Private wells in Ireland and elsewhere have been shown to be prone to microbial contamination with the main suspected sources being practices associated with agriculture and domestic wastewater treatment systems (DWWTS). While the microbial quality of private well water is commonly assessed using faecal indicator bacteria, such as Escherichia coli, such organisms are not usually source-specific, and hence cannot definitively conclude the exact origin of the contamination. This research assessed a range of different chemical contamination fingerprinting techniques (ionic ratios, artificial sweeteners, caffeine, fluorescent whitening compounds, faecal sterol profiles and pharmaceuticals) as to their use to apportion contamination of private wells between human wastewater and animal husbandry wastes in rural areas of Ireland. A one-off sampling and analysis campaign of 212 private wells found that 15% were contaminated with E. coli. More extensive monitoring of 24 selected wells found 58% to be contaminated with E. coli on at least one occasion over a 14-month period. The application of fingerprinting techniques to these monitored wells found that the use of chloride/bromide and potassium/sodium ratios is a useful low-cost fingerprinting technique capable of identifying impacts from human wastewater and organic agricultural contamination, respectively. The artificial sweetener acesulfame was detected on several occasions in a number of monitored wells, indicating its conservative nature and potential use as a fingerprinting technique for human wastewater. However, neither fluorescent whitening compounds nor caffeine were detected in any wells, and faecal sterol profiles proved inconclusive, suggesting limited suitability for the conditions investigated.

Babitonga Bay is a South Atlantic estuary with significant ecological function; it is part of the last remaining areas of mangrove communities in the Southern Hemisphere. The aim of this study was to determine the spatial distribution of the faecal sterols and linear alkylbenzenes (LABs) in surface sediments and to perform an integrated evaluation of several molecular marker indices to assess the sewage contamination status in the study area. The highest observed concentrations of faecal sterols (coprostanol + epicoprostanol) and LABs were 6.65 μg g−1 and 413.3 ng g−1, respectively. Several faecal sterol indices were calculated and correlated with coprostanol levels; these analyses showed that the index limits presented in the current literature could underestimate the sewage contamination in this study area. For the overall estuarine system, a low sewage impact may be assumed based on the low total mass inventories calculated for coprostanol (between 1.4% and 4.8%).

Numerous studies have established that acute or chronic exposure to environmental pollutants like particulate matter (PM) leads to the development of accelerated aging related pathologies including pulmonary and cardiovascular diseases, and thus air pollution is one of the major global threats to human health. Air pollutant particulate matter 2.5 (PM₂.₅)-induced cellular dysfunction impairs tissue homeostasis and causes vascular and cardiopulmonary damage. To test a hypothesis that elevated plasminogen activator inhibitor-1 (PAI-1) levels play a pivotal role in air pollutant-induced cardiopulmonary pathologies, we examined the efficacy of a drug-like novel inhibitor of PAI-1, TM5614, in treating PM₂.₅-induced vascular and cardiopulmonary pathologies. Results from biochemical, histological, and immunohistochemical studies revealed that PM₂.₅ increases the circulating levels of PAI-1 and thrombin and that TM5614 treatment completely abrogates these effects in plasma. PM₂.₅ significantly augments the levels of pro-inflammatory cytokine interleukin-6 (IL-6) in bronchoalveolar lavage fluid (BALF), and this also can be reversed by TM5614, indicating its efficacy in amelioration of PM₂.₅-induced increases in inflammatory and pro-thrombotic factors. TM5614 reduces PM₂.₅-induced increased levels of inflammatory markers cluster of differentiation 107 b (Mac3) and phospho-signal transducer and activator of transcription-3 (pSTAT3), adhesion molecule vascular cell adhesion molecule 1 (VCAM1), and apoptotic marker cleaved caspase 3. Longer exposure to PM₂.₅ induces pulmonary and cardiac thrombosis, but TM5614 significantly ameliorates PM₂.₅-induced vascular thrombosis. TM5614 also reduces PM₂.₅-induced increased blood pressure and heart weight. In vitro cell culture studies revealed that PM₂.₅ induces the levels of PAI-1, type I collagen, fibronectin (Millipore), and sterol regulatory element binding protein-1 and 2 (SREBP-1 and SREBP-2), transcription factors that mediate profibrogenic signaling, in cardiac fibroblasts. TM5614 abrogated that stimulation, indicating that it may block PM₂.₅-induced PAI-1 and profibrogenic signaling through suppression of SREBP-1 and 2. Furthermore, TM5614 blocked PM₂.₅-mediated suppression of nuclear factor erythroid related factor 2 (Nrf2), a major antioxidant regulator, in cardiac fibroblasts. Pharmacological inhibition of PAI-1 with TM5614 is a promising therapeutic approach to control air pollutant PM₂.₅-induced cardiopulmonary and vascular pathologies.

The quality and availability of water has become a pressing issue worldwide, being particularly important in semi-arid regions, where climate change has aggravated the problem. The use of anthropogenic chemicals, classified as emerging pollutants, adds to the problem representing a treat, since they are not regulated and have a potential impact on human and environmental health. This pressing problem has not been studied widely in complex environments like the one we present here. Distribution and seasonal variability of fecal sterols, alkylphenols, pesticides (emerging pollutants) and nutrients were determined in 35 wells used for agriculture and human consumption in the Valley of Maneadero, located in the semi-arid region of Baja California, Mexico. The presence of the tested pollutants in the saline aquifer was heterogeneous, showing important differences in concentration and distribution. Wells destined for household use showed the highest variability. In these wells, anthropogenic fecal sterols were detected and, alkylphenols, such as octyphenol and nonylphenol had maximum concentrations (2.7 ng/mL). In agriculture and urban wells, we identified DDT and organochlorine pesticides, as well as myclobutanil, which is considered a modern pesticide. Nitrates were identified in concentrations above international standards, mainly during the dry season, in both the agricultural and urban areas. As emerging pollutants represent a negative effect on environmental and human health, this is the first paper showing the importance of measuring this type of pollutant in agricultural/semi-urban areas, especially in aquifers that have been overexploited and communities that have relied on the use of septic tanks for decades.

In this work, source pollution tracing of the sediments of the Danube River and its tributaries in Serbia was performed using sterol ratios. Improved liquid chromatography-tandem mass spectrometry method, which enabled complete chromatographic separation of four analytes with identical fragmentation reactions (epicoprostanol, coprostanol, epicholestanol and cholestanol), was applied for the determination of steroid compounds (hormones, human/animal and plant sterols). A widespread occurrence of sterols was identified in all analyzed samples, whereas the only detected hormones were mestranol and 17α-estradiol. A human-sourced sewage marker coprostanol was detected at the highest concentration (up to 1939 ng g−1). The ratios between the key sterol biomarkers, as well as the percentage of coprostanol relative to the total sterol amount, were applied with the aim of selecting the most reliable for distinction between human-sourced pollution and the sterols originated from the natural sources in river sediments. The coprostanol/(cholesterol + cholestanol) and coprostanol/epicoprostanol ratios do not distinguish between human and natural sources of sterols in the river sediments in Serbia. The most reliable sterol ratios for the sewage pollution assessment of river sediments in the studied area were found to be coprostanol/(coprostanol + cholestanol), coprostanol/cholesterol and epicoprostanol/coprostanol. For the majority of sediments, human-derived pollution was determined. Two sediment samples were identified as influenced by a combination of human and natural biogenic sources.

Copper oxide nanoparticles (CuO NP) have been produced on a large scale due to their economically interesting thermophysical properties. This heightens the concern about risks they may pose on their release into the environment, possibly affecting non-target organisms. Microalga are important organisms in ecotoxicological studies as they are at the base of the aquatic food chain, but information about their biochemical and photosynthetic changes in response CuO NP are still scarce. We studied the effects of CuO NP in Raphidocelis subcapitata using morphological, photosynthetic and biochemical biomarkers. Our results showed that the NP affected microalgal population growth with 0.70 mg Cu L⁻¹ IC₅₀–₉₆ ₕ (inhibition concentration). Based on predicted environmental concentrations of Cu NPs in aquatic environments, our results indicate potential risks of the NP to microalgae. Algal cell size, granularity and photosynthetic efficiencies were affected by the CuO NP at 0.97 and 11.74 mg Cu L⁻¹. Furthermore, lipid metabolism was affected mostly at the highest NP concentration, but at environmentally relevant values (0.012 and 0.065 mg Cu L⁻¹) the production of sterols (structural lipids) and triacylglycerols (reserve lipid) increased. Moreover, we found evidence of cell membrane impairment at the highest CuO NP concentration, and, as a photosynthetic response, the oxygen evolving complex was its main site of action. To the best of our knowledge, this is the first study to date to investigate microalgal lipid composition during CuO NP exposure, showing that it is a sensitive diagnostic tool. This research demonstrated that CuO NP may affect the physiology of R. subcapitata, and because they were observed in a primary producer, we foresee consequences to higher trophic levels in aquatic communities.

Microplastics are widespread emerging contaminants that have been found globally in the marine and freshwater ecosystem, but there is limited knowledge regarding its impact on coral reef ecosystem and underpinning mechanism. In the present study, using Pocillopora damicornis as a model, we investigated cytological, physiological, and molecular responses of a scleractinian coral to acute microplastic exposure. No significant changes were observed in the density of symbiotic zooxanthellae during the entire period of microplastic exposure, while its chlorophyll content increased significantly at 12 h of microplastic exposure. We observed significant increases in the activities of antioxidant enzymes such as superoxide dismutase and catalase, significant decrease in the detoxifying enzyme glutathione S-transferase and the immune enzyme alkaline phosphatase, but no change in the other immune enzyme phenoloxidase during the whole experiment period. Transcriptomic analysis revealed 134 significantly up-regulated coral genes at 12 h after the exposure, enriched in 11 GO terms mostly related to stress response, zymogen granule, and JNK signal pathway. Meanwhile, 215 coral genes were significantly down-regulated at 12 h after exposure, enriched in 25 GO terms involved in sterol transport and EGF-ERK1/2 signal pathway. In contrast, only 12 zooxanthella genes exhibited significant up-regulation and 95 genes down-regulation at 12 h after the microplastic exposure; genes regulating synthesis and export of glucose and amino acids were not impacted. These results suggest that acute exposure of microplastics can activate the stress response of the scleractinian coral P. damicornis, and repress its detoxification and immune system through the JNK and ERK signal pathways. These demonstrate that microplastic exposure can compromise the anti-stress capacity and immune system of the scleractinian coral P. damicornis, despite the minimal impact on the abundance and major photosynthate translocation transporters of the symbiont in the short term.

Southeast Brazilian bays have been increasingly degraded by untreated organic loads. Therefore, to assess fecal contamination status, sediment quality regarding polycyclic aromatic hydrocarbons (PAHs), and sources of organic matter (OM), we have determined fine-grained and total organic carbon (TOC) content and concentrations of PAHs and sterols in twenty-six surface sediment samples in Sepetiba Bay. The fine-grained (1–26 %), TOC (0.20–3.45 %), PAHs (<LQ – 78.27 ng g⁻¹) and sterols (0.10–21.58 μg g⁻¹) results showed a decreasing trend from the internal to the external sector of the study area. The diagnostic ratios of selected PAHs and sterols indicated a mixture with significant contribution from continental and pyrolytic OM in all stations. The fecal contamination is significant to the internal sector of Sepetiba Bay. Considering a site-specific sediment quality guidelines (SQGs) the PAHs levels with more restricted benchmark values indicate the harbor and the internal sector as contaminated.

The discharge of sewage into the Antarctic marine environments by scientific stations has resulted in local changes in these pristine sites. To assess the distribution and concentration of sewage indicators from the Brazilian Antarctic station, sediments were sampled during the 2009/10 austral summer at four points (water depth of 20 and 60m). Concentrations of faecal sterols and linear alkylbenzenes (LABs) ranged from <0.01 to 0.17μgg⁻¹ and <1.0 to 46.5ngg⁻¹ dry weight, respectively. Maximum concentration of faecal sterols was similar to the value previously calculated as the background level for this area (0.19μgg⁻¹), and it is lower than the concentration observed in previous studies (1997–2008), whereas the LABs concentrations remained practically constant (35ngg⁻¹). Despite the low concentrations of sewage markers, the permanent human activities in the region require monitoring programs to determine continuing trends and prevent the increase of anthropogenic impacts.

Polycyclic aromatic hydrocarbons (PAHs) most likely derived from natural sources were observed in two sediment cores covering the last 100 years in an Amazon estuarine region. A considerable change in the PAHs main source was observed in the 1960s. Before the 1960s, the sources of PAHs seem to be related to biogenic and/or early-diagenetic processes. Concentrations of perylene were higher before the 1960s and suggest that its primary source to the sediments in the Amazon region is linked to a short-term diagenetic transformation of their biogenic precursors. The natural formation of alkylated PAHs in sediments was linked to the methylation of the parental aromatic hydrocarbons due to sediment maturation processes and the dehydrogenation of sterols in the sediments. The relatively rapid reaction occurring in recent sediments of the Amazon region suggests the importance of the microbial community in the transformation of biogenic precursors to alkylated-PAHs in the sediments.
