Sediment grain size determines microplastic exposure landscapes for sandy beach macroinfauna

Despite the global occurrence of microplastic contamination on sandy beaches, evidence of microplastic distribution within beaches remains contradictory. When conflicting evidence is used to inform sampling surveys, it increases uncertainty in resulting data. Moreover, it hampers spatially explicit risk characterization of microplastic pollution to intertidal fauna. We aimed to guide sampling designs for microplastic monitoring on beaches, and to quantify macroinfauna exposure to microplastics. Microplastic abundance, quantified between 5 mm–66 μm, lacked a significant zonation across the top sediment layer of sub-terrestrial, upper and lower midlittoral, and swash zones at two sites with varying anthropogenic influence on a microtidal dissipative beach in Uruguay. Microplastic abundance decreased exponentially with increasing grain size, as revealed by Bayesian Poisson regression, although the decrease was less steep compared to prior knowledge regarding sediment – plastic interactions obtained for large (millimeter-sized) industrial pellets. Significant differences in microplastic contamination between the two sites with varying anthropogenic influence likely related to their proximity to a freshwater canal. Corresponding field measurements of body burdens of fibers and irregular particles were significantly lower for the polychaete Euzonus (Thoracophelia) furcifera, despite its preference for finer sediments with higher microplastic loads, compared to the isopods Excirolana braziliensis and Excirolana armata. Results provide critical insights toward representative sampling of microplastics within beach sites. Specifically, we caution against sampling limited to the drift line, and instead recommend: 1) reporting beach morphodynamic characteristics; 2) using clearly defined, ecologically-informed zonation schemes; and 3) accounting for sediment grain size as a covariate to normalize among reported contamination levels. The results contribute valuable baseline data toward realistic exposure landscapes relative to the sediment grain size preferences of macroinfauna, needed to inform laboratory experiments.

A laboratory bioassay was conducted to investigate the ecotoxicity of a chromium-enriched superfood, Spirulina platensis, on the meiofauna collected from the Ghar El Melh lagoon, Tunisia. After 1 month of exposure, the abundances of meiobenthic taxa and the taxonomic and morpho-functional diversity of nematodes showed significant differences between the Spirulina and Spirulina + chromium groups. The nematodes were more tolerant of all types of stressors compared to harpacticoids, polychaetes, and oligochaetes, and the lowest taxonomic and morpho-functional diversity of nematodes was observed in the highest sedimentary concentration of S. platensis (50% DW). The mixed treatments may have been richer in micro-habitats and subject to low selective pressure, thereby hosting nematodes with a wide range of adaptations. The responses of the nematode species differed depending on their functional traits. Spirulina enriched with chromium induced two responses for the same feeding group: high toxicity for Daptonema fallax and low toxicity for two Theristus species (T. flevensis and T. modicus). The ecotoxicity of the Spirulina/chromium mixtures were lower than that of Spirulina alone, suggesting mutual neutralization between these two elements. The association between functional traits and taxonomic diversity showed that the effects of the mixtures were not additive and that one of the stressors camouflaged the effect of the other. Our findings should encourage the commercialization of chromium-enriched S. platensis owing to its lower ecotoxicity than Spirulina alone.

Global concern about floating marine debris and its fundamental role in shaping coastal biodiversity is growing, yet there is very little knowledge about debris-associated rafting communities in many areas of the world's oceans. In the present study, we examined the encrusting assemblage on different types of stranded debris (wood, plastic, glass, and metal cans) along the Iranian coast of the Persian Gulf. In total, 21 taxa were identified on 132 items. The average frequency of occurrence (±SE) across all sites and stranded debris showed that the barnacle Amphibalanus amphitrite (68.9 ± 1.1%), the oyster Saccostrea cucullata (40.9 ± 0.7%), the polychaete Spirobranchus kraussii (27.3 ± 0.5%), green algae (22 ± 0.5%) and the coral Paracyathus stokesii (14.4 ± 0.7%) occurred most frequently. Relative substratum coverage was highest for A. amphitrite (44.3 ± 2.7%), followed by green algae (14.4 ± 1.5%), Spirobranchus kraussii (9.3 ± 1.3%), Saccostrea cucullata (7.6 ± 1.3%) and the barnacle Microeuraphia permitini (5.8 ± 0.9%). Despite the significant difference in coverage of rafting species on plastic items among different sites, there was no clear and consistent trend of species richness and coverage from the eastern (Strait of Hormuz) to the western part of the Persian Gulf. Some rafting species (bryozoans and likely barnacles) were found to be non-indigenous species in the area. As floating marine debris can transport non-indigenous species and increase the risk of bio-invasions to this already naturally- and anthropogenically-stressed water body, comprehensive monitoring efforts should be made to elucidate the vectors and arrival of new invasive species to the region.

The increasing consumption of anticancer drugs through single and/or combinatory chemotherapy worldwide raised concern regarding their toxicity burden in coastal zones. The toxicity of a mixture of three compounds involving the drugs cisplatin (CisPt), cyclophosphamide (CP) and tamoxifen (TAM) was determined on the marine polychaete Nereis diversicolor exposed to an increasing range of their concentrations, respectively: Mix A: 0.1 + 10 + 0.1 ng L−1; Mix B: 10 + 100 + 10 ng L−1; Mix C: 100 + 500 + 25 ng L−1; Mix D: 100 + 1000 + 100 ng L−1. Different endpoints were assessed, including disturbance in the burrowing behaviour, neurotoxicity (acetylcholinesterase – AChE activity), antioxidant enzymes (superoxide dismutase – SOD; catalase – CAT; selenium-dependent glutathione peroxidase – Se-GPx and total glutathione peroxidases T-GPx activities), biotransformation metabolism (glutathione-S-transferases - GST), lipid peroxidation (LPO) and genotoxicity (DNA damage). Biological effects of the mixtures of anticancer compounds on N. diversicolor were compared with previous studies about effects on the same biological model under single-drug exposure conducted with the same molecules. Regarding SOD activity, TAM showed an antagonist effect over CisPt and CP in mixtures C and D. In Mix D, there was a synergistic effect of TAM and CisPt that inhibited CAT activity and an additive interaction of CisPt and CP on the Phase II biotransformation enzyme. Drugs in Mix A also suppressed polychaetes' GST activity, although different from the respective single-drug responses, besides able to induce T-GPx activity, that was not sufficient to avoid oxidative damage and mid-grade DNA damage. Due to the absence of burrowing impairment in Mix A, mechanisms involved in neurotoxicity were other than the one driven by AChE alterations. At the intermediary concentrations (Mix B and C), only LPO occurred. Data from drugs individually may not predict the risks provided by mixtures.

Anthropogenic activities including coastal industries, urbanization, extensive agriculture and aquaculture as well as their cumulative impacts represent major sources of perturbation of marine coastal systems. Macrobenthic communities are useful ecological indicators for monitoring the health status of marine environments (or polluted environments). The present study reports, for the first time, the response of benthic macrofauna sampled during two years survey (2015–2016) to multiple anthropogenic pressures on the coastal zone south of Sfax (Tunisia). A total of 12 stations were monitored seasonally at locations downstream from the main potential sources of disturbance. 106 macrobenthos taxa, belonging to six animal phyla and 70 families, were identified with a dominance of polychaetes (42%), crustaceans (35%) and molluscs (18%). We used an ANOVA test and cluster analysis to identify spatial gradient linked to environmental and anthropogenic factors, including depth, sedimentary texture and anthropogenic activities (i.e. phosphogypsum discharges).The macrofauna present lowest species number and abundance on stations undergoing anthropogenic inputs, which are extremely polluted by heavy metals (Cd, Cu, F and N) and excess of organic matter. Univariate parameters reveal a general trend of increasing species diversity with increasing distance from the pollution source. The polluted stations are strongly dominated by carnivores, and selective deposit feeders, and more closely linked to the availability of trophic resources than to anthropogenic constraints. The seasonal changes in macrobenthic abundance, diversity indices and community structure are mainly linked to the biological cycle (e.g. recruitment events) of the dominant species. Biotic indices (AMBI and BO2A) classified the coastal zone south of Sfax as moderate and good ecological status. This study suggests that initiating a long-term monitoring programme would improve our understanding of the temporal changes of macrobenthic communities of this ecosystem, contributing to the assessment of effective management and conservation measures in this disturbed area.

The exponential production and use of plastics has generated increasing environmental release over the past decades, and microplastics (MPs) have been reported across all the oceans. Field studies have documented the occurrence of MPs in several species, but important knowledge gaps still remain. In the present study, we characterized the distribution of MPs in ten sediment-dwelling and epibenthic species representative of different habitat, feeding modes and trophic levels within the inner Oslofjord (Oslo, Norway), an area subjected to moderate anthropogenic pressures. Analysed species included fish, bivalves, echinoderms, crustaceans and polychaetes. MPs were present in all the species with a frequency up to 65% of positive individuals for some species. In most cases, 1 or 2 MPs were found per individual, but some organisms contained up to 7 particles. A total of 8 polymer typologies were identified, with PE and PP being the most common according to our extraction protocol. MP sizes ranged from 41 μm to lines as long as 9 mm. Our results indicate that occurrence of MPs in analysed biota is not influenced by organism habitat or trophic level, while characteristics and typology of polymers might be significantly affected by feeding mode of organisms.

The distribution, feeding ecology and microplastic contamination were assessed in different ontogenetic phases of Haemulidae species inhabiting the Goiana Estuary, over a seasonal cycle. Pomadasys ramosus and Haemulopsis corvinaeformis are estuarine dependent species that use habitats with specific environmental conditions each season. Pomadasys ramosus was found in the upper and middle estuaries during the rainy season, when salinity showed the lowest values. Haemulopsis corvinaeformis was found in the lower estuary during the dry season, when salinity increased in the estuary. Juveniles of P. ramosus are zooplanktivores, feeding mainly on calanoid copepods. Sub-adults and adults are zoobenthivores, feeding on invertebrates associated to the bottom, mainly Polychaeta. Juveniles of H. corvinaeformis were not found in the main channel, but sub-adults and adults showed a zoobenthivore habit, feeding mainly on Anomalocardia flexuosa (Mollusca: Bivalvia). Dietary shifts along the life cycle and the spatio-temporal relationship between their distribution and the availability of microplastics along the estuary seem to have a strong influence in the ingestion of microfilaments. The highest average ingestion of microfilaments by P. ramosus coincided with the peak of ingestion of Polychaeta by sub-adults in the upper estuary during the late rainy season. For H. corvinaeformis the highest ingestion of microfilaments coincided with the peak of ingestion of A. flexuosa by adults in the lower estuary during the late dry season. Such contamination might be attributed to the time when these phases shifted to a more diverse diet and began to forage on benthic invertebrates. Research on microplastic contamination must consider species-specific behaviour, since the intake of microplastics is dependent on patterns of distribution and trophic guild within fish assemblages.

High concentrations of DDT and metabolites (ΣDDT) have been detected in sediment and the demersal flatfish hornyhead turbot (Pleuronichtys verticalis) collected from Palos Verdes (PV), California, USA, a site contaminated with over 100 metric tons of DDT throughout 1960s–70s. This study was conducted to assess the transfer of ΣDDT from PV-sediment into polychaetes (Neanthes arenaceodentata) and hornyhead turbot, and to investigate if the responses in turbots from two different laboratory exposures mimic those in turbots caught in PV (PV-turbot). Turbot fed PV-sediment-contaminated polychaete for 7 days had liver concentrations of ΣDDT similar to PV-turbot. After 28 days, ΣDDT also accumulated in livers of turbot gavaged with a ΣDDT mixture. In vitro cell bioassays indicated significant increases of 17β-estradiol equivalents (EEQ) in turbot bile extracts as compared to the control in the 7-day study. These responses corresponded to those measured in PV-fish. Glucocorticoid receptor (GR), anti-androgen receptor (anti-AR), estrogen receptor (ER) or aryl hydrocarbon receptor (AhR) activities were also observed in extracts of PV-sediment, and PV-sediment-exposed worm. Anti-AR, AhR and GR activities were significantly higher in PV-sediment than reference sediment (San Diego, SD). Higher transcripts of hepatic VTG, ERα and ERβ were found in PV-turbot than SD-turbot, but were unaltered in fish exposed to sediment-contaminated worms for the 7-day study. In contrast, liver extracts from the 28-day treatment of ΣDDT showed lower EEQ but similar hepatic VTG and ERβ transcripts relative to those of PV-turbot. These data indicated that trophic transfer of sediment-associated DDT in 7-day exposures corresponded to field measurements of DDT residues and in vitro ER bioactivities, but failed to mimic in vivo biological effects observed in field fish. In contrast, treatment with ΣDDT alone for 28 days mimicked in vivo biological effects of DDTs in PV fish, but did not correspond to liver concentrations or in vitro bioactivities.

In the last decade studies have reported the presence of several pharmaceutical drugs in aquatic environments worldwide and an increasing effort has been done to understand the impacts induced on wildlife. Among the most abundant drugs in the environment is caffeine, which has been reported as an effective chemical anthropogenic marker. However, as for the majority of pharmaceuticals, scarce information is available on the adverse effects of caffeine on marine benthic organisms, namely polychaetes which are the most abundant group of organisms in several aquatic ecossystems. Thus, the present study aimed to evaluate the biochemical alterations induced by environmentally relevant concentrations of caffeine on the polychaete species Diopatra neapolitana and Arenicola marina. The results obtained demonstrated that after 28 days exposure oxidative stress was induced in both species, especially noticed in A. marina, resulting from the incapacity of antioxidant and biotransformation enzymes to prevent cells from lipid peroxidation. The present study further revealed that D. neapolitana used glycogen and proteins as energy to develop defense mechanisms while in A. marina these reserves were maintained independently on the exposure concentration, reinforcing the low capacity of this species to fight against oxidative stress.

The sediment-dwelling ragworm, Nereis diversicolor was exposed to sediment spiked with aqueous Cu (CuAq, CuCl2), CuO nanoparticles (CuONP) or CuO microparticles (CuOMicro) at 150 μg Cu g−1 dw sediment for 10d. Exposures to CuAq and CuOMicro caused mortality (62.5 and 37.5%, respectively), whereas mean burrowing time increased during exposure to CuAq and CuONP from 0.12 h (controls) to 19.3 and 12.2 h, respectively. All Cu treatments bioaccumulated, especially CuAq (up to 4 times more than the other treatments). Cu was roughly equally distributed among the five subcellular fractions in controls and worms exposed to CuONP or CuOMicro. In contrast, ≈50% of accumulated Cu in CuAq exposed worms was found in metal rich granules and significantly more Cu was present in heat-denatured proteins and organelles than in worms exposed to CuOMicro or in controls. Our results suggest that Cu form affects its bioaccumulation and subsequent toxicity and detoxification in a polychaete like N. diversicolor.
