Effects of long-term exposure of Mytilus galloprovincialis to thiacloprid: A multibiomarker approach 全文

Thiacloprid is a neonicotinoid insecticide widely exploited in agriculture and easily mobilized towards aquatic environments by atmospheric agents. However, little information about its toxicological effects on aquatic invertebrate bioindicators is available. In this study, specimens of the mussel Mytilus galloprovincialis were exposed to thiacloprid at environmental (4.5 μg L⁻¹) and 100 times higher than environmental (450 μg L⁻¹) concentrations for 20 days. Thiacloprid affected haemolymph biochemical parameters, cell viability in the digestive gland, antioxidant biomarkers and lipid peroxidation in the digestive gland and gills at environmentally relevant concentrations (4.5 μg L⁻¹). In addition, thiacloprid exposure caused histological damage to the digestive gland and gills. Interestingly, the pesticide was detected at levels equal to 0.14 ng g⁻¹ in the soft tissues of sentinels exposed for 20 days to 450 μg L⁻¹ thiacloprid in seawaterμ. Due to its harmful potential and cumulative effects after long-term exposure of M. galloprovincialis, thiacloprid may pose a potential risk to nontarget aquatic organisms, as well as to human health. This aspect requires further in-depth investigation.

We exposed twelve mesocosm stream channels and four instream channels to one, two, and four pulses of the insecticide lambda-cyhalothrin (0.1 μg L⁻¹) applied at two day intervals, each pulse lasting 90 min. Unexposed controls were included. We monitored macroinvertebrate taxonomic composition in the channels and in deployed leaf packs one day before and 29 days after the first exposure. Further, we measured drift in and out of the channels and leaf litter decomposition. Lambda-cyhalothrin exposures induced significantly increased drift in both experiments especially for Gammarus pulex, Amphinemura standfussi, and Leuctra spp. Macroinvertebrate taxonomic composition increasingly changed with increasing number of lambda-cyhalothrin exposures being most pronounced in the mesocosm channels. Further, leaf decomposition significantly decreased with increasing number of exposures in the mesocosm channels. Our study showed that species with predicted highest sensitivity to lambda-cyhalothrin were primary drivers of significant changes in taxonomic composition lasting for at least one month despite continuous recolonization of exposed channels from upstream parts of the natural stream and from the water inlet in the mesocosm channels. The overall results highlight the importance of sequential exposures to insecticides for understanding the full impact of insecticides on macroinvertebrates at the community level in streams.

After the exploitation of coal mines in the 19th and 20th centuries in northern France, many mining slag heaps (SH) were left without any particular management or monitoring. Currently, the influence of these SHs on the quality of surrounding wetlands is hardly known.The purpose of this work is to determine the water quality in the neighbourhood of two SHs located near the city of Douai and its influence on the distribution of aquatic invertebrates in local wetlands. Our approach involves (1) the spatial and temporal characterization of the water composition (anions, major elements, sulphide, DOC and alkalinity) and of the biological diversity (aquatic invertebrates) and (2), based on this chemical and biological screening, the establishment of relationships between water quality and biodiversity distribution through multivariate data analysis. The results clearly indicate that substantial leaching from the slag heaps occurs, given the very high concentrations of dissolved sulphates (in the range of 2 g L⁻¹). While the pH remains weakly basic, indicating that the leaching water has been neutralized by the highly carbonated regional substratum, high levels of biodegradable organic matter and sulphate contents have been noticed. They sporadically cause significant drops in dissolved oxygen and the occurrence of dissolved sulphides that massively reduce biodiversity, qualitatively and quantitatively. In Summer, oxygen saturation is generally lower due to the higher rate of organic matter degradation, and the risk of anoxic episodes therefore increases. Finally, as wetlands are vulnerable environments, these preliminary results suggest that monitoring and management of these sites must be attempted quickly to avoid the degradation of those valuable habitats.

Urban wastewater effluents bring large amounts of nutrients, organic matter and organic microcontaminants into freshwater ecosystems. The effects of this complex mixture of pollutants on freshwater invertebrates have been studied mainly in temperate rivers and streams with high dilution capacities. In contrast, Mediterranean streams and rivers have lower dilution capacities especially during the seasonal drought, and are therefore exposed to high concentrations of pollutants. Here, we assess the effects of urban wastewater pollution on invertebrate communities from Mediterranean streams under different hydrological conditions. Specifically, we assessed the invertebrates taxonomic composition and functional biological traits in 12 streams, differing in stream and substrate size (sand or cobbles), under low (2 surveys) and baseflow (1 survey) conditions. In each stream, we selected reaches both upstream and downstream of the wastewater discharge point. Our results indicate that urban wastewater pollution favours the most tolerant invertebrate taxa and homogenises functional trait composition over time. Changes in functional traits were more evident during the seasonal drought, when the low flow conditions at the upstream and downstream sites were more severe and, pollutant concentrations downstream were at their highest. However, the effects of urban wastewater pollution were not uniform in the downstream sites; as local invertebrate communities differed in according to the river substrate and stream size (i.e., width and discharge). Overall, urban pollution caused by wastewater enhanced both, taxonomic and functional differences between the invertebrate communities. Such an absence of homogenisation among wastewater pollution impacted sites was probably related to the relevant role of stream substrate-size as well flow conditions in the rivers receiving the impact. These are attributes that need to be considered when setting the pollutant discharge limits in rivers and streams receiving effluents.

This study examined to what degree bioaccumulated pesticides in transplanted zebra mussels can give an insight to pesticide bioavailability in the environment. In addition, it was investigated if pesticide body residues could be related to ecological responses (changes in macroinvertebrate community composition). For this at 17 locations, 14 pesticide concentrations and nine dissolved metals were measured in translocated zebra mussels and the results were related to the structure of the macroinvertebrate community. Critical body burdens in zebra mussel, above which the ecological status was always low, could be estimated for chlorpyrifos, terbuthylazine and dimethoate being respectively 8.0, 2.08 and 2.0 ng/g dry weight.With multivariate analysis, changes in the community structure of the macroinvertebrates were related to accumulated pesticides and dissolved metals. From this analysis, it was clear that the composition of the macroinvertebrate communities was not only affected by pesticides but also by metal pollution. Two different regions could be clearly separated, one dominated by metal pollution, and one where pesticide pollution was more important.The results of this study demonstrated that zebra mussel body burdens can be used to measure pesticide bioavailability and that pesticide body burdens might give insight in the ecological impacts of pesticide contamination. Given the interrelated impacts of pesticides and heavy metals, it is important to further validate all threshold values before they can be used by regulators.

Glyphosate (GLY) is one of the most used herbicide worldwide. Considering that information concerning the impact of GLY on bivalves is scarce, in this study we evaluated for the first time the effects of environmentally realistic concentrations of GLY (10, 100 and 1000 μg/L) to the mussel Mytilus galloprovincialis. Mussels were exposed for 7, 14 and 21 days and several biomarkers were measured in haemocytes/haemolymph (total haemocyte counts, haemocyte diameter and volume, haemolymph pH, haemolymph lactate dehydrogenase activity, haemocyte lysate lysozyme and acid phosphatase activities), as well as in gills and digestive gland (antioxidant enzyme and acetylcholinesterase activities). The concentrations of GLY and its main metabolite aminomethylphosphonic acid in the experimental tanks were also measured. The MANOVA analysis demonstrated that the experimental variables considered (exposure concentration, exposure duration, and their interaction) affected significantly biomarker responses. In addition, the two-way ANOVA analysis indicated that GLY was able to affect most of the cellular parameters measured, whereas antioxidant enzyme activities resulted to be influenced moderately. Interestingly, exposure to GLY reduced significantly acetylcholinesterase activity in gills. Although preliminary, the results of this study demonstrated that GLY can affect both cellular and biochemical parameters in mussels, highlighting a potential risk for aquatic invertebrates.

Microplastic has been ubiquitously detected in freshwater ecosystems. A variety of freshwater organisms were shown to ingest microplastic particles, while a high potential for adverse effects are expected. However, studies addressing the effect of microplastic in freshwater species are still scarce compared to studies on marine organisms. In order to gain further insights into possible adverse effects of microplastic particles on freshwater invertebrates and to set the base for further experiments we exposed the mud snail (Potampoyrgus antipodarum) to a large range of common and environmentally relevant non-buoyant polymers (polyamide, polyethylene terephthalate, polycarbonate, polystyrene, polyvinylchloride). The impact of these polymers was tested by performing two exposure experiments with irregular shaped microplastic particles with a broad size distribution in a low (30%) and a high microplastic dose (70%) in the food.First, possible effects on adult P. antipodarum were assessed by morphological and life-history parameters. Second, the effect of the same mixture on the development of juvenile P. antipodarum until maturity was analyzed.Adult P. antipodarum showed no morphological changes after the exposure to the microplastic particles, even if supplied in a high dose. Moreover, although P. antipodarum is an established model organism and reacts especially sensitive to endocrine active substances no effects on embryogenesis were detected. Similarly, the juvenile development until maturity was not affected.Considering, that most studies showing effects on marine and freshwater invertebrates mostly exposed their experimental organisms to very small (≤20 μm) polystyrene microbeads, we anticipate that these effects may be highly dependent on the chemical composition of the polymer itself and the size and shape of the particles. Therefore, more studies are necessary to enable the identification of harmful synthetic polymers as some of them may be problematic and should be declared as hazardous whereas others may have relatively moderate or no effects.

Information on antibiotic use was collected through interviewing 29 farmers, and the concentrations of the most commonly used antibiotics, oxytetracycline (OTC) and enrofloxacin (ENR), were monitored in river water and sediment samples. Moreover, we assessed the toxicity of OTC and ENR on tropical freshwater invertebrates and performed a risk assessment for aquatic ecosystems. All interviewed tilapia farmers reported to routinely use antibiotics. Peak water concentrations for OTC and ENR were 49 and 1.6 μg/L, respectively. Antibiotics were most frequently detected in sediments with concentrations up to 6908 μg/kg d.w. for OTC, and 2339 μg/kg d.w. for ENR. The results of this study indicate insignificant short-term risks for primary producers and invertebrates, but suggest that the studied aquaculture farms constitute an important source of antibiotic pollution.

The goal of this study was to investigate the effects of ambient concentrations of pesticides combined with abiotic factors on the key aquatic species Daphnia magna, Chydorus sphaericus and Asellus aquaticus by means of 21 days field exposure experiments. In situ bioassays were deployed in ditches around flower bulb fields during spring and autumn 2011–2012. The results showed that phosphate was the most variable parameter followed by pesticides expressed as toxic units, as the main factors explaining differences between sites. Variation in reproduction and growth of cladoceran D. magna was largely explained by nutrients, whereas dissolved oxygen contributed mostly to variations in reproduction of C. sphaericus. Dissolved organic carbon contributed to variations in growth of the detrivore A. aquaticus. It is concluded that abiotic stressors rather than pesticides contributed significantly to the performance of aquatic invertebrates.

The São Domingos mine (Portugal) is, potentially, a good site for ecotoxicological studies, due to a pH and metal gradient of acid mine drainage. In this study, the toxicity of several mine sediments was evaluated using the aquatic oligochaete Lumbriculus variegatus as a test organism. Our hypothesis was that exposure to contaminated sediments would cause behavioural early warning responses in L. variegatus. Five sites, with pH ranging from 2.5 to 6.5, and with associated metals, were investigated. The results showed poor sediment quality in most of the collected sediments and Fe, S and As were the dominant elements in the samples. High mortalities were observed, ranging from 32.6 to 100%, indicating severe contamination. The collected sediments did not support good L. variegatus growth and significantly changed its behaviour. Early warning responses consisted of decreased locomotion and decreased peristaltic movements. A behaviour inhibition will affect the ecosystem balance by limiting the organisms' ability to avoid capture, which leads to a higher risk of predation. Behavioural responses of the aquatic oligochaeta Lumbriculus variegatus may be used to detect early warning responses.
