B-type esterases in the snail <em>Xeropicta derbentina</em>: an enzymological analysis to evaluate their use as biomarkers of pesticide exposure

International audience | The study was prompted to characterize the B-type esterase activities in the terrestrial snail <em>Xeropicta derbentina</em> and to evaluate its sensitivity to organophosphorus and carbamate pesticides. Specific cholinesterase and carboxylesterase activities were mainly obtained with acetylthiocholine (Km =77.2 mM; Vmax= 38.2 mU/mg protein) and 1-naphthyl acetate (Km= 222 mM, Vmax= 1095 mU/mg protein) substrates, respectively. Acetylcholinesterase activity was concentration-dependently inhibited by chlorpyrifos-oxon, dichlorvos, carbaryl and carbofuran (IC50 =1.35 x 105–3.80 x 108 M). The organophosphate-inhibited acetylcholinesterase activity was reactivated in the presence of pyridine-2- aldoxime methochloride. Carboxylesterase activity was inhibited by organophosphorus insecticides (IC50 =1.20 x 105–2.98 x 108 M) but not by carbamates. B-esterase-specific differences in the inhibition by organophosphates and carbamates are discussed with respect to the buffering capacity of the carboxylesterase to reduce pesticide toxicity. These results suggest that B-type esterases in<em> X. derbentina</em> are suitable biomarkers of pesticide exposure and that this snail could be used as sentinel species in field monitoring of Mediterranean climate regions

The Mediterranean environment, and most of the Italian peninsula, presents some peculiarities in terms of crop response to O3 since most physiological mechanisms activated upon O3 exposure, such as stomatal closure, often overlap and interact with those that underlie plant adaptation to drought and hyperosmotic stress, which are typical of these environments. OTC and EDU experiments have demonstrated that O3 causes strong yield losses when crops are grown without water limitations. However, exposure to water or saline stress significantly reduced O3 effects on crop yield. In this review, we present the methodological approaches that have been used to study plant-ozone interactions in Italy as well as biochemical, physiological and agronomic responses for representative cropping systems of the Mediterranean climate. Is the 22% yield loss due to ambient ozone in non-limiting water conditions a realistic estimate for moderately stressed crops, typical of most Mediterranean regions?

An account of histo-cytological and ultrastructural studies on ozone effect on crop and forest species in Italy is given, with emphasis on induced cell death and the underlying mechanisms. Cell death phenomena possibly due to ambient O3 were recorded in crop and forest species. In contrast, visible O3 effects on Mediterranean vegetation are often unclear. Microscopy is thus suggested as an effective tool to validate and evaluate O3 injury to Mediterranean vegetation. A DAB-Evans blue staining was proposed to validate O3 symptoms at the microscopic level and for a pre-visual diagnosis of O3 injury. The method has been positively tested in some of the most important crop species, such as wheat, tomato, bean and onion and, with some restriction, in forest species, and it also allows one to gain some very useful insights into the mechanisms at the base of O3 sensitivity or tolerance. Ozone-induced cell death is a frequent phenomenon in Mediterranean conditions, not only in the most sensitive crops but also in forest species.

The impact that microplastics have on baleen whales is a question that remains largely unexplored. This study examined the interaction between free-ranging fin whales (Balaenoptera physalus) and microplastics by comparing populations living in two semi-enclosed basins, the Mediterranean Sea and the Sea of Cortez (Gulf of California, Mexico). The results indicate that a considerable abundance of microplastics and plastic additives exists in the neustonic samples from Pelagos Sanctuary of the Mediterranean Sea, and that pelagic areas containing high densities of microplastics overlap with whale feeding grounds, suggesting that whales are exposed to microplastics during foraging; this was confirmed by the observation of a temporal increase in toxicological stress in whales. Given the abundance of microplastics in the Mediterranean environment, along with the high concentrations of Persistent Bioaccumulative and Toxic (PBT) chemicals, plastic additives and biomarker responses detected in the biopsies of Mediterranean whales as compared to those in whales inhabiting the Sea of Cortez, we believe that exposure to microplastics because of direct ingestion and consumption of contaminated prey poses a major threat to the health of fin whales in the Mediterranean Sea.

High O3 levels, driving uptake and challenging defense, prevail on the Canary Islands, being associated with the hot and dry summers of the Mediterranean-type climate. Pinus canariensis is an endemic conifer species that forms forests across these islands. We investigated the effects of ozone on photosynthesis and biochemical parameters of P. canariensis seedlings exposed to free-air O3 fumigation at Kranzberg Forest, Germany, where ambient O3 levels were similar to those at forest sites in the Canary Islands. The twice-ambient O3 regime (2xO3) neither caused visible injury-like chlorotic or necrotic spots in the needles nor significantly affected violaxanthin, antheraxanthin and zeaxanthin levels and the de-epoxidation state of the xanthophyll cycle. In parallel, stomatal conductance for water vapour, net photosynthesis, intercellular CO2 concentration, chlorophyll fluorescence parameters, as well as antioxidant levels were hardly affected. It is concluded that presently prevailing O3 levels do not impose severe stress on P. canariensis seedlings.

Concentrations of CO2 and CH4 measured over 3 years at a rural site in the Spanish northern plateau were investigated together with vegetation and meteorological variables. Two procedures were implemented to study the annual evolution. Kernel estimation provided a detailed time description, and the harmonic model may be fitted easily. The site was characterised by grass from autumn to spring. However, vigorous growth was observed during the latter season due to the biological cycle of plants under favourable meteorological conditions. A CO2 peak was observed a fortnight before the time of maximum NDVI, and was attributed to the prevalence of respiration over photosynthesis. A pronounced trough was apparent in summer and was explained by the death of vegetation and active dispersion in a highly developed boundary layer. CH4 evolution was characterised by a deficit period from May to October, indicating that meteorological evolution played a key role. The harmonic model showed that annual and half-annual cycles evidenced a similar contribution for CO2, whereas said weight for the half-annual cycle was considerably smaller for CH4.

In order to increase the knowledge about crop tolerance to air pollutants in the different agroclimatic zones of the world, so that they can be efficiently considered for improving peri-urban agriculture, increasing the success of restoration projects, or enhancing air quality in polluted sites, the suitability of four economical valuable tree crops of the Mediterranean agriculture were studied under field conditions: date palm tree (Phoenix dactylifera L.), pomegranate (Punica granatum L.), fig tree (Ficus carica L.), and olive tree (Olea europaea L.). The measurement of biochemical markers such as ascorbic acid content, leaf relative water content, leaf total chlorophyll and leaf extract pH, at two contrasted air quality sites, a polluted site located around Gabes (Tunisia) industrial area and a control site, allowed the assessment of the air pollution tolerance index (APTI) and anticipated performance index (API) for the assayed species. Results showed obvious differences between the evergreen and the caducifolious tree crops assayed. Phoenix dactylifera tree (API = 6) was classified as an excellent performer for growing under poor air quality, followed by Olea europaea tree (API = 2) which was classified as a moderate performer. Both of trees can be recommended for successful results in peri-urban agriculture and restoration projects of polluted areas in the Mediterranean climate; on the contrary, the suitability of the Punica granatum (API = 1) was very poor, but still potentially interesting as a biological indicator of air pollution. Regarding the Ficus carica tree (API = 0), this species is not suitable for growing in air-polluted areas.

This study belongs to the biomonitoring program of soil qualities using a land snail, Cantareus aspersus, as bioindicator. The metal-soil contamination in some sites (National Park of El Kala (NPK), El Bouni, Sidi Amar, Nechmaya, and Guelma) located in Northeast Algeria were determined during two seasons (winter and spring 2015, 2016). Glutathione (GSH) content and acetylcholinesterase (AChE) activity were significantly decreased in snails collected during spring as compared with those noticed during winter under bioclimate change. In addition, a significant difference between various sites was observed, depending on the proximity to pollution sources. The significant variation of biomarker levels is a function of the physic-chemical properties of soils when they positively correlated with EC, H, and OM, and negatively correlated with all metallic elements. Moreover, Fe and Al₂O₃ are the most abundant in all the sites, and the most polluted site was found as that of El Bouni, followed by Sidi Amar, Nechmaya, and Guelma, since NPK is the less polluted site and considered a reference site. The tested biomarkers are sensitive oxidative parameters in snails exposed to pollution correlated significantly with the soil physic-chemical properties and metallic element contents in soil. Indeed, C. aspersus could be used as sentinel species in field monitoring of Mediterranean climate regions.

One of the most common anthropogenic impacts on river ecosystems is the effluent discharge from wastewater treatment plants. The effects of this contamination on stream biota may be intensified in Mediterranean climate regions, which comprise a drought period that leads to flow reduction, and ultimately to stagnant pools. To assess individual and combined effects of flow stagnation and sewage contamination, biofilm and gastropod grazers were used in a 5-week experiment with artificial channels to test two flow velocity treatments (stagnant flow/basal flow) and two levels of organic contamination using artificial sewage (no sewage input/sewage input). Stressors’ effects were determined on biofilm total biomass and chlorophyll (Chl) content, on oxygen consumption and growth rate of the grazers (Theodoxus fluviatilis), and on the interaction grazer-biofilm given by grazer’s feeding activity (i.e., biofilm consumption rate). The single effect of sewage induced an increase in biofilm biomass and Chl-a content, simultaneously increasing both grazers’ oxygen consumption and their feeding activity. Diatoms showed a higher sensitivity to flow stagnation, resulting in a lower content of Chl-c. Combined stressors interacted antagonistically for biofilm total biomass, Chl-b contents, and grazers’s feeding rate. The effect of sewage increasing biofilm biomass and grazing activity was reduced by the presence of flow stagnation (antagonist factor). Our findings suggest that sewage contamination has a direct effect on the functional response of primary producers and an indirect effect on primary consumers, and this effect is influenced by water flow stagnation.

The goal of the present work was to increase our knowledge on the behavior of manganese and nickel in soil within a Mediterranean environment. The study assessed the concentration levels of Mn and Ni (heavy metals selected for their essential role in the development of plants) in 250 soil horizon samples within 125 soil profiles of undisturbed soils in La Rioja (Spain). The study was undertaken to investigate and predict Mn and Ni concentrations on a regional scale. The analysis of spatial distribution of the elements was found to be affected by the nature of bedrock and, to a lesser extent, the anthropogenic origin. The variation of vertical distributions can be related, first, to natural sources—mainly the bed rocks—and, second, to soil processes. The geographical distribution of soil Mn is important to agriculture, nutrition, and health. Soil Mn and Ni maps of the area were elaborated, using geostatistics and geographic information systems. Mapping of geographical distributions will be useful in future research to determine regional patterns of Mn and Ni bioavailability, Mn and Ni deficiencies, and the possible consequences of land disposal of Mn- and Ni-laden wastes.
