Multicompartment and cross-species monitoring of contaminants of emerging concern in an estuarine habitat 全文

The fate of many chemicals in the environment, particularly contaminants of emerging concern (CEC), have been characterised to a limited extent with a major focus on occurrence in water. This study presents the characterisation, distribution and fate of multiple chemicals including pharmaceuticals, recreational drugs and pesticides in surface water, sediment and fauna representing different food web endpoints in a typical UK estuary (River Colne, Essex, UK). A comparison of contaminant occurrence across different benthic macroinvertebrates was made at three sites and included two amphipods (Gammarus pulex &Crangon crangon), a polychaete worm (Hediste diversicolor) and a gastropod (Peringia ulvae). Overall, multiple contaminants were determined in all compartments and ranged from; <LOQ – 386 ng L⁻¹ in surface water (n = 59 compounds), <LOQ – 146 ng g⁻¹ in sediment (n = 39 compounds) and <LOQ – 91 ng g⁻¹ biota (n = 33 compounds). H. diversicolor and P. ulvae (sediment dwellers) showed greater chemical body burden compared with the two swimming amphipod species sampled (up to 2.5 - 4-fold). The most frequently determined compounds in biota (100%, n = 36 samples) included; cocaine, benzyoylecgonine, carbamazepine, sertraline and diuron. Whilst some of the highest concentrations found were in species H. diverscolor and P. ulvae for psychoactive pharmaceuticals including citalopram (91 ng g⁻¹), sertraline (69 ng g⁻¹), haloperidol (66 ng g⁻¹) and the neonicotinoid, imidacloprid (33 ng g⁻¹) Sediment was noted as an important exposure route for these benthic dwelling organisms and will be critical to monitor in future studies. Overall, the analysis of multiple species and compartments demonstrates the importance of including a range of exposure pathways in order to appropriately assess chemical fates and associated risks in the aquatic environment.

Global concern about floating marine debris and its fundamental role in shaping coastal biodiversity is growing, yet there is very little knowledge about debris-associated rafting communities in many areas of the world's oceans. In the present study, we examined the encrusting assemblage on different types of stranded debris (wood, plastic, glass, and metal cans) along the Iranian coast of the Persian Gulf. In total, 21 taxa were identified on 132 items. The average frequency of occurrence (±SE) across all sites and stranded debris showed that the barnacle Amphibalanus amphitrite (68.9 ± 1.1%), the oyster Saccostrea cucullata (40.9 ± 0.7%), the polychaete Spirobranchus kraussii (27.3 ± 0.5%), green algae (22 ± 0.5%) and the coral Paracyathus stokesii (14.4 ± 0.7%) occurred most frequently. Relative substratum coverage was highest for A. amphitrite (44.3 ± 2.7%), followed by green algae (14.4 ± 1.5%), Spirobranchus kraussii (9.3 ± 1.3%), Saccostrea cucullata (7.6 ± 1.3%) and the barnacle Microeuraphia permitini (5.8 ± 0.9%). Despite the significant difference in coverage of rafting species on plastic items among different sites, there was no clear and consistent trend of species richness and coverage from the eastern (Strait of Hormuz) to the western part of the Persian Gulf. Some rafting species (bryozoans and likely barnacles) were found to be non-indigenous species in the area. As floating marine debris can transport non-indigenous species and increase the risk of bio-invasions to this already naturally- and anthropogenically-stressed water body, comprehensive monitoring efforts should be made to elucidate the vectors and arrival of new invasive species to the region.

The distribution, feeding ecology and microplastic contamination were assessed in different ontogenetic phases of Haemulidae species inhabiting the Goiana Estuary, over a seasonal cycle. Pomadasys ramosus and Haemulopsis corvinaeformis are estuarine dependent species that use habitats with specific environmental conditions each season. Pomadasys ramosus was found in the upper and middle estuaries during the rainy season, when salinity showed the lowest values. Haemulopsis corvinaeformis was found in the lower estuary during the dry season, when salinity increased in the estuary. Juveniles of P. ramosus are zooplanktivores, feeding mainly on calanoid copepods. Sub-adults and adults are zoobenthivores, feeding on invertebrates associated to the bottom, mainly Polychaeta. Juveniles of H. corvinaeformis were not found in the main channel, but sub-adults and adults showed a zoobenthivore habit, feeding mainly on Anomalocardia flexuosa (Mollusca: Bivalvia). Dietary shifts along the life cycle and the spatio-temporal relationship between their distribution and the availability of microplastics along the estuary seem to have a strong influence in the ingestion of microfilaments. The highest average ingestion of microfilaments by P. ramosus coincided with the peak of ingestion of Polychaeta by sub-adults in the upper estuary during the late rainy season. For H. corvinaeformis the highest ingestion of microfilaments coincided with the peak of ingestion of A. flexuosa by adults in the lower estuary during the late dry season. Such contamination might be attributed to the time when these phases shifted to a more diverse diet and began to forage on benthic invertebrates. Research on microplastic contamination must consider species-specific behaviour, since the intake of microplastics is dependent on patterns of distribution and trophic guild within fish assemblages.

The exponential production and use of plastics has generated increasing environmental release over the past decades, and microplastics (MPs) have been reported across all the oceans. Field studies have documented the occurrence of MPs in several species, but important knowledge gaps still remain. In the present study, we characterized the distribution of MPs in ten sediment-dwelling and epibenthic species representative of different habitat, feeding modes and trophic levels within the inner Oslofjord (Oslo, Norway), an area subjected to moderate anthropogenic pressures. Analysed species included fish, bivalves, echinoderms, crustaceans and polychaetes. MPs were present in all the species with a frequency up to 65% of positive individuals for some species. In most cases, 1 or 2 MPs were found per individual, but some organisms contained up to 7 particles. A total of 8 polymer typologies were identified, with PE and PP being the most common according to our extraction protocol. MP sizes ranged from 41 μm to lines as long as 9 mm. Our results indicate that occurrence of MPs in analysed biota is not influenced by organism habitat or trophic level, while characteristics and typology of polymers might be significantly affected by feeding mode of organisms.

In the last decade studies have reported the presence of several pharmaceutical drugs in aquatic environments worldwide and an increasing effort has been done to understand the impacts induced on wildlife. Among the most abundant drugs in the environment is caffeine, which has been reported as an effective chemical anthropogenic marker. However, as for the majority of pharmaceuticals, scarce information is available on the adverse effects of caffeine on marine benthic organisms, namely polychaetes which are the most abundant group of organisms in several aquatic ecossystems. Thus, the present study aimed to evaluate the biochemical alterations induced by environmentally relevant concentrations of caffeine on the polychaete species Diopatra neapolitana and Arenicola marina. The results obtained demonstrated that after 28 days exposure oxidative stress was induced in both species, especially noticed in A. marina, resulting from the incapacity of antioxidant and biotransformation enzymes to prevent cells from lipid peroxidation. The present study further revealed that D. neapolitana used glycogen and proteins as energy to develop defense mechanisms while in A. marina these reserves were maintained independently on the exposure concentration, reinforcing the low capacity of this species to fight against oxidative stress.

Biological stress responses in individuals are used as indicators of pollution in aquatic ecosystems, but detecting ecologically relevant responses in whole communities remains a challenge. We developed an experimental approach to detect the effects of pollution on estuarine communities using field-based mesocosms. Mesocosms containing defaunated sediments from four estuaries in southeastern Australia that varied in sediment contamination were transplanted and buried in sediments of the same four estuaries for six weeks. Mesocosm sediment properties and metal concentrations remained representative of their source locations. In each estuary, fauna communities associated with sediments derived from the site with the highest metal concentrations were significantly different from other communities. This pattern was evident for some of the individual taxa, in particular the polychaete Capitella sp. Consistent responses across estuaries suggest numbers of individuals, and especially Capitella sp., could be used to identify contaminated sediments in estuaries with similar fauna and site characteristics.

Fitness, (biometric measurements, reproduction) and behaviour that are ecologically relevant biomarkers in assessing the quality of estuarine sediments were studied by comparing the responses of the polychaete worm Nereis diversicolor – a key species in estuaries – along a pollution gradient. Intersite differences were shown for all the measured parameters: size–weight relationships, energy reserves as glycogen and lipids, sexual maturation patterns, total number of oocytes per female, total and relative fecundity, burrowing behaviour. The physiological and behavioural status of N. diversicolor was consistently disturbed in the larger, most contaminated estuaries (Loire and Seine, Fr.) compared to reference sites (Bay of Bourgneuf, Goyen estuary, Fr.). Many classes of potentially toxic chemicals present in these estuaries most likely contribute to these impairments but food availability may act as a confounding factor, interfering with the potential impact of contaminants. Fitness, and behaviour in Nereis diversicolor are affected by anthropogenic pressure.

Desalination is a growing activity that has introduced a new impact, brine discharge, which may affect benthic communities. Although the role of polychaetes as indicators to assess organic pollution is well known, their tolerance to salinity changes has not been examined to such a great extent. The aim of this study was to examine the effect of brine discharge over soft bottom polychaete assemblage along the Alicante coast (Southeast Spain) over a two year period. Changes in the polychaete assemblage was analysed using univariate and multivariate techniques. We compared a transect in front of the discharge with two controls. At each transect we sampled at three depths (4, 10 and 15 m) during winter and summer. We have observed different sensitivity of polychaete families to brine discharges, Ampharetidae being the most sensitive, followed by Nephtyidae and Spionidae. Syllidae and Capitellidae showed some resistance initially, while Paraonidae proved to be a tolerant family. The Polychaete assemblage is affected by the brine discharge of the Alicante desalination plant and we detect different sensitivity levels in polychaete families to brine impact.

Estuaries are subject to intense human use globally, with impacts from multiple stressors, such as metal contaminants. A key challenge is extending beyond traditional monitoring approaches to understand effects to biota and system function. To explore the metabolic effects of complex metal contaminants to sediment dwelling (benthic) fauna, we apply a multiple-lines-of-evidence approach, coupling environmental monitoring, benthic sampling, total metals analysis and targeted metabolomics.We characterise metabolic signatures of metal exposure in three benthic invertebrate taxa, which differed in distribution across sites and severity of metal exposure: sipunculid (very high), amphipod (high), maldanid polychaete (moderate). We observed sediment and tissue metal loads far exceeding sediment guidelines where toxicity-related adverse effects may be expected, for metals including, As, Cd, Pb, Zn and Hg.Change in site- and taxa-specific metabolite profiles was highly correlated with natural environmental drivers (sediment total organic carbon and water temperature). At the most metal influenced sites, metabolite variation was also correlated with sediment metal loads. Using supervised multivariate regression, taxa-specific metabolic signatures of increased exposure and possibility of toxic effects were characterised against multiple reference sites. Metabolic signatures varied according to each taxon and degree of metal exposure, but primarily indicated altered cysteine and methionine metabolism, metal-binding and elimination (lysosomal) activity, coupled to change in complex biosynthesis pathways, responses to oxidative stress, and cellular damage.This novel multiple-lines-of-evidence approach combining metabolomics with traditional environmental monitoring, enabled detection and characterisation of chronic metal exposure effects in situ in multiple invertebrate taxa. With capacity for application to rapid and effective monitoring of non-model species in complex environments, these approaches are critical for improved assessment and management of systems that are increasingly subject to anthropogenic drivers of change.

Although the hazards of microplastics (MPs) have been quite well explored, the aberrant metabolism and the involvement of the autophagy pathway as an adverse response to environmental MPs in benthic organisms are still unclear. The present work aims to assess the impact of different environmental MPs collected from the south coast of the Mediterranean Sea, composed by polyethylene (PE), polyethylene vinyl acetate (PEVA), low-density polyethylene (LDPE), high-density polyethylene (HDPE), polypropylene (PP) and polyamide (PA) on the metabolome and proteome of the marine polychaete Hediste diversicolor. As a result, all the microplastic types were detected with Raman microspectroscopy in polychaetes tissues, causing cytoskeleton damage and induced autophagy pathway manifested by immunohistochemical labeling of specific targeted proteins, through Tubulin (Tub), Microtubule-associated protein light chain 3 (LC3), and p62 (also named Sequestosome 1). Metabolomics was conducted to further investigate the metabolic alterations induced by the environmental MPs-mixture in polychaetes. A total of 28 metabolites were differentially expressed between control and MPs-treated polychaetes, which showed elevated levels of amino acids, glucose, ATP/ADP, osmolytes, glutathione, choline and phosphocholine, and reduced concentration of aspartate. These novel findings extend our understanding given the toxicity of environmental microplastics and unravel their underlying mechanisms.
