Traffic-related air pollution and endurance exercise: Characterizing non-targeted serum metabolomics profiling

Although the exposure to traffic-related air pollution (TRAP) has emerged as one of main problem worldwide to inhabitants’ health in urban centers, its impact on metabolic responses during exercise is poorly understood. The aim of study was to characterize the profile of non-target serum metabolomics during prolonged exercise performed under TRAP conditions. Ten healthy men completed two 90 min constant-load cycling trials under conditions of either TRAP or filtered air. Experimental trials were performed in a chamber located on an avenue with a high volume of vehicle traffic. Blood samples were taken at 30 min, 60 min, and 90 min of exercise. Based on Nuclear Magnetic Resonance metabolomics, the non-target analysis was used to assess the metabolic profile. Twelve, 16 and 18 metabolites were identified as discriminants. These were: at 30 min of exercise, the coefficient of determination (R²) 0.98, the predictive relevance, (Q²) 0.12, and the area under the curve (AUC) 0.91. After 60 min of exercise: (R²: 0.99, Q²: 0.09, AUC: 0.94); and at 90 min of exercise (R²: 0.91, Q²: <0.01, AUC: 0.89), respectively. The discriminant metabolites were then considered for the target analysis, which demonstrated that the metabolic pathways of glycine and serine metabolism (p = 0.03) had been altered under TRAP conditions at 30 min of exercise; arginine and proline metabolism (p = 0.04) at 60 min of exercise; and glycolysis (p = 0.05) at 90 min of exercise. The present results suggest that exposure to TRAP during prolonged exercise leads to a significant change in metabolomics, characterized by a transitional pattern and lastly, impairs the glucose metabolism.

Ammonia (NH₃) emissions, the majority of which arise from livestock production, are linked to high concentration of PM₂.₅ and lower air quality in China. NH₃ mitigation options were well studied at the small-scale (laboratory or pilot), however, they lack of a large-scale test in China. This study fills this crucial gap by evaluating the cost-benefit of pioneering NH₃ mitigation projects carried out for a whole county – Sheyang, Jiangsu province, China. Measures were implemented in 2019 following two distinct strategies, improved manure treatment for industrial livestock farms, and collection and central treatment for traditional livestock farms. Emission reductions of 16% were achieved in a short time. While this is remarkable, it falls short of expectations from small-scale studies. If measures were fully implemented according to purpose and meet expectations from the small scale, higher emission reductions of 42% would be possible. The cost benefit analysis presented in this study demonstrated advantages of central manure treatment over in-farm facilities. With improved implementation of mitigation strategies in industrial livestock farms, traditional livestock farms may play an increasing role in total NH₃ emissions, which means such farms either need to be included in future NH₃ mitigation policies or gradually replaced by industrial livestock farms.The study found an agricultural NH₃ reduction technology route suitable for China's national conditions (such as the “Sheyang Model”).

As a result of historical industrial activity, the sediments in the inner harbor of Owen Sound Bay in the northeastern part of Lake Huron in Ontario, Canada are contaminated with organic compounds. The present study showed that the concentrations of Ʃ PAH₁₆₋EPA in all sediments in the inner harbor were above the sediment quality guidelines for the province of Ontario, Canada, with mean Ʃ PAH₁₆₋EPA concentrations at the most contaminated site of 46,000 μg/kg dry weight. The concentrations of polychlorinated biphenyls, brominated diphenyl ethers, and organochlorine compounds were all below sediment quality guidelines. The patterns of PAH and alkyl-PAH compounds in sediment cores indicated that contamination is from mixed sources, with a strong indication of pyrogenic contamination from industries that used to operate in the area, including a coal gasification plant. Other areas of the bay are impacted by petrogenic contamination, potentially from spills of fuel. The even distribution of PAH and alkyl-PAH compounds throughout core profiles at depths up to 25 cm indicates that this is a dynamic system and contaminated sediments are not being covered by deposition of less contaminated sediments. This study illustrates the value of determining the patterns of both PAH and alkyl-PAH compounds in sediments for regulatory purposes and also for forensic source tracking.

The co-occurrence of enhancement in aerosol concentration, temperatures, and ozone mixing ratio was observed between June 29 and July 4, 2018 (enhanced period, EP) on Long Island (LI) and the greater NYC metropolitan area during part of the 2018 Long Island Sound Tropospheric Ozone Study (LISTOS). Two aerosol formation pathways were identified during the EP, the first being the condensation of semi- and intermediate volatility oxidation products of anthropogenic volatile organic compounds (AVOCs) under stagnant synoptic flow conditions, high temperatures and afternoon sea-breeze circulation. While this first pathway was prevalent, the most abundant organic aerosol factor was less oxidized oxygenated organic aerosol or LO-OOA. The second formation pathway occurred during a period of more persistent (synoptic) on-shore flow transporting more aged aerosol which consisted of an internal mixture of more oxidized oxygenated organic aerosol (MO-OOA), methanesulfonic acid (MSA) and sulfate. It was estimated that 35% of the sulfate observed during the mature period (an average of about 1.2 μg m⁻³) originated from oceanic dimethyl sulfide (DMS) emissions. These two formation pathways helped elucidate the sources of fine particle pollution, highlighted the interaction between human emissions and natural DMS emission, and will help our understanding of pollution affecting other urban areas adjacent to large bodies of water during hot and stagnant periods.

Deca-brominated diphenyl ether (BDE-209) is shown to be persistent in soil and it's urgent to find an effective method to remove BDE-209 from contaminated soil. In this study, the investigation of BDE-209 degradation in three different rice-vegetable rotation soils induced by electron beam (E-beam) irradiation under flooded and non-flooded conditions was conducted. The reductive efficiency of ¹⁴C-BDE209 reached the highest level at 50 kGy and the values in flooded soils of rice-eggplant rotation soil (RES), rice-peanut soil (RPS), and rice-chili pepper soil (RCS) were 93.5%, 87.2%, and 73.8%, respectively. The reductive efficiencies in non-flooded soils of RES, RPS, and RCS were 73.4%, 81.0%, and 78%, respectively. The D₀.₅ values (dose required for reducing 50% BDE-209) of BDE-209 in non-flooded soils were lower than those in flooded soils, suggesting greater degradation efficiency of BDE-209 in non-flooded soils than in flooded soils. The BDE-209 was degraded into higher-brominated PBDEs and lower-brominated PBDEs by E-beam irradiation. The results demonstrate that BDE-209 in the soil can be degraded by E-beam irradiation, non-flooded condition is better than flooded condition for the removal of BDE-209, and the main degradation mechanism of BDE-209 by E-beam irradiation is debromination. This study provides a rapid and effective method for degrading BDE-209 that is persistent in soils, and has important implications for the remediation of soil contaminated by PBDEs in and around E-waste dismantling areas.

Mine tailings sites are considered as a continuous source of discharged metal(loid)s and residual organic flotation reagents. They are extremely toxic environments representing unique ecological niches for microbial communities. Mine tailings as a source of multi-resistance genes have been poorly investigated. Metagenomic analysis for four active nonferrous metal(loid) tailings sites with different environmental parameters was conducted. The abundance of Thiobacillus, able to tolerate acidity and showing iron- and sulfur/sulfide oxidation capacities, was significantly different (p < 0.05) between acid and neutral tailings sites. Correlation analyses showed that Zn, Pb, TP, Cd, and Cu were the main drivers influencing the bacterial compositions. Multi-metal resistance genes (MRGs) and antibiotic resistance genes (ARGs), such as baca and copA, were found to be co-selected by high concentrations of metal(loid)s tailings. The main contributors to different distributions of MRGs were Thiobacillus and Nocardioides genus, while genera with low abundance (<0.1%) were the main contributors for ARGs. Functional metabolic pathways related to Fe–S metabolism, polycyclic aromatic hydrocarbons (PAHs) degradation and acid stress were largely from Altererythrobacter, Lysobacter, and Thiobacillus, respectively. Such information provides new insights on active tailings with highly toxic contaminants. Short-term metal(loid) exposure of microorganism in active nonferrous metal(loid) tailings contribute to the co-occurrence of ARGs and MRGs, and aggravation of tailings acidification. Our results recommend that the management of microorganisms involved in acid tolerance and metal/antibiotic resistance is of key importance for in-suit treatment of the continuous discharge of tailings with multiple metal(loid) contaminants into impoundments.

Common plasticizers and their alternatives are environmentally ubiquitous and have become a global problem. In this study, common plasticizers (phthalates and metabolites) and new alternatives [bisphenol analogs, t-butylphenyl diphenyl phosphate (BPDP), and bisphenol A bis(diphenyl phosphate) (BDP)] were quantified in urine and hair samples from children in Hong Kong, drinking water (tap water/bottled water) samples, and airborne particle samples from 17 kindergartens in Hong Kong. The results suggested that locally, children were exposed to various plasticizers and their alternatives. High concentrations of BPDP and BDP were present in urine, hair, tap water, bottled water, and air particulate samples. The geometric mean (GM) concentrations of phthalate metabolites in urine samples (126–2140 ng/L, detection frequencies < 81%) were lower than those detected in Japanese and German children in previous studies. However, a comparison of the estimated daily intake values for phthalates in tap water [median: 10.7–115 ng/kg body weight bw/day] and air particles (median: 1.23–7.39 ng/kg bw/day) with the corresponding reference doses indicated no risk. Bisphenol analogs were detected in 15–64% of urine samples at GM concentrations of 5.26–98.1 ng/L, in 7–74% of hair samples at GM concentrations of 57.5–2390 pg/g, in 59–100% of kindergarten air samples at GM concentrations of 43.1–222 pg/m³, and in 33–100% of tap water samples at GM concentrations of 0.90–3.70 ng/L. A significant correlation was detected between the concentrations of bisphenol F in hair and urine samples (r = 0.489, p < .05). The estimated daily urinary excretion values of bisphenol analogs suggest that exposure among children via tap water intake and airborne particle inhalation in kindergartens cannot be ignored in Hong Kong.

The well-known toxicity of conventional chemical oil spill dispersants demands the development of alternative and environmentally friendly dispersant formulations. Therefore, in the present study we have developed a pair of less toxic and green dispersants by combining lactonic sophorolipid (LS) biosurfactant individually with choline myristate and choline oleate ionic liquid surfactants. The aggregation behavior of resulted surfactant blends and their dispersion effectiveness was investigated using the baffled flask test. The introduction of long hydrophobic alkyl chain with unsaturation (attached to choline cation) provided synergistic interactions between the binary surfactant mixtures. The maximum dispersion effectiveness was found to be 78.23% for 80:20 (w/w) lactonic sophorolipid-choline myristate blends, and 81.15% for 70:30 (w/w) lactonic sophorolipid-choline oleate blends at the dispersant-to-oil ratio of 1:25 (v/v). The high dispersion effectiveness of lactonic sophorolipid-choline oleate between two developed blends is attributed to the stronger synergistic interactions between surfactants and slower desorption rate of blend from oil-water interface. The distribution of dispersed oil droplets at several DOR were evaluated and it was observed that oil droplets become smaller with increasing DOR. In addition, the acute toxicity analysis of developed formulations against zebra fish (Danio rerio) confirmed their non-toxic behavior with LC₅₀ values higher than 400 ppm after 96 h. Overall, the proposed new blends/formulations could effectively substitute the toxic and unsafe chemical dispersants.

Heavy fuel oil (HFO) accounts for approximately 80% of the fuel consumption of ocean-going ships in the world. Multiple toxic species are found in HFO exhaust, however, carbonaceous substances emitted from low-speed marine engine exhaust at different operating loads have not been thoroughly addressed. Therefore, a bench test for a low-speed marine engine with HFO fuel under different operating modes was carried out in this study. Emission factors and characteristics of CO₂, CO, organic carbon (OC), elemental carbon (EC), as well as OC and EC fragments, organic matters of n-alkanes and polycyclic aromatic hydrocarbons (PAHs) are given and discussed. Combined with the correlation analysis results among the measured species and engine technical parameters, the formation processes and influence factors of carbonaceous components are also inferred in this study. Besides, together with OC to EC ratio, n-alkanes to PAHs ratio, etc., EC1 to soot-EC ratio in PM can be considered as tracer characteristic of high-sulfur-content HFO ship distinguished from diesel fuel ships. Profiles of n-alkanes and PAHs in PM can be used to distinguish shipping emission source from other combustion sources. Moreover, characteristics of carbonaceous components in size-segregated particles are also discussed, including OC, EC, OC and EC fragments, as well as organic matters. Results show that most of the particle mass, OC, EC, and organic matters are concentrated in fine particles with size of less than 1.1 μm, indicating the significance of ultrafine particles. Formation processes of OC and EC fragments, EC1 and soot-EC are also deduced and proved combined with the characteristics of OC and EC fragments, organic matters, and especially PAHs. Besides, the large variations of OC to EC ratios and speciated profiles of n-alkanes and PAHs in different particle size bins indicate that particle size should be considered when they are used as characteristic tracer in source apportionment studies.

N6-methyladenosine (m⁶A) mRNA methylation plays a role in various brain functions. Exposure to chronic constant light (CCL) has been reported to impair cognition, yet whether the underlying mechanism involves m⁶A remains unknown. In this study, mice exposed to CCL for 3 weeks show impaired cognitive behavior, which was associated with increased m⁶A level in hippocampus. Accordingly, the m⁶A demethylase FTO was inhibited while the methyltransferases METTL3, METTL14 and WTAP, as well as the reader protein YTHDF2, were elevated in the hippocampus of CCL-exposed mice. CCL exposure significantly activated hippocampal expression of circadian regulator cryptochrome 1 and 2 (CRY1 and 2). Meanwhile, hippocampal neurogenesis was impaired with suppression of BDNF/TrκB/ERK pathway. To further delineate the signaling pathway and the role of m⁶A, we altered the expression of CRY1/2 in hippocampus neuron cells. CRY1/2 overexpression inhibited FTO and increased m⁶A levels, while CRY1/2 knockdown led to opposite results. Luciferase reporter analysis further confirmed CRY1/2-induced FTO suppression. Furthermore, FTO knockdown increased m⁶A on 3′UTR of TrκB mRNA, and decreased TrκB mRNA stability and TrκB protein expression, in a YTHDF2-dependent manner. These results indicate that CCL-activated CRY1/2 causes transcriptional inhibition of FTO, which suppresses TrκB expression in hippocampus via m⁶A-dependent post-transcriptional regulation and contributes to impaired cognitive behavior in mice exposed to constant light.
