Organohalogen compounds in blubber of Indo-Pacific bottlenose dolphin (Tursiops aduncus) and spinner dolphin (Stenella longirostris) from Zanzibar, Tanzania

Blubber samples of Indo-Pacific bottlenose (Tursiops aduncus) and spinner (Stenella longirostris) dolphins from Zanzibar, East Africa, were analyzed for a wide range of organohalogen compounds. Methoxylated polybrominated diphenyl ethers (MeO-BDEs), presumably biogenic, were found at higher concentrations than anthropogenic organochlorine pesticides (OCPs). Only traces of industrial pollutants, such as polychlorinated biphenyls, were detected. The OCP levels found off Zanzibar were lower than those reported from other regions while MeO-BDE levels were higher. The relative composition of the OCPs indicated recent use of lindane (γ-hexachlorocyclohexane) and aged residues of DDT and technical HCH. Placental transfer was estimated to 2.5% and 0.5% of the total burden of OCPs and MeO-BDEs, respectively. Overall transfer from mother to calf in Indo-Pacific bottlenose dolphins was estimated to 72% and 85% for the OCPs and MeO-BDEs burdens, respectively. Health effects of MeO-BDEs are not known, but structural similarities with well-known environmental toxins are cause for concern.

Environmental water quality guidelines often work under the assumption that the toxicity of environmental pollutants is identical when present in isolation or in a complex chemical mixture. Thus, there is a crucial gap in our knowledge regarding how these toxicants interact and alter the toxicological effects in aquatic organisms. The present study examined the effects of acute (72-hr) aqueous exposures of Cadmium (Cd), a highly toxic non-essential trace metal, and Benzo[a]Pyrene (B[a]P), a prototypical polycyclic aromatic hydrocarbon (PAH) in adult zebrafish. Following a range-finding series of individual single-toxicant exposures, a second series was carried out using select concentrations in binary mixture exposures (using 5.8 or 22 μg/L for Cd; 0.44 or 1.07 μg/L for B[a]P). Our results demonstrated that tissue accumulation of both toxicants increased significantly in the presence of the second toxicant relative to single-toxicant exposures. Cd-only and B[a]P-only single toxicant exposures caused a significant downregulation of cytochrome p4501a (CYP1A1) and metallothionein-2 (MT2) mRNA in the gills, respectively, however binary co-exposures using both toxicants resulted in strong up-regulation of CYP1A1 and MT2. Additionally, co-exposures caused a strong induction of SOD1 and CAT mRNA transcript levels in the gill. The observed increase in body burden and transcript modulation did not translate into additive or more-than-additive toxic effects (oxidative stress) in zebrafish.

Over the past decade, microscopic plastic debris, known as microplastics, emerged as a contaminant of concern in marine and freshwater ecosystems. Although regularly detected in aquatic environments, the toxicity of those synthetic particles is not well understood. To address this, we investigated whether the exposure to microplastics adversely affects the amphipod Gammarus pulex, a key freshwater invertebrate.Juvenile (6–9 mm) and adult (12–17 mm) individuals were exposed to irregular, fluorescent polyethylene terephthalate fragments (PET, 10–150 μm; 0.8–4,000 particles mL⁻¹) for 24 h. Results show that body burden after 24 h depends on the dose and age of G. pulex with juveniles ingesting more microplastics than adults. After chronic exposure over 48 d, microplastics did not significantly affect survival, development (molting), metabolism (glycogen, lipid storage) and feeding activity of G. pulex.This demonstrates that even high concentrations of PET particles did not negatively interfere with the analyzed endpoints. These results contradict previous research on marine crustaceans. Differences may result from variations in the exposure regimes (e.g., duration, particle concentrations), plastic characteristics (e.g., type, size, shape, additives) as well as the species-specific morphological, physiological and behavioral traits. As a detritivorous shredder G. pulex is adapted to feed on non-digestible materials and might, therefore, be less sensitive towards exposure to synthetic particles. Accordingly, we argue that the autecology needs to be taken into account and that research should focus on identifying traits that render species susceptible to microplastic exposure.

Baseline characterizations of estuarine sediments in Chukchi and Beaufort Seas, were conducted. Concentrations of 194 organic and elemental chemicals were analyzed in sediment and fish, plus stable isotopes of carbon and nitrogen. The estuaries are shallow embayments, with little shoreline relief. The water columns were turbid, high salinity, and not stratified. Concentrations of arsenic and nickel were elevated throughout the region. Arsenic in fish tissue was elevated. Concentrations of PAHs were relatively high for pristine locations, but did not include petroleum hydrocarbons. Characteristics of PAHs indicate large contributions of terrestrial organic matter. With the exception of Peard Bay, all the estuaries reflected the strong influence of terrestrial plant input with low δₒ/ₒₒ values for carbon and nitrogen. Chlorinated pesticides and PCBs were uniformly low, but detectable in fish tissue. PCB and cyclodiene concentrations were half that seen in southeast Bristol Bay. Hexachlorobenzene was detected in all fish samples.

Although polychlorinated biphenyls (PCBs) have been banned in many countries for more than three decades, exposures to PCBs continue to be of concern due to their long half-lives and carcinogenic effects. In National Institute for Occupational Safety and Health studies, we are using semiquantitative plant-specific job exposure matrices (JEMs) to estimate historical PCB exposures for workers (n = 24,865) exposed to PCBs from 1938 to 1978 at three capacitor manufacturing plants. A subcohort of these workers (n = 410) employed in two of these plants had serum PCB concentrations measured at up to four times between 1976 and 1989. Our objectives were to evaluate the strength of association between an individual worker’s measured serum PCB levels and the same worker’s cumulative exposure estimated through 1977 with the (1) JEM and (2) duration of employment, and to calculate the explained variance the JEM provides for serum PCB levels using (3) simple linear regression. Consistent strong and statistically significant associations were observed between the cumulative exposures estimated with the JEM and serum PCB concentrations for all years. The strength of association between duration of employment and serum PCBs was good for highly chlorinated (Aroclor 1254/HPCB) but not less chlorinated (Aroclor 1242/LPCB) PCBs. In the simple regression models, cumulative occupational exposure estimated using the JEMs explained 14–24 % of the variance of the Aroclor 1242/LPCB and 22–39 % for Aroclor 1254/HPCB serum concentrations. We regard the cumulative exposure estimated with the JEM as a better estimate of PCB body burdens than serum concentrations quantified as Aroclor 1242/LPCB and Aroclor 1254/HPCB.
