Mercury Stoichiometric Relationships in a Subtropical Peatland

Spatially variable areas, or hotspots, of elevated mercury (Hg) concentrations in soil, water, and wildlife occur throughout the Everglades wetland ecosystem. This study investigates the stoichiometric controls of Hg relative to soil, water, and biotic components. Surface water, porewater, soil, periphyton, and Gambusia spp. (mosquitofish) were collected from hotspots and non-spot stations and analyzed for various parameters, including total mercury (THg), organic carbon (OC), total carbon (TC), total phosphorus (TP), and total nitrogen (TN) between late 2010 and early 2013. Soil nutrient ratios were significantly different between hotspot and non-hotspot stations, indicating a difference in trophic status and position along the decay continuum or differences in limiting nutrients. Overall, soil total Hg concentrations were negatively correlated with soil TC/TN, while soil TC/TP and soil TN/TP molar ratios and soil THg were negatively correlated at hotspot stations. Meanwhile, mosquitofish THg was negatively correlated with soil TC/TN molar ratio and positively correlated with soil TC/TP and TN/TP molar ratios, suggesting trophic truncation. Soil, surface water, and porewater THg, TC, and OC interactions resulted in significant differences between hotspot and non-hotspot stations and between molar ratios of C, N, and P. Periphyton-surface water THg/OC homeostasis and soil nutrient ratios significantly explained mosquitofish THg concentrations, further indicating a trophic influence on mosquitofish THg and potential hotspot dynamics. Several factors and processes including bottom-up trophic interaction and vegetation influence on Hg accumulation dynamics and food-chain length explain the development and persistence of Hg hotspot formation within the Everglades system.

Polycyclic aromatic hydrocarbons (PAHs) are petroleum components that, when dissolved in the aquatic environment, can disrupt normal animal physiological functions and negatively affect species populations. Gambusia yucatana is an endemic fish of the Yucatán Peninsula that seems to be particularly sensitive to the presence of PAHs dissolved in the water. Here, we examined PAH effects on gene expressions linked to endocrine disruption and biotransformation in this species. Specifically, we examined the expression of vitellogenin I (vtg1), vitellogenin II (vtg2), oestrogen receptor α (esr1), oestrogen receptor β (esr2), aryl hydrocarbon receptor (AhR) and the cytochrome P4503A (CYP3A) genes. We exposed G. yucatana to different concentrations of PAHs (3.89, 9.27, 19.51 μg/L) over a period of 72 h and found changes associated with reproduction, such as increases in hepatic expression of vtg, esr, AhR and CYP3A, mainly at concentrations of 9.27 and 19.51 μg/L. Our results also indicate that benzo[a]pyrene was probably the main PAH responsible for the observed effects. The genes examined here can be used as molecular markers of endocrine-disrupting compounds, as the PAHs, present in the environment, as gene expression increases could be observed as early as after 24 h. These biomarkers can help researchers and conservationists rapidly identify the impacts of oil spills and improve mitigation before the detrimental effects of environmental stressors become irreversible.

The disruption of endocrine systems due to environmental contaminants potentially impacts on the developmental, behavioural, regulatory and reproductive systems of animals. A major source of exposure of animals (terrestrial and aquatic) to endocrine-disrupting compounds is through contact with contaminated surface waters contaminated with sewage effluent and/or stormwater discharge. We studied the response to endocrine-disrupting compounds of Gambusia holbrooki mosquito fish resident of wetlands that were used for the storage of either treated sewage effluent or stormwater runoff. We found that fish from wetlands that received polluted waters directly from the source (treated sewage effluent or stormwater runoff) demonstrated a morphological response consistent with endocrine disruption. In contrast, fish in the second in the series of wetlands that housed treated sewage effluent did not show evidence of such response. However, those from the second in the series of stormwater receiving wetlands did display a morphological response, although it could be considered milder than was observed in fish from the first in this effluent stream. Fish were also smaller in the wetland that received sewage effluent directly from the sewage treatment plant than elsewhere. Although the differences were not statistically significant, the average size of fish in the first in the series of stormwater wetlands were also small and second only to those in the first sewage effluent wetland.

Zn is an essential trace metal in living beings. However, excessive concentrations can cause toxic effects even in the aquatic biota. Zn is widely used in different industrial sectors, which has increased its presence in aquatic environments. To assess the acute toxicity of Zn, bioassays were performed with the fish Gambusia sexradiata for a 96-h exposure using ZnCl₂ (0 and 15 salinity) and ZnO nanomaterials (0 salinity). The mean lethal concentrations (LC₅₀–96 h) for ZnCl₂ were 25.36 (19.64–32.76) and 177.91 (129.39–244.63) mg Zn L⁻¹ to 0 and 15 salinity, respectively. The increased concentration of ZnCl₂ showed a dose-response relationship; similarly, the increase in salinity significantly reduces the toxicity of Zn. Characterisation of ZnO nanomaterials was carried out by FTIR, DRX, SEM, DLS and zeta potential. The FTIR spectra showed the characteristic band of Zn-O vibration at 364 cm⁻¹, while DRX presents the hexagonal wurtzite structure with an average crystallite size of 40 nm. SEM micrographs reveal rod-like shapes with lengths and diameters of 40–350 nm and 90 nm, respectively. Agglomerates of 423 nm in water suspension were obtained by DLS and zeta potential of + 14.4 mV. Under these conditions, no mortality was observed due to the rapid flocculation/precipitation of ZnO nanomaterials, which involved brief interaction periods of Zn in the water column with the fish. Gambusia sexradiata is affected by increased Zn concentrations in hard water conditions, and salinity changes modified Zn toxicity, placing it as a suitable model for toxicity tests for this type of particles.

The use of synthetic pesticides to control vector populations is detrimental to human health and the environment and may lead to the development of resistant strains. Plants can be alternative sources of safer compounds effective on mosquito vectors. In this study, the mosquito larvicidal activity of Boswellia ovalifoliolata leaf essential oil (EO) was evaluated against Anopheles stephensi, Anopheles subpictus, Aedes aegypti, Aedes albopictus, Culex quinquefasciatus, and Culex tritaeniorhynchus. GC-MS revealed that the B. ovalifoliolata EO contained at least 20 compounds. The main constituents were β-pinene, α-terpineol, and caryophyllene. In acute toxicity assays, the EO was toxic to larvae of An. stephensi, Ae. aegypti, Cx. quinquefasciatus, An. subpictus, Ae. albopictus, and Cx. tritaeniorhynchus with LC₅₀ values of 61.84, 66.24, 72.47, 82.26, 89.80, and 97.95 μg/ml, respectively. B. ovalifoliolata EO was scarcely toxic to mosquito fishes, backswimmers, and water bugs predating mosquito larvae with LC₅₀ from 4186 to 14,783 μg/ml. Overall, these results contribute to develop effective and affordable instruments to magnify the reliability of Culicidae control programs.
