DNA integrity assessment in hemocytes of soft-shell clams (Mya arenaria) in the Saguenay Fjord (Québec, Canada)

The purpose of this study was to examine the effects of pollution on DNA integrity in the feral soft-shell clam (Mya arenaria) in the Saguenay Fjord. Intertidal clams were collected downstream and upstream of the fjord at sites under anthropogenic pollution. DNA integrity was assessed by following changes in single- and double-stranded breaks, variation in DNA content and micro-nuclei (MN) incidence in hemocytes. The results revealed that clams collected at polluted sites had reduced DNA strand breaks (lower DNA repair activity), increased DNA content variation and MN frequency in hemocytes. The data revealed that DNA content variation was closely related to MN frequency and negatively with DNA strand breaks formation. Water conductivity was also related to reduced MN frequency and DNA content variation, indicating that, in addition to the effects of pollution, the gradual dilution of saltwater could compromise mussel health.

The aim of this study is to investigate the use of keratin colloidal solution, which was obtained from wool, for the removal of Pb(II) from water. The addition of keratin colloidal solution (15 g L(-1), 0.30 mL) to a Pb(II) solution (1.0 mM, 0.90 mL, pH 5.0) resulted in the formation and precipitation of a Pb-keratin aggregate. Measurement of the Pb(II) and protein concentrations in the supernatant solution revealed that 88 and 99 % of the Pb(II) and keratin protein were removed from the solution, respectively. The maximum Pb(II) uptake capacity of keratin in the colloidal solution was 43.3 mg g(-1). In addition, the Pb-keratin aggregate was easily decomposed via the addition of nitric acid, which enabled the recovery of Pb(II). However, aggregation did not occur in solutions with Pb(II) concentrations below 0.10 mM. Therefore, we used a keratin colloidal solution encapsulated in a dialysis cellulose tube to remove Pb(II) from 0.10 mM solutions, which enabled the removal of 95 % of the Pb(II). From these results, we conclude that keratin colloidal solution is useful for the treatment of water polluted with Pb(II).

This research was executed between March 2009 and March 2010 to monitor particulate matter size distribution and its composition in Istanbul. Particulate matter composition was determined using ion chromatography and inductively coupled plasma optical emission spectrometry. The sampling point is adjacent to a crowded road and the Bosporus Strait. Two prevailing particulate modes are found throughout PM₁₀ by sampling with a nine-stage low-volume cascade impactor. First mode in the fine mode is found to be between 0.43 and 0.65 μm, whereas the other peak was observed between 3.3 and 4.7 μm, referring to the coarse mode. The mean PM₁₀ concentration was determined as 41.2 μg/m³, with a standard deviation of 16.92 μg/m³. PM₀.₄₃ had the highest mean concentration value of 10.67 μg/m³, making up nearly one fourth of the total PM₁₀ mass. For determining the effect of traffic on particulate matter (PM) composition and distribution, four different sampling cycles were applied: entire day, nighttime, rush hour, and rush hour at weekdays. SO ₄ ⁻² and organic carbon/elemental carbon proportions are found to be lower in night samples, representing a decrease in traffic. The long-range transports of dust storms were observed during the sampling periods. Their effects were determined analytically and their route models were run by the HYSPLIT model and validated through satellite photographs taken by the NASA Earth Observatory.

The adsorption of crystal violet from aqueous solution by NaOH-modified rice husk was investigated in a laboratory-scale fixed-bed column. A two-level three factor (2³) full factorial central composite design with the help of Design Expert Version 7.1.6 (Stat Ease, USA) was used for optimisation of the dynamic dye adsorption process and evaluation of interaction effects of different operating parameters: initial dye concentration (100–200 mg L⁻¹), flow rate (10–30 mL min⁻¹) and bed height (5–25 cm). A correlation coefficient (R ²) value of 0.999, model F value of 1,936.59 and its low p value (<0.0001) along with lower value of coefficient of variation (1.38 %) indicated the fitness of the response surface quadratic model developed during the present study. Numerical optimisation applying desirability function was used to identify the optimum conditions for a targeted breakthrough time of 12 h. The optimum conditions were found to be initial solution pH = 8.00, initial dye concentration = 100 mg L⁻¹, flow rate = 22.88 mL min⁻¹ and bed height = 18.75 cm. A confirmatory experiment was performed to evaluate the accuracy of the optimised procedure. Under the optimised conditions, breakthrough appeared after 12.2 h and the column efficiency was determined as 99 %. The Thomas model showed excellent fit to the dynamic dye adsorption data obtained from the confirmatory experiment. Thereby, it was concluded that the current investigation gives valuable insights for designing and establishing a continuous wastewater treatment plant.

The stringent regulations for discharging acid mine drainage (AMD) has led to increased attention on traditional or emerging treatment technologies to establish efficient and sustainable management for mine effluents. To assess new technologies, laboratory investigations on AMD treatment are necessary requiring a consistent supply of AMD with a stable composition, thus limiting environmental variability and uncertainty during controlled experiments. Additionally, biotreatment systems using live cells, particularly micro-algae, require appropriate nutrient availability. Synthetic AMD (Syn-AMD) meets these requirements. However, to date, most of the reported Syn-AMDs are composed of only a few selected heavy metals without considering the complexity of actual AMD. In this study, AMD was synthesised based on the typical AMD characteristics from a copper mine where biotreatment is being considered using indigenous AMD algal-microbes. Major cations (Ca, Na, Cu, Zn, Mg, Mn and Ni), trace metals (Al, Fe, Ag, Na, Co, Mo, Pb and Cr), essential nutrients (N, P and C) and high SO₄ were incorporated into the Syn-AMD. This paper presents the preparation of chemically complex Syn-AMD and the challenges associated with combining metal salts of varying solubility that is not restricted to one particular mine site. The general approach reported and the particular reagents used can produce alternative Syn-AMD with varying compositions. The successful growth of indigenous AMD algal-microbes in the Syn-AMD demonstrated its applicability as appropriate generic media for cultivation and maintenance of mining microorganisms for future biotreatment studies.

Landscape lakes in the city suffer high eutrophication risk because of their special characters and functions in the water circulation system. Using a landscape lake HMLA located in Tianjin City, North China, with a mixture of point source (PS) pollution and non-point source (NPS) pollution, we explored the methodology of Fluent and AQUATOX to simulate and predict the state of HMLA, and trophic index was used to assess the eutrophication state. Then, we use water compensation optimization and three scenarios to determine the optimal management methodology. Three scenarios include ecological restoration scenario, best management practices (BMPs) scenario, and a scenario combining both. Our results suggest that the maintenance of a healthy ecosystem with ecoremediation is necessary and the BMPs have a far-reaching effect on water reusing and NPS pollution control. This study has implications for eutrophication control and management under development for urbanization in China.

Bioavailable concentrations of polycyclic aromatic hydrocarbons (PAHs) were investigated in water of Three Gorges Reservoir (TGR) using semipermeable membrane devices during the period of completely impounding water. ∑PAH concentrations in water of TGR in the period of completely impounding water were 15-381 ng L(-1). ∑PAH concentrations increased from town or counties to big industrialized cities in TGR, indicating urbanization effects on PAH pollution in the water. Tributaries in TGR have a certain contribution of PAH pollution to the mainstream of Yangtze River and their pollution could not be neglected. An obvious decrease of PAH concentration was observed after 175-m water impounding in 2011 in TGR. Several factors may account for this decrease, including execution of comprehensive treatment and management measures in TGR, less rainfall in 2011, and sedimentation effect caused by the dam. Passive sampling method has been successfully applied in the investigation of trace PAH in water of TGR and proved to be a useful and efficient tool for the management and sustainable development of the big reservoir. The results of the study provide valuable information about PAH pollution in the whole reservoir including some tributaries, and the pollution status is dynamically related with human activities. Therefore, PAH could be used as a marker compound or indicator in the network monitoring system to surveil and trace the pollution status in TGR.

Bisphenol A (BPA), which is ubiquitous in the environment, is an example of an endocrine-disrupting compound (EDC). Ammonium assimilation has an important function in plant growth and development. However, insufficient information on the potential effect of BPA on ammonium assimilation in plants is available. In this study, the effects of BPA on ammonium assimilation in roots of soybean seedlings were investigated. During the stress period, 1.5 mg L(-1) of BPA improved glutamine synthetase (GS)/glutamate synthase (GOGAT) cycle and glutamate dehydrogenase (GDH) pathway in ammonium assimilation. The amino acid and the soluble protein contents increased in the soybeans. At 17.2 and 50.0 mg L(-1) of BPA, the GS/GOGAT cycle was inhibited and the GDH pathway was promoted. The amino acid content increased and the soluble protein content decreased. During the recovery period, the GS/GOGAT cycle and the GDH pathway recovered at 1.5 and 17.2 mg L(-1) of BPA but not at 50.0 mg L(-1) of BPA. The amino acid content continuously increased and the soluble protein content decreased compared with those in the control treatment. In summary, BPA treatment could affect the contents of soluble protein and amino acid in the soybean roots by regulating ammonium assimilation.

Industrial activity is one of the main sources of ambient pollution in developed countries. However, research analyzing its effect on birth outcomes is inconclusive. We analyzed the association between proximity of mother's municipality of residence to industries from 24 different activity groups and risk of very (VPTB) and moderate (MPTB) preterm birth, very (VLBW) and moderate (MLBW) low birth weight, and small for gestational age (SGA) in Spain, 2004-2008. An ecological study was defined, and a "near vs. far" analysis (3.5 km threshold) was carried out using Hierarchical Bayesian models implemented via Integrated Nested Laplace Approximation. VPTB risk was higher for mothers living near pharmaceutical companies. Proximity to galvanization and hazardous waste management industries increased the risk of MPTB. Risk of VLBW was higher for mothers residing near pharmaceutical and non-hazardous or animal waste management industries. For MLBW many associations were found, being notable the proximity to mining, biocides and animal waste management plants. The strongest association for SGA was found with proximity to management animal waste plants. These results highlight the importance of further research on the relationship between proximity to industrial sites and the occurrence of adverse birth outcomes especially for the case of pharmaceutical and animal waste management activities.

As part of a large study on assessing the impact of environmental contaminants in Indian avifauna, the presence of organochlorine pesticides (OCPs) in liver tissues of 16 species of birds collected from Ahmedabad, India during 2005–2007 was quantified. The higher concentrations of total organochlorine pesticides were detected in livers of shikra Accipiter badius (3.43 ± 0.99 μg/g wet wt) and the lower levels in white ibis Pseudibis papillosa (0.02 ± 0.01 μg/g wet wt). Marked differences in the concentrations of total OCPs occurred among species (p < 0.05). Concentrations of DDT and its metabolites, hexachlorocyclohexane (HCH) and isomers, dieldrin, and heptachlor epoxide were lower than the concentrations reported for various species of birds in India. Accumulation pattern of organochlorine pesticides in birds was, in general, in the order HCH > DDT > heptachlor epoxide > dieldrin. Among various pesticides analyzed, p,p′-DDE and β-HCH contributed maximum towards the total OCPs and study indicates the continuous use of lindane and DDT for agriculture and public health purpose, respectively. Although no serious threat is posed by any of the organochlorine pesticides detected in the present study species, continued monitoring is recommended.
