Phthalate ester pollution in the environment and food chain is frequently reported. Microbial treatment is a green and efficient method for solving this problem. The isolation and systematic investigation of microorganisms generally recognized as safe (GRAS) will provide useful resources. A GRAS Bacillus subtilis strain, BJQ0005, was isolated from Baijiu fermentation starter and efficiently degraded phthalate esters (PAEs). The half-lives for di-isobutyl phthalate, di-butyl phthalate and di-(2-ethylhexyl) phthalate were 3.93, 4.28, and 25.49 h, respectively, from the initial amount of 10 mg per 10 mL reaction mixture, which are records using wild-type strains. Genome sequencing and metabolic intermediate analysis generated the whole metabolic pathway. Eighteen enzymes from the α/β hydrolase family were expressed. Enzymes GTW28_09400 and GTW28_13725 were capable of single ester bond hydrolysis of PAEs, while GTW28_17760 hydrolyzed di-ester bonds of PAEs. Using molecular docking, a possible mechanism affecting enzymatic ester bond hydrolysis of mono-butyl phthalate was proposed of GTW28_17760. The carboxyl group generated by the first hydrolysis step interacted with histidine in the catalytic active center, which negatively affected enzymatic hydrolysis. Isolation and systematic investigation of the PAE degradation characteristics of B. subtilis will promote the green and safe treatment of PAEs in the environment and food industry.

The role of exposure time on wetland sediment-bound phosphorus (P) biogeochemical behavior is studied in Lake Poyang after the operation of the Three Gorges Dam (TGD). The multiple P compounds primarily include orth–P (88.3%), mono–P (8.9%), DNA–P (2.1%), and pyro–P (0.8%) in the exposed sediments. A significant decreasing trend of orth–P occurred after the operation of the Three Gorges Dam (TGD), with the mean concentration decreasing from 175.9 to 142.5 mg kg⁻¹ from 2007 to 2012 (ANOVA: P < 0.05), whereas the temporal change in biogenic P showed great variability. The plant distribution pattern and the increase in plant biomass due to decreased water levels might be the reason that caused variations in the P species. Furthermore, the content of orth–P, mono–P, DNA–P, and pyro–P showed increasing trends as sediment exposure time increased. However, the enzyme hydrolysis rate of DNA–P decreased with exposure time and may cause the bioavailability of biogenic P to decrease. Despite the fact that the bioavailability of biogenic P might decline in the short term, the favorable environmental conditions for P release in sediment rewetting processes, together with the increase in orth–P and biogenic P due to extended exposure time, indicate that these large additions of P would enter the overlying water and cause water quality decline once the sediment is submerged underwater during the next wet season. An environmental process analysis showed that the increased exposure time induced sediment environmental conditions changes that played an important role in the biogeochemical cycle of P and may be an important way of P replenishment in Lake Poyang. The results of this study help provide a better understanding of the role of sediment drying/wetting cycles in nutrient biogeochemical behavior and fates in wetland ecosystems.

This study aims to establish the relative importance of sediment organic phosphorus (Po) to the total P and the major classes of organic molecules that contribute to sediment Po, determined by measuring their susceptibility to enzymatic hydrolysis, across a suite of lakes ranging from oligotrophic to eutrophic status. The results showed that Po accounted for 21–60% of total P, and bioavailable Po accounted for 9–34% of Po in the sediments. The bioavailable Po includes mainly labile (H2O-Po) and moderately labile (NaOH-Po) P forms. For H2O-Po (accounting for only1.4% of Po), 53% (average) was labile monoester P, 28% was diester P and 17% was phytate-like P. For NaOH-Po (accounting for 9–33% of Po), 32% was labile monoester P, 33% was phytate-like P and 18% was diester P. The composition of bioavailable Po, determined by enzyme assays, was related to the lake nutrient levels, which implies that sediment bioavailable Po could act as an effective indicator for lake eutrophic status. With the increase of lake nutrient levels, bioavailable Po content and alkaline phosphatase activity in the sediment all increased, indicating that Po represents an important and bioavailable source of P that increases with eutrophication, and could contribute to internal loading and resistance of eutrophic lakes to remediation. This implies that eutrophic lakes would maintain long-term eutrophic status and algal bloom phenomena even after the external input of P was controlled and the total P concentration of water has declined. Thus, in order to reduce the release risk of sediment P more efficiently and effectively, sediment P control technique should focus not only on reducing the total P and inorganic P, but should also pay close attention to the removal of bioavailable Po.

A high demand exists in bisphenols (BPs) screening studies for quick, reliable and straightforward analytical methods that generate data faster and simultaneously. Herein, we describe a combination of enzymatic probe sonication (EPS) and ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) for quick extraction and simultaneous quantification of eight important BPs in animal-derived foods. Results obtained demonstrated that the ultrasonic probe power could not only enhance the enzymatic hydrolysis efficiency, but also accelerate the liquid-liquid extraction procedure. Under optimized EPS parameters, one sample could be exhaustively extracted within 120 s, as compared with 12 h needed for the conventional enzymatic extraction which is more suitable for high-throughput analysis. The method was successfully applied to analyze residual BPs in animal-derived foods collected from Beijing, China. Widespread occurrence of BPA, BPS, BPF, BPAF, BPP, and BPB were found, with detection frequencies of 65.2%, 42.4%, 33.7%, 29.4%, 28.3%, and 27.2%, respectively. The highest total concentration levels of BPs (sum of the eight BPs analyzed, ΣBPs) were found in chicken liver (mean 12.2 μg/kg), followed by swine liver (6.37 μg/kg), bovine muscle (3.24 μg/kg), egg (2.03 μg/kg), sheep muscle (2.03 μg/kg), chicken muscle (1.45 μg/kg), swine muscle (1.42 μg/kg), and milk (1.17 μg/kg). The estimated daily intake (EDI) of BPs, based on the mean and 95th percentile concentrations and daily food consumptions, was estimated to be 5.687 ng/kg bw/d and 22.71 ng/kg bw/d, respectively. The human health risk assessment in this work suggests that currently BPs do not pose significant risks to the consumers because the hazard index (HI) was <1.

Organic phosphorus (Pₒ) plays a very important role in the process of lake eutrophication, but there is still a lack of knowledge about the internal cycle of Pₒ in suspended particulate matter (SPM) dominated by algal debris. In this study, the characterization of bioavailable Pₒ by sequential extraction and enzymatic hydrolysis showed that 45% of extracted TP was Pₒ in SPM of Lake Dianchi, and 43–98% of total Pₒ in H₂O, NaHCO₃ and NaOH fractions was enzymatically hydrolyzable Pₒ (EHP, H₂O−EHP: 31–53%). Importantly, labile monoester P was the main organic form (68%) of EHP, and its potential bioavailability was higher than that of diester P and phytate-like P. According to the estimation of P pools in SPM of the whole lake, the total load of Pᵢ plus EHP in the H₂O extract of SPM was 74.9 t and had great potential risk to enhance eutrophication in the lake water environment. Accordingly, reducing the amount of SPM in the water during the algal blooming period is likely to be a necessary measure that can successfully interfere with or block the continuous stress of unhealthy levels of P on the aquatic ecosystem.

In this work, valorisation pathways of brewers’ spent grains (BSG) towards biofuels production under the biorefinery concept were studied utilizing experimental data that provide a common base for straightforward comparison. The dehydration and the recovery of used oil, bioethanol and biogas from BSG were studied. The process units involved were thoroughly investigated and optimized. The oil extraction efficiency reached up to 70% using solid-liquid extraction process with hexane as solvent. The optimal ethanol yield achieved was 45% after the application of acid pretreatment, enzymatic hydrolysis with CellicCTec2 and fermentation with S. Cerevisiae. As far as biogas potential is concerned, the raw BSG, defatted BSG and stillage presented values equal to 379 ± 19, 235 ± 21 and 168 ± 39 mL biogas/g for respectively. Through the combination of the proposed schemes, three biorefinery scenarios were set up able to produce biodiesel, bioethanol and/or biogas. Material flow diagrams were set up in order to assess these schemes. Given that BSG could ensure ‘green’ energy production in the range of 4.5–7.0 million MJ/y if the European BSG potential is fully valorised, BSG could substantially contribute to the biofuel energy strategy.

Phosphorus (P) is an essential nutrient element for biological growth that can contribute to eutrophication in aquatic ecosystems. Water trophic status and algae growth are primarily related to the content of bioavailable P, which is primarily related to enzymatically hydrolysable organic P(EHOP) and dissolved inorganic P(IP). In this study, soil samples from the water-level fluctuation zone (WLFZ) were collected from a tributary of the Three Gorges Reservoir (TGR) to characterize the properties of organic P(OP) fractions using solution ³¹P-nuclear magnetic resonance (NMR) and enzymatic hydrolysis. ³¹P-NMR showed that orthophosphate was the main part of the bioavailable P in the WLFZ soil and accounted for 80.4% of the NaOH-EDTA extractable total P (NaOH-EDTA TP), while phosphate monoester accounted for 60.5% of NaOH-EDTA extractable OP (NaOH-EDTA OP). The soil properties and replenishment from the mainstream of the Yangtze River to the Pengxi River have a certain effect on the content and distribution of P forms in the WLFZ soil of the tributary. The EHOP accounted for 28.1% of the NaOH-EDTA OP, and a significant positive correlation was observed between labile monoester P and EHOP and organic matter (OM). The water-soluble OP(H₂O-OP), bicarbonate-extractable OP(NaHCO₃-OP), and Fe- and Al-associated OP(Fe/Al-OP) were significantly hydrolyzed by phosphatase and thus exhibited great release potential. The ranking of the bioavailability of OP was Fe/Al-OP > H₂O-OP > NaHCO₃-OP. Phytate-like P were mainly found in H₂O-OP and NaHCO₃-OP, which indicated that periodic submersion–emersion cycles promoted the release of phytate-like P from Fe/Al-OP into the water column of the TGR. These observations suggest that when the external P input was effectively controlled, a huge risk of release of the internal OP from the WLFZ soil, and the biogeochemical cycling of the bioavailable P played an important role in maintaining the eutrophication of the reservoir.

PURPOSE OF THE REVIEW: Freshwater wetlands are found in various climatic zones ranging from tropics to tundra, and their roles from groundwater recharge and flood control to water quality management and biodiversity protection are well recognized. Phosphorus (P) is a limiting nutrient for algal growth in freshwater systems, including wetlands. Various physico-chemical and biological characteristics of wetlands regulate cycles of nutrients such as P. Thus, estimating internal loading of P in wetlands would be crucial in the formulation of effective P management strategies in the wetland systems. This review and limnological data presented may offer needed knowledge/evidence for the effective control of P inputs in wetlands and provide insights on possible ways for interventions in controlling eutrophication and saving the ecosystem from collapse. RECENT FINDINGS: Various ways of P losses such as agriculture, urbanization, etc., to the water bodies have severely impacted water quality of wetlands by altering physical and chemical nature of the P compounds and release bound P to the water columns. Studies indicate that P sorption–desorption dynamic, mineralization, and enzymatic hydrolysis of P in freshwater wetlands’ soils/sediments are crucial in causing internal loading or sink of P in wetland systems. Thus, extensive studies on abovementioned arenas are crucial to restore natural freshwater wetlands or to increase the efficiency of constructed wetlands in retaining P. In general, researchers have elucidated significant amounts of limnological data to understand eutrophication processes in freshwater wetlands; however, studies on the interactions of P stability and hydro-climatic changes are not well understood. Such changes could significantly influence localized limnology/microenvironments and exacerbate internal P loading in freshwater wetlands; thus, studies in such direction deserve the attention of scientific communities.

The hydrolyzed protein derived from seafood waste is regarded as a premium and low-cost nitrogen source for microbial growth. In this study, optimization of enzymatic shrimp waste hydrolyzing process was investigated. The degree of hydrolysis (DH) with four processing variables including enzyme/substrate ratio (E/S), hydrolysis time, initial pH value and temperature, were monitored. The DH values were used for response surface methodology (RSM) optimization through central composite design (CCD) and for training artificial neural network (ANN) to make a process prediction. Results indicated that the optimum levels of variables are: E/S ratio at 1.64%, hydrolysis time at 3.59h, initial pH at 9 and temperature at 52.57°C. Hydrocarbon-degrading bacteria Bacillus subtilis N3-1P was cultivated using different DHs of hydrolysate. The associated growth curves were generated. The research output facilitated effective shrimp waste utilization.

Lignocellulosic biomass is considered as a recalcitrant substrate for anaerobic digestion due to its complex nature that limits its biological degradation. Therefore, suitable preprocessing for the improvement of the performance of conventional anaerobic digestion remains a challenge in the development of anaerobic digestion technology. The physical and chemical characteristics of wheat straw (WS), as a representative lignocellulosic biomass, have a significant impact on the anaerobic digestion process in terms of quantity and quality of the produced biogas. This study aimed at investigating the enzymatic saccharification and detoxification of straw prior to anaerobic digestion with the final objective of enhancing the performance of conventional anaerobic systems of recalcitrant fractions of agricultural waste. The experimental activity was performed in lab and pilot scale treating WS. Alkaline delignification of straw using sodium hydroxide (NaOH) was studied prior to enzymatic hydrolysis for the production of easily biodegradable sugars. After defining the optimum conditions for the pretreatment scheme, the anaerobic digestability of the effluents produced was measured. Finally, the final liquid effluents were fed to a pilot scale anaerobic digester of 0.5 m³ volume, applying an increasing organic loading rate (OLR) regime (in terms of chemical oxygen demand (COD) from 0.2 to 15 kg COD/m³/day). The optimum conditions for the delignification and enzymatic hydrolysis of WS were defined as 0.5 M NaOH at 50 °C for 3–5 h and 15 μL Cellic CTec2/g pretreated straw at 50 °C. It was proven that the resulting liquid effluents could be fed to an anaerobic digester in the ratio that they are produced with satisfactory COD removal efficiencies (over 70%) for OLRs up to 10 kg COD/m³/day. This value is correspondent to a hydraulic retention time of around 7.5 days, much lower than the respective one for untreated straw (over 12 days).
