Metagenomic analysis reveals the diversity and distribution of antibiotic resistance genes in thermokarst lakes of the Yellow River Source Area 全文

Thermokarst lakes form as the results of ice-rich permafrost thawing and act as important water resources in cold regions. However, the distributions of antibiotic resistance genes (ARGs) in thermokarst lakes are far less studied. Using metagenomic sequencing approach, we provided the first study to document ARGs in thermokarst lakes of the Yellow River Source Area on the Qinghai-Tibet Plateau (QTP). The results revealed that both sediment and water of the thermokarst lakes harbor diverse ARGs. Multidrug resistance genes were the most diverse, while rifamycin resistance genes were the most abundant with rpoB2 and rpoB genes having the highest proportion. Sediment samples contained more ARGs than water samples, but their composition differed between the two types of samples. However, the composition variations of sediment and water ARGs were closely correlated. The Sorensen dissimilarities of ARGs were controlled by strong turnover processes in sediment samples, and by turnover and nestedness in water samples. High contributions of nestedness were found between sediment and water samples. Moreover, ARGs in water had more significant relationships with environmental variables than that in sediment. Given the role of thermokarst lakes as important water resources in permafrost landscape, as well as intensifying influences of climate change and anthropogenetic activities, thermokarst lakes could bring potential ARG risks, warranting further investigation and evaluation.

Organophosphate ester (OPE) levels, profiles, sources, spatial distribution, and partitioning were firstly studied in the rivers of the Shandong Peninsula. A total of 53 water samples and 45 sediment samples were collected from the rivers and the sewage treatment plant in the peninsula to quantitate levels of 13 targeted OPEs. Total OPE concentrations ranged from 263 to 6676 ng L⁻¹ in the water, and 39.3–360 ng g⁻¹ in the sediment. TEP, TCPP, and TCEP together contributed more than 90% of total OPE content. TCEP and TCPP concentrations in the Xiaoqing River sediment were increased by approximately two and seven times from 2014 to 2019, respectively. Total OPE concentrations generally increased from upstream regions to the estuaries. The main OPE sources were municipal effluent in the Jiaozhou Bay (JZB) watershed and chemical industrial wastewater in the Laizhou Bay (LZB) watershed. TCPP, TEP, and TCEP were generally approaching equilibrium between sediment and overlying water, while TNBP, TIBP, and TBOEP effectively transferred from the overlying water to the sediment. The riverine OPE flux was 0.66 ton/year to JZB and 3.58 ton/year to the LZB. TCPP and TCEP in municipal effluent, and TEP in chemical industrial wastewater should be regulated to protect Shandong Peninsula waters.

The emergence of resistance genes is a global phenomenon that poses a significant threat to both animals and humans. Lakes are important reservoirs of genes that confer resistant to antibiotics and metals. In this study, we investigated the distribution and diversity of antibiotic resistance genes (ARGs) and metal resistance genes (MRGs) in the sediment of Daihai Lake using high-throughput sequencing and metagenomic analysis. The results indicated that all sampling sites had similar bacterial community structures, with Proteobacteria, Actinobacteria, Firmicutes, and Bacteroidetes being the most abundant. A total of 16 ARG types containing 111 ARG subtypes were deposited in the sediment. Among the resistance genes to bacitracin, multidrug, macrolide-lincosamide-streptogramin (MLS), tetracycline, beta-lactam, and sulfonamide were the dominant ARG types, accounting for 89.9–94.3% of the total ARGs. Additionally, 15 MRG types consisting of 146 MRG subtypes were identified. In all samples, MRGs of the same type presented resistance to Pb, Ni, Hg, W, Zn, Ag, Cr, Fe, As, Cu, and multimetals. Overall, the distribution and diversity of antibiotic and metal resistance genes showed no significant differences in the samples. Plasmids (91.03–91.82%) were the most dominant mobile genetic elements in the sediments of Daihai Lake. Network analysis indicated that the target ARGs and MRGs were significantly positively correlated with the microorganisms. Potential hosts for various ARGs and MRGs include Proteobacteria, Euryarchaeota, Actinobacteria, Chloroflexi, and Bacteroidetes.

Algal blooms (ABs) can affect the migration of phosphorus (P) among sediments, interstitial water and overlying water. It is important to analyze the characteristics of P and their interactions in the three media during ABs. A 5-month field study (June to October in 2016) was conducted in Zhushan Bay of Lake Taihu. P fractions, P adsorption characteristics and P diffusion fluxes at the sediment-water interface (SWI) were investigated. During the outbreak period of ABs from June to August, labile P concentrations in the sediment measured by diffusive gradients in thin films (DGT-labile P) and its diffusion fluxes across the SWI increased significantly. The equilibrium P concentration (EPC₀) of the sediment was higher than the PO₄³⁻-P concentration in the overlying water. During the period of decline of ABs from September to October, the concentrations and diffusion fluxes of DGT-labile P sharply decreased. However, the sediment total P (TP), overlying water TP, total dissolved P (TDP) and PO₄³⁻-P concentrations increased. These results show that the ability of sediment solids to supplement interstitial water labile P was significantly enhanced by the outbreak of ABs. Labile P was then intensively released into the overlying water by interstitial water. Degraded algae became a crucial P source during the period of decline of ABs. P from the degraded algae was re-released to the sediment and overlying water. The observed DGT-labile P and DGT-labile Fe coupling in June, September and October confirmed the Fe redox-driven P release mechanism in sediment during these periods. The decoupling of DGT-labile P and DGT-labile Fe was observed in July and August and was probably caused by algal decomposition, labile organic P degradation and/or sulfate reduction in sediment stimulated by the ABs outbreak.

As reservoirs for pollutants transported via the Yangtze and Yellow Rivers, the Bohai Sea (BS) and Yellow Sea (YS) play an important role in transporting microplastics (MPs) to the Pacific Ocean. The fate, sources and mass budget of MPs in the BS and the YS were investigated by Pearson correlation, principal component analysis-multilinear regression analysis (PCA-MRLA) and a mass balance model to sedimentary MPs data. Average MP abundances were 137 and 119 items kg⁻¹ in the Bohai and Yellow Seas, respectively. MPs <1000 μm exhibited similar distribution patterns to total organic carbon and fine-grained sediments, while MPs >1000 μm were confined in the BS and exhibited a strong positive correlation with chlorophyll-a and polyethylene terephthalate, suggesting that larger MPs might deposit faster due to biofouling or when comprised of high density polymers. PCA-MLRA analysis indicated land-based inputs (packing materials, textile material and daily commodities) were dominant in the BS, while maritime activities (fishing and mariculture) were the main source of MPs in the YS. The mass balance model revealed that the total MP input and output to the BS and the YS was 3396.92 t yr⁻¹ and 3814.81 t yr⁻¹, respectively. The major input pathway of MPs to the BS and the YS were river discharge and air deposition, respectively. Notably, 94% of MPs in the BS and the YS were deposited to sediments. This study revealed that BS and YS sediments play an important role in preventing MPs from being further transported to the Pacific Ocean, thus more attention should be paid to local ecological risk assessment.

To reduce direct discharges of surface runoff to receiving waters, separate sewer systems have been implemented, with runoff retention basins (RRB) for pollutant pretreatment by sedimentation and infiltration. However, due to frequent and intense precipitation events, most RRBs are overwhelmed by runoff resulting in overflow into the receiving freshwater bodies. Hence, the present study evaluates the impact of traffic-related runoff overflow on metal concentrations in sediment and Gammarus sp. Downstream of the RRB outfall in the receiving stream. Samples were collected from the RRB, upstream (reference site) and at different distances downstream from the RRB outfall in the stream. The samples were analyzed for the presence and distribution of metals using ICP-MS. Furthermore, ecotoxicological effects of the overflow on benthic species were assessed using Lumbriculus variegatus exposed to the field sediments. Our findings reveal that overflow of the RRB results in elevated traffic-related metal concentrations in sediment and biota of the stream. Within the first 50 m downstream increased sediment metal concentrations were found. The gammarids downstream of the RRB outfall showed an increased accumulation of several metals. Similarly, the metals were found to be taken up by the endobenthic L. variegatus under laboratory conditions and the bioaccumulation pattern was related to the sediment concentrations. Bioaccumulation by both organisms is an indication that overflow of the RRB also leads to uptake of increased element amounts in organisms downstream. Laboratory-based studies addressing standard toxicity endpoints showed no clear toxic effects on growth and reproduction. However, elevated levels of metallothioneins were measured in the annelids during the test period. This indicates a physiological response induced by increased metal concentrations due to RRB overflow. Hence, the results of this study show that discharges by the RRB increase the metal concentration in the receiving stream with the possibility of adverse effects on organisms.

Estuaries provide critical habitat for food webs supporting fish and shellfish consumed by humans, but estuarine ecosystem health has been threatened by increases in nitrogen loading as well as inputs of the neurotoxin, mercury (Hg), which biomagnifies in food webs and poses risk to humans and wildlife. In this study, the effects of nutrient loading on the fate of Hg in shallow coastal estuaries were examined to evaluate if their interaction enhances or reduces Hg bioavailability in sediments, the water column, and concentrations in lower trophic level fish (Fundulus heteroclitus and Menidia menidia). Multiple sites were sampled within two human impacted coastal lagoons, Great South Bay (GSB) and Jamaica Bay (JB), on the southern coast of Long Island, NY, United States of America (U.S.A.). Carbon (C), nitrogen (N), sulfur (S), Hg, and methylmercury (MeHg) were measured in surface sediments and the water column, and total Hg (THg) was measured in two species of forage fish. Minimal differences were found in dissolved and particulate Hg, dissolved organic carbon (DOC), and salinity between the two bays. Across lagoons, concentrations of chlorophyll-a were correlated with total suspended solids (TSS), and water column THg and MeHg was largely associated with the particulate fraction. Methylmercury concentrations in particulates decreased with increasing TSS and chlorophyll-a, evidence of biomass dilution of MeHg with increasing productivity at the base of the food chain. Water column Hg was associated with THg concentrations in Atlantic silversides, while mummichog THg concentrations were related to sediment concentrations, reflecting their different feeding strategies. Finally, higher nutrient loading (lower C:N in sediments) while related to lower particulate concentrations coincided with higher bioaccumulation factors (BAF) for Hg in both fish species. Thus, in shallow coastal lagoons, increased nutrient loading resulted in decreased Hg concentrations at the base of the food web but resulted in greater bioaccumulation of Hg to fish relative to its availability in algal food.

Microplastics can have several negative consequences on a variety of organisms, and their prevalence in marine ecosystems has become a major concern. Researchers have recently focused their attention on the world's largest gulf, the Gulf of Mexico (GoM), to determine and assess the impact of microplastic pollution on various environmental compartments (i.e., water, sediment, and biota). This paper critically reviews the analytical methodologies as well as summarizes the distribution, accumulation, sources, and composition of microplastics in a handful of studies (n = 14) conducted in the Gulf of Mexico (GoM) covering countries like the USA (n = 10) and Mexico (n = 4). Current quality control measures with respect to sampling and microplastic extraction are summarized. Of 14 studies reviewed, 47% primarily focused on examining sediments for microplastics, with biota and water comprising 35% and 18%, respectively. The abundance ranged from 31.7 to 1392 items m⁻² and 60–1940 items kg⁻¹ in sediment, 12–381 particles L⁻¹ in water, and 1.31–4.7 particles per fish in biota. Irregular shaped fragments were the most abundant, followed by fiber, film, foam, hard, and beads etc. Different polymer types of microplastics have been found, including polyethylene, polypropylene, polystyrene, polyamide, nylon, and rayon etc. According to published research, 46 out of 100 fish thriving in this region are susceptible to microplastic ingestion. Although microplastic concentration in the GoM is among the highest found worldwide, the determination of microplastic contamination is still a growing field of research and methodological discrepancies largely limit the realization of establishing a baseline information on the microplastic abundance of the GoM. In this respect, considerable efforts must be dedicated towards evaluating their distribution and exposure levels; thereby, major challenges and future research directions are briefly discussed.

Phosphorus (P) cycling present in sediments associated with iron (Fe), manganese (Mn) and sulfur (S) geochemical processes may cause secondary pollution in overlying water. Understanding the mechanisms of P release from sediments should help to restore water quality. This study used the diffusive gradients in thin film (DGT) technique to investigate the seasonal variation in the lability, remobilization mechanisms, and release characteristics of sediment P in the uncontaminated Xizhi River and the severely contaminated Danshui River, South China. P accumulation in sediments contributed to higher DGT-labile P concentrations in contaminated reaches, and the highest labile P concentrations were generally observed in summer season at each site. The significant positive relationships (p < 0.05) between labile Fe and P confirmed the Fe–P coupling release mechanism in uncontaminated sediments. Stronger relationships between labile Mn and P at contaminated sites indicated that Mn oxides played an important role in P remobilization. However, sulfate reduction associated with microbial activities (crucial genera: Desulfobulbus, Desulfomicrobium and Desulforhabdus) was considered to decouple the Fe & Mn–P cycling relationship, promoting P release at contaminated sites. The effluxes of sediment P were much higher in the Danshui River (mean 0.132 mg cm⁻²·d⁻¹) than in the Xizhi River (mean 0.038 mg cm⁻²·d⁻¹). And hot season led to growth in P effluxes that was much greater in contaminated river.

Studies of microplastics (MPs) in remote, trans-boundary and alpine rivers are currently lacking. To understand the sinks and transport mechanisms of MPs, this study investigated the distributions and sources of MPs in the surface waters and sediments of five tributaries of the Koshi River (KR), a typical alpine river in the Himalayas between China and Nepal. Mean abundances of MPs in water and sediment were 202 ± 100 items/m³ and 58 ± 27 items/kg, dry weight, respectively. The upstream tributary, Pum Qu in China, had the smallest abundance of MPs, while the middle tributary, Sun Koshi in Nepal, had the greatest abundance. Compared to international values in rivers, contamination of the KR with MPs was low to moderate. Fibers represented 98% of all MP particles observed, which consisted of polyethylene, polyethyleneterephthalate, polyamide, polypropylene, and polystyrene. Blue and black MPs were prevalent, and small MPs (<1 mm) accounted for approximately 60% of all MPs. Atmospheric transmission and deposition were considered to be the principal sources of MPs in the upstream tributary. The results imply that point sources associated with mostly untreated sewage effluents and solid wastes from households, major settlements, towns, and cities were most important sources of MPs in the KR. Non-point sources from agricultural runoff and atmospheric transport and deposition in the middle stream tributaries also contribute a part of microplastics, while the least amount was from fishing in the downstream tributary. Urbanization, agriculture, traffic, and tourism contributed to pollution in the KR by MPs. Equations to predict abundances of MPs based on river altitudes revealed that different trends were affected by both natural and human factors within the KR basin. This study presents new insights into the magnitude of MP pollution of a remote alpine river and provides valuable data for developing MP monitoring and mitigation strategies in similar environments worldwide.
