Cadmium (Cd) is a potentially hazardous element with substantial biological toxicity, adversely affecting plant growth and physiological metabolism. Therefore, it is necessary to explore practical and environment-friendly approaches to reduce toxicity. Jasmonic acid (JA) is an endogenous growth regulator which helps plants defend against biological and abiotic stresses. To determine how JA help relieve Cd toxicity in rice, both laboratory and field experiments were implemented. In the seedling stage, the role of JA in mediating rice Cd tolerance was investigated via a fluorescent probe in vivo localization, Fourier Transform Infrared Spectroscopy (FTIR), and colorimetry. At the mature growth stage of rice, field experiments were implemented to research the effects of JA on the Cd uptake and translocation in rice. In the seedling stage of rice, we found that JA application increased the cell wall compartmentalization of Cd by promoting the Cd combination on chelated-soluble pectin of rice roots and inhibited Cd movement into protoplasts, thereby reducing the Cd content in the roots by 30.5% and in the shoots by 53.3%, respectively. Application of JA reduced H₂O₂ content and helped relieve Cd-induced peroxidation damage of membrane lipid by increasing the level of catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), and glutathione (GSH), but had no significant effect on the superoxide dismutase (SOD) activity. Additionally, field experiments showed that foliar spraying of JA inhibited rice Cd transport from the stalk and root to the grain and reduced Cd concentration in grain by 29.7% in the high-Cd fields and 28.0% in the low-Cd fields. These results improve our understanding of how JA contributes to resistance against Cd toxicity in rice plants and reduces the accumulation of Cd in rice kernels.

Antimony (Sb) and arsenic (As) are chemical analogs, but their behaviors in plants are different. To investigate the Sb uptake, translocation and speciation in As-hyperaccumulator P. cretica, a hydroponic experiment was conducted. In this study, P. cretica was exposed to 0.2-strength Hoagland nutrient solution, which contained 0.5 or 5 mg/L antimonite (SbIII) or antimonate (SbV). After 14 d exposure, P. cretica took up 1.4–2.8 times more SbIII than SbV. Since P. cretica was unable to translocate Sb, its roots accumulated >97% Sb with the highest at 7965 mg/kg. In both SbIII and SbV treatments, SbIII was the predominant species in P. cretica, with 90–100% and 46–100% SbIII in the roots. As the first barrier against Sb to enter plant cells, more Sb was accumulated in cell wall than cytosol or organelles. The results suggest that P. cretica may detoxify Sb by reducing SbV to SbIII and immobilizing it in root cell walls. Besides, the presence of SbIII significantly reduced the concentrations of dissolved organic C including organic acids in P. cretica root exudates. Further, increasing Sb levels promoted P accumulation in the plant, especially in the fronds, which may help P. cretica growth. The information from this study shed light on metabolic transformation of Sb in As-hyperaccumulators P. cretica, which helps to better understand Sb uptake and detoxification by plants.

The accumulation of arsenic in crop plants has become a worldwide concern that affects millions of people. The major source of arsenic in crop plants is irrigation water and soil. In this study, Serendipita indica, an endophytic fungus, was used to investigate the protection against arsenic and its accumulation in the tomato plant. We found that inoculation of S. indica recovers seed germination, plant growth and improves overall plant health under arsenic stress. A hyper-colonization of fungus in the plant root was observed under arsenic stress, which results in reduced oxidative stress via modulation of antioxidative enzymes, glutathione, and proline levels. Furthermore, fungal colonization restricts arsenic mobilization from root to shoot and fruit by accumulating it exclusively in the root. We observed that fungal colonization enhances the arsenic bioaccumulation factor 1.48 times in root and reduces the arsenic translocation factor by 2.96 times from root to shoot and 13.6 times from root to fruit compared to non colonized plants. Further, investigation suggests that S. indica can tolerate arsenic by immobilizing it on the cell wall and accumulating it in the vacuole. This study shows that S. indica may be helpful for the reduction of arsenic accumulation in crops grown in arsenic-contaminated agriculture fields.

Selenium engineered nanomaterials (Se ENMs)-enabled agriculture has developed rapidly, however, the roles of surface charge in the bioavailability and enrichment efficiency of Se ENMs are still unknown. Herein, various Se ENMs of homogenous size (40–60 nm) and different surface charges (3.2 ± 0.7, −29.0 ± 0.4, and 45.5 ± 1.3 mV) were prepared to explore the Se content and nutritional quality in Brassica chinensis L. The results demonstrated that soil application of various Se ENMs (0.05 mg kg⁻¹) displayed different bio-availabilities via modulating the secretion of root exudates (e.g., tartaric, malic, and citric acids), microbial community composition (e.g., Flavobacterium, Pseudomonas, Paracoccus, Bacillus and Rhizobium) and root cell wall. Negatively charged Se ENMs (Se (−)) showed the highest Se content in the shoot of B. chinensis (3.7-folds). Se (−) also significantly increased yield (156.9%) and improved nutritional quality (e.g., ascorbic acid, amino acids, flavonoids, fatty acids, and tricarboxylic acid) of B. chinensis. Moreover, after harvest, the Se (−) did not lead to significant change in Se residue in soil, but the amount of Se residue in soil was increased by 5.5% after applying the traditional Se fertilizer (selenite). Therefore, this study provides useful information for producing Se-fortified agricultural products, while minimizing environmental risk.

Root cell wall (RCW) modification is a widespread important defense strategy of plant to cope with trace metals. However, mechanisms underlying its remolding in cadmium (Cd) accumulation are still lacking in hyperaccumulators. In this study, changes of RCW structures and components between nonhyperaccumulating ecotype (NHE) and hyperaccumulating ecotype (HE) of Sedum alfredii were investigated simultaneously. Under 25 μM Cd treatment, RCW thickness of NHE is nearly 2 folds than that of HE and the thickened cell wall of NHE was enriched in low-methylated pectin, leading to more Cd trapped in roots tightly. In the opposite, large amounts of high-methylated pectin were assembled around RCW of HE with Cd supply, in this way, HE S. alfredii decreased its root fixation of Cd and enhanced Cd migration into xylem. TEM and AFM results further confirmed that thickened cell wall was caused by the increased amounts of cellulose and lignin while root tip lignification was resulted from variations of sinapyl (S) and guaiacyl (G) monomers. Overall, thickened cell wall and methylated pectin have synchronicity in spatial location of roots, and their coordination contributed to Cd accumulation in S. alfredii.

Plant leaf cuticles play a critical role in the accumulation and transport of atmospheric polycyclic aromatic hydrocarbons (PAHs). The relationship between the distribution and retained amount of PAHs on the leaf cuticles and the leaves micro-zone structures is still unclear. In this study, a confocal microscopic fluorescence spectral analysis (CMFSA) system with a spatial resolution of 200 nm was established as a direct and noninvasive means to determine the microscopic distribution and quantify the retained amount of benzo[a]pyrene (B[a]P) at Aegiceras corniculatum (Ac), Kandelia obovata (Ko) and Avicennia marina (Am) leaf cuticle micro-zones (0.096 mm²). The linear ranges for the established method were 10–1900 ng spot⁻¹ for Ac, 15–1700 ng spot⁻¹ for Ko and 30–1800 ng spot⁻¹ for Am, and the detection limits were 0.06 ng spot⁻¹ for Ac, 0.06 ng spot⁻¹ for Ko and 0.07 ng spot⁻¹ for Am. Notably, B[a]P formed clusters and unevenly distributed at the leaf cuticles. On the adaxial cuticles, B[a]P was mainly accumulated unevenly along the epidermis cell wall, and it was also distinctively distributed in the secretory cells around salt glands for Ac and Am. On the abaxial leaf cuticles, B[a]P was concentrated in the salt glands and stomata apart from being unevenly distributed in the epidermis cell wall. Moreover, the amount of B[a]P retained presented a negative correlation with the polarity of leaf cuticles, which resulted in the amount of B[a]P retained on the adaxial leaf cuticles being significantly higher than that on abaxial leaf cuticles. Our results provide a potential in situ method for investigating the distribution and retained amount of PAHs at plant leaf surface micro-zones, which would contribute to further studying and understanding the mechanism of migration and transformation of PAHs by plant leaves from a microscopic perspective.

Though the interaction between humic acid (HA) and heavy metals has been widely reported, the effects of HA on the toxicity of heavy metals to plants are still in debate. In this study, the regulation mechanisms of HA on Pb stress in tea plant (Camellia sinensis L.) was investigated through hydroponic experiments, and the experimental results were explained by using transmission electron microscope (TEM), scanning transmission X-ray microscopes (STXM) and isobaric tags for relative and absolute quantitation (iTRAQ) differential proteomics. Significant alleviation of Pb stress was found with HA coexistence. TEM results showed that HA greatly mitigated the damage of cells caused by Pb stress. Compared with sole Pb treatment, the addition of HA increased the contents of pectin and pectic acid in the cell wall by 10.5% and 30.5%, while arabinose (Ara) and galactose (Gal) decreased by 20.5% and 15.9%, respectively, which were beneficial for increasing Pb adsorption capacity of the cell wall and promoting cell elongation. Moreover, iTRAQ differential proteomics analysis proved that HA strengthened the antioxidant system, promoted the synthesis of cell wall, and stabilized protein and sulfur-containing substance metabolism in molecular level. Notably, the concentration of calcium (Ca) in the cell wall of HA coexistence treatment was 47.4% higher than Pb treatment. STXM results also indicated that the distribution of Ca in the cell wall was restored with the presence of HA. This might promote the formation of the egg-box model, thus alleviating Pb stress in cells. Our results reveal the regulation mechanisms of HA on Pb detoxification in plants and provide useful information for improving the safety of agricultural products.

Cell wall acts as a major metal sink in plant roots, while a few studies focused on root cell wall binding in plants for the phytostabilization of multi-metal contaminated soils. A pot experiment was performed to characterize root cell wall properties of the mining ecotype (ME) and non-mining ecotype (NME) of Athyrium wardii (Hook.) in response to Cd and Pb. The cell wall was found to be the major sink for Cd (41.3–54.3%) and Pb (71.4–73.8%) accumulation in roots of the ME when exposed to Cd and/or Pb. The ME showed more Cd and Pb accumulation in root cell walls when exposed to Cd and Pb simultaneously, compared with those exposed to single Cd or Pb as well as the NME, suggesting some modifications for cell walls. The uronic acid contents of pectin and hemicellulose 1 (HC1) in root cell walls of the ME increased significantly when exposed to Cd and Pb simultaneously, suggesting enhanced cell wall binding capacity, thus resulting in more Cd and Pb bound to pectin and HC1. In particular, pectin was found to be the predominant binding site for Cd and Pb. Greater pectin methylesterase activity along with a lower degree of methylesterification were observed in the cell walls of the ME when exposed to Cd and Pb simultaneously. Furthermore, the ME present more O–H, N–H, C–OH, C–O–C, C–C and/or Ar–H in root cell walls when exposed to Cd and Pb simultaneously. These changes of root cell wall properties of the ME lead to enhanced cell wall binding ability in response to the co-contamination of Cd and Pb, thus could be considered a key process for enhanced Cd and Pb accumulation in roots of the ME when exposed to Cd and Pb simultaneously.

High sensitivity towards Cu toxicity is problematic when using some hyperaccumulator plants for phytoremediation of soils with mixed contamination of Cu. Sedum alfredii, a Cd/Zn co-hyperaccumulator and Pb accumulator, is widely used for remediation of Cd, Zn, and Pb co-contaminated soils in China. In this paper, the tolerance and accumulation ability of S. alfredii towards Cu stress and its potential for phytoremediation of multi-metal polluted soils have been studied. Both the hyperaccumulating ecotype (HE) and non-hyperaccumulating ecotype (NHE) of S. alfredii accumulated high Cu in the roots and translocated minimal Cu to the shoots, and Cu in the stems and leaves mostly restricted in the vascular tissues (phloem zone). The HE plants, however, exhibited high Cu resistance with stimulated lateral root growth and increased chlorophyll content under 10 μM Cu treatment. XANES analysis showed that Cu in HE roots comprised Cu²⁺ (46.7%), Cu-histidine (35.2%) and Cu-cell wall (18.1%). The NHE under Cu stress showed decreased biomass, reduced leaf chlorophyll content, altered root architecture, and higher Cu localized to root cell wall as compared with the HEs. Potted HE plants thrived six months in multi-metal contaminated soils including 3897 mg kg⁻¹ available Cu. In conclusion, HE S alfredii is highly tolerant toward Cu due to metal homeostasis in root cells. Therefore, this plant has great potential to remediate Zn, Cd, and Pb contaminated soils those also contain high levels of Cu.

The synergistic cooperation of microbial cells and their extracellular polymeric substances (EPS) in biofilms is critical for the biofilm’s resistance to heavy metals and the migration and transformation of heavy metals. However, the effects of different components of biofilms have not been fully understood. In this study, the spatial distribution and speciation of copper in the colloidal EPS, capsular EPS, cell walls and membranes, and intracellular fraction of unsaturated Pseudomonas putida (P. putida) CZ1 biofilms were fully determined at the subcellular level. It was found that 60–67% of copper was located in the extracellular fraction of biofilms, with 44.7–42.3% in the capsular EPS. In addition, there was 15.5–20.1% and 17.2–21.2% of copper found in the cell walls and membranes or the intracellular fraction, respectively. Moreover, an X-ray absorption fine structure spectra analysis revealed that copper was primarily bound by carboxyl-, phosphate-, and hydrosulfide-like ligands within the extracellular polymeric matrix, cell walls and membranes, and intracellular fraction, respectively. In addition, macromolecule quantification, fourier-transform infrared spectroscopy spectra and sulfur K-edge x-ray absorption near edge structure analysis further showed the carboxyl-rich acidic polysaccharides in EPS, phospholipids in cell walls and cell membranes, and thiol-rich intracellular proteins were involved in binding of copper in the different components of biofilm. The full understanding of the distribution and chemical species of heavy metals in biofilms not only promotes a deep understanding of the interaction mechanisms between biofilms and heavy metals, but also contributes to the development of effective biofilm-based heavy metal pollution remediation technologies.
