Identification and Quantification of Antibiotic Residues and Evaluation of Microbial Resistance to Antibiotics in Huatanay River Waters in Peru 全文

The Huatanay River in Cusco-Peru, is affected by wastewater discharges along its course. In order to evaluate this impact, we evaluate antibiotic residues and their impact on the increase of bacterial resistance in the city of Cusco treatment plant. For this purpose, water samples from the influent and effluent of the treatment plant were analyzed by chromatographic methods; additionally, sensitivity tests were performed with three bacterial strains (Escherichia coli, Salmonella sp., and Klebsiella sp.), which were isolated from the same place. Six antibiotic residues were identified (ceftriaxone, amoxicillin, trimethoprim, sulfamethoxazole, dicloxacillin, and lincomycin). Those found in the highest concentration were: amoxicillin (91495 and 0 µg/L) and lincomycin (33970 and 10800 µg/L) in the influent and effluent, respectively. There is more resistance in the effluent than the influent in the case of E. coli shows resistance in the effluent to cephalexin (30 µg) and azithromycin (15 µg). Salmonella sp. is resistant to amoxicillin (15 µg), dicloxacillin (1 µg), lincomycin (2 µg), ceftriaxone (30 µg), cephalexin (30 µg), and ciprofloxacin (5 µg). Finally, Klebsiella sp. is sensitive to ceftriaxone (30 µg), amoxicillin (15 µg), and cephalexin (30 µg). This confirms that the antibiotic residues contained in the wastewater of Cusco generate resistance in the isolated bacteria.

This review aims to gather and summarize information about the occurrence of emerging contaminants and antibiotic resistance genes in environmental matrices in Latin America. We aim to contribute to future research by compiling a list of priority pollutants adjusted to the needs and characteristics of Latin America, according to the data presented in this study. In order to perform a comprehensive research and secure a representative and unbiased amount of quality data concerning emerging contaminants in Latin America, the research was performed within the Scopus® database in a time frame from 2000 to July 2019. The countries with higher numbers of published articles were Brazil and México, while most studies were performed in the surroundings of Mexico City and in Southern and Southeastern Brazil. The main investigated environmental matrices were drinking water and surface water. The presence of antibiotic resistance was frequently reported, mainly in Brazil. Monitoring efforts should be performed in other countries in Latin America, as well as in other regions of Brazil and México. The suggested priority list for monitoring of emerging contaminants in Latin America covers: di(2-ethylhexyl) phthalate (DEHP), bisphenol-A (BP-A), 4-nonylphenol (4-NP), triclosan (TCS), estrone (E1), estradiol (E2), ethinylestradiol (EE2), tetracycline (TC), amoxicillin (AMOX), norfloxacin (NOR), ampicillin (AMP) and imipenem (IMP). We hope this list serves as a basis for the orientation of the future research and monitoring projects to better understand the distribution and concentration of the listed emerging substances.

This work reports the architecture of a novel class of membrane-supported 1D MOF hollow superstructures, by using the bio-inspired polydopamine (PDA) mediated contra-diffusion synthetic strategy, for facile and efficient separation of uranium in a flow-through mode. PDA chemistry was firstly employed to modify the inner surfaces of the cylindrical pore channels of polycarbonate track-etched membrane (PCTM), thereby regulating the heterogeneous nucleation and interfacial growth of ZIF-8 crystals. ZIF-8 hollow superstructures embedded in membrane matrix with well-defined 1D channels were obtained. These membrane-supported MOF hollow superstructures then, for the first time, served as integrated chromatographic micro-column arrays for effective entrapment of uranium from aqueous solutions. It is highlighted that the PCTM supported ZIF-8 superstructures exhibited outstanding uranium entrapment ability in both traditional batch mode (capacity 62.3 mg/g) and fast flow-through mode (removal rate over 90% for 3 level). Moreover, new insights into the interaction between ZIF-8 and uranyl ions were obtained, suggesting that an ion-exchange mechanism involved synergistic effect was responsible for uranium binding, especially in a long-term exposure. The membrane-supported 1D MOF hollow superstructures developed in this work represent a new category of organic-inorganic composite membrane. And, it is envisioned that the methodology established in this work would be versatile for preparing more MOF superstructures with deployable form for separation applications.In summary, a novel class of membrane-supported ZIF-8 hollow superstructure was fabricated for effective separation of uranyl ions.

A highly selective sample cleanup procedure combined with molecularly imprinted solid-phase extraction (MISPE) was developed for the isolation of gonyautoxins 2,3 (GTX2,3) from Alexandrium tamarense sample. The molecularly imprinted polymer microspheres (MIPMs) were prepared by suspension polymerization using caffeine as the dummy template molecule, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linker and polyvinyl alcohol as the dispersive reagent. The polymer microspheres were used as a selective sorbent for the solid-phase extraction of gonyautoxins 2,3. An off-line MISPE method followed by high-performance liquid chromatography (HPLC) with fluorescence detection for the analysis of gonyautoxins 2,3 was established. Finally, the extract samples from Alexandrium tamarense were analyzed. The results showed the imprinted polymer microspheres exhibited high affinity and selectivity for gonyautoxins 2,3. The interference matrix in the extract were obviously cleaned by MISPE and the extraction efficiency of gonyautoxins 2,3 in the sample ranged from 81.74% to 85.86%.

A comprehensive analytical procedure for a reliable identification of nontarget polar contaminants in aquatic sediments was developed, based on the application of ultra-high-pressure liquid chromatography (UHPLC) coupled to hybrid quadrupole time-of-flight mass spectrometry (QTOFMS). The procedure was applied for the analysis of freshwater sediment that was highly impacted by wastewater discharges from the pharmaceutical industry. A number of different contaminants were successfully identified owing to the high mass accuracy of the QTOFMS system, used in combination with high chromatographic resolution of UHPLC. The major compounds, identified in investigated sediment, included a series of polypropylene glycols (n = 3–16), alkylbenzene sulfonate and benzalkonium surfactants as well as a number of various pharmaceuticals (chlorthalidone, warfarin, terbinafine, torsemide, zolpidem and macrolide antibiotics). The particular advantage of the applied technique is its capability to detect less known pharmaceutical intermediates and/or transformation products, which have not been previously reported in freshwater sediments.

Element load, conventional biomarkers and altered protein expression profiles were studied in Carcinus maenas crabs, to assess contamination of “Domingo Rubio” stream, an aquatic ecosystem that receives pyritic metals, industrial contaminants, and pesticides. Lower antioxidative activities – glucose-6-phosphate and 6-phosphogluconate dehydrogenases, catalase – were found in parallel to higher levels of damaged biomolecules – malondialdehyde, oxidized glutathione –, due to oxidative lesions promoted by contaminants, as the increased levels of essential – Zn, Cu, Co – and nonessential – Cr, Ni, Cd – elements. Utility of Proteomics to assess environmental quality was confirmed, especially after considering the six proteins identified by de novo sequencing through capLC-μESI-ITMS/MS and homology search on databases. They include tripartite motif-containing protein 11 and ATF7 transcription factor (upregulated), plus CBR-NHR-218 nuclear hormone receptor, two components of the ABC transporters and aldehyde dehydrogenase (downregulated). These proteins could be used as novel potential biomarkers of the deleterious effects of pollutants present in the area.

Eels are an ideal biomonitor for persistent organic pollutants (POPs) because of their high lipid content, longevity and tendency to remain within a defined range during their freshwater life phase. This study investigated concentrations of POPs in eels (Anguilla anguilla) from 30 sites across Scotland, including polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (BDEs), DDT (and metabolites), hexachlorocyclohexanes (α, β, γ-HCH), hexachlorobenzene (HCB), hexachlorobutadiene (HCBD) and pentachlorobenzene. Despite its EU-wide ban ∼30 years ago, DDT and its derivatives were detected in almost all samples. PCB 153 and 138 were the most widely detected PCB congeners, while BDE 47 was the dominant BDE. Pentachlorobenzene was not detected, while HCBD was detected once only. α-HCH, β-HCH and HCB concentrations were very low (generally <3 μg/kg or below detection). When compared with 1986 and 1995 data, the results revealed considerable decreases in p,p′-DDE concentrations. More drastic reductions were evident for γ-HCH, reflecting the tightening restrictions on pesticide use imposed over the previous decades.

The aim of this study was to validate lichens as biomonitors of PAH atmospheric deposition; for that, an inter-comparison between the PAH profile and concentrations intercepted in lichens with those of air, soil and pine needles was performed. The study was conducted in a petro-industrial area and the results showed that PAH profiles in lichens were similar to those of the air and pine needles, but completely different from those of soils. Lichens accumulated higher PAH concentrations when compared to the other environmental compartments and its concentrations were significantly and linearly correlated with concentrations of PAHs in soil; we showed that a translation of the lichen PAHs concentrations into regulatory standards is possible, fulfilling one of the most important requirements of using lichens as biomonitors. With lichens we were then able to characterize the air PAHs profile of urban, petro-industrial and background areas. Lichen PAH concentrations can identify geographic areas that may be out of compliance with regulatory standards.

Two types of integrative sampling approaches (passive samplers and biomonitors) were tested for their sampling characteristics of selected endocrine disrupting compounds (EDCs). Chemical analyses (LC/MS/ MS) were used to determine the amounts of five EDCs (nonylphenol, bisphenol A, estrone, 17b-estradiol and 17a-ethinylestradiol) in polar organic chemical integrative samplers (POCIS) and freshwater mussels (Unio pictorum); both had been deployed in the influent and effluent of a municipal wastewater treatment plant (WWTP) in Genoa, Italy. Estrogenicity of the POCIS samples was assessed using the yeast estrogen screen (YES). Estradiol equivalent values derived from the bioassay showed a positive correlation with estradiol equivalents calculated from chemical analyses data. As expected, the amount of estrogens and EEQ values in the effluent were lower than those in the influent. Passive sampling proved to be the preferred method for assessing the presence of these compounds since employing mussels had several disadvantages both in sampling efficiency and sample analyses.

Inorganic arsenic is a chronic exposure carcinogen. Analysis of UK baby rice revealed a median inorganic arsenic content (n = 17) of 0.11 mg/kg. By plotting inorganic arsenic against total arsenic, it was found that inorganic concentrations increased linearly up to 0.25 mg/kg total arsenic, then plateaued at 0.16 mg/kg at higher total arsenic concentrations. Inorganic arsenic intake by babies (4-12 months) was considered with respect to current dietary ingestion regulations. It was found that 35% of the baby rice samples analysed would be illegal for sale in China which has regulatory limit of 0.15 mg/kg inorganic arsenic. EU and US food regulations on arsenic are non-existent. When baby inorganic arsenic intake from rice was considered, median consumption (expressed as μg/kg/d) was higher than drinking water maximum exposures predicted for adults in these regions when water intake was expressed on a bodyweight basis. Median consumption of organic arsenic levels for UK babies from baby rice is above threshold considered safe.
