Identification and Quantification of Antibiotic Residues and Evaluation of Microbial Resistance to Antibiotics in Huatanay River Waters in Peru

The Huatanay River in Cusco-Peru, is affected by wastewater discharges along its course. In order to evaluate this impact, we evaluate antibiotic residues and their impact on the increase of bacterial resistance in the city of Cusco treatment plant. For this purpose, water samples from the influent and effluent of the treatment plant were analyzed by chromatographic methods; additionally, sensitivity tests were performed with three bacterial strains (Escherichia coli, Salmonella sp., and Klebsiella sp.), which were isolated from the same place. Six antibiotic residues were identified (ceftriaxone, amoxicillin, trimethoprim, sulfamethoxazole, dicloxacillin, and lincomycin). Those found in the highest concentration were: amoxicillin (91495 and 0 µg/L) and lincomycin (33970 and 10800 µg/L) in the influent and effluent, respectively. There is more resistance in the effluent than the influent in the case of E. coli shows resistance in the effluent to cephalexin (30 µg) and azithromycin (15 µg). Salmonella sp. is resistant to amoxicillin (15 µg), dicloxacillin (1 µg), lincomycin (2 µg), ceftriaxone (30 µg), cephalexin (30 µg), and ciprofloxacin (5 µg). Finally, Klebsiella sp. is sensitive to ceftriaxone (30 µg), amoxicillin (15 µg), and cephalexin (30 µg). This confirms that the antibiotic residues contained in the wastewater of Cusco generate resistance in the isolated bacteria.

In this study, we demonstrate that partial least-squares regression analysis with full cross-validation of spectral reflectance data estimates the amount of polycyclic aromatic hydrocarbons in petroleum-contaminated tropical rainforest soils. We applied the approach to 137 field-moist intact soil samples collected from three oil spill sites in Ogoniland in the Niger Delta province (5.317°N, 6.467°E), Nigeria. We used sequential ultrasonic solvent extraction–gas chromatography as the reference chemical method. We took soil diffuse reflectance spectra with a mobile fibre-optic visible and near-infrared spectrophotometer (350–2500 nm). Independent validation of combined data from studied sites showed reasonable prediction precision (root-mean-square error of prediction = 1.16–1.95 mg kg−1, ratio of prediction deviation = 1.86–3.12, and validation r2 = 0.77–0.89). This suggests that the methodology may be useful for rapid assessment of the spatial variability of polycyclic aromatic hydrocarbons in petroleum-contaminated soils in the Niger Delta to inform risk assessment and remediation.

Microcystin-LR (MC-LR), one of the most common cyanotoxins, is produced by harmful cyanobacteria. The current study focuses on the photosensitized transformation of MC-LR in dissolved organic matter (DOM) enriched solutions under solar simulated irradiation. It appears that the direct energy transfer of triplet excited state DOM with MC-LR plays a key role and leads to photosensitized isomerization of Adda side chain. Furthermore a micro-heterogeneous mechanism has been proposed. Size exclude chromatograph (SEC) has been applied to explore the adsorption of MC-LR on the DOM. The adsorption phenomenon supported the fact that the pseudo first-order photodegradation rates showed positive correlation with the adsorption. The photo-transformation rate of MC-LR increases as pH decreases which is also the result of the adsorptive interaction of MC-LR with DOM. Finally two bulk water parameters (TOC and UV350 nm) have been applied to predict the photodegradation rates of MC-LR in the varied water matrixes.

Selective supercritical fluid extraction (SSFE) was used as a measurement of compound chemical accessibility and as a predictor of compound bioavailability from three natural soils and artificial analogues prepared to have comparable total organic carbon content. Soils spiked with phenanthrene, pyrene, PCB 153, lindane, and p,p′-DDT were aged for 0, 14, 28, or 56 days and then selectively extracted by supercritical fluid extraction. Compounds exhibited decreasing extractability with increasing pollutant–soil contact time and increasing total organic carbon content in tested soils. However, the different extractability of compounds from artificial and natural pairs having comparable TOC indicates the limitations of using TOC as an extrapolation basis between various soils. The comparison of extractability with bioaccumulation by earthworms (Eisenia fetida) previously published by Vlčková and Hofman (2012) showed that only for PAHs it was possible to predict their bioaccumulation by means of selective SFE.

Chlorinated paraffins (CPs) were found in the biodegradable fraction of source separated waste from Uppsala, Sweden. We identified and quantified the CPs by multivariate evaluation of gas chromatography-electron capture detection chromatograms. Using principal component analyses (PCA) we identified different types of CP-formulations and also obtain quantitative data. PCA yielded better identifications of individual CP-formulations than visual comparison of chromatograms. Partial least squares regression gave good calibration curves of the standards, but did not work for the waste samples. No source of CPs could be identified in the waste collection chain, and as the waste samples seemed to contain at least two different CP-formulations the source was probably to be found in the waste material itself. The method was used to determine CPs in additional environmental samples, demonstrating that multivariate methods may be developed into a powerful tool for identification and quantification of complex mixture.

Hexabromocyclododecanes (HBCDs) have been considered candidate persistent organic pollutants, however, environmental data on their presence in China, a major world market, are limited. In this study, airborne particle-bound HBCDs in Shanghai, China were quantified with liquid chromatography–tandem mass spectrometry, and their total concentrations varied from 3.21 to 123 pg/m³. The industrial area showed about three times higher HBCD levels than the urban areas. Gamma-HBCD dominated in particle-associated HBCDs in the industrial area, while α-HBCD was the major diastereomer of the urban areas, which is quite different from the pattern of technical products. Besides α-, β- and γ-HBCDs, δ-HBCD was also detected in all samples (0.09–6.31 pg/m³), while no ɛ-HBCD was found. Airborne β- and γ-HBCDs were racemic in most studied areas due to their chiral signatures showing no significant difference from commercial mixture. Fractions of (+)-α-HBCD (0.417–0.467), however, suggested the enrichment of (−)-α-HBCD and the involvement of enantioselective bioprocesses.

A comprehensive analytical procedure for a reliable identification of nontarget polar contaminants in aquatic sediments was developed, based on the application of ultra-high-pressure liquid chromatography (UHPLC) coupled to hybrid quadrupole time-of-flight mass spectrometry (QTOFMS). The procedure was applied for the analysis of freshwater sediment that was highly impacted by wastewater discharges from the pharmaceutical industry. A number of different contaminants were successfully identified owing to the high mass accuracy of the QTOFMS system, used in combination with high chromatographic resolution of UHPLC. The major compounds, identified in investigated sediment, included a series of polypropylene glycols (n = 3–16), alkylbenzene sulfonate and benzalkonium surfactants as well as a number of various pharmaceuticals (chlorthalidone, warfarin, terbinafine, torsemide, zolpidem and macrolide antibiotics). The particular advantage of the applied technique is its capability to detect less known pharmaceutical intermediates and/or transformation products, which have not been previously reported in freshwater sediments.

Metabolic profiling in plants can be used to differentiate between treatments and to search for biomarkers for exposure. A methodology for processing Ultra-High-Performance Liquid Chromatography–Diode-Array-Detection data is devised. This methodology includes a scheme for selecting informative wavelengths, baseline removal, retention time alignment, selection of relevant retention times, and principal component analysis (PCA). Plant crude extracts from rapeseed seedling exposed to sublethal concentrations of glyphosate are used as a study case. Through this approach, plants exposed to concentrations down to 5 μM could be distinguished from the controls. The compounds responsible for this differentiation were partially identified and were different from those specific for high exposure samples, which suggests that two different responses to glyphosate are elicited in rapeseed depending on the level of exposure. The PCA loadings indicate that a combination of other metabolites could be more sensitive than the response of shikimate to detect glyphosate exposure.

Four fluoroquinolone antibiotics (norfloxacin, ciprofloxacin, lomefloxacin, and enrofloxacin) in tap water in Guangzhou and Macao were analyzed using high performance liquid chromatography fluorescence detection. The results showed that all target antibiotics were detected in high rate both in Guangzhou (77.5%) and Macao (100%), ranging from 1.0 to 679.7 ng/L (SD ≤ 37.6) in Guangzhou, and from 2.0 to 37.0 ng/L (SD ≤ 2.5) in Macao. The fluoroquinolone antibiotics pollution in tap water widely distributes in Guangzhou and Macao. In addition, the effect of rainfall on concentration of fluoroquinolone antibiotics in south China was also investigated. Our result indicates that the antibiotic concentration in tap water in Guangzhou tends to obviously reduce at the beginning of rainy season, even decreases below the limit of quantification immediately. Thus, it was clarified that the heavy rain in south China has the function of reducing the fluoroquinolone antibiotics concentrations in tap water.

Eels are an ideal biomonitor for persistent organic pollutants (POPs) because of their high lipid content, longevity and tendency to remain within a defined range during their freshwater life phase. This study investigated concentrations of POPs in eels (Anguilla anguilla) from 30 sites across Scotland, including polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (BDEs), DDT (and metabolites), hexachlorocyclohexanes (α, β, γ-HCH), hexachlorobenzene (HCB), hexachlorobutadiene (HCBD) and pentachlorobenzene. Despite its EU-wide ban ∼30 years ago, DDT and its derivatives were detected in almost all samples. PCB 153 and 138 were the most widely detected PCB congeners, while BDE 47 was the dominant BDE. Pentachlorobenzene was not detected, while HCBD was detected once only. α-HCH, β-HCH and HCB concentrations were very low (generally <3 μg/kg or below detection). When compared with 1986 and 1995 data, the results revealed considerable decreases in p,p′-DDE concentrations. More drastic reductions were evident for γ-HCH, reflecting the tightening restrictions on pesticide use imposed over the previous decades.
