B-vitamin supplementation ameliorates anxiety- and depression-like behavior induced by gestational urban PM2.5 exposure through suppressing neuroinflammation in mice offspring

PM₂.₅ exposure is an emerging environmental concern and severe health insult closely related to psychological conditions such as anxiety and depression in adolescence. Adolescence is a critical period for neural system development characterized by continuous brain maturation, especially in the prefrontal cortex. The etiology of these adolescent conditions may derive from fetal origin, probably attributed to the adverse effects induced by intrauterine environmental exposure. Anxiety- and depression-like behavior can be induced by gestational exposure to PM₂.₅ in mice offspring which act as a useful model system. Recent studies show that B-vitamin may alleviate PM₂.₅-induced hippocampal neuroinflammation- and function-related spatial memory impairment in adolescent mice offspring. However, cortical damage and related neurobehavioral defects induced by gestational PM₂.₅ exposure, as well as the potential reversibility by interventions in mice offspring require to be elucidated. Here, we aimed to investigate whether B-vitamin would protect mice offspring from the adverse effects derived from gestational exposure to urban PM₂.₅ on cortical areas to which anxiety and depression are closely related. Pregnant mice were divided into three groups: control group (treated with PBS alone), model group (treated with both PM₂.₅ and PBS), and intervention group (treated with both PM₂.₅ and B-vitamin), respectively. The mice offspring were then applied to comprehensive neurobehavioral, ultrastructural, biochemical, and molecular biological analyses. Interestingly, we observed that gestational PM₂.₅ exposure led to neurobehavioral defects including anxiety- and depression-like behavior. In addition, neuroinflammation, oxidative damage, increased apoptosis, and caspase-1-mediated inflammasome activation in the prefrontal cortex were observed. Notably, both behavioral and molecular changes could be significantly alleviated by B-vitamin treatment. In summary, our results suggest that the anxiety- and depression-like behavior induced by gestational PM₂.₅ exposure in mice offspring can be ameliorated by B-vitamin supplementation, probably through the suppression of apoptosis, oxidative damage, neuroinflammation, and caspase-1-mediated inflammasome activation.

Rainwater chemistry plays an important role in the earth-surficial ecosystem, but studies on rainwater chemical composition of karst agro-ecosystem are rare. To explore the rainwater alkalization and the provenance of components responsible for neutralization, two-years chemical monitoring of rainwater was carried out in a karst agricultural catchment in Southwest China. The main findings suggest that SO₄²⁻, NO₃⁻, Ca²⁺, and NH₄⁺ are the principal ions. All the ionic contents show distinctly seasonal variation (highest in winter) in response to variations in seasonal precipitation because the rain-scour process can efficiently remove atmospheric materials. Source identification indicates that Cl⁻ and Na⁺ are mainly derived from marine input whereas SO₄²⁻ and NO₃⁻ are controlled by anthropogenic emission, in particular, fixed emission sources. The source of NH₄⁺ is attributed to intense agricultural production, while Ca²⁺ and Mg²⁺ are mainly derived from calcite dissolution. The rainwater alkalization caused by the seasonal acid neutralization (via basic components, Ca²⁺ and NH₄⁺) is beneficial to crop growth but also reflect agricultural overfertilization. Sulfur controlled the total wet acid deposition (68%–94%) and could be a potential agent of weathering.

Polycyclic aromatic hydrocarbons (PAHs) pollution has become a worldwide environmental problem, and the spread of PAHs can cause carcinogenicity, mutagenicity, and toxicity to humans. However, the transfer and accumulation of PAHs in crop leaves has not been clearly understood. In this study, we first reported that the environmental temperature could induce phenanthrene transfer and accumulation in hydrocultured wheat, corn and soybean leaves via vacuum-infiltration-centrifugation method. Phenanthrene accumulation rises significantly (p < 0.05) in the first 8 h and reaches the maximum accumulation rate at the 4th h. Then the accumulation turns stable in both apoplast and sympalst of wheat, soybean and corn leaves. Temperature is positively correlated with phenanthrene accumulation in apoplast and sympalst of soybean and corn leaves, and phenanthrene accumulation increases under lower temperature in apoplast and sympalst of wheat leaves. Temperature also displays a positive correlation with the phenanthrene accumulation under gradient phenanthrene treatments in both apoplast and sympalst. In addition, the wheat, corn and soybean leaves have the same phenanthrene accumulation pathways and symplast pathway is major for phenanthrene accumulation with the contribution over 55% in total phenanthrene accumulation. Meanwhile, based on the Elovich equation, the symplast and apoplast processes of phenanthrene accumulation are endothermic. In the end, our findings will offer a new understanding for phenanthrene transfer and accumulation pathway in plant leaves and put forward a new biological reference of PAHs transfer in environmental science.

Trichlorfon is an organic phosphorus pesticide used to control different parasitic infections in aquaculture. The repeated, excessive use of trichlorfon can result in environmental pollution, thus affecting human health. This study aimed to determine the effects of different concentrations of trichlorfon (0, 0.1, 0.5 and 1.0 mg/L) on the intestinal barrier, oxidative stress, inflammatory response and intestinal microbiome of common carp. Trichlorfon exposure significantly reduced the height of intestinal villus and decreased the expression levels of tight junction genes, such as claudin-2, occludin and ZO-1, in common carp. Moreover, the activities of antioxidant enzymes, such as CAT, SOD and GSH-Px, exhibited a decreasing trend with increasing trichlorfon concentrations, while the contents of MDA and ROS elevated in the intestinal tissues of common carp. The mRNA and protein levels of pro-inflammatory cytokines TNF-α and IL-1β were significantly upregulated by trichlorfon exposure. The level of anti-inflammatory cytokine TGF-β was remarkably higher in 1.0 mg/L trichlorfon treatment group compared to control group. In addition, the results demonstrated that trichlorfon exposure could affect the microbiota community composition and decreased the community diversity in the gut of common carp. Notably, the proportions of some probiotic bacteria, namely, Lactobacillus, Bifidobacterium and Akkermansia, were observed to be reduced after trichlorfon exposure. In summary, the findings of this study indicate that exposure to different concentrations of trichlorfon can damage intestinal barrier, induce intestinal oxidative damage, trigger inflammatory reaction and alter gut microbiota structure in common carp.

We apply convolutional neural network (CNN) model for estimating daily 24-h averaged ground-level PM2.5 of the conterminous United States in 2011 by incorporating aerosol optical depth (AOD) data, meteorological fields, and land-use data. Unlike some of the recent supervised learning-based approaches, which only utilized the predictors from the location of which PM2.5 value is estimated, we naturally aggregate predictors from nearby locations such that the spatial correlation among the predictors can be exploited. We carefully evaluate the performance of our method via overall, temporally-separated, and spatially-separated cross-validations (CV) and show that our CNN achieves competitive estimation accuracy compared to the recently developed baselines. Furthermore, we develop a novel predictor importance metric for our CNN based on the recent neural network interpretation method, Layerwise Relevance Propagation (LRP), and identify several informative predictors for PM2.5 estimation.

Pesticides often occur as mixtures of complex compounds in water environments, while most of studies only focus on the toxic effects of individual pesticides with little attention to the joint toxic effects. In the present study, we aimed to the mixture toxicity of beta-cypermethrin (BCY) and thiacloprid (THI) to zebrafish (Danio rerio) employing multiple toxicological endpoints. Results displayed that the 96-h LC50 values of BCY to D. rerio at various developmental stages ranged from 2.64 × 10 (1.97 × 10–3.37 × 10) to 6.03 × 103 (4.54 × 103–1.05 × 104) nM, which were lower than those of THI ranging from 2.97 × 104 (1.96 × 104–4.25 × 104) to 2.86 × 105 (2.19 × 105–5.87 × 105) nM. Mixtures of BCY and THI exhibited synergistic response in embryonic zebrafish. Meanwhile, the enzyme activities of antioxidants (CAT and SOD) and detoxification enzyme (CarE), endogenous T-GSH and MDA contents, as well as gene expressions (tsh, crh, cxcl and bax) involved in oxidative stress, cellular apoptosis, immune system and endocrine system were obviously changed in the mixture exposure compared with the respective BCY or THI treatment. Consequently, the increased toxicity of pesticide mixture suggested that the toxicological data acquired from individual pesticide tests might underrate the toxicity risk of pesticides that actually arise in the real environment. Taken together, our present study provided evidence that mixture exposure of BCY and THI could induce additional toxic effect compared with their respective individual pesticides on D. rerio, offering valuable insights into the toxic mechanism of pesticide mixture.

Silver (Ag) is amongst the most well studied nanomaterials (NMs), although most studies have only dealt with a single AgNM at a time and one biological endpoint. We here integrate the results of various testing-tools (endpoints) using a terrestrial worm, the standard ecotoxicological model organism Enchytraeus crypticus. Exposure spanned both water and soil exposure, it covered all life stages (cocoons, juveniles and adults), varying exposure durations (1-2-3-4-5-21 days), and covered 5 biological endpoints: hatching success, survival, reproduction, avoidance and gene expression (qPCR target genes GABA and Acetyl cholinesterase). We tested 4 Ag materials: PVP coated (PVP-AgNM), non-coated (NC-AgNM), the JRC reference Ag NM300K and AgNO₃. Results showed that short-term exposure via water to assess impact on cocoons’ hatching predicted longer term effects such as survival and reproduction. Moreover, if we extended the exposure from 11 to 17 day this allowed discrimination between hatch delay and impairment. Exposure of juveniles and adults via water showed that juveniles were most sensitive with survival affected. Across materials the following toxic ranking was observed: AgNO₃ ≥ Ag NM300K ≫ NC-AgNM ≥ PVP-AgNM. E. crypticus avoided AgNO₃ in a dose-response manner, avoiding most during the first 24 h. Avoidance of Ag NM300K and NC-AgNM only occurred during the first 24 h and the PVP coated AgNM were not avoided at all. The up-regulation of the GABA triggering anesthetic effects, indicated the high ecological impact of Ag materials in soil: Ag affects the GABAergic system hence organisms were not able to efficiently avoid and became intoxicated, this caused impacts in terms of survival and reproduction.

The excessive application of manure has caused a high load of phosphorus (P) in the North China Plain. Having an understanding of how manure application affects soil P changes and its transport between different soil layers is crucial to reasonably apply manure P and reduce the associated loss. Based on our 28-year field experiments, the compositions and changes of P species and the risk of P loss under excessive manure treatments were investigated, i.e., no fertilizer (CK), mineral fertilizer NPK (NPK), NPK plus 22.5 t ha⁻¹ yr⁻¹ swine manure (LMNPK), and NPK plus 33.75 t ha⁻¹ yr⁻¹ swine manure (HMNPK). Manure application increased the content of orthophosphate and myo-inositol hexaphosphate (myo-IHP), especially the orthophosphate content exceeded 95%. The amount of orthophosphate in manure and the conversion of organic P to inorganic P in soil were the main reasons for the increased soil orthophosphate. Compared with NPK treatment, soil microbial biomass phosphorus and alkaline phosphatase activity in LMNPK and HMNPK treatments significantly increased. Compared with NPK treatment, a high manure application rate under HMNPK treatment could increase the abundance of organic P-mineralization gene phoD by 60.0% and decrease the abundance of inorganic P-solubilization gene pqqC by 45.9%. Due to the continuous additional manure application, soil P stocks significantly increased under LMNPK and HMNPK treatments. Furthermore, part of the P has been leached to the 60–80 cm soil layer. Segmented regression analysis indicated that CaCl₂–P increased sharply when Olsen-P was higher than 25.1 mg kg⁻¹, however the content of Olsen-P did not exceed this value until 10 years after consecutive excessive manure application. In order to improve soil P availability and decrease the risk of P loss, the manure application rate should vary over time based on soil physicochemical conditions, plants requirements, and P stocks from previous years.

Perfluorooctanoic acid (PFOA) has applications in numerous industrial products and is an industrial waste that is persistently present in the environment. Exposure to PFOA results in nonalcoholic fatty liver disease (NAFLD). However, the underlying mechanisms remain unclear. In this study, male C57BL/6 mice were exposed to PFOA (1 mg/kg/day) for 4 weeks to evaluate the effect of PFOA, and the human liver cell line (L-02) was used to observe the direct effect of PFOA in vitro. After PFOA exposure, the expression of genes related to hepatic lipogenesis, the NLRP3 inflammasome, and autophagy were measured. We found that exposure to PFOA induced lipid accumulation and stimulated lipogenesis in both mouse livers and L-02 cells. In addition, increased NLRP3 aggregation and enhanced production of IL-1β occurred after PFOA treatment. We also found that PFOA exposure induced autophagosome formation and p62 accumulation, indicating blockage of autophagic flux. Rapamycin alleviated PFOA-induced lipid accumulation and NLRP3 inflammasome activation by activating autophagic flux. Conversely, chloroquine, an autophagic flux inhibitor, exacerbated PFOA-induced lipid accumulation and NLRP3 inflammasome activation. Collectively, these results provide evidence to show that PFOA-induced blockade of autophagic flux causes an increase in lipid synthesis and inflammation in vivo and in vitro.

Ingestion of microplastics by marine organisms has been well documented, but their interaction with chemical pollutants has not been sufficiently addressed. The aim of this study was to determine the individual and combined effects of chlorpyrifos (CPF) and polyethylene microplastics (MP) on the survival, fecundity, feeding and egg viability of Acartia tonsa, a calanoid copepod widely distributed in planktonic communities. The median lethal concentration obtained for CPF was higher (LC₅₀ = 1.34 μg/L) than for the combination with MP (LC₅₀ = 0.37 μg/L), or CPF-loaded MP (LC₅₀ = 0.26 μg/L). Significant effects were also observed for feeding and egg production (EC₅₀ = 0.77 and 1.07 μg/L for CPF, 0.03 and 0.05 μg/L for CPF combined with MP, 0.18 and 0.20 μg/L for CPF-loaded MP). No significant effects were observed in the exposure to ‘virgin’ MP. This study confirms the role of MP as vectors of pollutants to marine organisms and supports the increased availability of certain toxicants carried out by MP. The effects observed in fitness-related responses suggest potential damage to A. tonsa populations. The comparison of the results obtained here with environmental concentrations indicates that the combined exposure to CPF and MP could constitute a risk to A. tonsa in the natural environment.
