Limited effects of environmentally-relevant concentrations in seawater of dibutyl phthalate, dimethyl phthalate, bisphenol A, and 4-nonylphenol on the reproductive products of coral-reef organisms
2022
Vered, Gal | Shenkar, Noa
Plastic additives (PAs) are chemical compounds incorporated into the plastic during the manufacturing process. Phthalate acid esters, bisphenols, and nonylphenols are all PAs found in marine environments and associated with endocrine-disrupting processes. However, our knowledge regarding the impact of endocrine-disrupting PAs on coral-reef organisms is limited. As reef population structure is directly linked to reproduction and larval settlement processes, interference with hormonal systems can impact coral-reef community structure, particularly if the effects of PAs differ among species. In the current study we exposed the reproductive products of four tropical coral-reef invertebrates to environmentally-relevant concentrations of four prevalent PAs in seawater: dibutyl phthalate (DBP), dimethyl phthalate, (DMP), 4-nonylphenol (4-NP), and bisphenol A (BPA), as well as to 10³ higher laboratory concentrations of these PAs. Our results revealed that apart from the significant negative effect of the 1 μg/L of 4-NP on the settlement of the soft coral Rhytisma fulvum, none of the other tested materials demonstrated a significant effect on the exposed organisms at environmentally-relevant concentrations in seawater. The 4-NP high laboratory concentration (1000 μg/L), however, had significant negative effects on all the examined species. The high laboratory BPA concentration (1000 μg/L) significantly reduced fertilization success in the solitary ascidian Herdmania momus, up to its complete failure to reproduce. Moreover, the high laboratory DMP concentration (100 μg/L) had a significant negative effect on planulae settlement of the stony coral Stylophora pistillata. Our findings demonstrate the negative and selective effects of PAs on the development and reproduction of coral-reef organisms; and, specifically, the significant effect found following exposure to 4-NP. Consequently, if we aim to fully understand the impact of these contaminants on this endangered ecosystem, we suggest that the actual concentrations within the living organism tissues should be tested in order to produce relevant risk assessments for brooding-coral species.

Next-generation sequencing method employing the sphingomonads-specific and universal 16s rRNA primers were applied to analyze the response of soil bacterial community to continuous stress of nanoscale zero-valent iron (nZVI) and/or polycyclic aromatic hydrocarbons (PAHs) under suspended conditions. The group specific primer greatly improves taxonomic resolution, the overall OTU numbers increased from 12 by universal primer to 42 by sphingomonads-specific primer. It was found that nZVI and PAHs had opposite selection effects on bacterial community composition, furthermore, the abundance of Sphingopyxis was positively related to nZVI while negatively related to PAHs, whereas abundances of Sphingobium and Sphingosinicella were positively related to PAHs (p < 0.01). The improved taxonomic resolution helps identify the potential keystone taxa, which provides valuable information for future bioremediation technology such as synthetic microbiome.

Currently little is known about the adsorption behaviors of metalloids on microplastics (MPs) and their complex toxic effects on aquatic plants. Herein, we investigated the adsorption behaviors of arsenic (As(III) and As(V)) on three types of MPs (polystyrene, polyvinyl chloride, and polyethylene) with four different particle sizes (100, 10, 1, and 0.1 μm). Compared with the short-term exposure experiment, co-toxicity of polystyrene nanoplastics (PS-NPs) and As on two submerged macrophytes (Vallisneria denseserrulata and Potamogeton crispus) were explored through two relatively longer 14-day-cultivation experiments in summer and spring, respectively. The adsorption results showed that As entered the internal surface adsorption site of MPs at 24 h and fully combined to reach equilibrium. The adsorption capacity also enhanced with the increase of MPs concentrations, which generated more adsorption sites for binding with MPs. The presence of PS-NPs increased the absorption of As on macrophytes by 36.2–47.2%. More serious damage of leaf structure by combined PS-NPs and As was observed by transmission electron microscope. The larger harms by the co-toxicity of MPs and As were also reflected by the changes in physiochemical characteristics (e.g. photosynthesis) and the enhancement of oxidative damage of macrophytes. This work provides a clear theoretical basis for the behavior of PS-NPs as carrier with other contaminants on submerged macrophytes, and clearly evaluates the co-toxicity of NPs and metalloids in complex aquatic environments.

Water quality forecasting can provide useful information for public health protection and support water resources management. In order to forecast water quality more accurately, this paper proposes a novel hybrid model by combining data decomposition, fuzzy C-means clustering and bidirectional gated recurrent unit. Firstly, the original water quality data is decomposed into several subseries by empirical wavelet transform, and then, the decomposed subseries are recombined by fuzzy C-means clustering. Next, for each clustered series, bidirectional gated recurrent unit is applied to develop prediction model. Finally, the forecast result is obtained by the summation of the predictions for the subseries. The proposed forecast model is evaluated by the water quality data of Poyang Lake, China. Results show that the proposed forecast model provides highly accurate forecast result for all of the six water quality data: the average of MAPE of the forecast results for the six water quality datasets is 4.59% for 7 day ahead prediction. Furthermore, our model shows better forecast performance than the other models. Particularly, compared with the single BiGRU model, MAPE decreased by 32.86% in average. Results demonstrate that the proposed forecast model can be used effectively for water quality forecasting.

Elevated carbon dioxide (ECO₂) concentration has profound impacts on ecosystem carbon fluxes, with consequent changes in carbon sequestration and its feedback to climate change. Agroecosystem plays an essential role in global carbon sequestration. However, it is not well understood how the carbon fluxes of agroecosystem respond to increasing atmospheric CO₂ concentrations. In this study, an in-situ 2-year field experiment was conducted using open-top chamber with treatments including ambient CO₂ concentration (CK) and ambient plus 200 μmol mol⁻¹ (T) to investigate the characteristics and main factors influencing carbon fluxes during the 2017–2019 winter wheat growing seasons. Results showed that the dynamics of CO₂ fluxes under different treatments had similar seasonal trends, with the peak flux observed at the heading-filling stage. Compared to the CK, T treatment increased the cumulative amount of CO₂ (CAC) by 17.2% and 24.0% in 2017–2018 and 2018–2019 growing seasons, respectively. In addition, the seasonal CAC was highly dependent on treatment and varied with year, while there was no interactive effect of treatment and year (p > 0.05). ECO₂ concentration increased the biomass of wheat by an average of 8.28% over two growing seasons. There was a significant positive correlation between biomass and CAC, with biomass elucidating 52% and 76% of the variations in CAC under CK and T treatments, respectively. A good correlation was found between net ecosystem exchange (NEE) and environmental variables under different treatments. During the pre-milk ripening period, the NEE mainly depended on photosynthetically active radiation (PAR) and air temperature (Ta), while NEE was mainly controlled by PAR and soil water content (SWC) during the post-milk ripening period. Overall, the findings presented here demonstrate that the carbon exchange in wheat fields under different treatments serves as carbon sequestration, while ECO₂ concentration enhances the capacity of winter wheat fields to act as carbon sinks, which may have feedback to the climate system in the future.

Ambient air pollutants are well-known risk factors for childhood asthma and asthma exacerbation. It is unknown whether different air pollutants individually or jointly affect pathophysiological mechanisms of asthma. In this study, we aim to integrate transcriptome and untargeted metabolome to identify dysregulated genetic and metabolic pathways that are associated with exposures to a mixture of ambient and traffic-related air pollutants among adults with asthma history. In this cross-sectional study, 102 young adults with childhood asthma history were enrolled from southern California in 2012. Whole blood transcriptome was measured with 20,869 expression signatures, and serum untargeted metabolomics including 937 metabolites were analyzed by Metabolon, Inc. Participants’ exposures to regional air pollutants (NO₂, O₃, PM₁₀, PM₂.₅) and near-roadway air pollutants averaged at one month and one year before study visit were estimated based on residential addresses. xMWAS network analysis and joint-pathway analysis were performed to identify subnetworks and genetic and metabolic pathways that were associated with exposure to air pollutants adjusted for socio-characteristic covariates. Network analysis found that exposures to air pollutants mixture were connected to 357 gene markers and 92 metabolites. One-year and one-month averaged PM₂.₅ and NO₂ were associated with several amino acids related to serine, glycine, and beta-alanine metabolism. Lower serum levels of carnosine and aspartate, which are involved in the beta-alanine metabolic pathway, as well as choline were also associated with worse asthma control (p < 0.05). One-year and one-month averaged PM₁₀ and one-month averaged O₃ were associated with higher gene expression levels of HSPA5, LGMN, CTSL and HLA-DPB1, which are involved in antigen processing and presentation. These results indicate that exposures to various air pollutants are associated with altered genetic and metabolic pathways that affect anti-oxidative capacity and immune response and can potentially contribute to asthma-related pathophysiology.

Although the hazards of microplastics (MPs) have been quite well explored, the aberrant metabolism and the involvement of the autophagy pathway as an adverse response to environmental MPs in benthic organisms are still unclear. The present work aims to assess the impact of different environmental MPs collected from the south coast of the Mediterranean Sea, composed by polyethylene (PE), polyethylene vinyl acetate (PEVA), low-density polyethylene (LDPE), high-density polyethylene (HDPE), polypropylene (PP) and polyamide (PA) on the metabolome and proteome of the marine polychaete Hediste diversicolor. As a result, all the microplastic types were detected with Raman microspectroscopy in polychaetes tissues, causing cytoskeleton damage and induced autophagy pathway manifested by immunohistochemical labeling of specific targeted proteins, through Tubulin (Tub), Microtubule-associated protein light chain 3 (LC3), and p62 (also named Sequestosome 1). Metabolomics was conducted to further investigate the metabolic alterations induced by the environmental MPs-mixture in polychaetes. A total of 28 metabolites were differentially expressed between control and MPs-treated polychaetes, which showed elevated levels of amino acids, glucose, ATP/ADP, osmolytes, glutathione, choline and phosphocholine, and reduced concentration of aspartate. These novel findings extend our understanding given the toxicity of environmental microplastics and unravel their underlying mechanisms.

Bioremediation of real-time petroleum refining industry oily waste (PRIOW) is a major challenge due to the poor emulsification potential and oil sludge disintegration efficiency of conventional bioamphiphile molecules. The present study was focused on the design of a covalently engineered supramolecular bioamphiphile complex (SUBC) rich in hydrophobic amino acids for proficient emulsification of hydrocarbons followed by the concomitant degradation of total petroleum hydrocarbons (TPH) in PRIOW using the hydrocarbonoclastic microbial bio-formulation system. The synthesis of SUBC was carried out by pH regulated microbial biosynthesis process and the yield was obtained to be 450.8 mg/g of petroleum oil sludge. The FT-IR and XPS analyses of SUBC revealed the anchoring of hydrophilic moieties of monomeric bioamphiphilic molecules, resulting in the formation of SUBC via covalent interaction. The SUBC was found to be lipoprotein in nature. The maximum loading capacity of SUBC onto surface modified rice hull (SMRH) was achieved to be 45.25 mg/g SMRH at the optimized conditions using RSM-CCD design. The SUBC anchored SMRH was confirmed using SEM, FT-IR, XRD and TGA analyses. The adsorption isotherm models of SUBC onto SMRH were performed. The integrated approach of SUBC-SMRH and hydrocarbonoclastic microbial bio-formulation system, emulsified oil from PRIOW by 92.86 ± 2.26% within 24 h and degraded TPH by 89.25 ± 1.75% within 4 days at the optimum dosage ratio of SUBC-SMRH (0.25 g): PRIOW (1 g): mass of microbial-assisted biocarrier material (0.05 g). The TPH degradation was confirmed by SARA fractional analysis, FT-IR, ¹H NMR and GC-MS analyses. The study suggested that the application of covalently engineered SUBC has resulted in the accelerated degradation of real-time PRIOW in a very short duration without any secondary sludge generation. Thus, the SUBC integrated approach can be considered to effectively manage the hydrocarbon contaminants from petroleum refining industries under optimal conditions.

Widespread conversion of coastal wetlands into aquaculture ponds in coastal region often results in degradation of the wetland ecosystems, but its effects on sediment's potential to produce greenhouse gases remain unclear. Using field sampling, incubation experiments and molecular analysis, we studied the sediment CH₄ production potential and the relevant microbial communities in a brackish marsh and the nearby aquaculture ponds in the Min River Estuary in southeastern China. Sediment CH₄ production potential was higher in the summer and autumn months than in spring and winter months, and it was significantly correlated with sediment carbon content among all environmental variables. The mean sediment CH₄ production potential in the aquaculture ponds (20.1 ng g⁻¹ d⁻¹) was significantly lower than that in the marsh (45.2 ng g⁻¹ d⁻¹). While Methanobacterium dominated in both habitats (41–59%), the overall composition of sediment methanogenic archaea communities differed significantly between the two habitats (p < 0.05) and methanogenic archaea alpha diversity was lower in the aquaculture ponds (p < 0.01). Network analysis revealed that interactions between sediment methanogenic archaea were much weaker in the ponds than in the marsh. Overall, these findings suggest that conversion of marsh land to aquaculture ponds significantly altered the sediment methanogenic archaea community structure and diversity and lowered the sediment's capacity to produce CH₄.

Chlorpyrifos (CPF), as an organophosphate insecticide extensively used in the modern agricultural system, has been gradually banned in many countries due to its reported health risks to organisms, including humans. This study used simulated paddy field experiments and carbon-14 tracing to explore the possibility of reducing environmental risks of chlorpyrifos application through appropriate agronomic practice. Results showed ¹⁴C-CPF concentration in rice plants planted in the red soil (RS) was significantly higher than that in black soil (BS) and fluvo-aquic soil (FS). The application of biochar and chicken manure in RS reduced ¹⁴C-CPF accumulation in rice plants, and the content of ¹⁴C-CPF in rice grains decreased by 25% and 50%, respectively. Adding biochar to all three soils reduced the migration of ¹⁴C-CPF, especially in FS with the highest risk of ¹⁴C-CPF migration. The addition of chicken manure in FS reduced the migration of ¹⁴C-CPF and the total residual amount of ¹⁴C-CPF in the soil. In addition, chicken manure treatment increased the formation of ¹⁴C-bound residues (BRs) in soils and changed the distribution ¹⁴C-BRs in humus. The results indicated that the degree of environmental risks associated with the CPF application varies with soil types and could be reduced by introducing suitable exogenous organic matter into different soils, which is of great significance for guiding the scientific application of chlorpyrifos in agronomic practices.
