Linking cadmium and mercury accumulation to nutritional intake in common dolphins (Delphinus delphis) from Patagonia, Argentina

Bioaccumulation of Hg and Cd from food is a complex ecological process that has been oversimplified in the past. Common dolphins (Delphinus delphis) provide a powerful model to biomonitor metal concentrations in marine environments worldwide. We combined proportions-based nutritional geometry with metal analysis, stomach content analysis and the proximate composition of prey, to yield novel insights into the accumulation of Hg and Cd. Our analysis showed an age-related accumulation trend for Cd and Hg in kidney and liver, with highest concentrations found at 18 years of age. When viewed through the lens of nutritional ecology, Argentine anchovy (58.1 Mass %) and South American long-finned squid (22.7 Mass %), provided most of the dietary intake of protein (P) and lipids (L) (P:L ratio = 2.6:1.0) and also represented the main source for Cd and Hg levels accumulated in their bodies. This study presents unprecedented evidence on metal accumulation in relation to age and nutritional intake in a marine predator.

The wide ecological relevance of lipid homeostasis modulators in the environment has been increasingly acknowledged. Tributyltin (TBT), for instance, was shown to cause lipid modulation, not only in mammals, but also in fish, molluscs, arthropods and rotifers. In vertebrates, TBT is known to interact with a nuclear receptor heterodimer module, formed by the retinoid X receptor (RXR) and the peroxisome proliferator-activated receptor (PPAR). These modulate the expression of genes involved in lipid homeostasis. In the present work, we isolated for the first time the complete coding region of the Echinodermata (Paracentrotus lividus) gene orthologues of PPAR and RXR and evaluated the ability of a model lipid homeostasis modulator, TBT, to interfere with the lipid metabolism in this species. Our results demonstrate that TBT alters the gonadal fatty acid composition and gene expression patterns: yielding sex-specific responses in fatty acid levels, including the decrease of eicosapentaenoic acid (C20:5 n-3, EPA) in males, and increase of arachidonic acid (20:4n-6, ARA) in females, and upregulation of long-chain acyl-CoA synthetase (acsl), ppar and rxr. Furthermore, an in vitro test using COS-1 cells as host and chimeric receptors with the ligand binding domain (LBD) of P. lividus PPAR and RXR shows that organotins (TBT and TPT (Triphenyltin)) suppressed activity of the heterodimer PPAR/RXR in a concentration-dependent manner. Together, these results suggest that TBT acts as a lipid homeostasis modulator at environmentally relevant concentrations in Echinodermata and highlight a possible conserved mode of action via the PPAR/RXR heterodimer.

Environmental health is increasingly compromised by persistent toxic substances, which may have serious implications in food safety and, thus, in human health. Polybrominated diphenyl ethers (PBDEs) are anthropogenic contaminants with endocrine disruption abilities and are commonly found in seafood, the main route of human exposure. Growing evidence points out that the human gut microbiota interacts with xenobiotics, which may lead to impairment of host homeostasis if functions of microbiota become compromised. The aim of this study was to ascertain if the physiological balance of human gut microbiome is affected by the presence and degree of exposure to PBDEs. Fermentation was performed in a batch closed-system using an inoculum made from fresh human stool. The volatolomic profile was analysed by solid-phase microextraction coupled to gas chromatography-mass spectrometry. Mesophilic, Gram-negative bacteria and coliforms were quantified by classic plating methods. Changes in the gut microbiome were evaluated after DNA extraction followed by deep sequencing of the 16S rDNA region. The exposure to PBDEs resulted in an imbalance in sulfur, short-chain fatty acids and aromatic organic compounds, changing the microbial volatolome in a dose- and time-dependent manner. Slight deviations in the microbial structure of human gut occurred in the presence of PBDEs, especially for high doses of exposure. For the first time, the impact of PBDEs on the microbial homeostasis of human gut microbiota was taken into consideration, revealing noteworthy modifications with serious health implications even at oral exposure doses considered as safe by worldwide regulatory entities.

A substantial increase in the usage of organophosphate esters (OPEs) as flame retardants and plasticizers in rubbers, textiles, upholstered furniture, lacquers, plastics, building materials and electronic equipment has resulted in their increasing concentrations in the environment over time. However, little is known about the concentrations and fate of OPEs and their metabolites (mOPEs) in biota, including chicken eggs. The aim of this study was to understand the spatial variation in the concentrations in chicken eggs and the partitioning between yolk and albumin. In total, 153 chicken eggs were purchased across Australia and analysed for 9 OPEs and 11 mOPE. Most of the compounds were found to be deposited in egg yolk, where diphenyl phosphate (DPHP, 3.8 ng/g wet weight, median) and tris(2-chloroisopropyl) phosphate (TCIPP, 1.8 ng/g wet weight, median) were predominant mOPE and OPE, respectively. Moreover, no spatial differences in concentrations of OPEs and mOPEs in eggs purchased from different locations were found in this study. Although comparable levels of ∑OPEs were detected in egg yolk and albumin, much higher concentrations of ∑mOPEs were found in yolk than albumin. Meanwhile, a negative correlation (R² = 0.964, p = 0.018) was found between the molecular mass of analytes and partitioning coefficient of Cyₒₗₖ/Cyₒₗₖ₊ₐₗbᵤₘᵢₙ (defined as chemical concentration in egg yolk divided by the sum of chemical concentrations in both yolk and albumin). These results indicate that n-octanol/water partition coefficients (log KOW) may not be a crucial factor in the distribution of OPEs and mOPEs between egg yolk and albumin, which is important in understanding distribution of emerging organic contaminants in biota.

Contamination of soil with heavy metals seriously harms the growth of crops. Silicon fertilizer is known to promote growth of crops and alleviate heavy metals stresses in vegetables. However, little is known about the effects of silicon fertilizer on pakchoi vegetable growth and soil microbial community in soil contaminated with multiple heavy metals. In order to elucidate this question, current study was designed to analyze the impact of different silicon fertilizer doses on the growth of pakchoi, heavy metals accumulation in pakchoi, and diversity and composition of bacterial community in heavy metals contaminated soil. Results of the study showed that, silicon fertilizer application significantly improved the yield of pakchoi and reduced the content of heavy metals in pakchoi. Moreover, the silicon fertilizer led to the heterogeneity of bacterial community structure in soil. Linear discriminant analysis (LDA) effect size (LEfSe) test showed the change of soil bacterial community structures under the higher silicon fertilizer doses (0.8–3.2%). Similarly, soil bacteria associated with heavy metal resistance and carbon/nitrogen metabolism showed a more active response to medium fertilizer dose (0.8% w/w). In addition, Mantel test and Redundancy analysis (RDA) showed that both the soil bacterial community structures and pakchoi growth were significantly correlated with soil EC, available K and pH. Study suggested that the application of silicon fertilizer provided richer bacteria associated with heavy metal resistance and plant growth, and more favorable soil physicochemical environment for the growth of pakchoi under multiple heavy metal contamination, and the impact was dependent on fertilizing dose.

The mechanisms behind nitrogen removal by the submersed macrophyte-biofilm complex in wetlands remain to be fully elucidated. This study investigated the role of Myriophyllum spicatum and the biofilm on their leaves in nitrogen removal in mesocosm experiments. ¹⁵N tracking showed that 61.9% and 30% of the ¹⁵N, respectively, was removed from the system and assimilated by the macrophyte-biofilm complex after loading with 5.4 mg L⁻¹¹⁵N labelled NH₄⁺ for 17 days. Nitrogen budget results showed that about 0.2%, 0.2% and 3.6% of the nitrogen were emitted as water-, HCl- and NaOH-soluble nitrogen-gas species, respectively. Bacteria (76.7–91.8%) were the predominant domain in all samples, followed by eukaryotes (8.0–23.0%), archaea and viruses. Network analyses showed that there were positive- and negative-correlative relationships among nitrogen-cycling genes and nitrifiers and denitrifiers. Our data highlight the important role of biofilm on submersed macrophytes for nitrogen removal.

Particulate matter (PM) released from the processes of livestock production has a negative impact on the health of animals and workers. Herein, the concentration, major chemical components, morphology and microbiological compositions of particulate matter 2.5 (PM2.5, particles with aerodynamic diameter less than 2.5 μm) in a broiler breeding house were investigated. The results showed that the PM2.5 distribution in the chicken house was affected by the illumination, draught fans, chicken frame structure and activity of the chickens in the broiler breeding house. Component analysis showed that organic carbon (OC) accounted for the largest proportion, and followed by element carbon (EC), SO42−, NO3−, NH4+, Na+, K+ and Ca2+. Ultrastructural observations revealed that the shape of PM2.5 had a round, rectangular, chain-like and irregular shape. The concentration of endotoxin was approximately 0.3 EU/m3. Microbiological analysis showed that at the genus level, the pathogenic bacteria included Staphylococcus, Corynebacterium, Enterococcus, Parabacteroides, Escherichia and Megamonas. The abundant harmful fungi were Aspergillus, Scopulariopsis, Wallemia, and Fusarium. Through redundancy analysis (RDA) analysis, we determined that OC, EC, Na+, K+, and NH4+ had strong correlations with Brachybacterium, Brevibacterium, Corynebacterium, Escherichia, Scopulariopsis and Microascus. SO42− was closely related to Scopulariopsis and Salinicoccus. Salinicoccus was also strongly correlated with NO3−. Our results indicated that feed, faeces, and outside soot are contributed to the increase in PM2.5 concentration in the chicken house, while the sources of the dominant bacterial and fungi might be feed, faeces, suspended outside soil and cereal crops.
显示更多 [+] 显示较少 [-]The role of miR-21 in nickel nanoparticle-induced MMP-2 and MMP-9 production in mouse primary monocytes: In vitro and in vivo studies

Exposure to metal nanoparticles causes both pulmonary and systemic effects. Nanoparticles can enter the circulation and act directly or indirectly on blood cells, such as monocytes. Monocytes/macrophages are among the first cells to home to inflammatory sites and play a key role in the immune response. Here we investigated the effects of nickel nanoparticles (Nano-Ni), partially [O]-passivated Nano-Ni (Nano-Ni-P), and carbon-coated Nano-Ni (Nano-Ni-C) on MMP-2 and MMP-9 production in mouse primary monocytes both in vitro and in vivo and explored the potential mechanisms involved. The dose- and time-response studies showed that exposure of primary monocytes from wild-type (WT) mice to 30 μg/mL of Nano-Ni for 24 h caused significant MMP-2 and MMP-9 production; therefore, these dose and time point were chosen for the following in vitro studies. Nano-Ni and Nano-Ni-P caused miR-21 upregulation, as well as MMP-2, MMP-9, TIMP-1 and TIMP-2 upregulation in monocytes from WT, but not miR-21 knock-out (KO), mice, indicating the important role of miR-21 in Nano-Ni-induced MMPs and TIMPs upregulation. However, Nano-Ni-C did not cause these effects, suggesting surface modification of Nano-Ni, such as carbon coating, alleviates Nano-Ni-induced miR-21 and MMPs upregulation. These results were further confirmed by in vivo studies by intratracheal instillation of nickel nanoparticles into WT and miR-21 KO mice. Finally, our results demonstrated that exposure of primary monocytes from WT mice to Nano-Ni and Nano-Ni-P caused downregulation of RECK, a direct miR-21 target, suggesting the involvement of miR-21/RECK pathway in Nano-Ni-induced MMP-2 and MMP-9 production.

Rapeseed (Brassica napus L.) is a winter oil crop and biodiesel resource that has been widely cultivated in the southern part of China. Applying rapeseed residue (RSD) to summer rice fields is a common agricultural practice under rice−rapeseed double cropping systems. However, in Cd−contaminated paddy fields, the influence mechanisms of this agricultural practice on the migration and distribution of Cd fractions in soil are not clear. Therefore, a field experiment was carried out to analyse the changes in soil pH, organic matter (OM), microbial biomass carbon (MBC) and nitrogen (MBN), enzyme activity (urease (UA), acid phosphatase (ACP), and dehydrogenase (DH)), Cd distribution fractions, and Cd concentration in rice tissues after RSD application. The results showed that RSD treatment significantly increased the soil OM and MBC concentrations and UA, ACP, and DH activities, decreased the soil acetic acid−extractable fraction of Cd (ACI–Cd), and increased the reducible fraction of Cd (Red–Cd). The formation of stable organic complexes and chelates upon application of RSD is a result of the high affinity of Cd for soil OM. The activities of soil ACP, DH and MBC can well reflect Cd ecotoxicity in soil, particularly the DH activity. In addition, RSD application was helpful in inducing iron plaque formation. The “barrier” effect of iron plaque resulted in reduced Cd accumulation in different tissues of rice. The health risk of rice consumption also decreased as a result of RSD application; it decreased by 0.89–30.0% and 24.1–51.7% in the two tested fields. Overall, the application of RSD was increased soil OM, microbial biomass, and enzyme activity, and these changes was instrumental in reduce the risk of cadmium pollution in rice fields.

In coastal ecosystems, top predators are exposed to a wide variety of nutrient and contaminant sources due to the diversity of trophic webs within inshore marine habitats. Mercury contamination could represent an additional threat to shark populations that are declining worldwide. Here we measured total mercury, carbon and nitrogen isotopes, as well as mercury isotopes, in two co-occurring shark species (the bull shark Carcharhinus leucas and the tiger shark Galeocerdo cuvier) and their potential prey from a coastal ecosystem of the western Indian Ocean (La Réunion Island). Our primary goals were to (i) determine the main trophic Hg sources for sharks and (ii) better characterize their diet composition and foraging habitat. Hg isotope signatures (Δ¹⁹⁹Hg and δ²⁰²Hg) of shark prey suggested that bull sharks were exposed to methylmercury (MeHg) produced in offshore epipelagic waters, while tiger sharks were exposed to offshore mesopelagic MeHg with additional microbial transformation in slope sediments. Δ¹⁹⁹Hg values efficiently traced the ecology of the two predators, demonstrating that bull sharks targeted coastal prey in shallow waters while tiger sharks were mainly foraging on mesopelagic species in the deeper waters of the island slope. Unexpectedly, we found a positive shift in δ²⁰²Hg (>1‰) between sharks and their prey, leading to high δ²⁰²Hg values in the two shark species (e.g. 1.91 ± 0.52‰ in bull sharks). This large shift in δ²⁰²Hg indicates that sharks may display strong MeHg demethylation abilities, possibly reflecting evolutionary pathways for mitigating their MeHg contamination.
