heart fat and yield-grade were unaffected. Conclusions: The initial arrival weight influences the growth performance, energetic efficiency, and carcass characteristics of Holstein steers of similar age. A sequence of BSP-1 peptides A0A0K1YXR5 from Bubalus bubalis (Domestic water buffalo) origin has been decided to be used to develop the peptide vaccine. Results: In this study, we succeeded in making synthetic vaccines from BSP-1 with a peptide sequence of LPEDSVPDEERVFPFTYRNRKHF. The three-dimensional theoretical prediction analy¬sis of the peptide binding pattern to its ligand, as well as the molecular docking, has also been revealed. Conclusions: A synthetic vaccine from the BSP-1 has been developed in this study with the amino acid sequence LPEDSVPDEERVFPFTYRNRKHF, which is buffer-soluble, and the three-dimensional theoretical prediction analysis of the peptide binding pattern of BSP-1 to its ligand, as well as molecular docking, has also been revealed. Objective: This study focuses on the orocutaneous fistula (OCF), a pathological channel between the buccal cavity and the outer surface (skin) of the face, causing leakage of saliva and fluid from the oral cavity to the face externally and also the ectopic incisors (EIs) erupted in a rare position in a day-old calf. The surgical interventions for these congenital defects were further described in this study. Materials and Methods: The calf was presented with an abnormal and external opening in the left submandibular region, having congenitally exposed jawbone and muscles with the resemblance of a linear groove, and was clinically examined based on the problem of swallowing milk due to lateral passing out of liquid through the unnatural tract before entering into the digestive tract. Clinical observation revealed an OCF with four EIs that abnormally erupted in the externally exposed groove of the fistula. Reconstructive surgery (RS) was performed along with a thorough guided approach to repair the defect, emphasizing normal functionality of the buccal cavity connecting to the pharynx and cranial esophagus, and smooth extraction of the EIs was ensured without making any deep wound. Results: After 2 weeks of postoperative care with supportive medications, the calf was quite recovered, having no further complications in the submandibular region along with no visible defect in swallowing. Conclusion: OCF in calves can be fruitfully treated by RS before getting severely infected and complicated. Outside the oral cavity, submandibular EIs can be easily removed, ensuring no further bleeding and defect. [J Adv Vet Anim Res 2022; 9(3.000): 509-515]

Objective: This study focuses on the orocutaneous fistula (OCF), a pathological channel between the buccal cavity and the outer surface (skin) of the face, causing leakage of saliva and fluid from the oral cavity to the face externally and also the ectopic incisors (EIs) erupted in a rare position in a day-old calf. The surgical interventions for these congenital defects were further described in this study. Objective: This study aims to develop and validate an Attenuated Total Reflectance–Fourier Transform Infrared Spectroscopy (ATR-FTIR) spectroscopic technique combined with a partial least squares regression (PLSR) model for rapid quantification and monitoring of aflatoxins in chicken feeds and food grains. Materials and Methods: A model of ATR-FTIR-PLSR was developed using ATR-FTIR spectra of mixed aflatoxin standards in 100% acetonitrile (112 samples) and 75% methanol (112 samples), validated by testing its prediction on 125 feed/food samples spiked with variable concentrations of aflatoxins, and applied to screen 660 samples of commercial chicken feeds and food grains from Nigerian and Malaysian markets for total aflatoxins, for which the dietary exposure risks to aflatoxins (DERA) and associated hepatocellular carcinoma (HCC) risks were evaluated for both countries. Results: The ATR-FTIR-PLSR model demonstrated excellent prediction power [R2 = 99.59%, p = 0.001, root mean square error of calibration (RMSEC) = 1.69, RMSE p = 1.98, bias = −0.26], sensi¬tivity (limit of quantitation and limit of the method < 5.0 ng/gm), precision (coefficient of variation = 0.97–1.72), and accuracy (% recovery of 88%–106%) in all the spiked samples. The model's pre¬diction was statistically reliable (R2 = 99.8%, p < 0.05) when compared with a high-performance liquid chromatography method. Levels of aflatoxins in the commercial samples signify high DERA (0.92–138.2 ng of aflatoxins/kg BW/day) and HCC risk (1.07%–159.91% of HCC/100,000 people/ year) in the exposed populations. Conclusions: Results feature the conceivable implementation of the proposed ATR-FTIR-PLSR model for rapid, accurate determination and monitoring of aflatoxins in commercial chicken feeds and food grains; and the need to strengthen aflatoxin control/prevention strategies in the study populations. Objective: Here, we developed and tested the efficacy of a vaccination protocol based on a commercially available live attenuated Salmonella enterica serovar Gallinarum (Salmonella galli¬narum) in layer chickens. Materials and Methods: 50 layer chickens of 16 weeks age were obtained and divided into two groups (n = 25), control and vaccinated. The vaccinated group received Salmonella vaccine at 0.2 ml/bird, s/c route at 16, 18, 30, and 42 weeks of age. At 21 weeks of age, birds from both groups were challenged with S. gallinarum orally at 4 × 107 colony-forming unit per bird. Results: Both rapid serum plate agglutination and enzyme-linked immunosorbent assay demonstrated a rising rate of seroconversion in vaccinated birds across the study period, with a 4% positive rate at 18 weeks, 56% at 21 weeks, 60% at 30 weeks, and 64% at each time point of 42 and 54 weeks. The vaccine showed 100% clinical protection and reduced the Salmonella shedding in the feces and eggs of the challenged birds. On the contrary, the unvaccinated challenged birds showed clinical signs and lesions typical of Salmonella infections with morbidity and mortality rates of 36% and 20%, respectively, and had high rates of Salmonella shedding in feces and eggs. Conclusions: With the proposed vaccination schedule Salmonella shedding was prevented, and a high seroconversion was confirmed. To prevent Salmonella infections in laying flocks, a 3-month interval immunization program is advised starting at the pre-laying stage. [J Adv Vet Anim Res 2022; 9(2.000): 301-309]

Objectives: The goal of this research was to figure out the effect of the local sorghum as silage on the performance of Bligon goats. Microbial protein synthesis, digestible nutrients, and average daily weight gain (ADWG) were measured to evaluate the goats' performance. Materials and Methods: The study was designed in a completely randomized design with a one-way pattern. Twelve female Bligon goats with 24.33 ± 2.83 kg (mean ± SEM) initial body weight were divided into three groups of total mixed ration (TMR) treatments. Group 1 received fresh Napier grass (FNG) as a control, group 2 received imported sorghum silage (ISS) of brown midrib resistance (BMR), and group 3 received local sorghum silage (LSS) of super-2. Analysis of variance was used to analyze the data on microbial protein synthesis and feed intake during the research. On the contrary, analysis of covariance was used to analyze ADWG with initial weight as a covariate. Results: Microbial protein synthesis, feed-intake, and ADWG of goats that received TMR based on silage of two varieties of sorghum, namely BMR (ISS) and super-2 (LSS), were lower (p < 0.05) than control. However, there was no significant difference between both TMR based on sorghum silages. ISS's feed conversion was better than LSS (p < 0.05), and FNG was the best. Sorghum silage as a basal ration in TMR had lower microbial protein synthesis but higher total digestible nutrient content than fresh forage, such as Napier grass. The sorghum varieties did not affect the microbial protein synthesis, digestible nutrients, and ADWG of Bligon goats. However, ISS treatment had higher feed efficiency than LSS. Conclusion: The local sorghum (super-2) silage can be used as ruminant feed as well as imported sorghum (BMR) offered as TMR. Objective: This study aimed to produce hyperimmune serum against genotype VII Newcastle disease virus (NDV) with several applications. Materials and Methods: Production of hyperimmune serum against genotype VII NDV was performed on eight New Zealand white rabbits divided into four groups. Rabbits were immunized three times on the 1st day, the 14th day, and the 30th day. Blood sampling was carried out on the 8th day after the third immunization. Results: All groups showed the same pattern of hemagglutination inhibition (HI) titer results. HI titers would peak on the 5th or the 9th day after the second immunization, then decrease until the 3rd day after the third immunization, and increase again on the 5th day after the third immu¬nization. Rabbits immunized intravenously showed higher HI titers than the other groups. These results indicate that the intravenous route for hyperimmune serum production against genotype VII Newcastle disease virus greatly affects the immune response result. Conclusions: The production of hyperimmune serum by intravenous immunization three times was able to produce the highest titer of 210 at 38 days. The agar gel precipitation test and the Western blot assay showed that the hyperimmune serum was specific for the Newcastle disease antigen. [J Adv Vet Anim Res 2022; 9(2.000): 211-220]

Objective: This study aimed to produce hyperimmune serum against genotype VII Newcastle disease virus (NDV) with several applications. Materials and Methods: Production of hyperimmune serum against genotype VII NDV was performed on eight New Zealand white rabbits divided into four groups. Rabbits were immunized three times on the 1st day, the 14th day, and the 30th day. Blood sampling was carried out on the 8th day after the third immunization. Results: All groups showed the same pattern of hemagglutination inhibition (HI) titer results. HI titers would peak on the 5th or the 9th day after the second immunization, then decrease until the 3rd day after the third immunization, and increase again on the 5th day after the third immu¬nization. Rabbits immunized intravenously showed higher HI titers than the other groups. These results indicate that the intravenous route for hyperimmune serum production against genotype VII Newcastle disease virus greatly affects the immune response result. Conclusions: The production of hyperimmune serum by intravenous immunization three times was able to produce the highest titer of 210 at 38 days. The agar gel precipitation test and the Western blot assay showed that the hyperimmune serum was specific for the Newcastle disease antigen. [J Adv Vet Anim Res 2022; 9(2.000): 211-220]
