Detoxification of aflatoxin B1 by Stenotrophomonas sp. CW117 and characterization the thermophilic degradation process 全文

Mycotoxins are high toxic, widely distributed contaminants in foodstuff. In this study, a aflatoxin B1 (AFB1) degrading strain S. acidoaminiphila CW117 was screened, and its detoxification characteristics were investigated. Substrate AFB1 at 45 μg/L was degraded by CW117 within 24 h; meanwhile, 4.1 mg/L AFB1 was almost degraded within 48 h. After 24 h degradation, the biotoxicity of the detoxified culture was eliminated. Strain CW117 efficient degradation to AFB1 (especially to low AFB1 concentrations) suggested its potential significance to detoxification development on food and feedstuff. The active degradation components present in the cell-free supernatant. The degradation ratio increased constantly with increasing incubation temperature raised (0–90 °C) and was even stable at 90 °C. Degradation was optimal at pH 6–7, and was only partially inhibited by metal-chelators (EDTA and EGTA), proteinase K, and a protein denaturant (sodium dodecyl sulfate, SDS). The recombinant laccase rLC1 (0.5 mg/mL) from CW117 degraded 29.3% of AFB1 within 24 h; however, the cell-free supernatant degraded 76.7% of the toxin in same time, with much lower protein content. The results indicated the CW117 degrades AFB1 via a combination of enzymes and micro-molecule oxides.

Estuaries are often considered to be the filters of pollutants from the land-derived outflows of freshwater to open seawater. Oysters are efficient bioaccumulators of metals in the estuarine environment, however, little information is available on the long-term tissue variability of metals in a large dynamic estuary under complex urbanized and anthropogenic impacts. Thus, an eight-year biomonitoring study of metals (Ag, Cd, Cr, Cu, Ni, Pb, and Zn) in the oysters from 10 sites were carried out to reveal the highly spatial-temporal variations in the Pearl River Estuary (PRE) of China during 2011–2018. Cd, Cu, and Zn in oysters were significantly correlated with the dissolved metals in seawater. Geographically, Ag, Cd, and Cr were higher in the western sites, and Cu, Ni, and Zn were higher in the eastern sites. High seasonal variations of Ag, Cu, and Zn were found in the wet season. The calculated annual change rates (vc) of Cd, Cu, Zn, Ag, Pb, Ni, and Cr in the oysters were −1.1, −45, −48, 0.338, −0.216, −2.2, and −2.8 μg/g/y, respectively. If such decreasing rates of vc (or natural logarithm rates v) were maintained, Cd, Cu, Zn, Pb, and Ni in oysters from PRE would be expected to recover to the national 50% concentrations in years 2022 (2024), 2045 (2079), 2073 (2110), 2021 (2023), and 2019 (2020), respectively. Long-term series observations of metals in organisms reflected the real bioavailability of metals, pollution status, and trends for environmental management and control in a large dynamic and contaminated estuary.

Microplastics are ubiquitous in natural waters and affect the environmental fate of hydrophobic organic micropollutants. This study evaluated the impacts of four microplastics, polypropylene (PP), polyethylene (PE), polystyrene (PS) and polymethyl methacrylate (PMMA), on the photodegradation of organotin compounds (OTCs) under UV₃₆₅ irradiation (2.3 ± 0.1 W m⁻²). The experiments were performed by mixing PP, PE, PS or PMMA microparticles with tri-organotins in artificial seawater. The photodegradation of OTCs in microplastic suspensions was influenced by the absorptivity onto microplastics. The decomposition rate of tributyltin (TBT) in UV-irradiated PP suspensions was greater than trimethyltin (TMT) and triphenyltin (TPhT) (p < 0.01). The adsorption capacities of OTCs (e.g., TBT) on PP particle surfaces were significantly lower than those on PE surfaces (p < 0.05) but similar with those on PMMA due to the different surface areas, shapes, and surface hydrophobicity of microplastics. TBT degraded faster (9.1%) in PS than in PMMA suspension (11.2%) within 240 min, respectively. However, only less than 5.4% was photodegraded in PP suspension due to the light scattering or absorption of the large sized PP particles. This study provided new insight into the impacts of microplastics on photodegradation of micropollutants in natural waters.

An increase in nutrient input may disturb the bacterioplankton communities in freshwater aquaculture ponds during the peak period of culture. Water samples were collected from ponds of three cultivated species. After the samples were filtered and total DNA was extracted, Illumina high-throughput sequencing was used to profile the spatiotemporal distributions in bacterioplankton communities, the belt diversity, and the influencing factors during this period of time. The results showed that Proteobacteria, Cyanobacteria, Bacteroidetes, and Actinobacteria were the dominant phyla. Biological differences in cultivated species were the main influencing factors that shaped bacterioplankton communities. Monthly variations and thermal stratification provided little and no contribution to bacterioplankton communities, respectively. CODmn, Chla, and TN were the most appropriate parameters to describe the environmental interpretation of bacterial ordinations, and CODmn was the predominant factor. In addition, the higher similarity between CODmn and Chla, shown by clustering analysis, indicated that the algae-bacteria symbiotic system may have an important role in material circulation in freshwater aquaculture pond water during the peak period of culture. The present study has helped to elucidate the biological characteristics of aquaculture tail water, and enriched the basic data provided by bacterioplankton studies.

Increasing levels of road noise are creating new anthropogenic soundscapes that may affect wildlife globally. Bats, which form about a third of all mammal species, are sensitive bioindicators, and may be particularly vulnerable because of their dependency on echolocation. Here we present the first controlled field experiment with free-living bats. Using a Before-After-Control-Impact phantom road experimental design, we examine the impacts of traffic noise on their activity and feeding behaviour. Disentangling the impacts of traffic noise from other co-varying exposures such as habitat quality, the experiment demonstrates a significant negative effect on the activity of each of the five, ecologically different, species (genus for Myotis spp.) examined. This suggests that the results are widely applicable. The negative effects are largely attributable to noise in the sonic spectrum, which elicited aversive responses in all bat species tested,whereas responses to ultrasoundwere restricted to a single species. Our findings demonstrate that traffic noise can affect bat activity at least 20m away from the noise source. For Pipistrellus pipistrellus and Pipistrellus pygmaeus, feeding behaviour, as well as overall activity, was negatively affected. Ecological Impact Assessments are therfore needed wherever there are significant increases in traffic flow, and not just when new roads are built. Further research is required to identify effective mitigation strategies, to delineate the zone of influence of road noise, and to assess whether there is any habituation over time.

Research on the atmospheric occurrence of short chain chlorinated paraffins (SCCPs) in industrialized areas is scarce. In this study, we investigated the concentrations, profile, and spatial distribution of SCCPs in the highly industrialized and developed areas of the Yangtze River Delta (YRD) in China using polyurethane foam passive air samplers. Sampling was performed during two separate periods in 2011. The concentrations of atmospheric SCCPs ranged from 6.1 to 63 ng m⁻³ in summer and 6.2–42 ng m⁻³ in winter. The C₁₀ and C₁₁ groups were the predominant carbon groups in all the samples. Different congener patterns between summer and winter were found, indicating that congeners in the air in winter may be influenced by local and external sources. The highest level of SCCPs was found in Suzhou, which is a highly industrialized area with many manufactories including electronic industries and plastic factories. Higher levels of SCCPs were found in the air than polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs), and polybrominated diphenyl ether (PBDEs), suggesting that the production and use of SCCPs were much higher than prohibited or restricted persistent organic pollutants (e.g., PCBs, OCPs, and PBDEs). Future studies should investigate the different sources of atmospheric SCCPs by conducting a comprehensive assessment of SCCP exposure.

Foamed plastic debris in aquatic systems has become one of the emerging global contaminants. In this study, the behavior of polystyrene foam (PSF) and microplastics (MPs) adhered on the PSFs were investigated on the Tuul River shore in Ulaanbaatar, the capital city of Mongolia. The micro-sized (<5 mm) PSF, which was the dominant PSF over 600 pieces in 100 m², have accumulated along the shoreline of Tuul River. Carbonyl index (CI) was calculated to evaluate the surface oxidation of macro-sized (20–100 mm), meso-sized (5–20 mm), and micro-sized PSFs and confirm the relative aging depending on photodegradation. CI ranged from 0.00 to 1.09 in the sampled PSFs, whereby the degraded PSFs with high CI were distributed on the shore of downstream of sewer drainage. Micro-sized PSFs showed a wide range of CI and a relatively high average value of CI as compared to those of meso- and macro-sized PSFs. Most of PSFs aggregated with MPs and the adhered MPs have been ubiquitously detected from the surface of PSFs. Adhered micro-sized plastics explored from the surface of PSFs with various sizes, except for mega-sized (>100 mm) PSF, ranged from 5 to 141 items per piece of PSF fragment. The aggregates of PSFs and MPs were common status of PSFs during their transportation. The present findings, which indicated a high concentration of adhered MPs, raise an environmental concern about the widespread aquatic plastic pollution.

Bitumen recovery from oil sands in northeastern Alberta, Canada produces large volumes of tailings, which are deposited in mining areas that must be reclaimed upon mine closure. A new technology of non-segregated tailings (NST) developed by Canadian Natural Resources Limited (CNRL) was designed to accelerate the process of oil sands fine tailings consolidation. However, effects of these novel tailings on plants used for the reclamation of oil sands mining areas remain to be determined. In the present study, we investigated the effects of NST on seedlings of three species of plants commonly planted in oil sands reclamation sites including paper birch (Betula papyrifera), white spruce (Picea glauca) and green alder (Alnus viridis). In the controlled-environment study, we grew seedlings directly in NST and in the two types of reclamation soils with and without added NST and we measured seedling growth, gas exchange parameters, as well as tissue concentrations of selected elements and foliar chlorophyll. White spruce seedlings suffered from severe mortality when grown directly in NST and their needles contained high concentrations of Na. The growth and physiological processes were also inhibited by NST in green alder and paper birch. However, the addition of top soil and peat mineral soil mix to NST significantly improved the growth of plants, possibly due to a more balanced nutrient uptake. It appears that NST may offer some advantages in terms of site revegetation compared with the traditional oil sands tailings that were used in the past. The results also suggest that, white spruce may be less suitable for planting at reclamation sites containing NST compared with the two studied deciduous tree species.

Planting leafy vegetables, especially pakchoi, in cadmium (Cd) polluted farmland is easy to lead to excessive Cd content in edible parts, which results in high risk of food chain. In this study, a field experiment was carried out to study the effects of intercropping of pakchoi with Cd hyperaccumulator Sedum alfredii Hance, and the roles of endophytic bacterium SaMR12 was also investigated. When intercropping with Sedum, the growth of pakchoi was not affected but their Cd concentration and accumulation were significantly increased, while which were obviously decreased by SaMR12 inoculation. After intercropping, the biomass of Sedum was significantly reduced, but their Cd concentration increased. SaMR12 inoculation significantly increased Cd accumulation of Sedum, and which increased to 3 times in Sedum monoculture. Those results showed that although intercropping with hyperaccumulator could lead to higher risk of pakchoi in Cd polluted field, intercropping with SaMR12 inoculated Sedum can decrease Cd concentration of pakchoi and promote Cd absorption of Sedum, which indicated that this endophyte can be made into a microbial inoculum as a soil additive for the safe production of vegetables and the soil Cd pollution remediation.

The ongoing transition to renewable fuel sources has led to increased use of wood and other biomass fuels. The physiochemical characteristics of biomass combustion derived aerosols depends on appliances, fuel and operation procedures, and particles generated during incomplete combustion are linked to toxicity. Frequent indoor wood burning is related to severe health problems such as negative effects on airways and inflammation, as well as chronic hypoxia and pathological changes in placentas, adverse pregnancy outcome, preterm delivery and increased risk of preeclampsia. The presence of combustion-derived black carbon particles at both the maternal and fetal side of placentas suggests that particles can reach the fetus. Air pollution particles have also been shown to inhibit trophoblast migration and invasion, which are vital functions for the development of the placenta during the first trimester. In this study we exposed a placental first trimester trophoblast cell line to wood smoke particles emitted under Nominal Burn rate (NB) or High Burn rate (HB). The particles were visible inside exposed cells and localized to the mitochondria, causing ultrastructural changes in mitochondria and endoplasmic reticulum. Exposed cells showed decreased secretion of the pregnancy marker human chorionic gonadotropin, increased secretion of IL-6, disrupted membrane integrity, disrupted proliferation and contained specific polycyclic aromatic hydrocarbons (PAHs) from the particles. Taken together, these results suggest that wood smoke particles can enter trophoblasts and have detrimental effects early in pregnancy by disrupting critical trophoblast functions needed for normal placenta development and function. This could contribute to the underlying mechanisms leading to pregnancy complications such as miscarriage, premature birth, preeclampsia and/or fetal growth restriction. This study support the general recommendation that more efficient combustion technologies and burning practices should be adopted to reduce some of the toxicity generated during wood burning.
