Metal-based nanoparticles (NPs) are considered detrimental to aquatic organisms due to their potential accumulation. However, little is known about the mechanisms underlying these effects and their species-specificity. Here we used stable silver (Ag) NPs (20 nm, from 10 to 500 μg/L) with a low dissolution rate (≤2.4%) to study the bioaccumulation and biological impacts in two freshwater gastropods: Lymnaea stagnalis and Planorbarius corneus. No mortality was detected during the experiments. Ag bioaccumulation showed a dose-related increase with an enhanced concentration in both species after 7d exposure. L. stagnalis displayed a higher accumulation for AgNPs than P. corneus (e.g., up to 18- and 15-fold in hepatopancreas and hemolymph, respectively) which could be due to the more active L. stagnalis having greater contact with suspended AgNPs. Furthermore, the hepatopancreas and stomach were preferred organs for bioaccumulation compared to the kidney, mantle and foot. Regarding biological responses, the hemolymph rather than hepatopancreas appeared more susceptible to oxidative stress elicited by AgNPs, as shown by significantly increasing lipid peroxidation (i.e., formation of malondialdehyde). Neurotoxicity was detected in L. stagnalis when exposed to high concentrations (500 μg/L). Comparison with impacts elicited by dissolved Ag revealed that the effects observed on AgNPs exposure were mainly attributable to NPs. These results highlighted the relationship between the physiological traits, bioaccumulation, and toxicity responses of these two species to AgNPs and demonstrated the necessity of species-specificity considerations when assessing the toxicity of NPs.

Nanosilver (NAg) is currently one of the major alternative antimicrobials to control microorganisms. With its broad-spectrum efficacy and lucrative commercial values, NAg has been used in medical devices and increasingly, in consumer products and appliances. This widespread use has inevitably led to the release and accumulation of the nanoparticle in water and sediment, in soil and even, wastewater treatment plants (WWTPs). This Article describes the physical and chemical transformations of NAg as well as the impact of the nanoparticle on microbial communities in different environmental settings; how the nanoparticle shifts not only the diversity and abundance of microbes, including those that are important in nitrogen cycles and decomposition of organic matters, but also their associated genes and in turn, the key metabolic processes. Current findings on the microbiological activity of the leached soluble silver, solid silver particulates and their respective transformed products, which underpin the mechanism of the nanoparticle toxicity in environmental microbes, is critically discussed. The Article also addresses the emerging evidence of silver-driven co-selection of antibiotic resistance determinants. The mechanism has been linked to the increasing pools of many antibiotic resistance genes already detected in samples from different environmental settings, which could ultimately find their ways to animals and human. The realized ecological impact of NAg calls for more judicial use of the nanoparticle. The generated knowledge can inform strategies for a better ‘risks versus benefits’ assessment of NAg applications, including the disposal stage.

Silver nanomaterials (AgNMs) are broadly used and among the most studied nanomaterials. The underlying molecular mechanisms (e.g. protein and metabolite response) that precede phenotypical effects have been assessed to a much lesser extent. In this paper, we assess differentially expressed proteins (DEPs) and metabolites (DEMs) by high-throughput (HTP) techniques (HPLC-MS/MS with tandem mass tags, reversed-phase (RP) and hydrophilic interaction liquid chromatography (HILIC) with mass spectrometric detection). In a time series (0, 7, 14 days), the standard soil model Enchytraeus crypticus was exposed to AgNM300K and AgNO₃ at the reproduction EC20 and EC50. The impact on proteins/metabolites was clearly larger after 14 days. NM300K caused more upregulated DEPs/DEMs, more so at the EC20, whereas AgNO₃ caused a dose response increase of DEPs/DEMs. Similar pathways were activated, although often via opposite regulation (up vs down) of DEPs, hence, dissimilar mechanisms underlie the apical observed impact. Affected pathways included e.g. energy and lipid metabolism and oxidative stress. Uniquely affected by AgNO₃ was catalase, malate dehydrogenase and ATP-citrate synthase, and heat shock proteins (HSP70) and ferritin were affected by AgNM300K. The gene expression-based data in Adverse Outcome Pathway was confirmed and additional key events added, e.g. regulation of catalase and heat shock proteins were confirmed to be included. Finally, we observed (as we have seen before) that lower concentration of the NM caused higher biological impact. Data was deposited to ProteomeXchange, identifier PXD024444.

Most legacy mines contributed to contamination of the environment before and after cessation of mining. Contamination from waste rock, slag and tailings can introduce large concentrations of metals and metalloids to the surface soil and downstream sediments. Since ants are able to accumulate metals in their bodies, we investigated the possibility of using the elemental compositions of ants as indicators of metals at legacy mines developed on ores rich in copper (Cu), zinc (Zn), arsenic (As), silver (Ag) and lead (Pb). Our results showed the concentrations of manganese (Mn) and Cu in ants were not significantly different between mine and reference samples and only Zn was significantly different between contaminated and reference areas. Crematogaster spp. and Notoncus spp. from reference areas accumulated larger concentrations of metals in their bodies compared to ants from the mine. Ants accumulated metals in different parts of their bodies. The abdomen was the main site for accumulation of Mn, iron (Fe) and Zn. Mandibles were only associated with accumulation of Zn. Copper and Pb showed no area of preferential accumulation and traces were detected in the whole body of the ants. Ants from five genera had similar regions for metal accumulation. The exoskeleton did not contribute to accumulation of metals; instead all metals were stored in internal organs. Not all genera were suitable for use as indicators; only Iridomyrmex spp. and Ochetellus spp. accumulated larger amount of metals in mine samples compared to reference samples.

Aerobic performance in fish is linked to individual and population fitness and can be impacted by anthropogenic contaminants. Exposure to some engineered nanomaterials, including silver nanoparticles (nAg), reduces rates of oxygen consumption in some fish species, but the underlying mechanisms remain unclear. In addition, their effects on swim performance have not been studied. Our aim was to quantify the impact of exposure to functionalized nAg on aerobic scope and swim performance in rainbow trout (Oncorhychus mykiss) and to characterize the contribution of changing rates of protein synthesis to these physiological endpoints. Fish were exposed for 48 h to 5 nm polyvinylpyrolidone-functionalized nAg (nAgPVP; 100 μg L⁻¹) or 0.22 μg L⁻¹ Ag⁺ (as AgNO₃), which was the measured quantity of Ag released from the nAgPVP over that time period. Aerobic scope, critical swimming speed (Ucᵣᵢₜ), and fractional rates of protein synthesis (Kₛ), were then assessed, along with indicators of osmoregulation and cardiotoxicity. Neither nAgPVP, nor Ag⁺ exposure significantly altered aerobic scope, its component parts, or swim performance. Kₛ was similarly unaffected in 8 tissue types, though it tended to be lower in liver of nAgPVP treated fish. The treatments tended to decrease gill Na⁺/K⁺-ATPase activity, but effects were not significant. The latter results suggest that a longer or more concentrated nAgPVP exposure may induce significant effects. Although this same formulation of nAgPVP is bioactive in other fish, it had no effects on rainbow trout under the conditions tested. Such findings on common model animals like trout may thus misrepresent the safety of nAg to more sensitive species.

The exposure risk of metal-based nanoparticles (NPs) to marine organisms and related food safety have attracted increasing attention, but the actual concentrations of these NPs in seawater and marine organisms are unknown. In this work, single particle inductively coupled plasma-mass spectrometry (spICP-MS) was used to quantify the concentrations and size distributions of NPs in different marine mollusks (oysters, mussels, scallops, clams, and ark shells) from an offshore aquaculture farm. Results showed that Ti, Cu, Zn, and Ag bearing NPs were detected in all the five mollusks with the mean sizes at 65.4–70.9, 72.2–89.6, 97.8–108.3, and 42.9–51.0 nm, respectively. The particle concentrations of Ti, Cu, Zn, and Ag bearing NPs in all mollusks (0.88–3.26 × 10⁷ particles/g fresh weight) were much higher than that in the seawater (0.46–0.79 × 10⁷ particles/mL), suggesting bio-accumulation of NPs. For all the five mollusks, Ag bearing NPs had the highest number-based bioconcentration factors (NBCFs) in all the tested NPs due to the smallest mean size of Ag bearing NPs in seawater (30.5 nm). In addition, the clams exhibited the lowest NBCFs of the four NPs than other mollusks. All four NPs were mainly accumulated in the gill and digestive gland, and could transfer to adductor muscle of all mollusks. Although all the four metals (Ti, Cu, Zn, Ag) in mollusks were safe for human consumption by the estimated daily intake (EDI) analysis, the risk of NPs remaining in the mollusks should be further considered when evaluating the toxicity of metals for human health. The findings could improve our understanding on the distribution and health risk of NPs in marine mollusks under offshore aquaculture.

With the burst of silver nanoparticles (AgNPs) applications, their potential entry into the environment has attracted increasing concern. To date, researches about the impacts of AgNPs on microbial communities have been scarcely conducted in the brackish waters. Here, the effects of interactions of AgNPs and Ag⁺ (as a positive control) with dissolved oxygen on natural brackish water microbial communities were investigated for 30 d. The introduction of AgNPs and Ag⁺ in natural brackish waters resulted in distinct bacterial community composition and structure as well as reduction of the richness and diversity, effects that were not eliminated completely during the tested periods. Anoxic conditions could attenuate the effects of AgNPs and Ag⁺ on the community, and dissolved oxygen made more contributions to community compositions for short-term exposure. High doses of AgNPs had more pronounced long-term impacts than Ag⁺ amendment. Compared with the controls, two general AgNP and Ag⁺ responses, namely, sensitivity and resistance, were observed. Sensitive species mainly included those of the genera Synechococcus and unclassified_f_Rhodobacteraceae, while resistant species mostly belonged to the phylum Bacteroidetes and participated in carbon metabolic processes. Our results indicated that the microbial communities that were involved in nutrient cycles (such as carbon, nitrogen, and sulfide) and photoautotrophic bacteria that contained bacteriochlorophyll were adversely affected by AgNPs and Ag⁺. In addition, dissolved oxygen could further change the microbial communities. These results implied that under different oxygen conditions AgNPs possibly resulted in varying microbial survival strategies and affected the biogeochemical cycling of nutrients in natural brackish waters.

Photovoltaic industry has shown tremendous growth among renewable energy sector. Though, this high installation rate will eventually result in generation of large volume of end-of-life photovoltaic waste with hazardous metals. In present study, reported leached metal contents from different photovoltaics in previous investigations were utilized for (i) potential fate and transport analysis to soil and groundwater and, (ii) estimating ecological and human health risks via dermal and ingestion pathways for child and adult sub-populations. The results indicate that the children are at highest risk, mainly due to lead (hazard quotient from 1.2 to 2.6). Metals, such as cadmium, lead, indium, molybdenum and tellurium pose maximum risks for child and adult sub-populations via soil-dermal pathway followed by soil-ingestion pathway. This is further proved by calculated high values of contamination factor and geo-accumulation index for cadmium (102.4), indium (238.9) and molybdenum (16.12). The estimated soil contamination is significant with respect to aluminium, silver, cadmium, iron, lead, however, groundwater contamination was insignificant. Exposure to polluted soils yields an aggregate hazard index (for non-cancer effects) > 1 for all four pathways, with soil dermal pathway as the major contributor. Lead poses significant cancer risk for all scenarios (average risk: 0.0098 to 0.047 (soil) and 2.1 × 10⁻⁵ to 3.5 × 10⁻⁵ (groundwater)), whereas acceptable non-cancer risk was observed for other metals from groundwater exposure. Further, variance contribution and spearman correlation coefficient analysis show that metal concentration, exposure frequency and ingestion rate are the main contributors towards overall uncertainty in risk estimates. More detailed assessment for environmentally-sensitive metals should be carried out by considering other field breakage scenarios also, although the assessment suggests low risk for majority of metals examined.

The widespread production and use of silver nanomaterials (AgNMs) in consumer and medical products have been raising environmental concerns. Once in the environment, the soil is one of the major sinks of AgNMs due to e.g. sewage sludge applications, and invertebrates are directly exposed. In this study, we investigate the potential of N-acetylcysteine (NAC) to reduce the toxic effects of Ag NM300 K (and AgNO3) on the soil invertebrate Enchytraeus crypticus. Ag NM300 K induces mortality, reproduction impairment, and avoidance. The addition of NAC to the soil showed a remarkable reduction in the toxicity of Ag, indicating that NAC can act as a detoxifying agent for terrestrial organisms exposed to Ag materials. That the reduction in toxicity likely is caused by thiol groups, was confirmed by GSH and GSSH studies. Identifying the mechanisms and hence alternatives that allow the recovery of contaminated soils is an important mitigation measure to promote environmental safety and reduce the associated risks to human health. Further, it may inform on strategies to implement in safe-by-design industry development.

Behavioural responses to contaminants are an important endpoint in ecotoxicology because they link effects at biochemical or cellular levels to impacts on individual fitness. Due to the increasing use of silver in nanomaterials, studies of its effects on the behaviour of aquatic organisms are important to assess the risks of silver nanoparticles (AgNP) released into the environment. The aim of this work was to evaluate the behavioural effects of silver on the marine amphipod Echinogammarus marinus after exposure to AgNO3 via water and AgCl or AgNP via food. Swimming activity of the amphipods was tracked during 6 min alternating dark and light conditions. Animals swam slower and responded less to light at higher concentrations of silver in the water. No differences were found in the behaviour of animals exposed via feeding up to 28 days, hence, longer exposure times may be required for the observation of effects. This is the first work to appraise behaviour effects of silver ions and AgNP on marine amphipods. Although the protocol has been successfully developed for this purpose, specimens appeared to habituate to test conditions during the experiments. Therefore, the need for further understanding of baseline behaviours in these model organisms is discussed.
