Bioactivity and safety evaluations of Cupressus sempervirens essential oil, its nanoemulsion and main terpenes against Culex quinquefasciatus Say

The essential oil (EO) of Cupressus sempervirens was obtained by hydrodistillation and analysed using gas chromatography–flame ionization detection (GC–FID) and gas chromatography–mass spectrometry (GC–MS). Two monoterpenes, α-pinene (49.1%) and δ-3-carene (21.4%), and one sesquiterpene hydrocarbon, α-cedrol (5.1%), were isolated as the EO major terpenes. An oil-in-water nanoemulsion (particle size 71.2 nm) was produced from the EO through a low-energy method. The EO, its nanoemulsion and its main constituents showed mosquitocidal and biochemical effects against Culex quinquefasciatus Say, the common vector of lymphatic filariasis parasites. All treatments showed dose-dependent bioactivity, and adults were more susceptible to the EO products than the larvae. The nanoemulsion showed superior activity, followed by the crude EO and α-cedrol. At 40 μg/ml, the nanoemulsion caused 100% larval mortality, while the EO and α-cedrol required twice this concentration to achieve the same larval mortality. The LC₅₀ values were 8.4, 16.1, 15.1, 30.7 and 53.4 μg/ml at 24 h after exposure for the nanoemulsion, crude oil, α-cedrol, δ-3-carene and α-pinene, respectively. For adults, 20.0 μl/l nanoemulsion caused 100% mortality, while twice this concentration of the EO was required to achieve the same effect. The LC₅₀’s against adults ranged between 6.2 and 40.4 μl/l. EO products prominently repelled mosquitoes at concentrations between 0.75 and 6.0 μl/cm². The EO products caused remarkable inhibition of Cx. quinquefasciatus acetylcholinesterase activity but were safer towards the non-target aquatic species Gambusia affinis. These results recommend the use of C. sempervirens EO, its nanoemulsion and main terpenes as natural tools to control Cx. quinquefasciatus.

We investigated the accumulation of Zn, Cu, Pb, and Cd in the soil and the leaves and bark of five common tree species (Eucalyptus occidentalis Endl., Acacia salicina Lindl., Cupressus sempervirens L., Casuarina equisetifolia L., and Tamarix aphylla (L.) Karst.) in the city of Gabès Tunisia to elucidate their bioaccumulation potential and determine their usefulness as biomonitors of metallic pollution in arid urban areas. Our results indicated that the bark had higher mean concentrations of Pb and Cd than leaves. In contrast, the leaves had higher mean concentrations of Zn and Cu than bark. No hyperaccumulation was detected for any of the analyzed metals in any of the studied species. E. occidentalis and T. aphylla had the highest mean concentrations of the investigated metals in leaves and bark. Based on the calculated metal accumulation index (MAI) values, these two species accumulated more metals than other studied tree species. Likewise, the concentrations of Zn, Cu, Pb, and Cd in soil had significant positive correlations with that in leaves and bark. Accordingly, E. occidentalis could be used for biomonitoring in arid areas subjected to industrial and traffic pollution. T. aphylla would be a good alternative when native species are a priority.

PURPOSE: The aim of this work was to investigate the possibility of using several bush and arboreal plant species, usually present as ornamental plants in street and parks, as environmental indicators of pollution. This is a research paper that evaluates the real possibility of using a fast and low-cost procedure to evaluate the pollution degree through data obtained from plant species growing within an urban environment. METHODS: Leaves of six different bush and arboreal species were collected from different parts of Madrid (Spain), ranging from highly polluted considered areas to medium and low contaminated ones. A total of 66 chemical elements, from major to minor and trace, were determined for every leaf sample by inductively coupled plasma-mass spectrometry. Statistical analyses were carried out using mainly box and whisker plots, linear discriminant analysis and cluster analysis. RESULTS: The pollution by different elements of the studied areas of Madrid cannot be considered generally dangerous for human health. The level detected for the contaminants, in general, is similar or lower than other urban cities. Pb and V concentrations in plant samples tend to increase as traffic density increases. The different studied plant species showed a different capability of accumulation of certain elements. Cedrus deodara accumulates specially Ag, Hg, Mo and V; Cupressus sempervirens, Zr; Pinus pinea, As and Sb; Nerium oleander Ni, Pb, Mo and Se; Ligustrum ovalifolium, Sc and V; and Pittosporum tobira, Ag, Cd, Rb and Sc. CONCLUSIONS: The leaves and needles collected from bush and arboreal plants common in this city have demonstrated to be useful to evaluate the level of pollution not only through the chemical analysis but also through the recognition of the visual injury symptoms. The application of multivariate statistical techniques combined with determining of element concentration and correlation analysis has been proved to be an effective tool for reach the objectives of the present work. This allows visualising quickly the damages and leading the sampling through the points of high-level pollution, saving analysis, time and money.
