Influence of isoprinosine on lymphocyte function in virus-infected feeder pigs

Pseudorabies is a porcine herpesvirus of major importance in the swine industry. Isoprinosine is an immunomodulating drug that has been shown to be beneficial in treating herpesvirus infections. Twenty-four 7-week-old pigs were allotted within litters to 1 of 4 groups: control, isoprinosine (ISO), pseudorabies virus (PRV), or isoprinosine and pseudorabies virus (ISO-PRV). Isoprinosine was administered daily for 16 days to the ISO and ISO-PRV groups (75 mg/kg of body weight/day, PO). Immunity in pigs in the PRV and ISO-PRV groups was challenged with pseudorabies virus (10(5) TCID50 units) on day 4. Rectal temperatures and viral excretion were monitored daily; total and differential leukocyte counts, lymphocyte response to mitogens, and interleukin-2 production were monitored every 4 days. Pigs challenge-inoculated with pseudorabies virus became ill, with the ISO-PRV group most severely affected. Rectal temperatures were high (P less than 0.05) in virally challenged pigs on days 5 to 12 and 14 to 16; isoprinosine did not alter this effect. Pseudorabies virus-infected pigs had leukocytosis (P less than 0.05) on days 12 and 16, primarily caused by neutrophilia. Concanavalin A-stimulated lymphocyte proliferation was decreased (P less than 0.06) in both PRV and ISO-PRV groups on day 12, compared with control pigs, but only in the PRV group on day 16. Pokeweed mitogen-stimulated lymphocyte proliferation was decreased (P less than 0.02) in ISO-PRV pigs on day 8 of the experiment. Interleukin-2 concentrations, pooled over all sampling days, were decreased (P less than 0.03) in pseudorabies virus-infected pigs. Viral excretion was not altered by isoprinosine treatment. These data suggest that pseudorabies virus infection decreased lymphocyte proliferative responses and interleukin-2 prodcution in pigs, and that isoprinosine did not mitigate these effects.

High-speed cinematography was used to record the movements of 12 cutting horses performing a standard test with a mechanical flag. Based on their previous competitive performances, horses were classified into 2 groups: group 1, composed of 5 moderately successful or average performers that had won less than $35,000 in purse money; and group 2, composed of 7 highly successful or elite performers that had amassed greater than $35,000 in competition earnings. Analysis of the results indicated that, compared with horses of the average group, the elite horses had faster reaction times in response to the start and cessation of flag movement (P less than 0.01), and were positioned closer to the flag during all stages of the trial (P less than 0.05). Discriminant analysis was used to construct a mathematical formula that could be used to classify an individual horse into 1 of the 2 alternative groups, based on the set of measurements. Two predictor variables were selected that described the maximal distance between the horse and the flag during the run and the part of the body that was moved first in response to the initial flag movement. The accuracy of the predicted group membership, compared with the actual group membership, was 100%.

A clinical trial examining the efficacy of 2 drugs for treatment of a natural epizootic of infectious bovine keratoconjunctivitis was performed. The study was conducted in 103 grazing Hereford calves during the summer of 1985. The calves were prospectively and randomly assigned to 1 of 3 groups at the beginning of the study on June 17, and were examined 3 times weekly thereafter until the final observation on August 6. Calves in group 1 (n = 34) were not treated and were used as controls. Calves of group 2 (n = 34) with corneal ulcers were treated with a long-acting oxytetracycline formulation (OTC group). The parenteral treatment was repeated in 72 hours. Affected calves of group 3 (n = 35) were treated topically with furazolidone spray when they developed new corneal ulcers, or when existing lesions worsened during subsequent examination periods (NFZ group). Healing times of the corneal ulcers were reported in 3 ways: the combined times for ulcers present in both eyes of a calf simultaneously (method A), independent times of each ulcer on a calf (method B), and time of the first ulcer for each calf (method C). Censored healing times were examined as left censored (ulcer present at the beginning of the study), right censored (ulcer not healed at the end of the study), or uncensored (true) healing times. The effect that the treatments had on healing times were investigated by use of notched box and whisker plots, life tables, and Cox regression models. The analysis indicated that treatment of calves with either antimicrobial reduced the healing time of corneal ulcers, compared with untreated controls. Calves treated with OTC had shorter periods with ulcers present on both eyes than did NFZ-treated calves. The healing time of the first ulcer on a calf was faster when treated with either antimicrobial than when not treated, but no significant difference between periods for OTC and NFZ treatments was found. Censored healing times were consistently longer than uncensored healing times. Box and whisker plots indicated that both treatments shortened healing times more than those for controls, and OTC shortened healing times more than did NFZ for responses A and B (but not C). Life tables showed that OTC healing times were shorter than those for controls, and NFZ shorter than controls for response B and C (but not A). Cox regression model (for response A) showed a borderline significant difference between times for OTC group and controls, and no significant difference between times for NFZ group and controls.

Lipopolysaccharide (LPS) fractions were obtained from smooth cultures of Brucella abortus strains 2308 and S-19 by butanol extraction procedures. The LPS from the initial butanol extraction contained 10 to 15% protein and was reduced to less than 1% protein by treatment with proteinase K. The LPS fractions were identified and characterized on the basis of the chemical analysis, sodium dodecyl sulfate gel electrophoresis, cesium chloride gradients, electron microscopy, and gel immunodiffusion. Results indicated that the butanol procedure is a reliable method in the extraction of LPS from Brucella abortus cells. Proteinase K-treated LPS containing less than 1% protein from strain 2308 was used to vaccinate BALB/cByJ mice. Immune and protective criteria for vaccinated and nonvaccinated mice were increased immunoglobulin (IgG and IgM) titers in sera of prechallenge-exposed mice, reduced colony-forming units/spleen, and splenomegaly in post-challenge-exposed mice. Results indicated that proteinase K-treated LPS was immunuogenic as well as protective for mice.

The normal B-scan ultrasonographic anatomic features of the eye and orbit of mesocephalic and dolichocephalic dogs were described. The B-scan appearance of ocular and orbital structures correlated well with the visual morphologic features of the specimens. The eyes of mesocephalic and dolichocephalic dogs were measured by use of ultrasonographic methods; those measurements were compared with direct measurements of the specimens. The 2-tailed Student t-test was used for all statistical analyses (P = 0.05). Measurements made included mid-cornea to anterior lens surface, lens thickness, vitreous body (posterior lens surface to retina), and axial globe length. The A-scan measurements of all 4 ocular distances were significantly different, compared with direct measurements. The B-scan measurements of mid-cornea to anterior lens surface, lens thickness, and vitreous body were significantly different from direct measurements; however, there was no significant difference between B-scan and direct measurements of axial globe length. There was no significant difference between A- and B-scan measurements. These findings suggest that A- and B-scan measurements are similar and that B-scan measurements are reasonably accurate for axial globe determination. Several variables were compared by B-scan and direct measurement methods. The axial globe length of dolichocephalic dogs was significantly longer than that of mesocephalic dogs. The axial globe length of male and female dogs was not significantly different in mescephalic or dolichocephalic dogs. There was no difference in the axial globe length of right and left eyes in mesocephalic or dolichocephalic dogs.

To evaluate the effect of exogenous testosterone on the development of T-2 toxin-induced necrosis of adrenal glands, mice were allotted to 3 treatment groups. Each treatment group contained castrated male, and castrated and sexually intact female mice. Each mouse in group 1 was given 0.16 mg testosterone propionate at 48-hour intervals for a total of 12 infections, group-2 mice were given similar injections of only the vehicle, and group-3 mice were given no treatment. Twenty-four hours after the last injection, the mice in all 3 groups were exposed for 10 minutes to an aerosol of T-2 toxin. All mice alive at 24 hours after exposure were euthanatized and the adrenal glands and thymuses were examined histologically. Necrosis of the adrenal cortex was not found in any of the mice given preexposure treatment with exogenous testosterone, whereas all mice given vehicle only or no treatment had T-2 toxin-induced necrosis of the inner portion of the adrenal cortex. Lymphocytolysis in the cortex of the thymus confirmed that each mouse of all 3 treatment groups had experienced systemic mycotoxicosis. The uniform severity of the lesion in all mice suggests that the thymus was not protected by exogenous testosterone administration or by the castration status of the mice. We propose that T-2 toxin-induced adrenal necrosis in mice is prevented by the presence of testosterone.

Immunochemical methods that are used to assess von Willebrand factor in human beings and dogs were used to assess von Willebrand factor in 3 cat species. Our findings indicated that the expression and multimeric composition of von Willebrand factor in plasma and platelets of cats were similar to those reported in human beings and dogs. We suggest that these methods may be used to evaluate von Willebrand disease in members of the cat family used in this study.

In a group of 65 dogs, radiographic liver length, the length of the axis from the most cranial part of the diaphragm to the apex of the liver tip, was correlated significantly (P less than 0.000001) with real liver volume. Within this group, radiograhic liver length, compared with the length of the eleventh thoracic vertebra, was correlated with liver volume per kilogram of body weight. In a group of 30 dogs, with histologically normal liver, this measurement was not affected by thoraic conformation. These findings suggest that radiographic liver length is a reliable measurement for estimating liver volume in dogs and that it is not influenced by thoracic conformation. For 60 of the 65 dogs, a method of assessment of liver volume was formulated that required 2 measurements to be made on the lateral abdominal radiograph and 1 to be made on the dog itself.

Measurement of liver size was made on nuclear scintigraphic images obtained from 16 clinically normal, anesthetized dogs in ventral, dorsal, right and left lateral, and left dorsal oblique positions after administration of technetium 99m-sulfur colloid. Linear measurements of liver length and width were make from each scintiscan, and liver surface area was determined by setting a region of interest manually and by use of a computer count of pixels above a minimal intensity (threshold method). All linear measurements had a statistically significant (P less than 0.05) correlation with liver and body weight, with the exception of the measurements of liver width made on dogs in dorsal and left lateral dorsal oblique positions. The highest correlation (r = 0.89) between the scintigraphic measurements and liver weight was the multiplication of measurements of liver width and length from the right lateral view. Although all area determinations were significantly (P less than 0.05) correlated with liver and body weight, for most views, the manual method of determining the region of interest had a slightly better correlation with the liver weight than did the threshold method.

The effects of Moraxella bovis on the morphologic features of purified bovine neutrophils and bovine corneal epithelial cells were examined, using transmission and scanning electron microscopy and light microscopy. Within 2 minutes after incubation of bovine neutrophils with living M bovis, electron microscopic cellular changes included vacuolation, swelling, and loss of microplicae. Most of the neutrophils were lysed by 10 minutes of incubation. Human neutrophils phagocytosed the M bovis and remained intact, even after 30 minutes of incubation with the bacteria. Living M bovis killed bovine corneal epithelial cells in vitro. Sterile filtrates prepared from 6-hour shaker cultures of M bovis also killed bovine corneal epithelial cells, but the cytotoxic activity was less than that produced by the living bacteria. Cellular changes were first observed in specimens collected 1 hour after corneal cell monolayers were inoculated with sterile culture filtrates. The changes in these cells included pit-like lesions on the cellular surface, cellular separation, and vacuolation.
