Mechanisms and uncertainties of Zn supply on regulating rice Cd uptake

Application of Zinc (Zn) is considered an effective measure to reduce Cadmium (Cd) uptake and toxicity in Cd-contaminated soils for many plant species. However, interaction between Zn and Cd in rice plant is complex and uncertain. In this study, four indica rice cultivars were selected to evaluate the effect of Zn exposure in an EGTA-buffered nutrient solution under varying Zn activities and a field level of Cd activity to characterize the interaction between Zn and Cd in rice. Severe depression in shoots’ biomass, tiller number, and SPAD (Soil and Plant Analyzer Development) value were found at both Zn deficiency and Zn phytotoxicity levels among four tested rice cultivars. There existed a strong antagonism interaction between Zn and Cd in both shoot and root from Zn deficiency to Zn phytotoxicity. The reduction of Cd accumulation in roots and shoots could be explained by the competition between Zn and Cd as well as the dilution effect of increasing biomass. The conflicting effect of Zn supply on Cd uptake may be attributed to the increasing transfer ratio of Cd from root to shoot with the increasing Zn²⁺ activities and the strong depression of Fe and Mn in shoots with the increasing Zn²⁺ activities as well as the variation of genotypes. Balance between Zn and Cd should be considered in field application.

Iron (Fe) is an essential nutrient for living organisms and Fe deficiency is a worldwide problem for the health of both rice and humans. Zinc (Zn) contamination in agricultural soils is frequently observed. Here, we studied Fe isotope compositions and transcript levels of Fe transporter genes in rice growing in nutrient solutions having a range of Zn concentrations. Our results show Zn stress reduces Fe uptake by rice and drives its δ⁵⁶Fe value to that of the nutrient solution. These observations can be explained by the weakened Fe(II) uptake through Strategy I but enhanced Fe(III) uptake through Strategy II due to the competition between Zn and Fe(II) combining with OsIRT1 (Fe(II) transporter) in root, which is supported by the downregulated expression of OsIRT1 and upregulated expression of OsYSL15 (Fe(III) transporter). Using a mass balance box model, we also show excess Zn reduces Fe(II) translocation in phloem and its remobilization from senescent leaf, indicating a competition of binding sites on nicotianamine between Zn and Fe(II). This study provides direct evidence that how Zn regulates Fe uptake and translocation in rice and is of practical significance to design strategies to treat Fe deficiency in rice grown in Zn-contaminated soils.

The impacts of iron oxide nanoparticles (γ-Fe2O3 NPs) and ferric ions (Fe³⁺) on plant growth and molecular responses associated with the transformation and transport of Fe²⁺ were poorly understood. This study comprehensively compared and evaluated the physiological and molecular changes of Citrus maxima plants as affected by different levels of γ-Fe2O3 NPs and Fe³⁺. We found that γ-Fe2O3 NPs could enter plant roots but no translocation from roots to shoots was observed. 20 mg/L γ-Fe2O3 NPs had no impact on plant growth. 50 mg/L γ-Fe2O3 NPs significantly enhanced chlorophyll content by 23.2% and root activity by 23.8% as compared with control. However, 100 mg/L γ-Fe2O3 NPs notably increased MDA formation, decreased chlorophyll content and root activity. Although Fe³⁺ ions could be used by plants and promoted the synthesis of chlorophyll, they appeared to be more toxic than γ-Fe2O3 NPs, especially for 100 mg/L Fe³⁺. The impacts caused by γ-Fe2O3 NPs and Fe³⁺ were concentration-dependent. Physiological results showed that γ-Fe2O3 NPs at proper concentrations had the potential to be an effective iron nanofertilizer for plant growth. RT-PCR analysis showed that γ-Fe2O3 NPs had no impact on AHA gene expression. 50 mg/L γ-Fe2O3 NPs and Fe³⁺ significantly increased expression levels of FRO2 gene and correspondingly had a higher ferric reductase activity compared to both control and Fe(II)-EDTA exposure, thus promoting the iron transformation and enhancing the tolerance of plants to iron deficiency. Relative levels of Nramp3 gene expression exposed to γ-Fe2O3 NPs and Fe³⁺ were significantly lower than control, indicating that all γ-Fe2O3 NPs and Fe³⁺ treatments could supply iron to C. maxima seedlings. Overall, plants can modify the speciation and transport of γ-Fe2O3 NPs or Fe³⁺ for self-protection and development by activating many physiological and molecular processes.

In this study, the effects of concentrations 0, 100, 250, 500, 750 and 1000 mg kg−1 of nanoscale zero-valent iron (nZVI) on germination, seedlings growth, physiology and toxicity mechanisms were investigated. The results showed that nZVI had no effect on germination, but inhibited the rice seedlings growth in higher concentrations (>500 mg kg−1 nZVI). The highest suppression rate of the length of roots and shoots reached 46.9% and 57.5%, respectively. The 1000mg kg−1 nZVI caused the highest suppression rates for chlorophyll and carotenoids, at 91.6% and 85.2%, respectively. In addition, the activity of antioxidant enzymes was altered by the translocation of nanoparticles and changes in active iron content. Visible symptoms of iron deficiency were observed at higher concentrations, at which the active iron content decreased 61.02% in the shoots, but the active iron content not decreased in roots. Interestingly, the total and available amounts of iron in the soil were not less than those in the control. Therefore, the plants iron deficiency was not caused by (i) deficiency of available iron in the soil and (ii) restraint of the absorption that plant takes in the available iron, while induced by (ⅲ) the transport of active iron from the root to the shoot was blocked. The cortex tissues were seriously damaged by nZVI which was transported from soil to the root, these were proved by scanning electron microscopy (SEM), transmission electron microscopy (TEM) and energy dispersive spectroscopy (EDS). This current study shows that the mechanism of iron deficiency in rice seedling was due to transport of active iron from the root to the shoot blocked, which was caused by the uptake of nZVI.

Arsenic (As) and cadmium (Cd), two major carcinogenic heavy metals, enters into human food chain by the consumption of rice or rice-based food products. Both As and Cd disturb plant-nutrient homeostasis and hence, reduces plant growth and crop productivity. In the present study, As/Cd modulated responses were studied in non-basmati (IR-64) and basmati (PB-1) rice varieties, at physiological, biochemical and transcriptional levels. At the seedling stage, PB-1 was found more sensitive than IR-64, in terms of root biomass; however, their shoot phenotype was comparable under As and Cd stress conditions. The ionomic data revealed significant nutrient deficiencies in As/Cd treated-roots. The principal component analysis identified NH₄⁺ as As-associated key macronutrient; while, NH₄⁺/NO₃⁻ and K⁺ was majorly associated with Cd mediated response, in both IR-64 and PB-1. Using a panel of 21 transporter gene expression, the extent of nutritional deficiency was ranked in the order of PB-1(As)<IR-64(As)<PB-1(Cd)<IR-64(Cd). A feed-forward model is proposed to explain nutrient deficiency induced de-regulation of gene expression, as observed under Cd-treated IR-64 plants, which was also validated at the level of sulphur metabolism related enzymes. Using urea supplementation, as nitrogen-fertilizer, significant mitigation was observed under As stress, as indicated by 1.018- and 0.794-fold increase in shoot biomass in IR-64 and PB-1, respectively compared to that of control. However, no significant amelioration was observed in response to supplementation of urea under Cd or potassium under As/Cd stress conditions. Thus, the study pinpointed the relative significance of various macronutrients in regulating As- and Cd-tolerance and will help in designing suitable strategies for mitigating As and/or Cd stress conditions.

The accurate assessment of soil selenium (Se) bioavailability is crucial for Se biofortification in Se-deficient areas and risk assessment in selenosis areas. However, a universally accepted approach to evaluate Se bioavailability in soil is currently lacking. This research investigated Se bioavailability in six soils treated with selenite (Se(IV)) or selenate (Se(VI)) by comparing diffusive gradients in thin-films (DGT) technique and chemical extraction methods through pot experiments. A bioindicator method was used to evaluate Se concentrations in pak choi and compare the results with the Se concentration measured by other methods. Results showed that chemical extraction methods presented different extraction efficiencies for available Se over a range of soil types, and the same extraction method had various extraction efficiencies for different Se species in the same soil. DGT measured Se concentrations (CDGT−Se) for Se(VI) treatment were 2.3–34.1 times of those for Se(IV) treatment. KH2PO4–K2HPO4 and AB-DTPA extractable Se could predict the bioavailability of soil Se, but they were disturbed by soil properties. HAc extraction was unsuitable for evaluating Se bioavailability in different Se(IV)-treated soils. By contrast, DGT technique was preferable for predicting plant uptake of Se(IV) over chemical extraction methods. Although DGT technique was independent of soil properties, KH2PO4–K2HPO4 extraction provided the best fitting regression equation for Se(VI) when it was dependent on soil organic matter. Thus, KH2PO4–K2HPO4 extraction may be preferred to assess Se(VI) bioavailability in different soil types on a large scale.

Amaranth (Amaranthus mangostanus L.) has superior capability for accumulating cadmium (Cd) and has the potential to be used for phytoremediation of Cd contaminated soils. Iron (Fe) is chemically similar to Cd and may mediate Cd-induced physiological or metabolic impacts in plants. The purpose was to investigate the model of time-dependent and concentration-dependent kinetics of Cd absorption under Fe deficiency, understanding the physiological mechanism of Cd absorption in amaranth roots. The kinetic characteristics of Cd uptake by amaranth grown in Cd enriched nutritional solution with or without Fe addition and with methanol-chloroform, carbonyl cyanide 3-chlorophenylhydrazone (CCCP), and lanthanum chloride (LaCl3) were compared using 109Cd2+ isotope labeling technique. The results showed that Cd uptake was time-dependent and about 90–93% of uptake occurred during the first 150 min. The kinetics of Cd uptake showed that two stages were involved. The saturation stage fitted the Michaelis-Menten model when concentrations of Cd were lower than 12.71 μmol/L and then the absorption of Cd by roots was increased linearly during the second stage. Only linear absorption was observed with methanol-chloroform treatment while the metabolic inhibitor CCCP inhibited only the saturation absorption process, and the Ca channel inhibitor LaCl3 partially inhibited the two stages of absorption. These results indicated that the root absorption of 109Cd2+ was enhanced under Fe deficiency which induced more Fe transporters in the root cell membrane, and the Ca channel, apoplastic and symplastic pathways enhanced the Cd absorption in roots.

Iron fertilizers are worthy to be studied due to alleviate the Fe deficiency. Different forms of iron oxide nanoparticles are selected to better understand possible particle applications as an Fe source for crop plants. In this study, we assessed the different effects of γ-Fe2O3 and Fe3O4 NPs on the physiology and fruit quality of muskmelon plants in a pot experiment for five weeks. Results showed that no increased iron content was found under NPs treatment in root, stem, leaf and fruit, except 400 mg/L Fe3O4 NPs had a higher iron content in muskmelon root. With the extension of NPs exposure, both γ-Fe2O3 and Fe3O4 NPs began to promote plant growth. In addition, γ-Fe2O3 and Fe3O4 NPs could increase chlorophyll content at a certain stage of exposure. Happily, 200 mg/L γ-Fe2O3 NPs and 100, 200 mg/L Fe3O4 NPs significantly increased fruit weight of muskmelon by 9.1%, 9.4% and 11.5%. It is noteworthy that both γ-Fe2O3 and Fe3O4 NPs caused positive effects on VC content, particularly 100 mg/L Fe3O4 NPs increased the VC content by 46.95%. To the best of our knowledge, little research has been done on the effect of nanoparticles on the whole physiological cycle and fruit quality of melon. The assessment of physiology and fruit quality of muskmelon plants in vitro upon γ-Fe2O3 and Fe3O4 NPs exposure could lay a foundation for NPs potential impact at every growth period of muskmelon plants.

There is an increasing evidence linking protective effect of selenium (Se) against Pb toxicology; however, Pb exposure risk assessments usually consider only the environmental Pb contamination and dietary intake. Based on the current understanding of mechanisms of SePb interactions, the physiological function/toxicology of Se and the toxicology of Pb, a new criterion for Se and Pb exposure assessment is developed. Additionally, seven existing criteria were also used to assess the resident health risks around a smelter in China. The Pb concentrations in locally-produced foods exceeded the national tolerance limits of China and the Se in the foods were similar to those in areas with adequate Se levels. In accordance with the illustrated assessments of the new criterion and seven existing criteria, we found a large knowledge gap between the new and traditional assessments of exposure to Pb and/or Se. The new assessment criteria suggested that almost all the residents were facing the Se deficiency and 58% of the residents not only had the adverse health of Se deficiency, but also had the health risks of Pb toxicity. The Pb and Se in the hair and urine may partly support the new criterion. This study suggested that the process of Se counteracting the Pb toxicity may result in Se deficiency. Pb exposure combined Se intake should be considered in future assessments of Pb exposure (or Se intake).

Cadmium uptake in rice is believed to be mediated by the Fe transport system. Phyto-available Cd can be changed by Fe fertilization of substrates. This work investigated whether and how Fe fertilization affects mitigation of Cd accumulation in paddy rice. A 90-d soil column experiment was conducted to study the change of Cd and Fe availability in soil after Fe fertilization (ionic and chelated Fe). A low-Cd accumulating cultivar (TY116) and a high-Cd accumulating cultivar (JY841) were grown in two Cd-polluted paddy soils amended with chelated Fe fertilizers. Additionally, both cultivars were grown in hydroponics to compare Fe-related gene expression in EDDHAFe-deficient and EDDHAFe-sufficient roots.The column experiment showed that EDTANa2Fe(II) and EDDHAFe(III) fertilization had a better mitigation effect on soil Cd availability compared to FeSO4·7H2O. Moreover, the field experiment demonstrated that these two chelated fertilizations could reduce Cd concentrations in brown rice by up to 80%. Iron concentrations in the brown rice were elevated by Fe chelates. Compared to EDDHAFe(III), EDTANa2Fe(II) fertilization had a stronger mitigation effect by generating more EDTANa2Cd(II) in the soil solution to decrease phyto-available Cd in the soil. While EDDHAFe(III) fertilization could increase soil pH and decrease soil Eh which contributed to decreasing phyto-available Cd in a contaminated soil. In the hydroponic experiment, Fe sufficiency significantly reduced Cd concentrations in above-ground organs. In some cases, the expression of OsIRT1, OsNRAMP1 and OsNRAMP5 was inhibited under Fe sufficiency relative to Fe deficiency conditions. These results suggest that mitigation of rice Cd by Fe chelate fertilization results from a decrease in available Cd in substrates and the inhibition of the expression of several Fe-related genes in the IRT and NRAMP families.
