Potential mechanisms of phthalate ester embryotoxicity in the abalone Haliotis diversicolor supertexta

The effects and associated toxicological mechanisms of five phthalate esters (PAEs) on abalone embryonic development were investigated by exposing the embryos to a range of PAEs concentrations (0.05, 0.2, 2 and 10μg/mL). The results showed that PAEs could significantly reduce embryo hatchability, increase developmental malformations, and suppress the metamorphosis of abalone larvae. The possible toxicological mechanisms of PAEs to abalone embryos included, affecting the Na⁺–K⁺-pump and Ca²⁺–Mg²⁺-pump activities, altering the peroxidase (POD) level and the malondialdehyde (MDA) production, damaging the extraembryonic membranes structure, as well as disrupting endocrine-related genes (gpx, cyp3a, and 17β-hsd 12) expression properties. Taken together, this work showed that PAEs adversely affected the embryonic ontogeny of abalone. The abilities of PAEs affecting the osmoregulation, inducing oxidative stress, damaging embryo envelope structure, and causing physiological homeostasis disorder, are likely to be a part of the common mechanisms responsible for their embryonic toxicity.

This study examined the physiological responses of the larval stages of Haliotis tuberculata, an economically important abalone, to combined temperature (17 °C and 19 °C) and pH (ambient pH and −0.3 units, i.e., +200% increase in seawater acidity) in a full factorial experiment. Tissue organogenesis, shell formation, and shell length significantly declined due to low pH. High temperature significantly increased the proportion of fully shelled larvae at 24 h post-fertilization (hpf), but increased the proportion of unshelled larvae at 72 hpf. Percentage of swimming larvae at 24 hpf, 72 hpf and 96 hpf significantly declined due to high temperature, but not because of low pH. Larval settlement increased under high temperature, but was not affected by low pH. Despite the fact that no interaction between temperature and pH was observed, the results provide additional evidence on the sensitivity of abalone larvae to both low pH and high temperature. This may have negative consequences for the persistence of abalone populations in natural and aquaculture environments in the near future.

Tidal current survey as well as geochemical and benthic foraminiferal analyses of sediment cores were conducted in an abalone farm and a Zostera bed to understand the degree to which the abalone farm facilities installed along a channel in a shallow sea affect the benthic environment and ecology. In the abalone farm, Ammonia beccarii-Pseudoparrella naraensis-Elphidium somaense-Rosalina globularis-Trochammina hadai and P. naraensis-E. somaense-A. beccarii-T. hadai assemblages appeared owing to an increase in the total nitrogen content from the biodeposits. The Zostera bed consisted of A. beccarii-P. naraensis-Buccella frigida-T. hadai assemblage owing to the gradual expansion of a brackish shallow-water environment by the rapidly decreasing current speed, and it may have flourished. Moreover, the total sulfur, Zn, Cr, and Cu contents in the sediments decreased remarkably more than those of the pre-abalone farming did, caused by the vigorous activity of Zostera marina physiology.

The increasing use of nanotechnology highlights the need to understand and clarify the environmental impacts of nanomaterials. In this study, the acute toxicity and oxidative stress of titanium dioxide nanoparticles (nTiO₂) in mature marine abalone (Haliotis diversicolor supertexta) were assessed. No acute effect was found in any of the treatment groups with nTiO₂ concentration gradients ranging from 0.1 to 10mg/L. However, the activity of an antioxidant enzyme superoxide dismutase (SOD) significantly increased in the group that was exposed to 1.0mg/L nTiO₂. The content of a non-enzymatic antioxidant, glutathione (GSH), significantly decreased in the groups with an nTiO₂ concentration ⩾1.0mg/L. The level of lipid peroxidation (LPO) was found to increase as the nTiO₂ dose increased. Furthermore, NO was produced in excess in abalone. These results demonstrated that, although nTiO₂ is not acutely toxic to abalone, it does exert oxidative stress on abalone.

This study reports molecular markers potentially associated with resistance or sensitivity to the impact of copper in juvenile red abalone, Haliotis rufescens, in the north of Chile under experimental conditions. Genomic analysis was made applying subtractive hybridization libraries (SSH) to identify genes up-and down regulated during cooper exposure in abalone over periods of 12 and 168h exposed to 2.5 and 10μg/L of Cu⁺². Results obtained from the SSH library revealed 368 different sequences regulated by copper, that correspond to eight major physiological functions. The validation of these sequences obtained by SSH as well as their expression kinetics were made by PCR in real time on 14 potential genes regulated by metal stress. This study provides information for the characterization of potential genomic markers that may be used in future environmental monitoring and to investigate new mechanisms of stress to copper in this commercially important marine species.

The abalone aquaculture industry in South Korea has grown rapidly since the 2000s. In this study, we investigated the sedimentary pollution at four major abalone farms responsible for ~60% of all South Korean abalone produced. We also surveyed the current statuses of cage facilities, abalone mass mortality, and current velocities within and outside farm cages. The concentrations of total organic carbon in the study area were 7.92 ± 2.09 mg g⁻¹, indicating a mild level of sedimentary pollution. We observed higher mortality rates in rectangular-shaped shelter cages than in triangular shelters. With increases in the number and size of abalone farming facilities, current velocities inside the cages declined by an average of 45% relative to those outside the cages, leading to poor habitat conditions for farmed abalone. Our results provide insights into the current status of the benthic environments and major causes of mass mortality in the abalone farms of South Korea.

To investigate the effects of mariculture on the sources, distribution and preservation of sedimentary organic carbon (SOC), sediments from an mariculture area in Ailian Bay, China, and a control area were analyzed for grain size composition, total organic carbon (TOC), total nitrogen (TN), carbon/nitrogen (C/N) ratio, and stable carbon and nitrogen isotopic composition (δ13C and δ15N). The sedimentary type of sediments in study area was clay silt. TOC, TN, C/N, δ13C, and δ15N ranged from 0.58 to 1.21%, 0.06–0.17%, 6.29–9.82, −23.20 to −18.50‰, and 6.17–7.38‰, respectively, and followed similar spatial patterns. TOC, TN and δ13C were higher in mariculture area than in control area. Biodeposit and kelp OC contributions greater of SOC in mariculture area than the control area (biodeposits: 20.10 ± 4.84 to 6.2 ± 1.3%; kelp: 15.3 ± 6.63 to 5.2 ± 0.84%). Overall, mariculture activities significantly influence the sources, distribution and preservation of SOC.

546 Vibrio isolates from rearing seawater (292 strains) and intestines of abalone (254 strains) were tested to ten antibiotics using Kirby–Bauer diffusion method. Resistant rates of abalone-derived Vibrio isolates to chloramphenicol (C), enrofloxacin (ENX) and norfloxacin (NOR) were <28%, whereas those from seawater showed large fluctuations in resistance to each of the tested antibiotics. Many strains showed higher resistant rates (>40%) to kanamycin (KNA), furazolidone (F), tetracycline (TE), gentamicin (GM) and rifampin (RA). 332 isolates from seawater (n=258) and abalone (n=74) were resistant to more than three antibiotics. Peaked resistant rates of seawater-derived isolates to multiple antibiotics were overlapped in May and August. Statistical analysis showed that pH had an important effect on resistant rates of abalone-derived Vibrio isolates to RA, NOR, and ENX. Salinity and dissolved oxygen were negatively correlated with resistant rates of seawater-derived Vibrio isolates to KNA, RA, and PG.

The embryos of a marine abalone, Haliotis diversicolor supertexta, were exposed to a typical environmental estrogen, 17α-ethynylestradiol (EE2), for 96 h, to examine the acute toxicity of EE2 to the embryogenesis of the abalone. A marine diatom, Navicula incerta, was used in the test media as settlement substrate and food for the abalone larvae. During the embryo culture, more than 30 % of EE2 could be removed from the test media by the diatom, mainly via biodegradation, leading to a decrease of water-borne exposure dose. Further, the exposure concentrations of EE2 around the living microenvironment of the abalone larvae could be magnified 350–468 times after the larvae settled on the diatom, as indicated by the bioconcentration factors of EE2 in the diatom. Increased bioaccumulation of EE2 in the diatom caused greater inhibition on the metamorphosis of the abalone larvae by enhancing the uptake of EE2 in the larvae via dietary exposure, while declined water-borne exposure dose did not affect the embryonic toxicity of EE2 and its uptake in the abalone larvae. The 96-h median effective concentration of EE2 to the metamorphosis of the abalone larvae was 10.01 μg L⁻¹, when the exposure doses in both the test media and the diatom were controlled stable. The 96-h hazard concentration for 5 % of the species was 1.20 μg L⁻¹, which was still higher than but close to the reported upper contamination level of EE2 and could be employed as the safety threshold for the metamorphosis of the abalone embryos.

In this study, the 96-h LC₅₀ at 22 and 26 °C values was 28.591 and 11.761 mg/L, respectively, for NiCl₂ exposure in the abalone. The alteration of physiological and immune–toxicological parameters such as the total hemocyte count (THC), lysozyme, phenoloxidase (PO), and phagocytosis activity was measured in the abalone exposed to nickel (200 and 400 μg/L) under thermal stress for 96 h. In this study, Mg and THC decreased, while Ca, lysozyme, PO, and phagocytosis activity increased in the hemolymph of Pacific abalone exposed to NiCl₂ when compared to a control at both 22 and 26 °C. However, these parameters were not affected by a rise in temperature from 22 to 26 °C in non-exposed groups. Our results showed that NiCl₂ below 400 μg/L was able to stimulate immune responses in abalone. However, complex stressors, thermal changes, or NiCl₂ can modify the immunological response and lead to changes in the physiology of host–pollutant interactions in the abalone.
