Silver (Ag) is amongst the most well studied nanomaterials (NMs), although most studies have only dealt with a single AgNM at a time and one biological endpoint. We here integrate the results of various testing-tools (endpoints) using a terrestrial worm, the standard ecotoxicological model organism Enchytraeus crypticus. Exposure spanned both water and soil exposure, it covered all life stages (cocoons, juveniles and adults), varying exposure durations (1-2-3-4-5-21 days), and covered 5 biological endpoints: hatching success, survival, reproduction, avoidance and gene expression (qPCR target genes GABA and Acetyl cholinesterase). We tested 4 Ag materials: PVP coated (PVP-AgNM), non-coated (NC-AgNM), the JRC reference Ag NM300K and AgNO₃. Results showed that short-term exposure via water to assess impact on cocoons’ hatching predicted longer term effects such as survival and reproduction. Moreover, if we extended the exposure from 11 to 17 day this allowed discrimination between hatch delay and impairment. Exposure of juveniles and adults via water showed that juveniles were most sensitive with survival affected. Across materials the following toxic ranking was observed: AgNO₃ ≥ Ag NM300K ≫ NC-AgNM ≥ PVP-AgNM. E. crypticus avoided AgNO₃ in a dose-response manner, avoiding most during the first 24 h. Avoidance of Ag NM300K and NC-AgNM only occurred during the first 24 h and the PVP coated AgNM were not avoided at all. The up-regulation of the GABA triggering anesthetic effects, indicated the high ecological impact of Ag materials in soil: Ag affects the GABAergic system hence organisms were not able to efficiently avoid and became intoxicated, this caused impacts in terms of survival and reproduction.

The Lippia alba essential oil (EO) is a fish anesthetic immiscible in water and commonly used diluted in ethanol. We evaluated the effectiveness of surfactant use with Lippia alba EO in the anesthesia of Oreochromis niloticus, as well as its toxicity in fish and mammals. The EO was extracted by hydrodistillation and the fish were exposed to anesthesia at the concentration of 250 μL/L for 10 min with the surfactants polysorbate 20 (T20), polysorbate 80 (T80), polyethylene glycol (PEG), and ethanol. We also evaluated fish recovery and anesthetic safety margin after exposure for 10, 20, and 30 min. To assess the surfactants’ toxicity in mammals, Mus musculus (mice) received the same treatments by gavage. The main constituents of the Lippia alba EO were linalool (42.36%), geraniol (12.46%), neral (10.7%), and limonene (7.45%). Deeper anesthesia was faster in the T20 (60 ± 2.9 s) and T80 (272 ± 21 s) treatment groups, while recovery time for T80 was longer (596 ± 47 s). All treatments showed a good safety margin, without mortality. The genotoxic effects caused by surfactants in mammals and fish were at similar levels to those found in the ethanol treatment. Therefore, this study demonstrated that the use of surfactants T20 and T80 in Oreochromis niloticus anesthesia presented neither a reduction nor a considerable increase of the toxicity when compared to the commonly used ethanol; however, an increase in anesthetic effectiveness was observed throughout the experiment.

Adsorption together with size exclusion and charge attraction/repulsion has to be taken into account when considering removal of pharmaceuticals as emerging contaminants from water by reverse osmosis and nanofiltration membranes. Glucocorticosteroids (hydrocortisone (HYDRO), dexamethasone (DEXA)), anesthetics (procaine, lidocaine) with relatively weak hydrophobicities (1 < log K O/W < 3), and membranes (XLE, LFC–1, CPA3, SWC1, NF90, and NF270) have been investigated in this study. Adsorption was studied by measuring the concentration of compounds in feed and permeate and by monitoring changes in membrane flux in the batch mode operation during 24 h. A decrease in the feed concentrations for HYDRO and DEXA (log K O/W < 2) was observed. The loss of these compounds in feed was associated with irreversible adsorption onto an NF270 and a CPA3 membrane. Therefore, when considering removal of pharmaceuticals with lower hydrophobicity, adsorption has to be particularly taken into account for membranes with bigger pores in the selective layer. Also, a high dipole moment and low water solubility affected adsorption on the membranes. For smaller and slightly more hydrophobic pharmaceuticals (log K O/W > 2), an increase in the feed concentration was obtained. Firstly, these compounds instantly adsorbed to the membrane. Secondly, the compounds diffused through the polymer matrix and desorbed to the permeate side after equilibrium had been reached.

Occupational exposure to inhalational anesthetics has been associated with health problems, including reproductive issues. Considering the scarcity and outdated nature of reviews concerning this relevant topic, which has implications for indoor pollution/environmental science/public health, this critical review aimed to systematically evaluate whether exposure to inhalational anesthetics is associated with abortion. Seven databases were searched with no language or year restrictions. Of the 3881 search results, 18 observational studies were included. Some studies demonstrated a significant association between occupational exposure to inhalational anesthetics and spontaneous abortion, especially among professionals who work for longer periods and/or in an environment without gas scavenging/ventilation systems, which may favor the occurrence of abortion in this population. Due to considerable heterogeneity and limitations, it cannot be concluded whether an association exists between occupational exposure to anesthetics and the occurrence of abortion. However, more well-designed studies should be performed, especially in less economically developed countries that do not have access to quality anesthetic gas scavenging/ventilation systems, thereby bringing this issue into sharp focus. This review highlights the need for scientific knowledge in this area and the extensive use of scavenging equipment and in the workplace to minimize exposure and reduce the risk of abortion.

We found an error in the materials and methods section. Since our team used two methodsfor anesthesia in rats and the anesthesia method used in this paper was 3.5% chloralhydrate anesthesia, we mistakenly wrote the anesthetic as 3% sodium pentobarbital.

PURPOSE: Some of the pharmaceuticals that are not extensively investigated in the aquatic environment are the anesthetic lidocaine (LDC), the analgesic tramadol (TRA), and the antidepressant venlafaxine (VEN). LDC metabolizes to 2,6-xylidine (2,6-DMA) and monoethylglycinexylidine (MEGX), TRA to O-desmethyltramadol (ODT), and VEN to O-desmethylvenlafaxine (ODV). Within this study, the distribution and behavior of these compounds in German wastewater treatment plants (WWTPs) were investigated. METHODS: Samples of influents and effluents from WWTPs in Hesse, Germany were collected between January and September 2010. Analytes were extracted from wastewater samples by solid-phase extraction and from solid samples by sonication. Extracts were measured using gas chromatography/mass spectrometry. RESULTS: LDC, TRA, VEN, ODT, and ODV were detected in all analyzed influent and effluent samples. 2,6-DMA could not be identified. MEGX was not detected. TRA and ODV were present in untreated wastewater at the highest concentrations (max, 1,129 (TRA) and 3,302 ng L−1 (ODV)), while the concentrations of LDC and VEN were all significantly lower (mean, 135 (LDC) and 116 ng L−1 (VEN)). All of the analytes were only partially removed in the WWTPs. The mean ratios between the concentrations of the metabolites and their respective parent compounds in influents were 4.7 (ODV/VEN) and 0.7 (ODT/TRA). These values remain approximately constant comparing influents and effluents. CONCLUSIONS: LDC, TRA, VEN, ODT, and ODV are only partially removed from sewage water by WWTPs and thus are continuously discharged in respective recipient rivers. A further transformation of TRA and VEN into the known metabolites during treatment in the WWTPs is not observed.

Despite the fact that there are tens of thousands of thermal baths in existence, knowledge about the occurrence of pharmaceutically active compounds (PhACs) in untreated thermal wastewater is very limited. Because used thermal water is typically legally discharged into surface waters without any treatment, the effluent poses environmental risks for the receiving water bodies. The aim of this study was to show the occurrence patterns and spatiotemporal characteristics of 111 PhACs in thermal wastewater. Six thermal water outflows of different thermal baths were tested in different seasons in the Budapest metropolitan region (Hungary), and diurnal analysis was performed. After solid-phase extraction, the samples were analysed and quantified by coupling supercritical fluid chromatography and mass spectrometry to perform simultaneous multi-residue drug analysis. The results confirm that water discharge pipes directly transport pharmaceuticals into surface water bodies; 34 PhACs were measured to be over the limit of quantification at least once, and 21 of them were found in more than one water sample. The local anaesthetic drug lidocaine, antiepileptic carbamazepine, analgesic derivative tramadol and illicit drug cocaine were detected in more than half of the samples. Caffeine, metoprolol and bisoprolol (cardiovascular drugs), benzoylecgonine (cocaine metabolite), diclofenac (NSAID), citalopram (antidepressant) and certain types of hormones also have a significant frequency of 30-50%. However, the occurrence and concentrations of PhACs vary according to the season and number/types of visitors. As demonstrated by the diurnal fluctuation, drug contamination of thermal waters can significantly vary, even for similar types of baths; furthermore, the quantity and types of some pollutants rapidly change in the discharged thermal wastewater.

Mosquitoes threaten the lives of humans, livestock, pets and wildlife around the globe, due to their ability to vector devastating diseases. Aglaia elaeagnoidea, commonly known as Priyangu, is widely employed in Asian traditional medicine and pest control. Medicinal activities include anti-inflammatory, analgesic, anticancer, and anesthetic actions. Flavaglines, six cyclopenta[b]benzofurans, a cyclopenta[bc]benzopyran, a benzo[b]oxepine, and an aromatic butyrolactone showed antifungal properties, and aglaroxin A and rocaglamide were effective to control moth pests. Here, we determined the larvicidal action of A. elaeagnoidea leaf aqueous extract. Furthermore, we focused on Priyangu-mediated synthesis of Ag nanoparticles toxic to Culex quinquefasciatus, Aedes aegypti and Anopheles stephensi. The plant extract and the nanolarvicide were tested on three mosquito vectors, following the WHO protocol, as well as on three non-target mosquito predators. Priyangu-synthesized Ag nanoparticles were characterized by spectroscopic (UV, FTIR, XRD, and EDX) and microscopic (AFM, SEM, and TEM) analyses. Priyangu extract toxicity was moderate on Cx. quinquefasciatus (LC₅₀ 246.43; LC₉₀ 462.09 μg/mL), Ae. aegypti (LC₅₀ 229.79; LC₉₀ 442.71 μg/mL), and An. stephensi (LC₅₀ 207.06; LC₉₀ 408.46 μg/mL), respectively, while Priyangu-synthesized Ag nanoparticles were highly toxic to Cx. quinquefasciatus (LC₅₀ 24.91; LC₉₀ 45.96 μg/mL), Ae. aegypti (LC₅₀ 22.80; LC₉₀ 43.23 μg/mL), and An. stephensi (LC₅₀ 20.66; LC₉₀ 39.94 μg/mL), respectively. Priyangu extract and Ag nanoparticles were found safer to non-target larvivorous fishes, backswimmers, and waterbugs, with LC₅₀ ranging from 1247 to 37,254.45 μg/mL, if compared to target pests. Overall, the current research represents a modern approach integrating traditional botanical pesticides and nanotechnology to the control of larval populations of mosquito vectors, with negligible toxicity against non-target including larvivorous fishes, backswimmers, and waterbugs.

In this work, a multi-class analytical method for determination of 22 frequently used pharmaceuticals was developed and validated. Analytes were from different classes for example macrolides, fluoroquinolones, tetracyclines, sulfonamides, anthelmintics, anesthetics, and others. Method was intended for analysis of aqueous samples so the sample preparation was done using solid-phase extraction (SPE). Different sorbents (C8, C18, polymeric, and ion exchange sorbents) combining different eluents (methanol, ethanol, acetonitrile, acetone, ethyl acetate) were investigated during development of sample preparation step. Samples were analyzed using HPLC-MS/MS, and therefore, chromatographic and mass spectrometer conditions were investigated. Optimal extraction efficiencies for most of the investigated analytes were obtained with Oasis HLB polymeric sorbents with acetonitrile as eluent. A study of matrix effect was carried out for wastewater treatment plant (WWTP) influent and effluent. The method was validated for linearity, detection limits and quantification limits, repeatability, and reproducibility. Method detection limits were in the range of 2.0–204.0 ng L⁻¹ for WWTP influent except for sulfaguanidine and dexamethasone. Also, method detection limits for WWTP effluent were from 1.0 to 115.4 ng L⁻¹. Method was successfully applied for analysis of real wastewater samples from municipal wastewater treatment plant. In the influent, pharmaceuticals from all investigated groups were present and the concentrations were from 50.0 to 4914.3 ng L⁻¹ for influent and 26.9 to 1699.2 ng L⁻¹ for effluent. It was also reported that some pharmaceuticals showed higher concentrations in the wastewater effluent than in the influent.
