Adaptive alterations in the fatty acids composition under induced oxidative stress in heavy metal-tolerant filamentous fungus Paecilomyces marquandii cultured in ascorbic acid presence

The ability of the heavy metal-tolerant fungus Paecilomyces marquandii to modulate whole cells fatty acid composition and saturation in response to IC₅₀ of Cd, Pb, Zn, Ni, and Cu was studied. Cadmium and nickel caused the most significant growth reduction. In the mycelia cultured with all tested metals, with the exception of nickel, a rise in the fatty acid unsaturation was noted. The fungus exposure to Pb, Cu, and Ni led to significantly higher lipid peroxidation. P. marquandii incubated in the presence of the tested metals responded with an increase in the level of linoleic acid and escalation of electrolyte leakage. The highest efflux of electrolytes was caused by lead. In these conditions, the fungus was able to bind up to 100 mg g⁻¹ of lead, whereas the content of the other metals in the mycelium was significantly lower and reached from 3.18 mg g⁻¹ (Cu) to 15.21 mg g⁻¹ (Zn). Additionally, it was shown that ascorbic acid at the concentration of 1 mM protected fungal growth and prevented the changes in the fatty acid composition and saturation but did not alleviate lipid peroxidation or affect the increased permeability of membranes after lead exposure. Pro-oxidant properties of ascorbic acid in the copper-stressed cells manifested strong growth inhibition and enhanced metal accumulation as a result of membrane damage. Toxic metals action caused cellular modulations, which might contributed to P. marquandii tolerance to the studied metals. Moreover, these changes can enhance metal removal from contaminated environment.

Phthalate acid esters (PAE), commonly named phthalates, are toxics classified as endocrine-disrupting compounds; they are primarily used as additives to improve the flexibility in polyvinyl chloride. Many studies have reported the occurrence of phthalates in different environmental matrices; however, none of these studies has yet established a complete overview for those compounds in the water cycle within an urban environment. This review summarizes PAE concentrations for all environmental media throughout the water cycle, from atmosphere to receiving waters. Once the occurrences of compounds have been evaluated for each environmental compartment (urban wastewater, wastewater treatment plants, atmosphere, and the natural environment), we reviewed data in order to identify the fate of PAE in the environment and establish whether geographical and historical trends exist. Indeed, geographical and historical trends appear between Europe and other countries such as USA/Canada and China, however they remain location dependent. This study aimed at identifying both the correlations existing between environmental compartments and the processes influencing the fate and transport of these contaminants into the environment. In Europe, the concentrations measured in waterways today represent the background level of contamination, which provides evidence of a past diffuse pollution. In contrast, an increasing trend has actually been observed for developing countries, especially for China.

The objective of the study was to determine the leachates toxicity from sewage sludge-amended soils (sandy and loamy). Samples originated from a plot experiment realized over a period of 29 months. Two types of soil were fertilized with sewage sludges at the dose of 3 % (90 t/ha). Soil samples were taken after 0, 7, 17, and 29 months from the application of sewage sludges. Leachates were obtained according to the EN 12457–2 protocol. The following commercial tests were applied for the estimation of the toxicity: Microtox (Vibrio fischeri), Microbial assay for toxic risk assessment (ten bacteria and one yeast), Protoxkit F™ (Tetrahymena thermophila), Rotoxkit F™ (Brachionus calyciflorus), and Daphtoxkit F™ (Daphnia magna). The test organisms displayed varied toxicity with relation to the soils amended with sewage sludges. The toxicity of the leachates depended both on the soil type and on the kind of sewage sludge applied. Notable differences were also observed in the sensitivity of the test organisms to the presence of sewage sludge in the soil. The highest sensitivity was a characteristic of B. calyciflorus, while the lowest sensitivity to the presence of the sludges was revealed by the protozoa T. thermophila. Throughout the periods of the study, constant variations of toxicity were observed for most of the test organisms. The intensity as well as the range of those variations depended both on the kind of test organism and on the kind of sludge and soil type. In most cases, an increase of the toxicity of soils amended with the sewage sludges was observed after 29 months of the experiment.

Nutrient load into the ocean can be retained during the process of plant uptake and sedimentation in marshes along the bay zone. Seasonal variations of biomass and nutrient concentration in three dominated plant assemblages and associated sediments were monitored in this study area to determine effects of salt marsh on nutrient retention. Results showed that plant aboveground biomass displayed a unimodal curve with nutrient concentration generally decreased from spring to winter. Belowground biomass was relatively low during the rapid growth period with nutrient concentration tending to decrease and then increase during this period. Plant total nitrogen (TN) pools are higher than total phosphorus (TP) pools, and both pools showed significant seasonal variations. Water purification coefficients (WPC) of nutrients by plant assimilation were 34.4/17.3, 19.3/24.0, and 5.14/6.04 t/(m(2) year) (TN/TP) for Phragmites australis, Spartina alterniflora, and Scirpus mariqueter, respectively. Overall, these results suggest that higher annual plant biomass and nutrient assimilation contribute to greater nutrient retention capacity and accumulation in sediments, thereby enabling reduced eutrophication in transitional waters.

Effect of nitric oxide donor (sodium nitroprusside, SNP, 500 μM) or hydrogen peroxide scavenger (dithiothreitol, DTT, 500 μM) on cadmium (Cd) or copper (Cu) uptake (150 μM solutions) and toxicity using Scenedesmus quadricauda was studied. Combined treatments (Cd or Cu + DTT or SNP) usually ameliorated metal-induced toxicity at the level of pigments, proteins, and mineral nutrients in comparison with respective metal alone. Viability tests (MTT and TTC) showed the lowest values preferentially in Cu treatments, indicating higher toxicity in comparison with Cd. Cd showed low impact on amino acids while strong Cu-induced depletion was mitigated by DTT and SNP. Amount of ROS and NO showed the most pronounced responses in SNP variants being rather reciprocal than parallel and regulated ascorbate peroxidase activity. Blot gel analyses of hsp70 protein did not reveal extensive changes after given exposure period. Phenols were elevated by DTT alone while all Cu treatments revealed depletion. Total Cu content decreased while total Cd content increased in metal + SNP or metal + DTT. Subsequent experiment using lower Cd, SNP or DTT doses (10 and 100 μM) revealed concentration-dependent impact on Cd uptake. Overall, DTT was found to be more suitable for the amelioration of metal-induced toxicity.

Standard species used in ecological risk assessment are chosen based on their sensitivity to various toxicants and the ease of rearing them for laboratory experiments. However, this mostly overlooks the fact that species in the field that may employ variable life-history strategies, which may have consequences concerning the vulnerability of such species to exposure with contaminants. We aimed to highlight the importance of copepods in ecology and to underline the need to include freshwater copepods in ecotoxicology. We carried out a literature search on copepods and Daphnia in ecology and ecotoxicology to compare the recognition given to these two taxa in these respective fields. We also conducted a detailed analysis of the literature on copepods and their current role in ecotoxicology to characterize the scale and depth of the studies and the ecotoxicological information therein. The literature on the ecology of copepods outweighed that in ecotoxicology when compared with daphnids. Copepods, like other zooplankton, were found to be sensitive to toxicants and important organisms in aquatic ecosystems. The few studies that were conducted on the ecotoxicology of copepods mainly focused on marine copepods. However, very little is known about the ecotoxicology of freshwater copepods. To enable a more realistic risk higher tier environmental risk assessment, we recommend considering freshwater copepods as part of the hazard assessment process. This could include the establishment of laboratory experiments to analyse the effects of toxicants on copepods and the development of individual-based models to extrapolate effects across species and scenarios.

Pterocarya stenoptera is a native deciduous tree species and a candidate for reforestation in the riparian zones of the Three Gorges Reservoir Region of Yangtze River in China. Water treatments of continuous flooding (CF) and periodic flooding–drought (PF) were applied to examine the growth dynamics of 4-month-old P. stenoptera seedlings and its effects on soil chemical properties. Results showed that P. stenoptera seedlings in both CF and PF significantly decreased leaf biomass accumulation and the height and diameter growth as compared to that in control (CK; treatment with well-watered, well-drained soil), respectively. There was no significant difference in stem biomass among the three groups, but root biomass in PF showed severe reduction compared to that in both CK and CF. Total biomass in PF was significantly decreased compared to that in CK, but comparable to that in CF. Furthermore, no significant difference was found between CF and CK in total biomass. Water treatments in the unplanted soil pots significantly influenced soil pH, soil organic matter (OM), total nitrogen (TN), and alkali hydrolysable nitrogen (AN) contents, in contrast to no significant effects in total phosphorus (TP), total potassium (TK), available phosphorus (AP), and available potassium (AK) contents. In P. stenoptera soils, there were significant effects by water treatment, time, and treatment × time in the eight tested soil chemical properties, except treatment in TK and time effect in OM content. Compared to unplanted soils, the growth of P. stenoptera seedlings significantly increased soil pH value and OM, TN, TP, and TK contents, while decreasing AN, AP, and AK contents in CK group, augmented the mean value of each of the tested soil chemical properties with an exception of AK content in CF group, and increased soil pH value and TN, AN, TP, and AP contents with no significant differences in OM, TK, and AK contents in PF group. Given the fact that TN and TP contents significantly increased in P. stenoptera soils as compared to those in unplanted soils, growth of P. stenoptera seedlings should be a successful candidate for restoration within the highly dynamic hydrologic zone of the riparian zones of the Three Gorges Reservoir Region.

Synthetic wastewaters are widely used in many fields of wastewater research and operational management. However, few comparative studies have been conducted on the large number of published formulations. Eleven synthetic formulations simulating municipal wastewaters were selected based on their frequency of use, relative complexity, ease of formulation and cost and compared to two real municipal wastewaters. Synthetic wastewaters vary significantly in their compositions and characteristics, especially in terms of alkalinity, BOD K ₁, SOUR, BODU, COD/BOD and C/N/P ratio, although they are portrayed as ‘typical’ in terms of characteristics and suitability for use. The pH, alkalinity and the presence of Ca and Mg should be considered in combination with the diluent used. Where the diluent is tap water, then the presence of treatment chemicals should also be considered. The effects of the micronutrients present are also an important factor. The study found that no single formulation is appropriate for all situations. Both the Syntho and Synthes formulations attempt to simulate real wastewater, whereas other formulations primarily act as readily biodegradable vectors for toxicity analyses, characterisation studies and treatment process evaluations. The criteria for choosing a particular synthetic wastewater very much depend on its intended application and require careful selection.

The science parks have helped shape Taiwan as a high-tech island with a good reputation worldwide. But some complaints on air pollution from the science parks have recently risen. To better understand the environmental effects of volatile organic compounds (VOCs) emitted from various high-tech factories in a science park, this study uses a source–receptor approach to characterize the environmental effects of VOCs from control device exhaust in Taichung Science Park. The chemical mass balance model (CMB8.2) of field measurements of 30 stacks and ambient air at nine sites was used to identify the source and relative contribution of ambient VOCs. The exhaust gas of various pollution control devices was also sampled by drawing a stream of the gases from the exhaust duct at its sampling port. The VOC source profile of each control device exhaust was determined using a database of noncharacteristic compounds. Monthly ambient concentrations of 167 VOCs were divided into monsoon datasets to investigate the effect of monsoon conditions on the emission of VOCs in the science park. This study also suggests a method for determining the optimum source profile in source–receptor modeling, and identifies and analyzes the sources of ambient VOCs at nine sites during southwest and northeast monsoons. Results show a direct relationship between the relative contribution of each source and its control device efficiency. The proposed source–receptor approach can characterize the environmental effect of air pollutants from various factories and successfully assess the efficiency of various control devices.

Sexually mature male mosquitofish (Gambusia holbrooki) were collected from various sites around Melbourne in 2009 to evaluate the performance of gonopodial indices as a biomarker for endocrine disruption in Melbourne's waterways. The mosquitofish indices assessed were body length (BL), gonopodial length (GL)/BL ratio, ray 4:6 ratio and the absence or presence of hooks and serrae, and these varied between sites. The study was complemented by measurements of estrogenic, retinoid, thyroid and aryl hydrocarbon (AhR) receptor activities of the water. Male mosquitofish were 16.3–21.5 mm in length, and although there was a statistically significant positive relationship showing that bigger fish had longer gonopodia than small fish (r2 = 0.52, p < 0.001), there were few significant differences in GL/BL ratio of fish between sites. Measured estrogenic activity was mostly in the range 0.1–1.7 ng/L EEQ, with one site having much higher levels (~12 ng/L EEQ). Aryl hydrocarbon (AhR) receptor activity was observed in all water samples (7–180 ng/L βNF EQ), although there was no consistent pattern in the level of AhR activity observed, i.e., ‘clean’ sites were as likely to return a high AhR activity response as urban or wastewater treatment plant (WWTP)-impacted sites. There was no correlation between measurements of receptor actvity and gonopodial length (GL):BL ratio and BL. We conclude that the mosquitofish gonopodia only fulfills part of the criteria for biomarker selection for screening. The mosquitofish indices assessed were cheap and easy-to-perform procedures; however, there is no baseline data from the selected sites to evaluate whether differences in the morpholical indices observed at a site were a result of natural selection in the population or due to estrogenic exposure.
