Detection of sea polluters with change in conditioned reflexive responses of fish

This work is based on biotests being experimented at sea organisms - fish Serranus scriba. We were studying the activity of the mentioned fish in sublethal concentrations of organochloric pesticide - lindane. Besides the classic biotests, we used some new methods for neurotoxic testing that include the influence of this pesticide on circadian activity of fish as well as the variations in obtaining the positive conditioned reflexive responses. The results of these experiments show that this kind of fish reacts at the subtile changes of toxic concentration with the change of quality and rhythm in its conditioned reflexive reactions, that appeared earlier than other global signs of intoxication.

The experiment was performed on marine Teleost Serranus scriba with lower concentrations of lindane then MPC (due to our valid law). The aim of this study was to find the effects of lindane to fish activities connected with nutrition. This motive is among the strongest. It includes parameters as: number of fish movements in the presence of prey or without it and the number of onslaughts to the prey during one hour. The results have shown that the increasing of lindane concentration caused considerable decreasing and even complete cessation of activity. Interest for prey and decreasing activity of predators in the presence of prey (typical for Serranus - as a fish of prey and the hunter) endures till the moment of convulsion.

Agricultural use of organochlorine pesticides (OCPs) increased during the twentieth century but many of them have been progressively banned several decades after their introduction. Nevertheless, these lipophilic chemical compounds may persist in soils and sediments. From sediment deposits, it is possible to reconstruct the chronology of OCP releases in relation to former applications through time. Nevertheless, long-term fate of OCPs i.e. source, transfer, and storage through the watershed, is also related to the OCPs-sediment characteristics interactions, and our study showed the significant links between OCPs and labile or refractory organic matter. From sediment cores collected in a mainly agricultural watershed, the Eure River watershed (France), aldrin and lindane widespread applications during the 1950s–1970s have been recorded. While lindane applications declined after that date, according to the temporal trend of the stable isomer of hexachlorocyclohexane (β-HCH), α-, and γ-HCH have been recorded at significant levels in the 2000s, suggesting first local post-ban applications. Nevertheless, the relationships between these OCPs and labile organic matter resulted in an overestimation of the post-ban releases. Also, the detection of stable metabolites of dichlorodiphenyltrichloroethane (DDT) (i.e. 4,4′-DDE) and heptachlor (i.e. heptachlor epoxide) several decades after their ban, revealed the role of old deep soils erosion in the chronology of OCP releases and thus the reemergence of stable transformation products from historical OCPs.

Fungi have an exceptional capability to flourish in presence of heavy metals and pesticide. However, the mechanism of bioremediation of pesticide (lindane) and multimetal [mixture of cadmium (Cd), chromium (Cr), copper (Cu), nickel (Ni), lead (Pb), zinc (Zn)] by a fungus is little understood. In the present study, Aspergillus fumigatus, a filamentous fungus was found to accumulate heavy metals in the order [Zn(98%)>Pb(95%)>Cd(63%)>Cr(62%)>Ni(46%)>Cu(37%)] from a cocktail of 30 mg L⁻¹ multimetal and lindane (30 mg L⁻¹) in a composite media amended with 1% glucose. Particularly, Pb and Zn uptake was enhanced in presence of lindane. Remarkably, lindane was degraded to 1.92 ± 0.01 mg L⁻¹ in 72 h which is below the permissible limit value (2.0 mg L⁻¹) for the discharge of lindane into the aquatic bodies as prescribed by European Community legislation. The utilization of lindane as a cometabolite from the complex environment was evident by the phenomenal growth of the fungal pellet biomass (5.89 ± 0.03 g L⁻¹) at 72 h with cube root growth constant of fungus (0.0211 g¹/³ L⁻¹/³ h⁻¹) compared to the biomasses obtained in case of the biotic control as well as in presence of multimetal complex without lindane. The different analytical techniques revealed the various stress coping strategies adopted by A. fumigatus for multimetal uptake in the simultaneous presence of multimetal and pesticide. From the Transmission electron microscope coupled energy dispersive X-ray analysis (TEM-EDAX) results, uptake of the metals Cd, Cu and Pb in the cytoplasmic membrane and the accumulation of the metals Cr, Ni and Zn in the cytoplasm of the fungus were deduced. Fourier-transform infrared spectroscopy (FTIR) revealed involvement of carboxyl/amide group of fungal cell wall in metal chelation. Thus A. fumigatus exhibited biosorption and bioaccumulation as the mechanisms involved in detoxification of multimetals.

Plant-specific root-microbe-soil interactions play an indisputable role in microbial adaptation to environmental stresses. However, the assembly of plant rhizosphere microbiomes and their feedbacks in modification of pollution alleviation under organochlorine stress condition is far less clear. This study examined the response of root-associated bacterial microbiomes to lindane pollution and compared the dissipation of lindane in maize-cultivated dry soils and rice-cultivated flooded soils. Results showed that lindane pollution dramatically altered the microbial structure in the rhizosphere soil of maize but had less influence on the microbial composition in flooded treatments regardless of rice growth, when the reductive dechlorination of lindane was actively coupled with natural redox processes under anaerobic conditions. After 30 days of plant growth, lindane residues dissipated much faster in anaerobic than in aerobic environments, with only 1.08 mg kg⁻¹ lindane remaining in flooded control compared to 12.79 mg kg⁻¹ in dry control soils. Compared to the corresponding unplanted control, maize growth significantly increased, but rice growth slightly decreased the dissipation of lindane. Our study suggests that opposite impacts would lead to the self-purification of polluted soils during the growth of xerophytic maize and hygrocolous rice. This was attributed to the contrasting belowground micro-ecological processes regarding protection of root tissues and thereby assembly of rhizosphere microbiomes shaped by the xerophytic and hygrocolous crops under different water managements, in response to lindane pollution.

The correlation between endocrine active contaminants in the environment and alterations in reproductive development of Sarotherodon melanotheron from Lagos lagoon has been investigated. Sediment and a total of 155 fish (74 males and 81 females) were collected between November 2014–March 2015 from selected contaminated sites (Ikorodu, Oworonshoki, Makoko and Idumota) and a putative control site (Igbore) along the lagoon. Sediment contaminant analysis revealed, significantly higher concentration of lindane, dieldrin, 4-iso-nonylphenol, 4-t-octylphenol and monobutyltin cation at the contaminated sites. Examination of gross morphological and histological changes of fish gonads showed a 27.4% prevalence of intersex in the sampled fish, of which 78% were males (testes-ova) and 22% were females (ovo-testis). Quantitative PCR (qPCR) of liver transcripts revealed the presence of vitellogenin (vtg) levels in male fish from contaminated sites. Zona radiata proteins (zrp) mRNA levels were significantly higher in females, compared to male fish. In general, significantly lower vtg and zrp transcripts levels were recorded at Igbore (control site), compared with contaminated sites. Principal component analysis (PCA) showed site and sex relationship in biological responses and contaminants, including trace metals, demonstrating that measured endocrine responses in fish were associated with contaminant burden in sediment. In addition, positive relationships were observed in male fish from Idumota, Oworonshoki and Ikorodu with vtg and dieldrin/4-iso-nonyphenol, with higher levels in male fish, compared to females. Further, contaminants from the Makoko, Oworonshoki and Ikorodu sites were positively associated with higher GSI and zrp in females. More importantly, the severity of intersex and changes in vtg transcripts imply a progressive feminization of male fish with concomitant alteration in the reproductive health of fish inhabiting the Lagos lagoon.

Black carbon and total organic carbon (TOC) along with organochlorines (OCs) were analyzed in soils from four sampling zones of Lesser Himalayan Region based on source proximity/anthropogenic influences along the altitude. CTO-375 method was used for BC analysis while OCs were analyzed by GC-MS/MS system. BC and TOC ranged between 0.16–1.77 and 6.8–41.3 mg g−1 while those of OCPs and PCBs ranged between 0.69 and 5.77 and 0.12–2.55 ng g−1, respectively. ∑DDTs were the dominant (87.9%) among OCPs while tri- and tetra- (65.5%) homologue groups among PCBs. Hexa-PCBs, however also showed higher contribution (20.4%) in the region. Source diagnostic ratios of DDE + DDD/DDT (0.1–1.53) indicated both fresh and old input while α-HCH/γ-HCH (0.19–2.49) showed presence of lindane in the region. Higher concentration of OCs were observed in Zone C at altitudinal range of 737–975 masl that are close to the human influences and potential sources of POPs. The results of linear regression analysis revealed potential input of BC in soil distribution of OC concentrations in the region.

Raptor feathers have been increasingly used to assess pollutants in ecotoxicological monitoring studies. However, the suitability of down feathers to detect pollutants has not yet been investigated. In this study, concentrations of persistent organic pollutants (POPs) and organophosphate ester flame retardants (OPEs) were assessed in down and juvenile contour feathers of Spanish cinereous vulture (Aegypius monachus) nestlings (circa 73 days old) and contaminant concentrations were compared between both types of feathers from the same individuals. Concentrations of polychlorinated biphenyls (PCBs: 1.30–6.16 ng g−1 dw feather), polybrominated diphenyl ethers (PBDEs: 0.23–1.35 ng g−1 dw feather), p,p'-dichlorodiphenyldichloroethylene (pp-DDE: 0.09–6.10 ng g−1 dw feather) and tris (1-chloro-2-propyl) phosphate (TCiPP: 0.86–48.96 ng g−1 dw feather) were significantly higher in down than in contour feathers. In contrast, contour feathers showed higher levels of the more volatile POP, lindane (0.25–3.12 ng g−1 dw feather). Concentrations of hexachlorobenzene (HCB) and OPEs (except TCiPP) were similar between the two types of feathers. By showing high accumulation of the most persistent POPs investigated, down feathers presented a contamination profile similar to that previously described in raptor eggs. As these feathers grow during the first days of a vulture chicks life, they probably reflect the contaminant burden of the chick due to maternal transfer to the egg. Overall, the present study provides the first indication that down feathers may be useful for biomonitoring studies. Further research is needed to confirm whether nestling down feathers reflect the concentrations in the egg.

Probabilistic environmental quality criteria for Naphthalene (Nap), Phenanthrene (Phe), Fluoranthene (Flu), Pyrene (Pyr), Triclosan (TCS), Tributyltin (TBT), Chlorpyrifos (CPY), Diuron (DUR), γ-Hexaclorocyclohexane (γ-HCH), Bisphenol A (BPA) and 4-Nonylphenol (4-NP) were derived from acute toxicity data using saltwater species representative of marine ecosystems, including algae, mollusks, crustaceans, echinoderms and chordates. Preferably, data concerns sublethal endpoints and early life stages from bioassays conducted in our laboratory, but the data set was completed with a broad literature survey. The Water Quality Criteria (WQC) obtained for TBT (7.1·10⁻³ μg L⁻¹) and CPY (6.6· 10⁻³ μg L⁻¹) were orders of magnitude lower than those obtained for PAHs (ranging from 3.75 to 45.2 μg L⁻¹), BPA (27.7 μg L⁻¹), TCS (8.66 μg L⁻¹) and 4-NP (1.52 μg L⁻¹). Critical values for DUR and HCH were 0.1 and 0.057 μg L⁻¹ respectively. Within this context, non-selective toxicants could be quantitatively defined as those showing a maximum variability in toxicity thresholds (TT) of 3 orders of magnitude across the whole range of marine diversity, and a cumulative distribution of the TT fitting to a single log-logistic curve, while for selective toxicants variability was consistently found to span 5 orders of magnitude and the TT distribution showed a bimodal pattern. For the latter, protective WQC must be derived taking into account the SSD of the sensitive taxa only.
显示更多 [+] 显示较少 [-]Effect of organochlorine pesticides exposure on the maize root metabolome assessed using high-resolution magic-angle spinning 1H NMR spectroscopy

1H-HRMAS NMR-based metabolomics was used to better understand the toxic effects on maize root tips of organochlorine pesticides (OCPs), namely lindane (γHCH) and chlordecone (CLD). Maize seedlings were exposed to 2.5 μM γHCH (mimicking basic environmental contaminations) for 7 days and compared to 2.5 μM CLD and 25 μM γHCH for 7 days (mimicking hot spot contaminations). The 1H-HRMAS NMR-based metabolomic profiles provided details of the changes in carbohydrates, amino acids, tricarboxylic acid (TCA) cycle intermediates and fatty acids with a significant separation between the control and OCP-exposed root tips. First of all, alterations in the balance between glycolysis/gluconeogenesis were observed with sucrose depletion and with dose-dependent fluctuations in glucose content. Secondly, observations indicated that OCPs might inactivate the TCA cycle, with sizeable succinate and fumarate depletion. Thirdly, disturbances in the amino acid composition (GABA, glutamine/glutamate, asparagine, isoleucine) reflected a new distribution of internal nitrogen compounds under OCP stress. Finally, OCP exposure caused an increase in fatty acid content, concomitant with a marked rise in oxidized fatty acids which could indicate failures in cell integrity and vitality. Moreover, the accumulation of asparagine and oxidized fatty acids with the induction of LOX3 transcription levels under OCP exposure highlighted an induction of protein and lipid catabolism. The overall data indicated that the effect of OCPs on primary metabolism could have broader physiological consequences on root development. Therefore, 1H-HRMAS NMR metabolomics is a sensitive tool for understanding molecular disturbances under OCP exposure and can be used to perform a rapid assessment of phytotoxicity.
