Nervous system disruption and concomitant behavioral abnormality in early hatched pufferfish larvae exposed to heavy oil

BACKGROUND AND OBJECTIVE: Spills of heavy oil (HO) over the oceans have been proven to have an adverse effect on marine life. It has been hypothesized that exposure of early larvae of sinking eggs to HO leads largely to normal morphology, whereas abnormal organization of the developing neural scaffold is likely to be found. HO-induced disruption of the nervous system, which controls animal behavior, may in turn cause abnormalities in the swimming behavior of hatched larvae. To clarify the toxicological effects of HO, we performed exposure experiments and morphological and behavioral analyses in pufferfish (Takifugu rubripes) larvae. EXPERIMENTAL PROCEDURE: Fertilized eggs of pufferfish were exposed to 50 mg/L of HO for 8 days and transferred to fresh seawater before hatching. The hatched larvae were observed for their swimming behavior, morphological appearance, and construction of muscles and nervous system. RESULTS AND DISCUSSION: In HO-exposed larvae, we did not detect any anomaly of body morphology. However, they showed an abnormal swimming pattern and disorganized midbrain, a higher center controlling movement. Our results suggest that HO-exposed fishes suffer developmental disorder of the brain that triggers an abnormal swimming behavior and that HO may be selectively toxic to the brain and cause physical disability throughout the life span of these fishes.

BACKGROUND, AIM, AND SCOPE: Assessment of environmental impacts from pesticide utilization should include genotoxicity studies, where the possible effects of mutagenic/genotoxic substances on individuals are assessed. In this study, the genotoxicity profile of the new formicide Macex® was evaluated with two genotoxicity tests, namely, the micronucleus test with mouse bone marrow and Vicia faba, and a mutagenicity test using the Ames Salmonella assay. MATERIALS AND METHODS: The bacterial reverse mutation test (Salmonella typhimurium strains TA97, TA98, TA100, TA102, and TA1535), the Vicia root tip and mouse micronucleus tests were conducted according to published protocols. RESULTS: In the range of the formicide Macex® concentrations tested from 0.06 to 1.0 g L−1 (or mgkg−1 in the mouse test), no genotoxicity was observed in the prokaryotic or eukaryotic test organisms. However, at Macex® concentrations of 0.5 g L−1 and above a significant decrease in the mitotic index (P ≤ 0.05) in the V. faba was observed. Micronucleus formation was likewise increased in the test organism at concentrations starting at 2.0 g L−1. CONCLUSIONS: These data allow us to classify this natural formicide preparation as a product with no geno-environmental-impact when applied at recommended concentrations.

INTRODUCTION: This article recalls and demonstrates that heterogeneous photocatalysis belongs to heterogeneous catalysis according to its initial history. There are criteria, which have to be imperatively taken into account to deal with true (photo-)catalytic reactions. The photocatalytic activity, chosen as the reaction rate, is governed by five physical parameters (mass of catalyst, wavelength, initial concentrations (or pressures) of the reactants, temperature (around room temperature), and radiant flux). DISCUSSION: Once performed in optimal conditions, the reaction has to be characterized with (a) its quantum yield <QY> (ratio of the reaction rate to the incident photon flux), (b) the turnover number <TON> (number of molecules converted per active site during a given time), and (c) the turnover frequency <TOF> (<TON> per second). The true catalytic nature demands that the conversion provides a number of converted molecules higher than the “stoichiometric threshold” defined as the number of the active sites initially present on the catalyst, i.e., that <TON> be much greater than 1, and possibly than at least two or three orders of magnitude. Eventually, a complete mass balance determination should be established for all elements. It is shown that the main reaction intermediates are radicals. All these concepts and principles are illustrated by various examples (selective mild oxidation reactions, water pollutants elimination, and air purification), leading to a list of recommendations for performing really true catalytic reactions.

INTRODUCTION: The goal of the present study was to investigate the effects of Cu contamination on the above-mentioned biochemical and physiological parameters in order to explore possible prevention strategies against heavy metal stress. MATERIALS AND METHODS: Effects of copper (Cu) on the roots of Sagittaria sagittifolia L. were studied after 10 days of treatment at five concentration levels. The accumulation of Cu, the generation rate of O₂ ·–, the contents of thiobarbituric acid reactive substances (TBARS) and polyamines, as well as the activities of arginine decarboxylase (ADC) and polyamine oxidase (PAO) in the roots were measured and analyzed. RESULTS AND DISCUSSION: It was observed that endogenous Cu content increased in roots of S. sagittifolia L. in a concentration-dependent manner, along with an increased production of O₂ ·–. TBARS content increased progressively up to 5 μmol l⁻¹ Cu. A constant increase in ADC activity was also observed. The results indicated that lower Cu concentrations (2.5 and 5 μmol l⁻¹, respectively) had greater enhancing effect on the contents of free Put and perchloric acid-soluble conjugated (PS-conjugated) putrescine (Put), while Cu treatments at different concentration levels had similar enhancing effect on the content of perchloric acid-insoluble bound Put. In total, Put content in each Cu-treated group was higher than that in the control group. PAO activity was inhibited up to 10 μmol l⁻¹ Cu but enhanced at higher Cu concentrations (20 and 40 μmol l⁻¹). This explained the initial rise and subsequent decline of the contents of all forms of spermine (Spm), free and PS-conjugated spermidine (Spd). However, with the increase of Cu concentration, total Spm content increased gradually while total Spd content decreased. Our results suggest that Cu is phytotoxic to the roots of S. sagittifolia L. at high concentrations, and that the increased Spm level is not sufficient to resist Cu-induced oxidative damages.

PURPOSE: The disinfection efficiency of water and secondary treated wastewater by means of photoelectrocatalytic oxidation (PEC) using reference strains of Enterococcus faecalis and Escherichia coli as faecal indicators was evaluated. Operating parameters such as applied potential (2–10 V), initial bacterial concentration (103–107 CFU/mL), treatment time (up to 90 min) and aqueous matrix (pure water and treated effluent) were assessed concerning their impact on disinfection. METHODS: PEC experiments were carried out using a TiO2/Ti film anode and a zirconium cathode in the presence of simulated solar radiation. Bacterial inactivation was monitored by the culture method and real-time SYBR green PCR. RESULTS: A 6.2 log reduction in E. faecalis population was achieved after 15 min of PEC treatment in water at 10 V of applied potential and an initial concentration of 107 CFU/mL; pure photocatalysis (PC) led to only about 4.3 log reduction, whilst negligible inactivation was recorded when the respective electrochemical oxidation process was applied (i.e. without radiation). PEC efficiency was generally improved increasing the applied potential and decreasing initial bacterial concentration. Regarding real wastewater, E. coli was more susceptible than E. faecalis during treatment at a potential of 5 V. Wastewater disinfection was affected by its complex composition and the contained mixed bacterial populations, yielding lower inactivation rates compared to water treatment. Screening the results obtained from both applied techniques (culture method and real-time PCR), there was a discrepancy regarding the recorded time periods of total bacterial inactivation, with qPCR revealing longer periods for complete bacterial reduction. CONCLUSIONS: PEC is superior to PC in terms of E. faecalis inactivation presumably due to a more efficient separation and utilization of the photogenerated charge carriers, and it is mainly affected by the applied potential, initial bacterial concentration and the aqueous matrix.

PURPOSE: The aim of this study was to analyze air concentrations of chemical and microbiological pollutants in the vicinity of an organic waste treatment plant, Ecoparc-2, located in Montcada i Reixac (Catalonia, Spain), as well as to determine the seasonal trends. The human health risks due to the presence of those agents were also assessed. METHODS: Air samples were collected at different distances and wind directions from the Ecoparc-2 in two campaigns (winter and summer of 2010). The levels of 19 volatile organic compounds (VOCs) were analyzed by GC-MS or HPLC-UV. In turn, the airborne amount of total bacteria, gram-negative bacteria, and fungi (including Aspergillus fumigatus) was also determined. RESULTS: Mean VOC concentrations were found to be 32.4 and 15.7 μg/m3 in winter and summer, respectively. Fungi at 25°C presented the highest geometric mean (1,126 and 863 cfu/m3 in winter and summer, respectively), while the concentrations of fungi at 37°C and total bacteria were also important in the hot season (332 and 250 cfu/m3, respectively). These results are in agreement with data obtained from the scientific literature. Anyhow, no significant differences were observed between both campaigns including those related to distances and wind directions. The current pollutant levels in the surrounding environment were also various orders of magnitude lower than those recently observed inside the facility. CONCLUSIONS: The human exposure to VOCs near the Ecoparc-2 was estimated to be low. Furthermore, the current environmental concentrations of those chemical and microbiological agents were clearly below threshold values recommended by regulatory organizations.

INTRODUCTION: The removal of azo Reactive Red 31(RR31) from synthetic dye solution using tannery sludge-developed activated carbon (TSC) was investigated. METHODS: TSC was prepared from a combination of physical and chemical activation. The developed TSC was characterized by FT-IR, SEM, TG–DTA, specific surface area and zero point charge of pH (pHzpc). The isotherm models, kinetic models and thermodynamic parameters were also analysed to describe the adsorptive behaviour of TSC. The effect of contact time, initial dye concentration, carbon dosage, agitation speed, initial pH and temperature were carried out for batch adsorption studies. The isotherm plot of the dye RR31 on TSC fitted better with the Langmuir adsorption isotherm than the Freundlich model. RESULTS: The maximum monolayer adsorption capacity of TSC in the removal of RR31 ranged from 23.15 to 39.37 mg/g. The thermodynamic parameters showed the endothermic and physical nature of the Reactive Red 31 adsorption on TSC. The entropy and enthalpy values were 181.515 J/Kmol and 5.285 kJ/mol, respectively. CONCLUSION: The developed cationic tannery sludge carbon was found to be an effective adsorbent in the removal of the anionic azo reactive dye RR31.

PURPOSE: The impact of CO₂ urban plume in a rural area was investigated by concentrations recorded near surface. METHODS: CO₂ dry concentrations at three levels near surface were recorded for about 8 months at a rural site. Daily cycles were obtained and directional analysis was made with percentiles. Several functions were used to fit background and plume concentrations and the goodness of fit was evaluated with different statistics, which were also compared. RESULTS: Daily cycle showed a difference of around 2 ppm during the night between the lowest (1.8 m) and the highest (8.3 m) levels. Weighting functions of the directional analysis revealed the influence of the Valladolid urban plume. Two regions were established, with local factors prevailing below 3 m s⁻¹ and transport dominating above 6 m s⁻¹. The best fit was achieved with a quadratic function for the background and a cubic function for the plume due to the lack of symmetry observed. Gamma and Weibull distributions were also successfully used. Some statistics, such as the root mean square error (RMSE), stood out when evaluating the goodness of fit, whilst others were discarded due to their extremely low values and the lack of sensitivity against the functions used. Finally, a comprehensive metric merging several statistics was also tested with slight differences against RMSE.

PURPOSE: In the ambit of a project searching for appropriate biological approaches for recovering a refinery soil contaminated with petroleum hydrocarbons (PHC), we compared results obtained in the absence and in the presence of the salt marsh plant Scirpus maritimus or Juncus maritimus or an association of these two plants, which were tested in the refinery environment. Synergistic effects caused by addition of a non-ionic surfactant and/or a bioaugmentation product were also investigated. Major challenges of this study were: field conditions and weathered contamination. METHODS: Transplants of the plants were carried out in individual containers filled with a weathered contaminated soil, which was recontaminated with turbine oil with two purposes: for increasing PHC level and allowing a comparison of the potential of plants for remediation of ancient and recent contamination. RESULTS: Analysis of total PHC led to the conclusion that, after 24-month exposure, neither J. maritimus nor the association caused any improvement in remediation. In contrast, S. maritimus revealed potential for PHC remediation, favoring degradation of both recent and older contamination (which was refractory to natural attenuation). About 15% of remediation improvement was found in the soil layer with higher root density (5–10 cm). A more marked improvement in that layer (28%) was observed when non-ionic surfactant amendment and bioaugmentation were used jointly. CONCLUSIONS: The fact that S. maritimus has demonstrated capability for PHC remediation, leads to admit that it has potential to be also used for recovering sediments that have suffered accidental oil spills.

INTRODUCTION: Meristematic mitotic cells of Allium cepa constitute an adequate material for cytotoxicity and genotoxicity evaluation of environmental pollutants, such as phenol, which is a contaminant frequently found in several industrial effluents. RESULTS AND DISCUSSION: In the present work, Brassica napus hairy roots (HR) were used for phenol removal assays. The toxicity of post-removal solutions (PRS) and phenol solutions was analyzed. These HR removed the contaminant with high efficiency (100–80% for phenol solutions containing 10–250 mg/L, respectively). Phenol solutions treated with B. napus HR showed a significant reduction of general toxicity compared to untreated phenol solutions, since the IC50 values were 318.39 and 229.02 mg/L, respectively. Moreover, PRS presented lower cytotoxicity and genotoxicity than that found in phenol solutions untreated. The mitotic index (MI) observed in meristematic cells treated with PRS (100 and 250 mg/L of phenol) showed an increase of 35% and 42%, whereas the chromosome aberrations showed a significant decrease. According to these results, B. napus HR cultures could be used for the treatment of solutions contaminated with phenol, since we observed not only high removal efficiency, but also an important reduction of the general toxicity, cytotoxicity, and genotoxicity.
